Özet

Objective: This study aimed to investigate the effect of \rhigh-fructose and high-fat diets on advanced glycation end \rproducts (AGE) and their receptors (RAGE) in rat liver immunohistochemically and ultrastructurally.\rMaterials and Methods: Rats were divided into three groups as \rthe control group, high-fructose group, and high-fat group. The \rhigh-fructose group was given 60% fructose diet, and the highfat group was fed 45% fatty diet. After 12 weeks, rats were sacrificed, and liver tissues were taken for light and electron microscopic examinations. Expressions of AGE, RAGE, nuclear factor \rkappa B (NF-κB), and 8–hydroxyguanosine (8-OhdG) were \rdetected immunohistochemically. Liver function enzymes, \rnamely, aspartate transaminase, alanine transaminase, alkaline \rphosphatase, and gamma-glutamyl transferase, were measured \rfrom serum samples.\rResults: Prominent degenerative changes were detected in \rthe liver sections of both diet groups. Immunostaining of AGE, \rRAGE, NF-κB, and 8-OHdG was more evident than that of the \rcontrol group. Activities of the liver function enzymes of these \rgroups were higher than those of the control group. Ultrastructurally, accumulations of glycogen and lipid droplets in hepatocytes were observed in the diet groups.\rConclusion: High-fructose and high-fat diets can cause ultrastructural changes in the liver, and the damage can be related \rwith increased AGE.
