Alemdar, Ali; Yilmaz, Ismayil; Ozcicek, Fatih; Bulut, Seval; Eden, Arif Onur; Gursul, Cebrail; Mendi, Ali Sefa; Kuzucu, Mehmet; Altuner, Durdu; Suleyman, Halis

Background and Objective: Ischemia-reperfusion (I/R) injury begins with tissue oxygen deprivation and continues with oxidative stress and inflammatory response. Mirtazapine is an antidepressant drug with antioxidant and anti-inflammatory properties. The current study was to investigate the effect of mirtazapine against I/R induced liver injury in rats. Materials and Methods: Albino Wistar-type male rats were divided into sham operation (SHAM), I/R (IR) and I/R+mirtazapine administrated (IRM) groups. One hour before anaesthesia, 20 mg kg(-1) mirtazapine was administered to the IRM group of the animals and distilled water was applied to the SHAM and IR groups as a solvent. I/R was achieved by clamping the hepatic artery (except for the SHAM group). Following I/R, all rat groups were killed with high-dose anaesthesia. Malondialdehyde (MDA), total glutathione (tGSH), nuclear factor kappa B (NF-kappa B), interleukin 1 beta (IL-1 beta) and tumour necrosis factor-alpha (TNF-alpha) were determined in liver tissues. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were measured in blood serum. In addition, histopathological examination was performed on liver tissue samples. Results: Mirtazapine prevented increased levels of MDA, NF-kappa B, IL-1 beta, TNF-alpha, ALT and AST in liver tissue with I/R and a decrease in tGSH. Furthermore, mirtazapine has alleviated I/R-associated severe hepatocyte degeneration, necrosis and other structural disorders in the liver. Conclusion: Biochemical and histopathological experimental results suggest that mirtazapine may be useful in the treatment of I/R-induced liver injury.

Delen, Leman Acun; Dişli, Zeliha Korkmaz; Taş, Hakan G.; Kuyrukluyildiz, Ufuk; Yazici, Gulce N.; Süleyman, Bahadir; Kuzucu, Mehmet; Gürsu, Cebrail; Suleyman, Halis

Propofol may cause an increase in reactive oxygen species in the body. In this study, we tested the effect of antioxidant thiamine pyrophosphate (TPP) on propofol-induced liver damage. The eighteen rats were split into three groups: HG, healthy; PP, propofol-treated (50 mg/kg) and PT, treated with propofol (50 mg/kg) and TPP (25 mg/kg). Total glutathione (tGSH), total oxidant (TOS), and total antioxidant (TAS) levels were tested together with aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH) and malondialdehyde (MDA). Histopathological examination of the tissues was performed. We have found that levels of MDA, TOS, ALT, AST, and LDH were all higher in PP group than in HG and PT groups (p < 0.05). In PP group, the TAS and tGSH levels were statistically substantially lower. The PT for oxidants levels showed a statistically significant reduction. In PT group, the levels of antioxidants were found to be considerably higher. The epitheliums, glands, and vascular structures of the PTs were histologically close to normal. By boosting antioxidants, TPP may help to reduce propofol-induced liver damage.

Facile synthesis of TiO2-coated cotton fabric and its versatile applications in photocatalysis, pH sensor and antibacterial activities

Özdemir, Agah Oktay; Caglar, Bulent; Çubuk, Osman; Coldur, Fatih; Kuzucu, Mehmet; Guner, Eda Keles; Doğan, Bilge; Caglar, Sema; Özdokur, Kemal Volkan

TiO2 nanoparticles-coated cotton fabric was readily prepared by the hydrolysis of TiCl4 solution over cotton fabric and characterized various techniques in comparison with pristine cotton fabric. Initially, the photocatalytic application of the obtained fabric was carried out by using it as a catalyst for the photodegradation of rhodamine B. A higher degradation ratio for rhodamine B in the presence of TiO2-coated cotton fabric as photocatalyst was observed under visible light compared to the one performed under UVA. The reactive species trapping experiments revealed that the superoxide radicals played a major role than other radical species and two different degradation mechanisms were proposed according the trapping results. Subsequently, considering that the fabricated TiO2-coated cotton fabric is a very suitable sensor material for the development of a wearable flexible pH sensor, a potentiometric solid state pH sensor prototype was constructed and tested for pH measurements. The proposed prototype exhibited a linear potentiometric response in the pH range from 2.0 to 9.0 with a slope of −23.9 mV/decade. The pH accuracies (as relative error %) calculated for the current electrode with respect to the commercial electrode in the studied buffer solutions were ranged between −1.8% and 1.9%. The presently proposed pH electrode allowed pH measurements highly coherent with commercially available pH electrode. Finally, the antibacterial activity of the TiO2 nanoparticles was evaluated by minimum bactericidal concentration (MBC), minimum inhibitory concentration (MIC) testing and also antibacterial property of the TiO2 nanoparticles-coated cotton fabric was demonstrated by GB/T 20944.3–2008 method using Staphylococcus aureus (ATCC 6538) and Escherichia coli (ATCC 25922) as representative strains of Gram-positive and Gram-negative bacteria, respectively. This study would be indicative to design of nano metal oxide-coated cotton fabrics in the application of photocatalyst, pH sensor and antibacterial agent against bacteria.

Effect of Usnea longisima Ethyl Acetate Extract on Staphylococcus aureus-Induced Experimental Rhinosinusitis in Rats

Dilber, Muhammet; Erhan, Ertugrul; Bulut, Seval; Kuzucu, Mehmet; Akyuz, Sumeyye; Suleyman, Halis

SUMMARY. The role of oxidant and proinflammatory cytokines in the pathogenesis of rhinosinusitis induced by Staphylococcus aureus has been demonstrated in studies. Usnea longissima ethyl acetate extract (ULE) was obtained from this lichen species. ULE is known to have antibacterial, antioxidant and anti-inflammatory effects. The objective of the study is to investigate the effect of ULE on S. aureus-induced experimental rhinosinusity in rats and compare it with cefazolin. Anesthesia was created through virtue of intraperitoneal injection of 25 mg/kg thiopental sodium to male albino Wistar rats. Then the rats were divided into groups as healthy (HG), ones inoculated with S. aureus (RSG) to their nostril, ones inoculated with S. aureus to their nostril + ULE (REA) applied and ones with S. aureus inoculated to their nostril + cefazolin (RCG) applied. Our biochemical test results revealed the fact that ULE was able to inhibit inflammatory mediators such as nuclear factor kappa B (NF-κB), tumor necrosis factor alpha (TNF-α), an interleukin 1 beta (IL-1β) and cyclooxygenase-2 (COX-2) increase with S. aureus in sinonasal tissue better than cefazolin. Moreover, while ULE significantly suppressed the increase in the amount of malondialdehyde (MDA), the oxidant parameter in the sinonasal tissue, and the decrease in the antioxidant total glutathione (tGSH) and cyclooxygenase-1 (COX-1), cefazolin could not. ULE and cefazolin were observed to be effective on standard bacteria. From our experimental results, it is understood that ULE prevents oxidative and inflammatory damage induced by S. aureus in the sinonasal tissue better than cefazolin.

Artichoke (Cynara Scolymus) Methanolic Leaf Extract Alleviates Diethylnitrosamine-Induced Toxicity in BALB/c Mouse Brain: Involvement of Oxidative Stress and Apoptotically Related Klotho/PPARγ Signaling

Cicek, Betul; Genc, Sidika; Yeni, Yesim; Kuzucu, Mehmet; Cetin, Ahmet; Yildirim, Serkan; Bolat, Ismail; Kantarci, Mecit; Hacimuftuoglu, Ahmet; Lazopoulos, Georgios

    (1) Background: Various epidemiological studies suggest that oxidative stress and disrupted neuronal function are mechanistically linked to neurodegenerative diseases (NDs), including Parkinson’s disease (PD) and Alzheimer’s disease (AD). DNA damage, oxidative stress, lipid peroxidation, and eventually, cell death such as NDs can be induced by nitrosamine-related compounds, leading to neurodegeneration. A limited number of studies have reported that exposure to diethylnitrosamine (DEN), which is commonly found in processed/preserved foods, causes biochemical abnormalities in the brain. Artichoke leaves have been used in traditional medicine as a beneficial source of bioactive components such as hydroxycinnamic acids, cynarine, chlorogenic acid, and flavonoids (luteolin and apigenin). The aim of this study is to investigate the favorable effects of exogenous artichoke (Cynara scolymus) methanolic leaf extract supplementation in ameliorating DEN-induced deleterious effects in BALB/c mouse brains. (2) Methods: This study was designed to evaluate DEN (toxicity induction by 100 mg/kg) and artichoke (protective effects of 0.8 and 1.6 g/kg treatment) for 14 days. All groups underwent a locomotor activity test to evaluate motor activity. In brain tissue, oxidative stress indicators (TAC, TOS, and MDA), Klotho and PPARγ levels, and apoptotic markers (Bax, Bcl-2, and caspase-3) were measured. Brain slices were also examined histopathologically. (3) Results: Artichoke effectively ameliorated DEN-induced toxicity with increasing artichoke dose. Impaired motor function and elevated oxidative stress markers (decreasing MDA and TOS levels and increasing TAC level) induced by DEN intoxication were markedly restored by high-dose artichoke treatment. Artichoke significantly improved the levels of Klotho and PPARγ, which are neuroprotective factors, in mouse brain tissue exposed to DEN. In addition, caspase-3 and Bax levels were reduced, whereas the Bcl-2 level was elevated with artichoke treatment. Furthermore, recovery was confirmed by histopathological analysis. (4) Conclusions: Artichoke exerted neuroprotective effects against DEN-induced brain toxicity by mitigating oxidant parameters and exerting antioxidant and antiapoptotic effects. Further research is needed to fully identify the favorable impact of artichoke supplementation on all aspects of DEN brain intoxication.

Cloning, Over-Expression, and Purification of β–Carbonic Anhydrasefrom an Extremophilic Bacterium: Deinococcus radiodurans

Taşkın Kafa, Ayşe Hümeyra; Cihan, Arzu Coleri; Cankaya, Murat; Kuzucu, Mehmet

In this study, the cloning, purification and initial characterization of carbonic anhydrase (DrCA) enzyme which we consider to be important in the resistance physiology from extremely radioresistant bacteria Deinococcus radiodurans is performed. In addition, the effect of increased gamma irradiation doses on pH-related DrCA enzyme activity was determined. DrCA activity after radiation treatment showed that the activity continuously increased by 6 fold, up to the first 800 Gy, which a decrease in activity was observed thereafter. The maximum $CO_2$ hydration activity for DrCA enzyme was observed at pH 7.0 and 40°C. DrCA enzyme, homo-dimer complex, is slightly thermostable. The activity of DrCA was significantly enhanced by several metal ions, especially $Zn^{2+}$, which resulted in 5-fold increases of $CO_2$ hydration activity. Also sulfonamide showed inhibitory effect on the pure enzyme. The apparent Km and Vmax for $CO_2$ as substrate were 8.4 mM and 637 WAU/mg for DrCA respectively. The $CO_2$ hydration assay demonstrated that the specific activity of purified recombinant enzymes (DrCA) was significantly high.

Ince, Sefa; Ozer, Mehmet; Kadioglu, Berrin Goktug; Kuzucu, Mehmet; Gundogdu, Betul; Gursul, Cebrail; Suleyman, Zeynep; Suleyman, Halis

Background and Objective: Interleukin 1 beta (IL-1 beta) and tumour necrosis factor-alpha (TNF-alpha) as the proinflammatory cytokines, whose production increases during reperfusion after ischemia (I/R), were reported to produce the Reactive Oxygen Species (ROS). It was reported that sunitinib has proinflammatory cytokine expression suppression and antioxidant effects. The study aims to investigate the effects of sunitinib on ovarian injuries created by I/R in albino Wistar female rats biochemically and histopathologically. Materials and Methods: Animals divided into 3 groups as sunitinib+ovarian tissues (SOIR), I/R ovarian tissues (OIR) and Sham operation (SG) groups. There were ischemia and reperfusion procedures for 2 hrs on the right ovaries of the OIR and SOIR group rats. Results: The Nuclear Factor kappa B (NF-kappa B), Tumour Necrosis Factor-alpha (TNF-alpha), Malondialdehyde (MDA) and Interleukin 1 beta (IL-1 beta) levels in the OIR group's ovarian tissue were high whereas their total glutathione (tGSH) level was found to be low compared to the SOIR and SG group. While distinct histopathologic injuries were seen in the ovarian tissue of the OIR group, histopathologic injuries in the SOIR group were mild. Conclusion: Sunitinib prevented the oxidant and proinflammatory cytokine increase and antioxidant decrease related to I/R. It is concluded that sunitinib might be used for the treatment of ovarian I/R injuries.

In Vitro Anticancer Activity of Novel Co(II) and Ni(II) Complexes of Non-steroidal Anti-inflammatory Drug Niflumic Acid Against Human Breast Adenocarcinoma MCF-7 Cells

Caglar, Sema; Altay, Ahmet; Kuzucu, Mehmet; Caglar, Bulent

Herein, we report the synthesis, characterization and anticancer activity of six novel complexes of non-steroidal anti-inflammatory drug niflumic acid with Co(II) and Ni(II). In vitro cytotoxicity screening in MCF-7, HepG2 and HT-29 cancer cell lines showed that the complex 3 [Co(nif)2(met)(4-pic)] and complex 6 [Ni(nif)2(met)(4-pic)] among all the complexes exhibited the highest cytotoxicity against MCF-7 cells with IC50 values of 11.14 µM and, 41.47 µM, respectively. Besides, all the complexes exhibited significantly higher selectivity towards mouse fibroblast 3T3L1 cells. Further mechanistic studies with both complexes on MCF-7 cells revealed their cytotoxic action through the mitochondrial-dependent apoptotic pathway causing an increase oxidative/nitrosative stress, decrease in mitochondrial membrane potential (ΔΨm), inducing the multicaspase activation and arresting the cell cycle at S phase. q-PCR analysis resulted in an increase in the expression of the apoptotic marker proteins bax, p53 and caspase-3 and -8 in MCF-7 cells, but a decrease in the expression of antiapoptotic bcl-2 gene. Moreover, both complexes induced the apoptosis through the inhibition of PI3K/Akt signaling pathway by decreasing the expression of PI3K and increasing dephosphorylation form of Akt protein. These results provide a significant contribution to the explanation of the anticancer mechanisms of these complexes in MCF-7 cells. [Figure not available: see fulltext.]

The effect of adenosine triphosphate on bevacizumab-induced ovarian damage and reproductive dysfunction in rats

Ince, Sefa; Ozer, Mehmet; Kadioğlu, Berrin Goktug; Kuzucu, Mehmet; Yilmaz, Sevil Karahan; Özkaraca, Mustafa; Gezer, Arzu; Suleyman, Halis

We investigated the effect of ATP’s protection against possible bevacizumab-induced ovarian damage and reproductive dysfunction in female albino Wistar rats. A total of 42 rats, 36 females, and 6 males were used in the experiment. Normal saline (0.9% NaCl) was injected as a solvent to the Bevacizumab (BVZ; n = 12) and Control (n = 6) groups. 25 mg/kg ATP was injected intraperitoneally (i.p.) to the ATP + bevacizumab (ABZ; n = 12) group. One hour after ATP and solvent administration, 10 mg/kg bevacizumab was i.p. injected to the ABZ and BVZ groups. Bevacizumab was administered once a day every two weeks; ATP was administered one a day for 30 days. At the end of this period, six rats from each group were sacrificed with high dose of anesthesia (thiopental sodium 50 mg/kg) and biochemical and histopathological examinations were performed in ovarian tissues. Mature male rats were kept in the laboratory for two months to breed the remaining female animals. The values showed that the oxidant parameters increased in the ovarian tissue of the BVZ group compared to the healthy controls and the ABZ group, while antioxidant parameters decreased. The number of breeding animals was significantly decreased in the BVZ group compared to the Control and the ABZ groups. This result suggests that ATP may be effective in preventing oxidative damage to the ovaries and infertility induced by bevacizumab.

The effect of taxifolin on oxidative ovarian damage and reproductive dysfunctions induced by antipsychotic drugs in female rats

Ince, Sefa; Ozer, Mehmet; Kadioglu, Berrin Goktug; Kuzucu, Mehmet; Ozkaraca, Mustafa; Gezer, Arzu; Suleyman, Halis; Cetin, Nihal

Aim: Typical antipsychotics (TAPs) are commonly used to treat schizophrenia and bipolar disorder. However, extrapyramidal disorders, hyperprolactinemia, and reproductive dysfunctions have been observed in women during the use of TAPs. For this reason, less toxic and prolactin-sparing atypical antipsychotic (AAP) drugs such as clozapine (CLN) have been developed. The aim of this study is to investigate the effect of taxifolin on possible ovarian and reproductive toxicity associated with CLN and haloperidol (HPL) in female Wistar albino rats. Methods: The rats were grouped as healthy control group (HCG), CLN, HPL, taxifolin + clozapine (TCL), and taxifolin + haloperidol (THL). Drugs were administered to the groups for 28 days. At the end of that time, ovarian tissues of six rats from each group were taken for histopathological and biochemical analyses. Remaining six rats in groups were examined for evaluation of reproductive dysfunctions. Results: Severe degeneration and vacuolization were observed in the primary, secondary, and primordial follicles of the ovarian tissues of CLN- and HPL-treated groups, of which malondialdehyde (MDA) level was high and total glutathione (tGSH) level was low. In the taxifolin-treated groups, taxifolin significantly prevented the increase of MDA level and decrease of tGSH level, and the severity of histopathological damage was found to be lower. In addition, it was found that taxifolin significantly prevented infertility and delay in pregnancy associated with CLN and HPL. Conclusions: The results of this experiment suggest that taxifolin can be beneficial in treating oxidative ovarian damage, infertility, and reproductive dysfunctions induced by CLN and HPL.

Effect of benidipine on experimental gastric ulcers in rats

Kuzucu, Mehmet; Suleyman, Bahadir; Mammadov, Renad; Ozcicek, Adalet; Ozcicek, Fatih; Altuner, Durdu; Süleyman, Zeynep

Peptic ulcer has been reported to increase acid secretion and lead to oxidative stress. Benidipine is an antihypertensive drug with antioxidant properties. This study aims to investigate the effects of benidipine on indomethacin-induced gastric ulcers in rats. Four groups were designed in this study. Gastric ulcer was occurred with indomethacin in the first three groups. These groups were treated with famotidine, benidipine and distilled water respectively 5 minutes before indomethacin administration. The fourth group rats didn’t have gastric ulcer and they received only distilled water. Six hours after drug administration gastric tissue was extracted and macroscopic and biochemical examinations were performed. Damaged areas in the stomach of benidipine and famotidine receiving animal groups were smaller than the indomethacin received animals. Indomethacin elevated the levels of malondialdehyde and myeloperoxidase in the stomach tissue (p<0.0001), and also decreased glutathione, glutathione related enzymes and superoxide dismutase (p<0.0001). Benidipine may be useful in preventing the toxic effect of indomethacin on the stomach.

The Effect of Taxifolin on Cisplatin-Induced Pulmonary Damage in Rats: A Biochemical and Histopathological Evaluation

Unver, Edhem; Tosun, Mustafa; Olmez, Hasan; Kuzucu, Mehmet; Cimen, Ferda Keskin; Suleyman, Zeynep; Xiao, Cheng

The effect of taxifolin on cisplatin-induced oxidative pulmonary damage was investigated biochemically and histopathologically in male albino Wistar rats. There were four groups, with six animals in each group: 50 mg/kg of taxifolin plus 2.5 mg/kg of cisplatin (TC) group, 2.5 mg/kg of cisplatin only (CIS) group, 50 mg/kg of taxifolin only (TG) group, and a healthy control group (HG). In terms of the experimental procedure, the animals in the TC and TG groups were first treated via oral gavage. The CIS and HG groups received distilled water as solvent, respectively. One hour later, the TC and CIS groups received cisplatin at a dose of 2.5 mg/kg (injected intraperitoneally). Taxifolin, cisplatin, and the distilled water were administered at the indicated dose and volume, using the same method daily for 14 d. At the end of this period, the animals were killed with a high dosage of thiopental anaesthesia (50 mg/kg). Blood and lung tissue samples were taken for biochemical (malondialdehyde (MDA), myeloperoxidase (MPO), total glutathione (tGSH), and 8-hydroxy-2 deoxyguanosine (8-OHdG)) analyses and histopathological examinations. The biochemical and histopathological results in the TC and HG groups were then compared with those in the CIS group. Cisplatin increased the levels of MDA, myeloperoxidase, and 8-OHdG, a marker of oxidative DNA damage, and reduced the amount of tGSH in the lung tissue. Moreover, severe alveolar damage, including oedema and extensive alveolar septal fibrosis, in addition to infiltration of polymorphic nuclear leucocytes and haemorrhagic foci, was observed in the CIS group. These histopathological findings demonstrate that taxifolin provides protection against pulmonary oxidative stress by preventing increases in oxidant parameters and decreases in antioxidants.

Objectives: We aimed to assess the effect of Hippophae rhamnoides extract (HR) on oxidative stress induced by cisplatin (Cis) in rat small intestine tissue by evaluating the biochemical and gene expression levels and histopathological changes. Materials and methods: The control group and Cis group received distilled water, while the HR25+cisplatin (HR25+Cis) group and the HR50+cisplatin (HR50+Cis) group were given HR 25 and 50 mg/kg, respectively, orally for seven days. HR25+Cis, HR50+Cis, Cis groups were injected with a single dose of intraperitoneal cisplatin on the first day. After sacrifice, the jejunum of each rat was removed for the assessment of oxidants and antioxidants. Analyses of the gene expression (for IL-1ß and TNF-α) and histopathological changes evaluated. Results: HR significantly inhibited the increase of oxidants and the decrease of antioxidants caused by cisplatin in the jejunal tissue. IL-1β and TNF-α gene expression levels were almost the same in both the HR50+Cis and the control groups. HR better prevented increase of the serum levels of IL-1β and TNF-α in animals at 50 mg/kg dose compared to 25 mg/kg dose. We confirmed that HR prevented the histopathological changes caused by cisplatin. Conclusions: It was concluded that oxidative stress caused by cisplatin may be preventable by co-administered HR.

Can endocan be a new biomarker in ventilator-associated pneumonia?

Kupeli, Ilke; Salcan, Sara; Kuzucu, Mehmet; Kuyrukluyıldız, Ufuk

Ventilator-Associated Pneumonia (VAP) is a hospital-acquired bacterial infection with high incidence and mortality rate. The aim of this study is to investigate the correlation between the Endocan level and development of VAP and whether or not this correlation was correlated with the clinical findings. Demographic data, white blood cell (WBC) count, procalcitonin (PCT), c-reactive protein (CRP), and fever levels of 60 patients were recorded in serial measurements for 5 days. When there was the presence of fever or elevated Endocan, alveolar lavage culture was taken and chest radiographies were taken. Correlations of the Endocan levels with the culture results and laboratory values were examined. The rate of VAP was found as 10.4/1000 mechanical ventilator days. Endocan levels were significantly higher in patients with VAP (p < 0.05). However, there was no significant difference among PCT, WBC, CRP measurements (p > 0.05). No correlation was found between Endocan levels and PCT, WBC and CRP levels in those with VAP (p > 0.05). A significant correlation was found between the Endocan level and the elevated fever 24 h later (p:0.001). The serum Endocan level on the day 3 had a specificity of 73.3%, a sensitivity of 68.9%, positive predictive value of 44%, and negative predictive value of 88.5% at the cut off level of 9.17 ng/mL. In this study, it was determined that high Endocan levels were associated with the development of VAP. The present study suggested that Endocan can be used as a screening tool for the development of VAP. Clinical Trials.gov ID: NCT02916277.

Purpose: To compare the preventive effects of N-acetyl cysteine (NAC), ozone preconditioning and ozone treatment against contrast-induced nephropathy (CIN) in an experimental rat model. Methods: Thirty adult male Wistar rats were randomly distributed into five groups (n=6 for each group). Group I served as control and Group II had only contrast agent, while Group III received NAC and Group IV received intraperitoneal ozone 6 hours before and 6 hours after introduction of contrast agent. Ozone treatment was applied for 5 days after the contrast agent was introduced in Group V. After induction of CIN, groups were compared in terms of serum levels of urea, creatinine, neutrophil gelatinase associated lipocalin, protein carbonyl, total antioxidant capacity (TAC) as well as degree of renal injury at histopathologic level. Results: Groups II-V displayed more obvious histopathological alterations such as hemorrhage and renal tubular injury compared with Group I. TAC (p=0.043) and creatinine (p=0.046) levels increased significantly in Group II after the intervention. In Group III, protein carbonyl level diminished remarkably (p=0.046), while creatinine level was increased (p=0.046) following the intervention. TAC level was higher in Group IV (p=0.028) and Group V (p=0.026) following the procedure. Conclusion: The N-acetyl cysteine and ozone treatment may alleviate the biochemical and histopathological deleterious effects of contrast-induced nephropathy via enhancement of total antioxidant capacity and decreasing oxidative stress.

In the treatment of heart disorders, metoprolol tartrates and digoxin drugs which act by way of blood are used widely. Being vital of using these drug substances, investigation of inhibitory effect on the activities of enzymes in the blood reveals the importance of this work. Aim of this study to investigate effects of metoprolol tartrate and digoxin on human erythrocyte carbonic anhydrase I isoenzyme in vitro conditions. CA-I isoenzyme from human blood has been purified using Sepharose-4B-l-tyrosine-sulfanilamide affinity chromatography method. The enzyme was purified ~116-fold with a yield of 62%. At the end of inhibition studies two substances were showed uncompetitive inhibition. Ki values were determined metoprolol tartrate 5.49±1.86 mM and digoxin 0.0914±0.0062 mM.
