Sunar, Mukadder; Yazici, Gulce Naz; Mammadov, Renad; Kurt, Nezahat; Arslan, Yusuf Kemal; Süleyman, Halis

Aim: In this study, it was aimed to investigate the effect of coenzyme Q10 (CoQ10) on cisplatin-induced oxidative retinal damage in rats biochemically and histopathologically. Materials and methods: Thirty male Wistar albino rats were divided into 3 groups randomly: untreated control (C group), only 2.5 mg/kg cisplatin daily administrated group for 2 weeks (CP group), 2.5 mg/kg cisplatin + 20 mg/kg orally CoQ10 daily administrated group for 2 weeks (CoQC group). At the end of experimental period, blood samples obtained before sacrification for the biochemical examination of serum malondialdehyde (MDA), total glutathione (tGSH), total oxidant system (TOS), total antioxidant systemic (TAS) levels and after eyes were removed for examined histopathology. Results: As a result of our study, severe histopathological damage was detected in the retinal tissue of the cisplatin group with serum malondialdehyde (MDA) and total oxidant system (TOS) levels were high and total glutathione (tGSH) and total antioxidant systemic (TAS) levels were low. However, it was observed that the histopathological damage associated with cisplatin was decreased in the retinal tissue of the CoQ10 group, which inhibited the increase in blood serum MDA/TOS levels and decrease in tGSH/TAS levels. Conclusion: The biochemical and histopathological results of our study were compatible with each other, so we concluded that the damage to the rat retinal tissue caused by cisplatin may be reversible with coenzyme.

Yuceli, Sahin; Suleyman, Bahadir; Yazici, Gulce Naz; Mammadov, Renad; Cankaya, Murat; Kunak, Celaleddin Semih; Bulut, Seval; Suleyman, Halis; Altuner, Durdu

Background: Ischemia is a condition in which blood flow to tissues is decreased or entirely stopped for various reasons. The reperfusion process exacerbates damage caused by ischemia in the organs and tissues. Reactive oxygen species (ROS) are mainly responsible for ischemia-reperfusion (IR) damage. ROS increase results in lipid peroxidation (LPO) and oxidative stress. In the literature, taxifolin reportedly suppresses ROS production. This study aimed to determine the effect of taxifolin, which is a flavonoid, on IR injury of the sciatic nerve in rats. Methods: This study divided 30 albino Wistar rats into 3 groups: IR without medication (IR) group, taxifolin applied IR (TAX+IR) group, and only dissection made to the sciatic nerve sham group (SHAM). Sciatic nerve injury was induced by applying 2 hours of ischemia and 3 hours of reperfusion to the abdominal aorta and iliolumbar arteries. Biochemical and histopathologic investigations then were performed on sciatic nerve tissues. Malondialdehyde, total glutathione, glutathione reductase, and glutathione peroxidase were analyzed as oxidative stress markers, and tumor necrosis factor–α and interleukin-1β levels were evaluated as inflammatory stress markers in biochemical tests. Results: The IR group has statistically significantly high oxidant and cytokine levels and low antioxidant levels compared with the TAX+IR group. Taxifolin treatment was also shown to cause significant histopathologic improvement. Conclusions: We suggest that taxifolin may be effective in preventing IR injury of the sciatic nerve.

Pazopanib is a tyrosine kinase inhibitor that is generally used for the treatment of metastatic renal cell cancer and advanced soft tissue sarcoma. It can cause various degrees of hepatotoxicity. Our study aimed to investigate the effect of taxifolin on pazopanib-induced liver toxicity. A total of 18 rats were divided into three groups: the pazopanib (PP), pazopanib plus taxifolin (TPP), and control (C) group. Taxifolin was administered to the TPP (n=6) group with a dose of 50 mg/kg. Distilled water was orally admnistered to the C (n=6) and PP (n=6) groups as a solvent. Subsequently, pazopanib 200 mg/kg was administered to the TPP and PP groups via the stomach. This procedure was repeated once a day for four weeks. Then, all rats were sacrificed, and their livers were removed. Malondialdehyde (MDA), total glutathione (tGSH), total oxidant status (TOS), and total antioxidant status (TAS) levels were evaluated. MDA and TOS levels were higher in the PP group compared with the levels of the other parameters (P<0.001). tGSH and TAS levels were lower in the PP group than in the TPP and C groups (P<0.001), and the aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) levels were higher. Furthermore, liver tissue damage, including hemorrhage, hydropic degeneration, and necrosis was observed in the PP group. Administration of taxifolin before pazopanib significantly improved degenerative changes. Our study demonstrated that the administration of taxifolin is significantly effective in preventing pazopanib-induced hepatotoxicity in rats.

The current study investigates the biochemical and histopathological effects of taxifolin on acrylamide-induced kidney damage. A 50 mg/kg dose of taxifolin was administered via oral gavage to the taxifolin + acrylamide (TACR) group (n-6) consisting of male albino Wistar rats. The same volume of distilled water used as solvent was orally administered to the acrylamide (ACR) (n-6) and healthy (HG) (n-6) groups. One hour after the administration of taxifolin and distilled water, a 20 mg/kg dose of acrylamide was orally administered to the TACR and ACR groups. This procedure was repeated once a day for 30 days. In the acrylamide group, malondialdehyde (MDA), tumour necrosis factor-alpha (TNF-α), and interleukin-1 beta (IL-1β) levels were found to be high, total glutathione (tGSH) levels were found to be low, and there was severe interstitial haemorrhage; additionally, tubular necrosis, tubular atrophy, leucocyte infiltration, and glomerular structures with expanded Bowman's space were observed. In the taxifolin group, where the increase of MDA, IL-1β, and TNF-α and the decrease of tGSH associated with acrylamide have been prevented, any histopathological finding other than mild necrosis and atrophic tubules was not found. This suggests that Taxifolin would prevent kidney tissue from acrylamide-induced damage would be effective in treating acrylamide-induced nephrotoxicity, inhibiting the increase of MDA, IL-1β and TNF-α, and decreasing tGSH associated with acrylamide.

Objective: To investigate preventive effects of taxifolin on ischemia-reperfusion induced oxidative ovarian damage in rats. Methods: A total of 18 female Wistar albino rats were randomly and equally divided into three groups: the sham group, the ovarian ischemia reperfusion group, and the 50 mg/kg taxifolin+ovarian ischemia reperfusion group. The ovarian ischemia reperfusion and taxifolin+ovarian ischemia reperfusion groups were exposed to ischemia for 2 h and then followed by two-hour reperfusion protocol. Biochemical and histopathologic examinations were performed on the extracted ovaries. Results: Levels of malondialdehyde and cyclooxygenase-2 were increased, while reduced-glutathione and cyclooxygenase-1 were decreased in the ovarian ischemia reperfusion group. However, these values were reversed in the taxifolin+ovarian ischemia reperfusion group. Similarly, the number of primordial and developing follicules decreased in the ovarian ischemia reperfusion group, while they were within normal range in the taxifolin+ovarian ischemia reperfusion group. Conclusions: Ischemia followed by reperfusion leads to oxidative stress-related ovarian injury, and taxifolin may be useful for protecting ovarian tissue from such injury.

Ozcicek, Fatih; Kara, Ali Veysel; Akbas, Emin Murat; Kurt, Nezahat; Yazici, Gulce Naz; Cankaya, Murat; Mammadov, Renad; Ozcicek, Adalet; Suleyman, Halis

Intestinal mucositis is an important problem in the patients receiving cancer treatment. We aimed to investigate the effect of anakinra, which is a well known anti-oxidant and anti-inflammatory agent, on methotrexate-induced small intestine mucositis in rats. Forty rats were divided into 4 groups with 10 in each group. The healthy group (HG) and the methotrexate group (MTXG) were given distilled water, while the methotrexate + anakinra 50 (MTX+ANA50) and the methotrexate + anakinra 100 (MTX+ANA100) groups were intraperitoneally administered 50 and 100 mg/kg of anakinra. After one hour, the MTXG, MTX+ANA50 and MTX+ANA100 groups were given oral methotrexate at a dose of 5 mg/kg. This procedure was repeated once a day for 7 days. After the rats had been sacrificed, the small intestine tissue of rats were removed for the assesment of biochemical markers, histopathological evaluation and gene expression analyze. Statistical analyses of the data were performed using one-way ANOVA. Malondialdehyde (MDA), myeloperoxidase (MPO) and interleukin-6 (IL-6) levels were significantly higher, whereas total glutathione (tGSH) levels were significantly lower in MTXG (P<0.001) compared to other groups. MTX also increased IL-1β and TNF-α gene expression levels in MTXG (P<0.001). Inflammatory cell infiltration and damage to the villus were observed histopathologically in the MTXG group, whereas only mild inflammation was seen in the MTX+ANA100 group. A dose of 100 mg/kg of anakinra prevented the increase of the biochemical markers and gene expression levels better than a dose of 50 mg/kg. Intestinal mucositis caused by MTX may be preventible by co-administered anakinra.

Purpose: Vandetanib is a wide spectrum tyrosine kinase inhibitor used for the treatment of metastatic medullary thyroid cancer (MTC) and various other cancer types. Although it is usually well-tolerated ıt has been linked to a variety of severe dermatologic reactions. Our study aimed was to investigate adenosine 5′-triphosphate (ATP) on vandetanib-induced skin damage. Materials and methods: A total number of 18 rats were divided into three equal groups as vandetanib group (VDB), vandetanib plus ATP group (VAT), and healthy group (HG); 25 mg/kg ATP was injected intraperitoneally (ip) to the VAT group. Normal saline was given to the HG and VDB groups as solvent via intraperitoneally. One hour later, 25 mg/kg vandetanib was applied orally via an orogastric catheter in the VAT and VDB groups. This procedure was repeated once daily for 4 weeks. After that period, all animals were sacrificed and their skin tissues removed. Malondialdehyde (MDA), total glutathione (tGSH), total oxidant status (TOS), total antioxidant status (TAS) levels in rats’ skin tissues were evaluated with histopathological analyses. Results: MDA and TOS levels measured higher in the VDB group compared to the VAT and HG groups (p < 0.001). tGSH and TAS levels of the VDB group measured lower than the VAT and HG groups (p < 0.001). The structure and morphology of skin tissue were normal in the control group. In the VDB group, skin tissue damage with thinner epitelium, ruptured and degenerated hair follicles, abnormal accumulation of abnormal keratin on the epithelium and oedematous areas in the dermis was observed. In the VAT group, these findings were significantly improved. Conclusion: We demonstrated that adenosine triphosphate can prevent vandetanib-induced skin toxicity in rats for the first time. The promising results denote that further studies testing this agent in other animal models and in humans are warranted.

Effect of rutin on cytarabine-associated pulmonary oedema and oxidative stress in rats

Bilgin, Asli O.; Mammadov, Renad; Suleyman, Bahadir; Unver, Edhem; Ozcicek, Fatih; Soyturk, Mehmet; Cimen, Ferda K.; Kurt, Nezahat; Suleyman, Halis

Cytarabine is effectively used in the treatment of adult acute leukemia, but it has a dose-limiting side effect of fatal pulmonary oedema because it increases the vascular permeability of the alveolar capillaries. The aim of the present study was to conduct a radiological, biochemical and histopathological investigation of the effect of rutin on cytarabine-associated pulmonary oedema in rats. Rats were treated with a combination of rutin+cytarabine by administering oral rutin at a dose of 50 mg/kg; other rat groups were orally administered the same volume of physiological saline. One hour after administration of rutin or saline, the rutin+cytarabine and cytarabine groups received an intraperitoneal injection of cytarabine (200 mg/kg). This administration procedure was repeated once a day for 14 days. Radiologically, 50% of the animals given cytarabine alone showed lung oedema, but the rutin+cytarabine group showed no oedema. The inclusion of rutin decreased the amounts of cytarabine-associated malondialdehyde, tumour necrosis factor-α, and nuclear factor-κB in the lung tissue. Rutin also inhibited the reduction of total glutathione by nitric oxide. These findings suggest that rutin may be a beneficial adjunct that can minimise the development of cytarabine-associated pulmonary oedema.

An experimental study on the use of lycopene to prevent infertility due to acute oxidative ovarian damage caused by a single high dose of methotrexate

Turkler, Can; Onat, Taylan; Yildirim, Engin; Kaplan, Selcuk; Yazici, Gulce Naz; Mammadov, Renad; Sunar, Mukadder

Background. Methotrexate (MTX) is an antineoplastic agent, which increases the level of reactive oxygen species (ROS) and decreases the level of antioxidants. Lycopene, is a potent antioxidant, which is used because of its protective effect against tissue damage. Objectives. The aim of this study was to determine the effect of lycopene on ovarian MTX-induced injury in rats. Material and methods. Rats (n = 36) were randomly divided into 3 equal groups: a group with MTX only (MG, n = 12), a group with lycopene and MTX (LMG, n = 12), and a healthy control group (HCG, n = 12). Then, malondialdehyde (MDA), myeloperoxidase (MPO) and total glutathione (tGSH) levels and histopathological findings were examined in the ovaries of rats. Apart from the histopathological and biochemical evaluation, the reproductive performance of the experimental groups was also examined. Results. Our study demonstrated that, in ovarian tissues of rats administered MTX, there was a decrease in the levels of tGSH, while MDA and MPO were increased, but it is observed that these ratios are reversed in the LMG (p < 0.05). It also has been proven that a single, high-dose use of MTX causes infertility in female rats, prolongs the gestation period and reduces the number of offspring. Conclusions. Lycopene pretreatment ameliorates the MTX induced ovarian injury and infertility in rats through its antioxidative activities.

Kuzucu, Mehmet; Suleyman, Bahadir; Mammadov, Renad; Ozcicek, Adalet; Ozcicek, Fatih; Altuner, Durdu; Süleyman, Zeynep

Peptic ulcer has been reported to increase acid secretion and lead to oxidative stress. Benidipine is an antihypertensive drug with antioxidant properties. This study aims to investigate the effects of benidipine on indomethacin-induced gastric ulcers in rats. Four groups were designed in this study. Gastric ulcer was occurred with indomethacin in the first three groups. These groups were treated with famotidine, benidipine and distilled water respectively 5 minutes before indomethacin administration. The fourth group rats didn’t have gastric ulcer and they received only distilled water. Six hours after drug administration gastric tissue was extracted and macroscopic and biochemical examinations were performed. Damaged areas in the stomach of benidipine and famotidine receiving animal groups were smaller than the indomethacin received animals. Indomethacin elevated the levels of malondialdehyde and myeloperoxidase in the stomach tissue (p<0.0001), and also decreased glutathione, glutathione related enzymes and superoxide dismutase (p<0.0001). Benidipine may be useful in preventing the toxic effect of indomethacin on the stomach.

No previous studies have examined the effect of Hippophae rhamnoides fruit extract (HRE), which contains both water and fat soluble vitamins, on cisplatin-induced ototoxicity. Therefore, the aim of the study was to investigate the effects of FIRE on cisplatin-induced ototoxicity in Guinea pigs and to evaluate the biochemical, gene expression and histopathological changes associated with cisplatin use. Experimental animals were divided into three groups: healthy (HG), HRE+cisplatin (HRC) and cisplatin control (CCG) groups. Biochemical, gene expression and histopathological examinations were carried on the removed inner ear tissues. In the HRC group, the oxidant parameter was lower and the anti-oxidant parameter was higher than in the CCG. These results are supported by gene expression levels and histopathological results. The use of FIRE against cisplatin-induced oxidative ototoxicity may be easier, more cost effective and more beneficial than the use of vitamins alone or in combination with each other.

Erhan, Ertugrul; Mammadov, Renad; Bilgin, Asli O.; Kunak, Celalettin S.; Cankaya, Murat; Cimen, Ferda K.; Altuner, Durdu

Oxidative stress is considered responsible for cisplatin autotoxicity. The aim of this study was to examine the protective effect of rutin on cisplatin-induced oxidative vestibular nerve damage in rats. Animals were divided into four groups (n = 6): healthy group (HG), 50 mg/kg rutin + cisplatin group (RG-50), 100 mg/kg rutin + cisplatin group (RG-100), and cisplatin group (CG). Rutin was administered orally for eight days, and 5 mg/kg cisplatin was injected intraperitoneally once every two days. Malondialdehyde, myeloperoxidase, and total glutathione levels were measured in serum and vestibular nerve tissue and examined histopathologically. Malondialdehyde and myeloperoxidase levels were lower in the serum and vestibular nerve tissue of RG-50 and RG-100 groups compared to the CG group; total glutathione was higher. Also, several histopathological changes were seen in the vestibular nerve tissue of the CG group. Rutin inhibited nerve tissue damage and may protect vestibular nerve tissue from cisplatin-induced oxidative damage.

Kara, Ali Veysel; Aldemir, Mehmet Naci; Ozcicek, Fatih; Mammadov, Renad; Yazıcı, Gulce Naz; Sunar, Mukadder; Gulaboglu, Mine

OBJECTIVE: To show the protective effects of taxifolin on cisplatin-induced oxidative renal injury. STUDY DESIGN: Rats were divided into 3 groups: the healthy group, the cisplatin group (2.5 mg/kg cisplatin only), and the taxifolin-cisplatin group (50 mg/kg taxifolin+2.5 mg/kg cisplatin). The rats were sacrificed after 14 days, and the kidneys were removed for examination of malondialdehyde (MDA), total glutathione (tGSH), and super oxide dismutase (SOD) levels. Blood urea nitrogen (BUN) and creatinine levels were measured in the blood samples taken before the animals were sacrificed. Kidney tissues were examined histopathologically. Finally, biochemical and histopathological results of the groups were compared. RESULTS: Cisplatin significantly increased MDA and decreased tGSH and SOD in the rat kidney tissue (p<0.001). Additionally, cisplatin caused an increase in serum creatinine and BUN levels (p<0.001). Taxifolin prevented the cisplatin-induced increase in MDA and decrease in tGSH and SOD (p<0.001). It also decreased serum creatinine and BUN levels significantly as compared with the cisplatin group (p<0.001). In the cisplatin group microscopic examinations showed obvious kidney damage including dilation, severe necrosis, and degenerative changes. Taxifolin prior to cisplatin markedly ameliorated these changes. CONCLUSION: We have demonstrated for the first time that taxifolin can prevent cisplatin-induced nephrotoxicity.

Yaşar, Hasan; Ersoy, Alevtina; Cimen, Ferda Keskin; Mammadov, Renad; Kurt, Nezahat; Arslan, Yusuf Kemal

Background. Cisplatin, used in cancer treatment, has toxic and apoptotic effects on the peripheral nervous system. Rutin, also known as vitamin P, has antioxidant and antiapoptotic activity. Objectives. The purpose of this study was to investigate the biochemical and histopathologic efficacy of rutin on neurotoxic and apoptotic effects caused by cisplatin in the peripheral nervous system. Material and methods. Twenty-four albino Wistar male rats were divided into the following 4 groups: control group (CG), only cisplatin-injected group (CIS), cisplatin and rutin 50 mg/kg (RG-50)-injected group, and cisplatin and rutin 100 mg/kg (RG-100)-injected group. Analyses were performed on sciatic nerve tissue of experimental animals. Analyses of malondialdehyde (MDA), total glutathione (tGSH), glutathione reductase (GSHRd), glutathione-s-transferase (GST), and superoxide dismutase (SOD) were performed. Caspase-3 expression in nerve tissue was also investigated. The analyzed groups were compared with CG. Results. Biochemical investigation shows that there is a statistically significant difference between CG and only CIS and RG-50. Control group and RG-100 were found to be similar. Cisplatin-induced changes were observed in histopathological analysis of the nerve tissue. The RG-100 and CG were found to be similar. The caspase-3 expression in the neural tissue was compared between groups. Control group and CIS were found to be different. Control group and RG-100 were found to be similar. Conclusions. Antioxidant and antiapoptotic effectiveness of rutin was detected against the toxic effects caused by cisplatin in the peripheral nerve tissue.

Purpose: To investigate the effects of the EtOAc extract of U. longissima which is uninvestigated previously on esophagogastric cancer induced in rats with N-methyl-N-nitro-N-nitrosoguanidin (MNNG). Methods: The anticancer activity of EtOAc extract of U. longissima was examined in the esophagogastric adenocarcinoma models induced in rats with MNNG. EtOAc extract of U. longissima, 50 and 100 mg/kg oral doses were administered once daily for six months. MNNG induced differentiated and undifferentiated type adenocarcinomas in the esophageal and gastric tissues of rats. Results: EtOAc extract of U. longissima obtained from U. longissima prevented gastric and esophageal cancerogenesis induced in rats with MNNG. EtOAc extract of U. longissima did not have a lethal effect at doses of 500, 1000 and 2000 mg/kg. The prominent anticarcinogenic activity of EtOAc extract of U. longissima 50 and 100 mg/kg suggests that it is not toxic and it is selective to the cancer tissue. Conclusion: This information may shed light on clinical implementation of EtOAc extract of U. longissima in future.

Can the negative effects of ketamine abuse on female genital organbe prevented by nimesulide? An experimental study

Turkler, Can; Onat, Taylan; Yildirim, Engin; Kaplan, Selcuk; Yazici, Gulce N.; Mammadov, Renad; Sunar, Mukadder

The objective of this study is to investigate the effects of nimesulide on ketamine-induced ovarian and uterine toxicity by biochemical and histopathological examinations. Ketamine is an anesthetic agent whose use leads to overproduction of catecholamines. Nimesulide is a cyclooxy-genase-2 inhibitor, which has also been reported to exert a significant antioxidant effect. Wistar albino female rats were randomly divided into three groups as follows: ketamine group (60 mg/ kg), ketamine (60 mg/kg)+nimesulide (50 mg/kg) group, and a healthy control group. Then, the biochemical levels and histopathological findings in the ovaries and uteri of the rats were examined for malondialdehyde, myeloperoxidase, total glutathione and superoxide dismutase. The study demonstrated that, in the uterine and ovarian tissues of rats that have been administered ketamine, there was a decrease in the levels of total glutathione and superoxide dismutase, while malondialdehyde and myeloperoxidase was increased: however it was observed that these ratios were reversed in the ketamine+nimesulide group. It was also proved that the negative effects of ketamine can be corrected with nimesulide when the myometrial and endometrial thicknesses are compared. Antioxidants such as nimesulide may protect against the damage caused by ketamine to the genital organs in young women.
