The objective of present study was to specify the antioxidant, antimicrobial, cytotoxic, anticholinesterase, and antityrosinase activities of Verbascum oocarpum Murb., together with its volatile components. The methanolic and aqueous extracts of aerial parts of V. oocarpum indicated substantial antioxidant activity derived from benzoic acid, sinapic acid, and quercetin compounds. The methanolic extract displayed moderate antimicrobial activity against Escherichia coli, Pseudomonas aeruginosa, and Mycobacterium smegmatis. It also exhibited a higher IC50 value on tyrosinase than kojic acid, and lower acetylcholinesterase and butyrylcholinesterase inhibitor activities than galantamin. MTT analysis revealed that high concentrations of V. oocarpum extract can result in cytotoxicity, with an IC50 of 0.444 mg/mL. Common volatile components included pentadecane, hex-2(E)-enal, limonene, phenylacetaldehyde, isophorone, 1-methoxy-4-(2-propenyl)-benzene, 5,6,7,7a-tetrahydro-4,4,7a-trimethyl-2(4h)-benzofuranone and hexadecane. V. oocarpummay be of potential benefit to the food, cosmetical, and pharmaceutical sectors owing to its potent anticholinesterase, antioxidant, and antimicrobial activities, volatile components, and limited cytotoxicity.
RDL Kalıcı Kaydıİncele - ArticleAçık Erişim2018Chemical applications, scarification and stratification effects on seed germination of rare endemic Verbascum calycosum hausskn. ex Murb. (Scrophulariaceae)
Hilooğlu, Muhip; Sözen, Emel; Yücel, Ersin; Kandemir, Ali
Verbascum calycosum is an endemic plant species having considerable narrow distribution in Erzincan (Turkey) region. This species is known from only a single population and its habitats are highly threatened due to intensive human activities and soil erosion. In this study, the germination behavior of V. calycosum under different concentrations of NaCl, HCl, KNO3, GA3 (100 and 200 μM), hot-cold stratification and mechanical scarification were investigated. Seeds were exposed to a photoperiod of 8 h light/16 h dark with a 23/18 °C thermoperiod. Germination rates increased with GA3-100 μM (39%), GA3-200 μM (54.5%), mechanical scarification (34.5%) and cold stratification treatments (+4 °C, 23.25%; -20 °C, 18.25%) on the other hand, KNO3, NaCl, HCl and stratification with hot water treatments have decreased germination rates significantly when compared to the control (12.25%). Increased germination after GA3 application and mechanical scarification indicated that seeds of V. calycosum exhibited both non-deep and intermediate physiological dormancy as well as physical dormancy due to its hard seed coat. The highest speed of germination index was obtained at cold stratification of +4 °C and -20 °C (10.3). This study represents first report about seed dormancy and germination characteristics of V. calycosum. Overall, these results will provide valuable data for ex situ conservation of this rare endemic plant.

Background and Objective: Teucrium leucophyllum is endemic plant species having considerable narrow distribution in Erzincan (Turkey) Region. The aim of this study was to determine genetic diversity levels of T. leucophyllum in order to obtain data for its conservation. Method: Genomic DNA was isolated from 81 plants using CTAB method. Genetic diversity in four natural populations of T. leucophyllum was investigated by ISSR markers. A binary matrix was produced by scoring each amplified fragment as present (1) or absent (0) from each individual. The matrix was used to produce an input file and analyzed using the software programs; POPGENE 1.32 and GenAlex. Results: Twenty primers amplified a total of 434 bright and discernible bands of 220-3000 bp, of which 431 bands (PPB: 99.31%) were polymorphic in 4 populations. The genetic diversity was high at the species level; the observed number of alleles (Na) was 1.993, the effective number of alleles (Ne) was 1.404, the Nei’s genetic diversity (H) was 0.263, and the Shannon’s information index (I) was 0.418. A high level of gene flow (Nm: 2.208) and low-moderate genetic differentiation (Gst: 0.184) were also observed. Analysis of molecular variance showed that 11% of the total genetic diversity resided among populations, while 89% within the populations. Conclusion: Our results indicated that the conservation efforts should aim to preserve the extant populations of this endangered species in its natural habitat. Seeds could be collected from Kayacik and Bagistas populations for ex situ conservation practices.

We investigated the antioxidant and enzyme inhibitory activities and chemical composition of the hydro-distilled essential oil (0.35% yield) from aerial parts of Thymus spathulifolius. Antioxidant capacity of the oil was assessed by different methods including free radical scavenging (DPPH and ABTS), reducing power (FRAP and CUPRAC) and phosphomolybdenum assay. Inhibitory activities were analyzed against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), α-amylase, α-glucosidase, and tyrosinase. Twenty-one constituents were identified representing 97.2% of the total oil with thymol (50.5%), borneol (16.7%) and carvacrol (7.7%) as the major components. The essential oil exhibited good antioxidant activity with IC50 values of 3.82 and 0.22 mg/mL determined by free radical scavenging DPPH and ABTS, respectively. EC50 values of FRAP and CUPRAC were found to be 0.12 and 0.34 mg/mL, respectively. The results of the present study support the uses of T. spathulifolius essential oil as a source of natural antioxidants and bioactivities for functional foods and phytomedicines.
RDL Kalıcı Kaydıİncele - ArticleYalnız künye2016Pastinaca erzincanensis (Apiaceae), A New Species from Eastern Turkey and Its Phylogenetic Position within Tordylieae
Menemen, Yusuf; Kandemir, Ali; Downie, Stephen R.
Pastinaca erzincanensis Menemen & Kandemir (Apiaceae) is described as a new species, restricted to a single location in Erzincan Province, Turkey. To describe the species and assess its phylogenetic placement, field-collected and herbarium specimens were examined for morphological, fruit anatomical/micromorphological and palynological variation and included in a molecular phylogenetic study. Comparisons of leaf morphology, palynology and fruit anatomy suggest an affinity with Pastinaca (Tordylieae tribe Tordyliinae), although the presence of vittae in the fruit ribs is unique within the genus. Maximum parsimony and maximum likelihood analyses of nrDNA ITS sequences from representative members of tribe Tordylieae show that P. erzincanensis nests within a well-supported clade comprising members of Pastinaca, Malabaila and Trigonosciadium. Within a poorly supported Pastinaca s. lato clade, P. erzincanensis is sister group to M. pastinacifolia, with P. pimpinellifolia, P. yildizii and M. brachytaenia placed as successive sister taxa. Pastinaca erzincanensis differs from M. pastinacifolia, P. pimpinellifolia, P. yildizii and M. brachytaenia by having nine vittae on its dorsal mericarp surface (four vallecular and five rib vittae) and no commissural vittae. Based on characters of leaf morphology, palynology and fruit anatomy, and the molecular phylogenetic analysis results, we recognize the new species as belonging to the genus Pastinaca.

Barbarea integrifolia DC. (Brassicaceae) is an endangered and endemic species located in Erzincan and Gümüşhane provinces of Turkey. In total, 27 individuals from 2 natural populations were assessed using the random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) method coupled with sequence analysis of the internal transcribed spacer (ITS1) rDNA region. Genetic accuracy of RAPD-PCR was tested with 25 RAPD primers, and it resulted in 115 clear and reproducible DNA fragments from 13 RAPD primers. Among these, 76 (66.1%) fragments were found to be polymorphic. According to the ITS1 analysis of B. integrifolia populations, the plants exhibited a relatively poor genetic diversity. In total, 60 ITS1 nucleotide sequences from the GenBank database with 2 newly identified ITS1 sequences from the Turkish populations were used to construct a maximum-likelihood tree. The lengths of ITS1 sequences of B. integrifolia were found to be 268 bp for population I and 271 bp for population II. ITS1 sequences of B. integrifolia species showed 97% sequence identity. Phylogenetic analysis revealed that Barbarea species were monophyletic and showed high bootstrap values. B. integrifolia had closer relationships with Cardamineae taxa, including Cardamine, Rorippa, and Armoracia. On the contrary, allied genus Nasturtium species were separated from Cardamineae taxa. The genetic information obtained from this study could be used for the development of conservation strategies not only for B. integrifolia but also for rare and endangered plant species.
