The previous studies formed the role of oxidants and proinflammatory cytokines in the pathogenesis of acute psychological stress-related cardiac damage leading to mortality and other complications. The aim of the study is to examine the protective effect of rutin against stress-induced cardiac damage. In the literature, no studies have been found analyzing the effects of rutin in acute stress-related oxidative damage induced by immobilization method in rats. Rutin was administered orally to rutin + stress applied (RSG) group albino rats at a dose of 50 mg/kg. For healthy control (HG) and stress applied to control (SAG) groups, distilled water as a solvent was orally administered at the same volume (0.5 mL). One hour after rutin and distilled water applications, all animals except for HG brought into a supine position and their legs and arms were ligated and kept in the same position for 24 h. The oxidant, cytokine and cardiac biomarker levels in blood serum and heart tissues of SAG animals were found to be significantly higher and total glutathione was lower than RSG and HG groups. Histopathologically dilated congested blood vessels and myocardial destruction, hemorrhage, edema, and polymorphonuclear leukocyte infiltration were observed in the SAG heart tissues. On the other hand, the histological heart tissue results of the RSG group was found to be similar compared to the healthy tissue except for the slightly dilated congested blood vessel. These results indicate that the rutin may be useful in the treatment of stress-related oxidative cardiac damage.

The effect of taxifolin on cisplatin-induced oxidative pulmonary damage was investigated biochemically and histopathologically in male albino Wistar rats. There were four groups, with six animals in each group: 50 mg/kg of taxifolin plus 2.5 mg/kg of cisplatin (TC) group, 2.5 mg/kg of cisplatin only (CIS) group, 50 mg/kg of taxifolin only (TG) group, and a healthy control group (HG). In terms of the experimental procedure, the animals in the TC and TG groups were first treated via oral gavage. The CIS and HG groups received distilled water as solvent, respectively. One hour later, the TC and CIS groups received cisplatin at a dose of 2.5 mg/kg (injected intraperitoneally). Taxifolin, cisplatin, and the distilled water were administered at the indicated dose and volume, using the same method daily for 14 d. At the end of this period, the animals were killed with a high dosage of thiopental anaesthesia (50 mg/kg). Blood and lung tissue samples were taken for biochemical (malondialdehyde (MDA), myeloperoxidase (MPO), total glutathione (tGSH), and 8-hydroxy-2 deoxyguanosine (8-OHdG)) analyses and histopathological examinations. The biochemical and histopathological results in the TC and HG groups were then compared with those in the CIS group. Cisplatin increased the levels of MDA, myeloperoxidase, and 8-OHdG, a marker of oxidative DNA damage, and reduced the amount of tGSH in the lung tissue. Moreover, severe alveolar damage, including oedema and extensive alveolar septal fibrosis, in addition to infiltration of polymorphic nuclear leucocytes and haemorrhagic foci, was observed in the CIS group. These histopathological findings demonstrate that taxifolin provides protection against pulmonary oxidative stress by preventing increases in oxidant parameters and decreases in antioxidants.

Background/Aims: This study aimed to investigate the effect of lutein on methotrexate (MTX)-induced pulmonary toxicity in rats biochemically and histopathologically. Methods: The rats in the MTX + lutein (MTXL, n = 6) group were given 1 mg/kg of lutein orally. A 0.9% NaCl solution was administered orally to the MTX (n = 6) group and the healthy group (HG, n = 6). One hour later, a single 20 mg/kg dose of MTX was injected intraperitoneally in the MTXL and MTX. Lutein or 0.9% NaCl solution was administered once a day for 5 days. At the end of this period, malondialdehyde (MDA), myeloperoxidase (MPO), total glutathione (tGSH), interleukin 1 beta (IL-1β), and tumor necrosis factor alpha (TNF-α) were measured in the lung tissues from the animals euthanized with 50 mg/kg thiopental sodium anesthesia. Subsequently, histopathological examinations were performed. Results: The levels of MDA, MPO, IL-1β, and TNF-α in the lung tissue of the MTX were significantly higher than those of the MTXL and HG groups (p < 0.0001), and the amount of tGSH was lower. The histopathological findings in the MTX group, in which the oxidants and cytokines were higher, were more severe. Conclusions: Lutein prevented the MTX-induced oxidative lung damage biochemically and histopathologically. This result indicates that lutein may be useful in the treatment of MTX-induced lung damage.

Background and Objective: Cisplatin is an antineoplastic, platinum derivative used in the treatment of various cancers. Mucositis is a notable side effect of cisplatin treatment. Rutin (vitamin P1) is a drug with antioxidant, anticancer, antidiabetic and antimicrobial properties. The aim of this study was to biochemically, histopathologically and immunohistochemically investigate the effect of rutin on cisplatin-induced mucositis of the small intestine (jejunum) in rats. Materials and Methods: Twenty four rats were divided into four groups with six animals in each group: healthy group (HG), cisplatin-only group (CCG), 50 mg kg-1 of rutin plus cisplatin group (RG-50) and 100 mg kg-1 of rutin plus cisplatin group (RG-100). Rutin or distilled water were administered via an oral gavage. One hour after the administration of rutin or distilled water, the CCG, RG-50 and RG-100 were intraperitoneally injected with 5 mg kg-1 of cisplatin once every 2 for 8 days. At the end of this period, all the animals were sacrificed using a high-dose anesthetic and their small intestines (jejunum) were removed for biochemical and histopathological procedures. Differences between the groups were analyzed using a one-way analysis of variance, followed by Dunnett’s multiple comparisons test. Results: The levels of oxidants increased in all the cisplatin-treated groups, whereas those of antioxidants decreased. Rutin administered at a dose of 100 but not 50 mg kg-1 reduced oxidant levels and increased antioxidant levels to those of healthy tissues. Histopathologically, tissue damage was observed in jejunal tissue of the rats administered only cisplatin, whereas treatment with 100 mg kg-1of rutin prevented cisplatin-induced histopathological damage. In the group administered 50 mg kg-1of rutin, jejunal tissue showed a near-normal appearance, except for mildly dilated congested blood vessels. As shown immunohistochemically, rutin prevented cisplatin-induced jejunal damage more effectively when administered at a dose of 100 mg kg-1 than 50 mg kg-1. Conclusion: Rutin may be useful in the prevention of cisplatin-induced jejunal mucositis.

The aim of this study was to investigate the effects of parecoxib on the ischemia/reperfusion (I/R) injury induced in rats by torsion-detorsion. Parecoxib was injected to all the rats intraperitoneally once prior to the torsion-detorsion. The ischemia/reperfusion and sham surgery groups were given the same volume of distilled water. One h later, torsion and detorsion were performed, each for two h, on the ischemia/reperfusion + 10 mg/kg parecoxib, ischemia/reperfusion + 20 mg/kg parecoxib, and ischemia/reperfusion groups. The results of the biochemical test showed that 20 mg/kg dose of parecoxib was more effective than the 10 mg/kg dose in inhibiting the increase in the MDA, COX-2, IL-1β and TNF-α levels and decrease in tGSH and COX-1 in the testicular tissue treated with torsion-detorsion. Furthermore, it was also histopathologically shown that the 20 mg/kg dose better prevented torsion-detorsion injury. The experimental results suggest that parecoxib will be clinically useful in the treatment of testicular I/R injury due to torsion-detorsion.
