Background/Objectives: The underlying molecular mechanisms of deep vein thrombosis (DVT), which continues to be a major global public health concern, remain unclear. A key component of anticoagulant therapy, heparin (HP) interacts with heparin-binding growth factors including pleiotrophin (PTN) and midkine (MK), both of which have basic amino acid-rich domains that have a strong affinity for HP. The purpose of this study was to determine if changes in the levels of circulating HP, MK, and PTN are linked to the onset of acute DVT. Methods: Thirty patients diagnosed with acute DVT by venous Doppler ultrasonography (VDU) and 28 healthy controls with normal VDU findings were enrolled. Serum HP, MK, and PTN concentrations were measured using ELISA. In DVT patients, blood samples were obtained before and after routine subcutaneous low-molecular-weight heparin treatment; controls provided a single blood sample. ROC curve analysis was used to assess diagnostic performance. Results: Prior to treatment, patients with acute DVT exhibited significantly lower serum HP levels (p < 0.05) and significantly higher MK and PTN levels compared with healthy controls (both p < 0.05). Following heparin administration, serum HP levels increased significantly (p < 0.05), while MK and PTN levels showed a decreasing trend that did not reach statistical significance (p > 0.05). ROC curve analysis demonstrated limited diagnostic performance for HP (sensitivity 10.3%, specificity 68.8%), PTN (62.1%, 54.2%), and MK (82.8%, 35.4%). Conclusions: Decreased circulating HP and increased MK and PTN levels are characteristics of acute DVT that may indicate endogenous HP sequestration through binding to these growth factors. This imbalance could lead to less free HP being available, which would encourage the formation of thrombus. Therapeutic approaches that target MK- and PTN-mediated HP interactions may constitute a unique approach for the therapy of acute DVT, as evidenced by the partial normalization seen after exogenous heparin delivery.

Background/Objectives: Sepsis remains one of the leading causes of mortality, yet its etiopathogenesis is still not fully understood. This study aimed to investigate the effects of cetirizine and dexamethasone (alone and in combination) on serum levels of Maresin-1 (MaR-1), TNF-α, IFN-γ, IL-1, IL-2, IL-6, IL-8, and IL-10 in a rat model of sepsis induced by the cecal ligation and puncture (CLP) method. Methods: Male Sprague Dawley rats aged 8–10 weeks were used and randomly divided into 7 groups, each containing 7 rats: Group 1 (Control), Group 2 (Sham), Group 3 (Sepsis), Group 4 (Sepsis + Saline), Group 5 (Sepsis + Cetirizine), Group 6 (Sepsis + Dexamethasone), and Group 7 (Sepsis + Cetirizine + Dexamethasone). Sepsis was induced via CLP in all groups except Control and Sham. Results: In the sepsis groups (G3–G7), neutrophil and white blood cell counts increased while lymphocyte counts decreased (p < 0.05). In groups treated with cetirizine and/or dexamethasone (G5–G7), a significant decrease in neutrophils and an increase in lymphocytes were observed. MaR-1 levels significantly decreased (p < 0.05) in all sepsis-induced groups compared to controls, while interleukin levels significantly increased. Cetirizine and dexamethasone supplementation significantly increased MaR-1 levels and decreased interleukin levels (p < 0.05). The combined treatment was more effective. Conclusions: This study is the first to highlight the potential of MaR-1 as a critical biomarker in sepsis diagnosis and monitoring, and cetirizine and dexamethasone, especially in combination, may represent a promising therapeutic option in sepsis management.

Demir, Sukru; Can, Bugra; Esmez, Omer; Aydin, Mustafa Ata; Gencer, Sevval Yagmur; Bildirici, Furkan; Koca, Süleyman Serdar; Sahin, İbrahim; Aydin, Suleyman
Objective – To investigate synovial fluid levels of maresin-1 (MaR-1) and maresin-2 (MaR-2) in patients with psoriatic arthritis (PsA), rheumatoid arthritis (RA), osteoarthritis (OA), and controls, and to assess their ability to differentiate PsA from RA. Methods – This prospective cross-sectional study included 88 patients undergoing knee arthrocentesis for joint effusion between September 2025 and February 2026. RA was classified according to the 2010 ACR/EULAR criteria, PsA using CASPAR criteria, and OA based on clinical and radiographic assessment. Synovial fluid MaR-1 and MaR-2 concentrations were measured by enzyme-linked immunosorbent assay (ELISA) and expressed as ng/mL. Group comparisons were performed using parametric or non-parametric statistical tests as appropriate. Receiver operating characteristic (ROC) curve analysis evaluated the diagnostic performance of MaR-1 and MaR-2 for distinguishing PsA from RA, including area under the curve (AUC), sensitivity, specificity, and optimal cut-off values. Logistic regression analysis assessed the association between MaR-1 levels and PsA. Results – Significant intergroup differences were observed for age, visual analogue scale (VAS) scores, and synovial MaR-1 concentrations (all p < 0.01). MaR-1 levels were significantly higher in PsA than RA (p < 0.01), whereas MaR-2 showed more modest differences among groups (p < 0.05). ROC analysis demonstrated good discriminatory performance of MaR-1 for differentiating PsA from RA, with an AUC of 0.815 (95% CI, 0.675–0.933). A cut-off value of 3.252 ng/mL provided 68.2% sensitivity and 86.4% specificity. Higher MaR-1 levels were significantly associated with PsA in unadjusted logistic regression analysis (OR = 1.62, 95% CI 1.18–2.22; p = 0.003). MaR-1 and MaR-2 levels were moderately positively correlated (Spearman’s r = 0.620, p < 0.001). Conclusion – Synovial MaR-1 concentrations were markedly elevated in PsA compared with RA and demonstrated good diagnostic discrimination, supporting the potential role of pro-resolving mediators as biomarkers in inflammatory arthropathies. The clinical significance of MaR-2 appears more limited and requires validation in larger studies.

Immunohistochemistry is a tissue-based technique that enables in situ visualization of specific molecules, most commonly proteins or peptides, within preserved cellular and tissue architecture through antigen–antibody specificity. By using labeled antibodies, immunohistochemistry allows spatially resolved and cell type–specific detection of target molecules and maintains the morphological context, thereby providing both localization and relative expression information at the light or electron microscopy level. Owing to this integrative capacity, immunohistochemistry has become an essential tool in biological research and diagnostic pathology for characterizing molecular distribution and tissue organization across animal and plant systems. The detection techniques based on fluorescence and enzymes (chromogenic) are compared in terms of their repeatability, multiplexing capability, sensitivity, and specificity. With a focus on protocol standardization and methodological rigor, important procedural processes are covered, including fixation, antigen extraction, antibody optimization, signal detection, and quality control. Additionally, the immunohistochemistry technique remains essential for locating and identifying immunogenic substances in a variety of biological settings. Therefore, drawing on more than two decades of laboratory experience, this narrative review summarizes the history of the immunohistochemistry assay, its underlying principles and procedures, common immunohistochemistry protocol, methodological process, essential reagents, infrastructure, and best-practice principles required for the establishment and optimization of a high-quality immunohistochemistry laboratory. © The Author(s) 2026. Objective: The neurological causes of suicide remain poorly understood. This study sought to ascertain whether there is a correlation between amygdala volume and suicidal behavior. Methods: This case–control retrospective study included 193 participants—108 healthy controls and 85 individuals with a history of suicide attempts. Sagittal magnetic resonance imaging was conducted for each participant, and cross-sectional areas of the left and right amygdala were determined independently using freehand tracing. The total volume of the amygdala was determined by multiplying the thickness of the slices by the sum of the regions. Results: Patients with a history of suicide attempts had significantly decreased amygdala volumes on both the right (p = 0.018) and left (p = 0.036) sides compared with controls. Receiver operating characteristic analysis determined a cutoff value of 1825 mm³ for both hemispheres. The cutoff sensitivity and specificity for the right amygdala were 30.6% and 88%, whereas the values for the left amygdala were 31.8% and 84.3%, respectively. Conclusions: This is the first study to show that individuals who had attempted suicide had considerably decreased left and right amygdala volumes, with receiver operating characteristic analysis indicating good specificity (84.3%–88%). According to these findings, a reduced amygdala volume might be a useful biomarker for identifying those who are at risk of attempting suicide. © The Author(s) 2025. Metabolic syndrome (MetS) is a prevalent public health problem. Uric acid (UA) is increased by MetS. We investigated whether administration of UA and 10% fructose (F) would accelerate MetS formation and we also determined the effects of irisin and exercise. We used seven groups of rats. Group 1 (control); group 2 (sham); group 3 (10% F); group 4 (1% UA); group 5 (2% UA); group 6 (10% F + 1% UA); and Group 7, (10% F + 2% UA). After induction of MetS (groups 3 -7), Group 3 was divided into three subgroups: 3A, no further treatment; 3B, irisin treatment; 3C, irisin treatment + exercise. Group 4, 1% UA, which was divided into three subgroups: 4A, no further treatment; 4B, irisin treatment; 4C, Irisin treatment + exercise. Group 5, 2% UA, which was divided into three subgroups: 5A, no further treatment; 5B, irisin treatment; 5C, irisin treatment + exercise. Group 6, 10% F + 1% UA, which was divided into three subgroups: 6A, no further treatment; 6B, irisin treatment; 6C, irisin treatment + exercise. Group 7, 10% F + 2% UA, which was divided into three subgroups: 7A, no further treatment; 7B, irisin treatment; 7C, irisin treatment + exercise., İrisin was administered 10 ng/kg irisin intraperitoneally on Monday, Wednesday, Friday, Sunday each week for 1 month. The exercise animals (in addition to irisin treatment) also were run on a treadmill for 45 min on Monday, Wednesday, Friday, Sunday each week for 1 month. The rats were sacrificed and samples of liver, heart, kidney, pancreas, skeletal muscles and blood were obtained. The amounts of adropin (ADR) and betatrophin in the tissue supernatant and blood were measured using an ELISA method. Immunohistochemistry was used to detect ADR and betatrophin expression in situ in tissue samples. The duration of these experiments varied from 3 and 10 weeks. The order of development of MetS was: group 7, 3 weeks; group 6, 4 weeks; group 5, 6 weeks; group 4, 7 weeks; group 3, 10 weeks. Kidney, liver, heart, pancreas and skeletal muscle tissues are sources of adropin and betatrophin. In these tissues and in the circulation, adropin was decreased significantly, while betatrophin was increased significantly due to MetS; irisin + exercise reversed this situation. We found that the best method for creating a MetS model was F + UA2 supplementation. Our method is rapid and simple. Irisin + exercise was best for preventing MetS.

Objectives: It has been recently reported that immune activity of some animal peptides/proteins has been found in some plants. In this study, the presence of immune reactivity of alarin, asprosin, paraoxonase, elabela, glucagon, and nesfatin-1 like molecules was investigated in two plant species related to Kastamonu Taşköprü garlic (an endemic cultivar of the Allium sativum L.) and Tunceli garlic (Allium tuncelianum). Methods: Ten Kastamonu and 10 Tunceli garlic were embedded in paraffin blocks and 4–5 μm transversal and longitudinal sections were taken and stained by immunohistochemical method. In addition, 500 mg of wet tissue samples were homogenized from these garlic and supernatants were obtained. The amounts of alarin, asprosin, elabela, glucagon and nesfatin-1 were studied by enzyme linked immunosorbent assay (ELISA) method and paraoxonase was studied by spectrophotometric method. Results: Studied molecules show immune reactivity in the cytoplasm of the bulb storage cells of the plants parenchyma. The ELISA supernatant results also confirmed the presence of these animal molecules in the plants. Conclusions: Although we cannot reveal the purpose of the presence of these animal molecules detected in plants with these existing data, we predict that these data will be a source of inspiration in the field of botany, that studies will shift in this direction and that they might contribute to the revision of the evolutionary processes of all living things, especially an ancestor of garlics.

Objective: The combination of diabetes mellitus and hypertension accelerates vascular complications and increases the risk of mortality and morbidity. The association of hypertension and diabetes mellitus is due to the common mechanism responsible for the pathogenesis of both diseases. The current study aimed to determine how catecholamine and cerebellin levels were affected both before and after treatment in the urine and blood of patients with hypertension and hypertension + diabetes mellitus. Methods: This cross-sectional study included 30 patients with newly diagnosed hypertension, 30 patients with previously diag- nosed diabetes mellitus and new hypertension, and 30 healthy controls. Serum and urinary levels of cerebelline, adrenaline, noradrenaline, metanephrine, and normetanephrines were mea- sured by ELISA. Other biochemical parameters (Fasting Blood Glucose [FBG], glycosylated hemoglobin A, low-density lipopro- tein, and triglyceride) were measured by autoanalyzer. In addi- tion, the clinical characteristics of the collected patients were also recorded including age, gender, blood pressure, and body mass index. Results: The gender was similar among the control, hyperten- sion, and hypertension+ diabetes mellitus groups (P > .05). There was a significant decrease in blood and urine cerebellin, meta- nephrine, and normetanephrine levels in hypertension + dia- betes mellitus patients compared with both newly diagnosed hypertension and controls (P < .001). There was no difference in cerebellin levels between newly diagnosed hypertension and controls (P = .395). There was a significant negative correlation between cerebellin and systolic and diastolic blood pressure and adrenaline. Conclusion: This result suggests that there is a relationship between cerebellin, catecholamine, and catecholamine metabolites in hypertension and hypertension + diabetes mellitus patients. In the future, there is a need for further studies on the possibility that these biomarkers can give an idea about the etiopathogenesis of diseases such as hypertension and diabetes mellitus.

Asprosin (ASP) and subfatin are hormones that regulate glucose metabolism. The role of ASP and subfatin in serous ovarian tumors has not been investigated. We investigated the expression of subfatin and asprosin in 30 serous benign, 30 serous borderline, 30 malignant and 30 control ovarian tissues. We investigated ASP and subfatin immunoreactivity and quantification was achieved using an ELISA method. ASP and subfatin were localized in the epithelial parts of normal ovarian tissues; however, in cancer tissues, immunoreactivity was detected in the parenchymal areas. Biochemical analysis of ovarian tissues revealed significantly decreased ASP and subfatin compared to the control. We propose that ASP and subfatin are promising candidates for biomarkers to distinguish serous benign, serous borderline and malignant ovarian cancers.

We investigated the presence of asprosin (ASP), betatrophin, elabela (ELA), glucagon and subfatin (SUB) in the milk of mothers with gestational diabetes mellitus (GDM) and compared their levels with blood levels. We also investigated whether these peptides are synthesized by the breast. We investigated 12 volunteer mothers with GDM and 14 pregnant non-GDM control mothers. The peptides were measured using ELISA and their tissue localization was determined using immunohistochemistry. Breast milk contains ASP, betatrophin, ELA, glucagon and SUB. The amount of the peptides ranged from highest to the lowest in colostrum, transitional milk and mature milk. The amount of peptides in the milk was greater than for blood. The peptides, except for ELA, were increased in milk and blood by GDM. Betatrophin and ELA are synthesized in the connective tissue of the breast. ASP, glucagon and SUB are synthesized in the alveolar tissue of the breast. These peptides in breast milk may contribute to the development of the gastrointestinal tract of newborns and infants.
RDL Kalıcı Kaydıİncele - Conference ObjectYalnız künye2022Basal blood concentrations of some orexigenic and anorexigenic hormones in obese and nonobese individuals according to blood groups
Karagoz, Z.K.; Aydin, S.; Aksoy, A.; Kalayci, M.; Ugur, K.; Kuloglu, T.; Cinar, V.; Yardim, M.; Aydin, Y.; Akbulut, T.
OBJECTIVE: Obesity is a serious public health problem associated with excessive food intake. Regulation of food intake in highly organized organisms is under the control of a large number of orexigenic and anorexigenic molecules. Therefore, the main purpose of this study has been to determine the relationship between obesity and some of the circulating orexigenic and anorexigenic peptides that have a role in appetite control and to determine whether the concentrations of these molecules differ according to blood groups. PATIENTS AND METHODS: The study included 400 individuals of whom 100 were obese women, 100 obese men, 100 healthy men and 100 healthy women. Obese women and men were divided into 4 groups, according to their blood groups. In the control group, healthy women and healthy men were similarly divided into 4 blood groups. Each blood group within the groups, therefore, had 25 participants. RESULTS: When leptin, nesfatin-1, obestatin and neuropeptide-Y, ghrelin and galanin levels of the control group and obese participants were compared, regardless of blood groups, leptin, nesfatin-1, obestatin and neuropeptide-Y were significantly higher, whereas only the ghrelin levels were significantly lower in obese patients. When the amounts of these hormones were measured according to gender, the situation was similar. When leptin, nesfatin-1, obestatin and neuropeptide-Y values of the control and obese participants’ blood groups were compared with each other; these hormones were high in all blood groups; however, leptin levels in A blood group, nesfatin-1 levels in AB and O blood group, obestatin levels in AB blood group, neuropeptide-Y levels in A, B, AB blood groups were significantly higher. When the ghrelin levels of the blood groups in the control group and obese participants were compared, it was only significantly lower in the AB blood group. The ghrelin levels in the other blood groups of the obese individuals were again low, but not significantly so. When the distribution of hormones according to gender was evaluated, a situation parallel to the above results was recorded. CONCLUSIONS: Leptin, nesfatin-1, obestatin and neuropeptide-Y and galanin levels of obese individuals were significantly higher than the control values, whereas the ghrelin values were significantly lower regardless of blood groups. Also, these hormones in blood partly varied with ABO blood groups. These different concentrations of hormones in ABO blood groups might be related with stimulation or suppression of appetite in human. However, further studies in other ethnic groups are needed to confirm these results.
RDL Kalıcı Kaydıİncele - ArticleYalnız künye2022Molecular communication between Apelin-13, Apelin-36, Elabela, and nitric oxide in gestational diabetes mellitus
Karagoz, Z.K.; Aydin, S.; Ugur, K.; Tigli, A.; Deniz, R.; Baykus, Y.; Sahin, İ.; Yalcin, M.H.; Yavuz, A.; Aksoy, A.
OBJECTIVE: Gestational diabetes mellitus (GDM) is a type of diabetes that affects from 3.8% to 6.9% of pregnancies worldwide, causing significant mortality and unfavorable obstetric outcomes, such as delivery trauma and macrosomia risk. The fundamental processes of this metabolic disorder that first appeared during pregnancy are still unknown. Tissue hormones, particularly adipokines, have aided in understanding the pathophysiology of numerous disorders in recent years. This study aims to determine if Apelin-13 (APLN-13), Apelin-36 (APLN-36), Elabela (ELA), and nitric oxide (NO) molecules have all a part in the pathophysiology of GDM. PATIENTS AND METHODS: The study included 30 pregnant control women and 30 pregnant women who had been diagnosed with GDM in the second trimester and whose body mass index and age were compatible with each other. Blood samples were collected from 60 participants during the second trimester (30 control pregnant women and 30 GDM pregnant women) and postpartum (17 controls vs. 14 GDM). In these blood samples, the amounts of APLN-13, APLN-36, ELA, and NO were studied using the ELISA method. In addition, the participants’ glucose, lipid profiles, and other parameters were obtained from the hospital record files. At postpartum, 29 pregnant women (13 control and 16 pregnant women with GDM) dropped out of the study without explanation. RESULTS: In the second trimester and postpartum plasma of mothers with GDM, APLN-13, APLN-36, NO, and ELA molecules were found to be significantly higher (< 0.05), compared to those of the control mothers, while APLN-13, APLN-36, NO values were significantly lower (0.05). While APLN-13, APLN-36, NO amounts in mothers with GDM were positively correlated with glucose amounts, they were negatively correlated with ELA amounts. Similarly, the triglyceride amounts in mothers with GDM were positively correlated with APLN-13, APLN-36 and NO, while they were negatively correlated with the ELA amounts. Due to gestational diabetes, APLN-13, APLN-36, NO, glucose, and triglyceride increased, and ELA decreased. CONCLUSIONS: It is predicted that the glucose increase in GDM is because Apelins reduce glucose transport to erythrocytes by inhibiting the sodium-dependent glucose transporter (SGLT) and that the increase in triglyceride and NO may be associated with high glucose levels in GDM. As a result, we believe that the above-mentioned chemicals may cause GDM Pathology by triggering one another.

Purpose: The molecules human interleukin (IL-18), the soluble cluster of differentiation (sCD40), platelet factor 4 variant 1 (PF4V1), and neutrophil gelatinase-associated lipocalin (NGAL) are all markers of inflammation in biological systems and are linked to prognosis in several inflammatory diseases as well. Since there is no study in which the above-mentioned molecules are studied together in ocular Behçet's disease (OBD), the aim of this study is to reveal whether these molecules are activity markers in active (OABD) and inactive (OIBD) disease. Methods: 30 OABD and 30 OIBD and 30 healthy individuals were included in the study. IL-18, sCD40, PF4V1, and NGAL molecules were studied in blood samples by the ELISA method. Results: When OABD and OIBD were compared to healthy individuals, the levels of IL-18, sCD40, PF4V1, and NGAL molecules were found to be statistically significant. These values were even more significantly higher in patients with OABD. Conclusion: When ROC values of IL-18, sCD40, PF4V1, and NGAL are evaluated, it is clear that these four molecules can be used as biomarkers to aid activity and diagnosis in OBD.

Aydin, Suna; Ugur, Kader; Yalcin, Hanifi; Sahin, İbrahim; Akkoc, Ramazan Fazil; Yakar, Burkay; Yucel, Dogan; Aydin, Suleyman
COVID-19 is the most devastating pandemic situation we have experienced in our age, affecting all systems. Although it affects all systems, it shows its most important effect through thrombophilia. Therefore, the possible cause of sudden death due to COVID-19 may be embolism caused by thrombophilia. D-dimer amounts increase due to COVID-19. The thrombosis is associated with sudden death in COVID-19 disease in populations. Since individuals with thrombophilia will be more prone to death due to COVID-19, it may be appropriate to administer low doses of Clexane (Enoxaparin sodium) or low-weight heparin for prophylactic purposes in order to consider these individuals at high risk and to prevent deaths. Moreover, in order not to risk the lives of healthcare professionals with thrombophilia, it would be appropriate to keep them away from individuals with COVID-19 disease and to employ them in different healthcare services according to their fields of expertise. It should also not be forgotten that different symptoms related to COVID-19 appear day by day, these different symptoms probably show that the virus has undergone mutations in order to survive, but no matter what, its effect on thrombophilia has not been eliminated yet. This compilation aims to present the reasons and causes of death due to COVID-19, possible treatment options, and thrombophilia panel tests and new parameters that may have a place in the meticulous interpretation of these tests and possible etiopathology in the light of current information. Therefore, presenting this information in a rational manner and keeping the parameters of the thrombophilia panel under strict control predict that the deaths due to the virus will be partially reduced.

The specialized resident-stem cells in gonads are tasked with restorating damaged ovarian cells following injury to maintain sequential reproductive events. When we talk about premature ovarian insufficiency (POI) we accept the existence of decreased stem cell and their regenerative abilities. The present study was to explain how restorating damaged ovarian cells following injury to maintain sequential reproductive events in evidence-based medicine indexed in PubMed and Web of Science. The exact mechanism is unclear stem cells transfer may improve compromised ovarian function and fertility outcome in women with POI. Soluble factors secreted by stem cell may rescue impaired mitochondrial function in oogonial stem cells, enhance metabolic capacity of resident stem cells, induce local neovascularization in the ovary, and activate gene shifting between transferred stem cells and germ cell precursors. This review may provide insight into how stem cells show some of their beneficial effects on compromised ovarian microenvironment and germ cell niche and paves the way for clinical trials for improving ovarian function of women with POI. We also had the opportunity to share our hypothesis about the design and development of induced oogonial stem cell (iOSC) and its use in POI.

Background: Apelin (APLN), elabela (ELA), and nitric oxide (NO) have effects on physiological and behavioural properties in biological systems. This study was designed to determine APLN, ELA and NO levels in schizophrenia patients and assess whether these molecules are of diagnostic value. Methods: A total of 33 schizophrenic patients and 32 ageand sex-adjusted healthy participants were included in the study. ELA, APLN and NO levels were measured using ELISA methods. Results: Although the ELA and NO levels of the patients were lower than the control group, APLN levels were higher (p = 0.039, p = 0.019, p = 0.048, respectively). There was a significant negative correlation between APLN levels and triglyceride (TG) and body mass index (BMI) levels (r =-0.426, p = < 0.001 and r =-0.330, p = 0.007, respec tively). Respectively, the areas under the receiver-operating characteristic (ROC) curves of the ELA/APLN, ELA/NO and APLN/NO ratios were 0.628, 0.590 and 0.709, 95% confident intervals (CI): 0.491–0.764, 0.450–0.730 and 0.579–0.840. Conclusions: Decreased levels of ELA and NO and increased APLN levels in schizophrenia suggest that these molecules may be involved in its etiopathology. The APLN/NO ratio also seems to show promise in the diagnosis of the disease and may be used in future.

Subfatin and spexin are two novel adipokines implicated in glucose homeostasis. This study was designed to investigate changes in blood subfatin and spexin levels during gestational diabetes mellitus (GDM) and childbirth, and define the mechanisms of these hormones in the physiopathology of GDM. A total of 60 pregnant women, comprising 30 diagnosed with GDM and 30 with normal gestation, were included in the study. The diagnosis of GDM was made through a 75-g oral glucose tolerance test (OGTT) administered between 24 and 28 weeks of pregnancy. The amounts of subfatin, spexin, and insulin were measured in blood samples by enzyme-linked immunosorbent assays; lipid profiles, glucose, and other biochemical parameters were measured by using an autoanalyzer. Levels of subfatin and spexin were significantly higher in blood samples drawn at baseline (before OGTT) in mothers with GDM compared to those with normal gestation. Similar observations were made in maternal and cord blood sampled at the end of pregnancy. However, at delivery, the increase in subfatin and spexin concentrations observed at baseline was abrogated in both groups of pregnant women, although levels in mothers with GDM were comparatively higher. These results show that levels of subfatin and spexin increased because of GDM and suggest that these hormones could be potential biomarkers for the diagnosis and management of GDM.

Purpose:Diabetic retinopathy (DRP) is the formation of edema and small vessels in the retina due to high blood glucose levels. Asprosin is a hormone that stimulates the release of glucose from the liver into the circulation. Considering the relationship between oxidative stress and DRP, our study aimed to determine the levels of the oxidative stress markers 4-hydroxynonenal (4-HNE) and 8-hydroxy-2′-deoxyguanosine (8-OHdG), as well as asprosin, in the blood and aqueous humor (Aq) of patients with and without DRP.Methods:Thirty patients with single-eye DRP and cataract (DRP + C), 30 patients with diabetes mellitus and cataract without DRP (DM + C), and 30 healthy control (CON) participants were enrolled into this retrospective study. Except for healthy controls, Aq and blood samples were taken from these patients during their cataract operation. Asprosin, 4-HNE, and 8-OHdG concentrations were analyzed using enzyme-linked immunosorbent assays. Results:In patients with DRP, the levels of asprosin, 4-HNE, and 8-OHdG were significantly higher in both Aq and blood samples compared with the group of patients without DRP.Conclusion:These findings suggest that the measurement of asprosin, 4-HNE, and 8-OHdG levels may support clinicians in determining the risk of DRP development.

Objectives: We compared the laboratory and clinical symptoms of coronavirus disease 2019 (COVID-19), other viral (beside COVID-19), and bacterial conjunctivitis patients with the values of controls. Methods: Twenty COVID-19, 15 other viral, 15 bacterial patients, and 15 control group were included in the study. COVID-19 was diagnosed with the real time reverse transcription–polymerase chain reaction while the indirect immunofluorescent antibody test was used in the detection of other viral agents. Bacterial agents were determined with the detection of the agent. The ophthalmologic examination of all cases was carried out by direct penlight, and the anterior segment evaluation was performed. The laboratory findings of all cases included in the study were obtained from the hospital records. Results: It was determined that 5% conjunctivitis and 5% diplopia developed in patients diagnosed with COVID-19, 20% conjunctivitis developed in patients due to other viral agents, and 66.6% conjunctivitis developed in patients due to bacterial agents. Conclusion: The incidence of conjunctivitis in COVID-19 patients was lower than in other viral and bacterial groups. Neutrophil/lymphocyte ratio and C-reactive protein come to the forefront as precious parameters with high specificity and sensitivity that might be useful to distinguish these diseases.
