Aim: In this study, it was aimed to investigate the effect of coenzyme Q10 (CoQ10) on cisplatin-induced oxidative retinal damage in rats biochemically and histopathologically. Materials and methods: Thirty male Wistar albino rats were divided into 3 groups randomly: untreated control (C group), only 2.5 mg/kg cisplatin daily administrated group for 2 weeks (CP group), 2.5 mg/kg cisplatin + 20 mg/kg orally CoQ10 daily administrated group for 2 weeks (CoQC group). At the end of experimental period, blood samples obtained before sacrification for the biochemical examination of serum malondialdehyde (MDA), total glutathione (tGSH), total oxidant system (TOS), total antioxidant systemic (TAS) levels and after eyes were removed for examined histopathology. Results: As a result of our study, severe histopathological damage was detected in the retinal tissue of the cisplatin group with serum malondialdehyde (MDA) and total oxidant system (TOS) levels were high and total glutathione (tGSH) and total antioxidant systemic (TAS) levels were low. However, it was observed that the histopathological damage associated with cisplatin was decreased in the retinal tissue of the CoQ10 group, which inhibited the increase in blood serum MDA/TOS levels and decrease in tGSH/TAS levels. Conclusion: The biochemical and histopathological results of our study were compatible with each other, so we concluded that the damage to the rat retinal tissue caused by cisplatin may be reversible with coenzyme.

Ozcicek, Fatih; Kara, Ali Veysel; Akbas, Emin Murat; Kurt, Nezahat; Yazici, Gulce Naz; Cankaya, Murat; Mammadov, Renad; Ozcicek, Adalet; Suleyman, Halis

Intestinal mucositis is an important problem in the patients receiving cancer treatment. We aimed to investigate the effect of anakinra, which is a well known anti-oxidant and anti-inflammatory agent, on methotrexate-induced small intestine mucositis in rats. Forty rats were divided into 4 groups with 10 in each group. The healthy group (HG) and the methotrexate group (MTXG) were given distilled water, while the methotrexate + anakinra 50 (MTX+ANA50) and the methotrexate + anakinra 100 (MTX+ANA100) groups were intraperitoneally administered 50 and 100 mg/kg of anakinra. After one hour, the MTXG, MTX+ANA50 and MTX+ANA100 groups were given oral methotrexate at a dose of 5 mg/kg. This procedure was repeated once a day for 7 days. After the rats had been sacrificed, the small intestine tissue of rats were removed for the assesment of biochemical markers, histopathological evaluation and gene expression analyze. Statistical analyses of the data were performed using one-way ANOVA. Malondialdehyde (MDA), myeloperoxidase (MPO) and interleukin-6 (IL-6) levels were significantly higher, whereas total glutathione (tGSH) levels were significantly lower in MTXG (P<0.001) compared to other groups. MTX also increased IL-1β and TNF-α gene expression levels in MTXG (P<0.001). Inflammatory cell infiltration and damage to the villus were observed histopathologically in the MTXG group, whereas only mild inflammation was seen in the MTX+ANA100 group. A dose of 100 mg/kg of anakinra prevented the increase of the biochemical markers and gene expression levels better than a dose of 50 mg/kg. Intestinal mucositis caused by MTX may be preventible by co-administered anakinra.

Bayram, Rana; Erhan, Ertugrul; Seckin, Ender; Salcan, Ismail; Mammadov, Renad; Kurt, Nezahat; Suleyman, Halis

In this study, we have examined the protective and proinflammatory effects of anakinra against paclitaxel-associated oxidative auditory nerve damage induced by paclitaxel in rats. Experimental animals were allocated into three groups: Paclitaxel administered controls, paclitaxel + anakinra administered group, and untreated health controls. At the end of this period, animals were sacrificed by high dose thiopental (50 mg/kg) anesthesia and auditory nerves were removed. Then, malondialdehyde, total glutathione, Interleukin-1β and nuclear factor kappa-B levels were measured in the auditory nerve tissue. Our biochemical test results showed that paclitaxel significantly increased the amount of malondialdehyde in auditory nerve tissue compared to anakinra and healthy group (P < 0.0001), whereas total glutathione level decreased. In addition, paclitaxel was found to increase interleukin-1β and nuclear factor kappa-B significantly compared to anakinra and healthy group (P < 0.0001). It was concluded that anakinra is effective in suppressing paclitaxel related oxidative stress and proinflammatory events in auditory nerve tissue.

Erhan, Ertugrul; Bayram, Rana; Suleyman, Bahadir; Kurt, Nezahat; Suleyman, Halis; Salcan, Ismail

One of the major side effects of dose-limiting cisplatin is ototoxicity, resulting in restricted clinical use. Cerebrolysin is a neuroprotective, neurotrophic and antioxidant drug derived from the brains of pigs. No studies have investigated the positive effects of Cerebrolysin on the damage to auditory nerves caused by cisplatin- induced oxidative stress. Eighteen rats were randomly divided into three groups of six: the Cerebrolysin and cisplatin group (CCG), the cisplatin group (CG) and the healthy group (HG). The CCG was administered 2.5 mL/kg of Cerebrolysin via subcutaneous injection daily, while the CG and HG were administered physiological saline solutions daily. Every other day, after receiving their dose of Cerebrolysin or physiological saline solution, the CCG and CG groups were administered 5 mg/kg of cisplatin via intraperitoneal injection (IP) every other day. The substances were administered for eight days. In acoustic nerve tissue, while the CCís oxidant parameters were significantly higher (p < 0.0001) than those of the CCG and HG groups, its antioxidant parameters were also significantly lower (p < 0.0001). Cerebrolysin antagonized the balance of oxidants and antioxidants in acoustic nerve tissue, resulting in higher concentrations of oxidants. The results suggested that Cerebrolysin may be useful in the treatment of cisplatin-induced oxidative acoustic nerve injury.

OBJECTIVE: To examine the effect of desloratadine on ovarian ischemia-reperfusion (I/R) injury in rats by biochemical and histopathological methods. STUDY DESIGN: I/R was induced in the right ovaries after distilled water was given to a group of female albino Wistar rats in this study. 15 mg/kg desloratadine was orally administered to another group and I/R was induced in their ovaries, whereas I/R was not induced in the sham operation group. After 2 hours of ischemia and 2 hours of reperfusion, malondialdehyde (MDA), total glutathione (tGSH), nuclear factor-κB (NF-κB), interleukin 1 beta (IL-1β), and tumor necrosis factor alpha (TNF-α) levels were measured in ovarian tissues extracted from the body of the animals sacrificed with a high dose of ketamine (120 mg/kg). Tissues were also evaluated histopathologically. RESULTS: It has been revealed that desloratadine significantly inhibited the increase of I/R-associated MDA, NF-κB, IL-1β, and TNF-α levels in the ovarian tissues of animals and the decrease in tGSH levels. In addition, desloratadine decreased severe I/R-associated histopathological damage. CONCLUSION: Our experimental results have shown that desloratadine is beneficial in terms of protecting ovarian tissue against I/R injury.

Effect of rutin on cytarabine-associated pulmonary oedema and oxidative stress in rats

Bilgin, Asli O.; Mammadov, Renad; Suleyman, Bahadir; Unver, Edhem; Ozcicek, Fatih; Soyturk, Mehmet; Cimen, Ferda K.; Kurt, Nezahat; Suleyman, Halis

Cytarabine is effectively used in the treatment of adult acute leukemia, but it has a dose-limiting side effect of fatal pulmonary oedema because it increases the vascular permeability of the alveolar capillaries. The aim of the present study was to conduct a radiological, biochemical and histopathological investigation of the effect of rutin on cytarabine-associated pulmonary oedema in rats. Rats were treated with a combination of rutin+cytarabine by administering oral rutin at a dose of 50 mg/kg; other rat groups were orally administered the same volume of physiological saline. One hour after administration of rutin or saline, the rutin+cytarabine and cytarabine groups received an intraperitoneal injection of cytarabine (200 mg/kg). This administration procedure was repeated once a day for 14 days. Radiologically, 50% of the animals given cytarabine alone showed lung oedema, but the rutin+cytarabine group showed no oedema. The inclusion of rutin decreased the amounts of cytarabine-associated malondialdehyde, tumour necrosis factor-α, and nuclear factor-κB in the lung tissue. Rutin also inhibited the reduction of total glutathione by nitric oxide. These findings suggest that rutin may be a beneficial adjunct that can minimise the development of cytarabine-associated pulmonary oedema.

Erhan, Ertugrul; Mammadov, Renad; Altuner, Durdu; Cimen, Ferda; Kurt, Nezahat; Hacimuftuoglu, Fazil; Turan, Fatih; Terzi, Suat; Celiker, Metin; Suleyman, Halis

No previous studies have examined the effect of Hippophae rhamnoides fruit extract (HRE), which contains both water and fat soluble vitamins, on cisplatin-induced ototoxicity. Therefore, the aim of the study was to investigate the effects of FIRE on cisplatin-induced ototoxicity in Guinea pigs and to evaluate the biochemical, gene expression and histopathological changes associated with cisplatin use. Experimental animals were divided into three groups: healthy (HG), HRE+cisplatin (HRC) and cisplatin control (CCG) groups. Biochemical, gene expression and histopathological examinations were carried on the removed inner ear tissues. In the HRC group, the oxidant parameter was lower and the anti-oxidant parameter was higher than in the CCG. These results are supported by gene expression levels and histopathological results. The use of FIRE against cisplatin-induced oxidative ototoxicity may be easier, more cost effective and more beneficial than the use of vitamins alone or in combination with each other.

The effect of taxifolin on cisplatin induced oxidative cochlear damage in rats: Biochemical and histopathological examination

Erhan, Ertugrul; Bayram, Rana; Cimen, Ferda Keskin; Altuner, Durdu; Seckin, Ender; Kurt, Nezahat; Suleyman, Halis

Ototoxicity of cisplatin is one of the major dose-limiting side effects. Cisplatin-dependent ototoxic-ity is known to be associated with reactive oxygen species. Protective effect of taxifolin, being a flavanone found in onions, milk thistle, French maritime, and Douglas fir bark, against cisplatin-associated ototoxicity will be examined in this study. There are no studies in the literature examining the protective effect of taxifolin against cisplatin-induced ototoxicity. 50 mg/kg taxifolin was orally administered to TXC group rats (n-6), and distilled water was orally administered as a solvent to CG (n-6) and HG (n-6) groups. One hour later, 5 mg/kg cisplatin was administered intraperitoneally (i.p) to TXC and CG groups. This procedure was repeated once a day for 7 days. At the end of this period, all animals were sacrificed by high-dose anesthesia (50 mg/kg thiopen-tal sodium) and biochemical and histopathological examinations were performed on the dissected cochlear tissue. Our biochemical test results showed that oxidant parameters were significantly increased whereas antiox-idant parameters were significantly decreased in the cisplatin group (CG) compared to taxifolin (TXC) and healthy (HG) groups (p < 0.0001 for both parameters). Histopathological results showed that severe cochlear vestibular membrane degeneration, dilated conjunctival blood vessels, edema and destruction developed in the cisplatin group. However, no pathological findings were found in the taxifolin-treated group except for mild degeneration and edema. This information suggests that taxifolin may be useful in the treatment of cisplatin-associated oxidative cochlear damage.

Ucak, Turgay; Karakurt, Yucel; Tasli, Gamze; Cimen, Ferda Keskin; Icel, Erel; Kurt, Nezahat; Ahiskali, Ibrahim; Süleyman, Halis

Background: We aimed to determine the protective effects of thiamine pyrophosphate on ethanol induced optic neuropathy in an experimental model. Methods: The rats were assigned into 4 groups, with 6 rats in each group as follows: healthy controls (HC group), only ethanol administered group (EtOH group), ethanol + thiamine pyrophosphate (20 mg/kg) administered group (TEt-20 group), and only thiamine pyrophosphate (20 mg/kg) (TPG group) administered group. To the rats in TEt-20 and TPG groups, 20 mg/kg thiamine pyrophosphate was administered via intraperitoneal route. To the rats in HC and EtOH groups, the same volume (0.5 ml) of distilled water as solvent was applied in the same manner. To the rats in TEt-20 and EtOH groups, one hour after application of thiamine pyrophosphate or distilled water, 32% ethanol with a dose of 5 g/kg was administered via oral gavage. This procedure was repeated once a day for 6 weeks. From the blood samples and tissues obtained from the rats, Malondialdehyde (MDA), reduced glutathione (GSH), interleukin 1 beta (IL-1β) and tumor necrosis factor alpha (TNF-α) levels were studied. Histopathological evaluations were performed to the optic nerve tissue. Results: Serum and tissue IL-1β, TNF-α and MDA levels were the highest in EtOH group which were significantly lower in thiamine pyrophosphate administered group (TEt-20 group) (p: 0.001). Serum and tissue reduced GSH levels were the lowest in EtOH group which were also significantly higher in TEt-20 group (p:0.001). In histopathological evaluations, in EtOH group there was obvious destruction and edema with hemorrhage and dilated blood vessels which were not present in any other groups. Conclusions: There was an apparent destruction in ethanol administered group in histopathological analyses with an augmented level of oxidative stress markers and all those alterations were prevented with concomitant thiamine pyrophosphate administration. These protective effects of thiamine pyrophosphate are extremely important in chronic ethanol consumption. Clinical studies are warranted to define the exact role of thiamine pyrophosphate in prevention of ethanol induced optic neuropathy.

Taşlı, Nurdan Gamze; Uçak, Turgay; Karakurt, Yücel; Keskin Çimen, Ferda; Özbek Bilgin, Aslı; Kurt, Nezahat; Süleyman, Halis

Aim: To determine the role of rutin in prevention of cisplatin induced retinal and optic nerve injury in an experimental study. Materials and methods: Totally 18 albino Wistar male rats were assigned into three groups, as follows: healthy controls (HC group), only cisplatin administered group for 14 days (CIS group), and rutin + cisplatin administered group for 14 days (RC group). Blood samples were obtained from animals just before the scarification. Serum malondialdehyde (MDA), myeloperoxidase (MPO), total glutathione (tGSH), superoxide dismutase (SOD), interleukin 1 beta (IL-1β) and tumour necrosis factor alpha (TNF-α) levels were investigated. The eyes were enucleated for histopathological evaluations of retina and optic nerve. Results: MDA, MPO, IL-1β and TNF-α levels were statistically significantly higher (p < 0.001) in CIS group compared with other two groups while tGSH and SOD levels were statistically significantly lower (p < 0.001). Regarding these parameters, in CIS group MDA, MPO, IL1β and TNF-α levels were statistically significantly increased with cisplatin administration and giving rutin concomitantly with cisplatin prevented this increase. On the other hand, tGSH and SOD levels were statistically significantly decreased with cisplatin administration and giving rutin concomitantly with cisplatin prevented this decrease. In qualitative analyses of histopathological findings of retina and optic nerve; the results of RC group were similar with the results of healthy controls; but there was statistically significant differences between CIS group and other two groups (p < 0.001). Conclusions: Concomitant rutin administration may prevent the detrimental effects of cisplatin on lipid peroxidation, oxidative stress and inflammation markers and may also avert the histopathological damage on retina and optic nerve. Further studies are warranted to determine the effects of cisplatin and rutin on eye.

Postoperative pain begins with tissue incision, continues with the production of free oxygen radicals, and is intensified by the inflammatory response. We investigated the effect of esomeprazole on postoperative pain and lung and heart complications of laparotomy. Albino Wistar-type rats were grouped into the paw scalpel incision (PSI), applied control (SIC), PSI + 20 mg/kg esomeprazole (SIE), PSI + 20 mg/kg meperidine (SIM), and healthy group (HG). For the laparotomy test, the rats were grouped as follows: laparotomy applied control (LTC), laparotomy + 20 mg/kg esomeprazole (LTE), laparotomy + 20 mg/kg meperidine (LTM), and healthy group (HG). Meperidine suppressed the PSI-induced pain. Esomeprazole decreased the level of oxidant and proinflammatory cytokines in the serum and lung and heart tissues of the LTE group and in the paw tissues of the SIE group, but meperidine did not. There were no histopathological findings in the lung and heart tissues of the LTE group, but tissue damage was found in the LTC and LTM groups. Esomeprazole may help reduce major surgery complications.

Nayki, Cenk; Nayki, Umit; Keskin Cimen, Ferda; Kulhan, Mehmet; Yapca, Omer Erkan; Kurt, Nezahat; Bilgin Ozbek, Aslı

The effect of rutin on ovarian ischemia-reperfusion (I/R) injury was investigated in this experimental study. Eighteen Wistar albino female rats were divided into three groups as follows: I/R group (IRG; n = 6), 50 mg/kg rutin + I/R group (RG; n = 6), and a healthy control group scheduled for a sham operation (SG; n = 6). 2 h of ischemia and following 2 h of reperfusion were created in the IRG and RG by using a torsion model involving atraumatic vascular clips. Rutin, a flavonoid glycoside, was injected intraperitoneally at the dose of 50 mg/kg to RG group 1 h before reperfusion. Then, rats were euthanized and their ovaries were removed for biochemical and histopathological examination and also assessment of the gene expressions. IRG group had a significant increase in malondialdehyde (MDA) levels, in the expressions of tumor necrosis factor alpha (TNF-α) and interleukin 1 beta (IL-1β), and also in the activity of cyclooxygenase 2 (COX-2) unlike the significant decrease in total glutathione (tGSH) levels and the activity of COX-1 when compared to the SG group. However, rutin significantly decreased MDA levels, the expressions of TNF-α and IL-1β, and also the activity of COX-2 while it increased significantly tGSH levels and the activity of COX-1 in the RG group in comparison with the IRG group. Rutin ameliorated the I/R-induced ovarian injury in rats via its possible antioxidative and anti-inflammatory effects.

Karakurt, Yücel; Uçak, Turgay; Tasli, NurdanGamze; Ahiskali, Ibrahim; Şipal, Sare; Kurt, Nezahat; Süleyman, Halis

Aim: Lutein is one of the most common carotenoids defined in human plasma as having potent anti-oxidant effects. We aimed to determine the biochemical and histopathological effects of lutein on cisplatin-induced oxidative retinal injury in rats. Materials and methods: Twenty-four rats were equally divided into four groups as healthy controls (HC group), only cisplatin (5 mg/kg) administered group (CIS group), Lutein (0.5 mg/kg) + cisplatin (5 mg/kg) administered group (LC group), and only Lutein (0.5 mg/kg) (LUT group) administered group. From the blood samples obtained, serum malondialdehyde (MDA), total glutathione (tGSH), interleukin 1 beta (IL-1β), and tumor necrosis factor alpha (TNF-α) levels were investigated. In histopathological analyses, the total retinal thickness, retinal pigment epithelium (RPE), photoreceptor layer (PL), outer nuclear layer (ONL), outer plexiform layer (OPL), inner nuclear layer (INL), inner plexiform layer (IPL), and ganglion cell layer (GCL) were evaluated. Results: MDA, IL-1β, and TNF-a levels were statistically significantly higher (p < 0.001) in CIS group compared with other three groups while tGSH levels were statistically significantly lower (p < 0.001). In subgroup analyses, there was no any statistically significant difference regarding all four parameters analyzed between HC, LC, and LUT groups. In histopathological analyses, cisplatin-induced retinal damage included atrophy and disorganization on outer segment, degeneration and detachment of RPE and PL from choroid, degeneration and edema of INL and IPL, total degeneration of GCL; while cisplatin-induced retinal damage was determined to be significantly prevented with 0.5 mg lutein treatment on histopathological evaluations. Conclusions: Lutein co-administration was highly effective in prevention of cisplatin-induced retinal damage due to the anti-oxidant and anti-inflammatory effects of lutein.

AIM: To determine the effects of Rutin on methanol induced optic neuropathy and compare the results with the effects of ethanol. METHODS: Totally 30 rats were divided into 5 groups, with 6 rats in each group as follows: healthy controls (C), methotrexate (MTX), methotrexate+methanol (MTM), methotrexate+methanol+ethanol (MTME) and methotrexate+ methanol+Rutin (MTMR). In all rabbits except those of the control group, MTX, diluted in sterile serum physiologic, 0.3 mg/kg per oral was applied for 7d by the aid of a tube. After this procedure to the rats of MTM, MTME and MTMR groups, 20% methanol with a dose of 3 g/kg per oral was given by the aid of a tube. In MTME group, 4h after the application of methanol, 20% ethanol was applied by the same way with a dose of 0.5 g/kg. On the other hand, in MTMR group 4h after the application of methanol, Rutin, which was dissolved in distilled water, was applied by the same way with a dose of 50 mg/kg. RESULTS: There were statistically significant differences in tissue 8-hydroxy-2 deoxyguanine (8-OHdG), interleukin-1β (IL-1β), tumor necrosis factor-alpha (TNF-α), malondialdehyde (MDA), myeloperoxidase (MPO). glutathione peroxidase (tGSH) and superoxide dismutase (SOD) levels between groups (P<0.001). In MTMR group tissue 8-OHdG, IL-1β, MDA, and MPO levels were similar with the healthy controls but significantly different than the other groups. In histopathological evaluations, in MTX group there was moderate focal destruction, hemorrhage and decrease in number of astrocytes and oligodendrocytes; in MTM group there was severe destruction and edema with decrease in number of astrocytes and oligodendrocytes; in MTME group there was mild hemorrhage, mild edema, mildly dilated blood vessels with congestion while in MTMR group, optic nerve tissue was resembling the healthy controls. CONCLUSION: Rutin may prevent methanol-induced optic neuropathy via anti-inflammatory effects and decreasing the oxidative stress. New treatment options are warranted in this disease to avoid loss of vision in patients.
