Ozbek Bilgin, Asli; Altuner, Durdu; Cankaya, Murat; Cakir, Tayfun; Mammadov, Renad; Bilgin, Yasin; Süleyman, Halis

The aim of this study was to examine the effects of Hippophae rhamnoides extract (HRE), known to have antioxidant properties, on ischemia reperfusion-induced brain injury in rats. Eighteen albino Wistar male rats were divided into three equal groups: 15 min of brain ischemia-3 h of reperfusion (BIR) group, 50 mg/kg HRE + brain ischemia-reperfusion (HRE + BIR) group, and control group for sham operation (SHAM). Malondialdehyde (MDA) level, myeloperoxidase (MPO) activity and 8-OHdG levels significantly increased, whereas total glutathione (tGSH) level, glutathione reductase (GSHRd), glutathione s-transferase (GST) activities decreased in the brain tissues of BIR group compared to SHAM group. HR application reversed these changes and brought the levels closer to the level of SHAM group. This study has revealed that HR inhibits IR-induced oxidative damage and inflammation in rat’s brain biochemically and it is effective in maintaining antioxidant activity and protecting against DNA damage in the tissue.

Preventive effect of nimesulide on ketamine induced cardiotoxicity and the useful interaction of nimesulide with ketamine

Mammadov, Renad; Akpinar, Erol; Coskun, Resit; Suleyman, Halis; Gul, Mehmet A.; Kuyrukluyildiz, Ufuk

The increased production of endogenous adrenaline (ADR) with ketamine is responsible for ketamine cardiotoxicity and the short duration of anaesthesia. The aim of this study was to investigate the effect of nimesulide on ketamine cardiotoxicity, anaesthesia duration and analgesic activity. Oxidant and anti-oxidant parameters were studied in the heart tissues, while ADR, creatine kinase-MB (CK-MB) and troponin-I (TP I) levels were measured in the blood. Malondialdehide (MDA), 8-hydroxyguanine (8-OH/Gua), CK-MB, TP I, and ADR levels in the heart tissue of the ketamine group were increased and the total glutathione (tGSH) level was decreased as compared to the healthy and nimesulide groups. However, it was found that the same parameters in the ketamine-administered adrenalectomised animals were almost identical to those of the healthy animals. Nimesulide may be useful in the reduction of ketamine cardiotoxicity, providing potent analgesia and extending the duration of anaesthesia.

Ahiskali, Ibrahim; Pinar, Can Lokman; Kiki, Murat; Mammadov, Renad; Ozbek Bilgin, Asli; Hacimuftuoglu, Ahmet; Cankaya, Murat; Keskin Cimen, Ferda; Altuner, Durdu

Purpose: Diabetic retinopathy (DR) is one of the leading causes of blindness. In DR patients, antioxidant defence is disrupted, and production of reactive oxygen species and pro-inflammatory cytokines such as interleukin 1β (IL-1β) and tumour necrosis factor alpha (TNF-α) increases. Taxifolin has been reported to suppress reactive oxygen species, IL-1β and TNF-α production. The aim of this study is to biochemically and histopathologically examine the protective effect of taxifolin against DR damage induced by alloxan. Materials and methods: Alloxan received rats with a blood glucose level of ≥250 mg/dL were divided into taxifolin-treated (TAX) (n = 6), diabetic control (DC) (n = 6) groups. There were rats received only saline in non-diabetic control (NC) group (n = 6). Taxifolin (50 mg/kg) was orally administered to the TAX group rats. DC and NC rats received the same volume of saline as a solvent. This procedure was repeated once a day for 3 months. At the end of this period, animals were killed by high dose thiopental sodium anaesthesia. Histopathological examinations were then performed on excised rat eyes. Malondialdehyde (MDA), total glutathione (tGSH), IL-1β and TNF-α levels were measured in obtained blood samples. Results: MDA, IL-1β and TNF-α levels were significantly increased in blood samples of DC group rats with hyperglycemia induced by alloxan compared with NC group (p < 0.0001), and decreased in the TAX group compared with the DC group (p < 0.0001). The levels of tGSH were significantly decreased in blood samples of DC group rats compared with NC group (p < 0.0001), and increased in the TAX group compared with the DC group (p < 0.0001). Histopathologically, retinal ganglion cells of the TAX group had a slightly dilated and congested blood vessel, and severe damage was inflicted to the retinal ganglion cell layer of the DC group. Conclusions: Experimental results suggest that taxifolin may be beneficial in the treatment of DR.

Mammadov, Renad; Suleyman, Bahadir; Akturan, Selcuk; Cimen, Ferda Keskin; Kurt, Nezahat; Suleyman, Zeynep; Malkoc, İsmail

Background/Aims: This study aimed to investigate the effect of lutein on methotrexate (MTX)-induced pulmonary toxicity in rats biochemically and histopathologically. Methods: The rats in the MTX + lutein (MTXL, n = 6) group were given 1 mg/kg of lutein orally. A 0.9% NaCl solution was administered orally to the MTX (n = 6) group and the healthy group (HG, n = 6). One hour later, a single 20 mg/kg dose of MTX was injected intraperitoneally in the MTXL and MTX. Lutein or 0.9% NaCl solution was administered once a day for 5 days. At the end of this period, malondialdehyde (MDA), myeloperoxidase (MPO), total glutathione (tGSH), interleukin 1 beta (IL-1β), and tumor necrosis factor alpha (TNF-α) were measured in the lung tissues from the animals euthanized with 50 mg/kg thiopental sodium anesthesia. Subsequently, histopathological examinations were performed. Results: The levels of MDA, MPO, IL-1β, and TNF-α in the lung tissue of the MTX were significantly higher than those of the MTXL and HG groups (p < 0.0001), and the amount of tGSH was lower. The histopathological findings in the MTX group, in which the oxidants and cytokines were higher, were more severe. Conclusions: Lutein prevented the MTX-induced oxidative lung damage biochemically and histopathologically. This result indicates that lutein may be useful in the treatment of MTX-induced lung damage.

Effect of Tianeptine on Oxidative Renal Damage Induced by Ischemia Reperfusion in Rats

Bilgin, Aslı Özbek; Süleyman, Bahadır; Mammadov, Renad

Renal ischemia-reperfusion (I/R) damage can develop following major surgical conditions such as shock, trauma, and renal transplantation and causes acute renal failure. Kidney I/R damage is known to be a pathological process that begins with the touch free of oxygen and continues with the production of free oxygen radicals. We will investigate the effect of renal I/R injury on tianeptine, an atypical antidepressant drug with antioxidant properties. There was no information on the effect of tianeptine on I/R-induced kidney damage in the literature. The aim of the present study was to investigate the effect of tianeptine on oxidative kidney damage induced by I/R in rats. Albino wistar male rats were divided into groups as renal I/R (RIR) group, 25 mg/kg of thianeptine+renal I/R (TIR-25) group and sham operation (SG) group. The I/R procedure (one hour ischemia, three hours reperfusion) was performed with 25 mg/kg intraperitoneal thiopental sodium anesthesia. Oxidative damage was assessed by malondialdehyde (MDA) and total glutathione (TGSH) measurements in kidney tissue. Biochemical test results showed that the I/R process increased the amount of MDA in the kidney tissue and decreased the tGSH. Tianeptine significantly prevented the increase of MDA and decrease of tGSH in kidney tissue after I/R. Our experimental results suggest that tianeptine may be useful in preventing I/Rinduced oxidative renal injury.

Effect of Taxifolin on Ethanol-Induced OxidativeBbrain Damage in Rats

Bilgin, Aslı Özbek; Süleyman, Bahadır; Mammadov, Renad

Studies have shown that alcohol exerts the most severe toxic effect on central nervous system by increasing free oxygen radical production and suppressing endogenous antioxidant production. We aim to biochemically investigate the effect of taxifolin on ethanol-induced oxidative brain damage in rats. ( Sakarya Med J, 2018, 8(3):638-643 ). Materials and Methods Experimental animals were divided into healthy (SG), ethanol-receiving control (EtOH) and taxifolin+ethanol receiving (TEtOH) groups. At the end of the administration of ethanol and taxifolin, the animals were sacrifi ced by high dose anesthesia and brain tissues were removed. Malondialdehyde (MDA), lipid peroxidation product, and total glutathione (tGSH), endogenous antioxidant, levels in brain tissues were measured. Results MDA levels were signifi cantly higher in the rats in the EtOH group than in those in the TEtOH group (p<0.001). In contrast, tGSH levels were signifi cantly lower in the rats in the EtOH group than in those in the TEtOH group (p<0.001). Conclusion Our results suggest that taxifolin protects brain tissue from ethanol-induced oxidative damage.

Onk, Didem; Coskun, Resit; Onk, Oruc A.; Cimen, Ferda K.; Kurt, Nezahat; Suleyman, Bahadir; Mammadov, Renad; Altuner, Durdu; Gul, Vahdet; Malkoc, Ismail

Postoperative pain begins with tissue incision, continues with the production of free oxygen radicals, and is intensified by the inflammatory response. We investigated the effect of esomeprazole on postoperative pain and lung and heart complications of laparotomy. Albino Wistar-type rats were grouped into the paw scalpel incision (PSI), applied control (SIC), PSI + 20 mg/kg esomeprazole (SIE), PSI + 20 mg/kg meperidine (SIM), and healthy group (HG). For the laparotomy test, the rats were grouped as follows: laparotomy applied control (LTC), laparotomy + 20 mg/kg esomeprazole (LTE), laparotomy + 20 mg/kg meperidine (LTM), and healthy group (HG). Meperidine suppressed the PSI-induced pain. Esomeprazole decreased the level of oxidant and proinflammatory cytokines in the serum and lung and heart tissues of the LTE group and in the paw tissues of the SIE group, but meperidine did not. There were no histopathological findings in the lung and heart tissues of the LTE group, but tissue damage was found in the LTC and LTM groups. Esomeprazole may help reduce major surgery complications.

Onk, Didem; Mammadov, Renad; Suleyman, Bahadir; Cimen, Ferda Keskin; Cankaya, Murat; Gul, Vahdet; Altuner, Durdu; Senol, Onur; Kadioglu, Yucel; Malkoc, Ismail

Thiamine pyrophosphate (TPP) is the active metabolite of thiamine. This study aimed to investigate the effects of thiamine and TPP on cisplatin-induced peripheral neuropathic pain (PNP). Male albino Wistar type Rattus norvegicus were divided into six groups (n=6) that received 2 mg/kg cisplatin (CIS), 25 mg/kg thiamine (TM), 2 mg/kg cisplatin+25 mg/kg thiamine (CTM), 25 mg/kg TPP (TPP), 2 mg/kg cisplatin+25 mg/kg TPP (CTPP), or distilled water (healthy group; HG) for 8 days intraperitoneally. Analgesic effect was measured with a Basile Algesimeter. IL-1β, malondialdehyde (MDA), total glutathione (tGSH), thiamine, and TPP were determined in blood samples. Histopathological examinations were performed on removed sciatic nerves. The percent analgesic effects of the CTM and CTPP groups were calculated to be 21.3% and 82.9%, respectively. Increased production of IL-1β and MDA by cisplatin was inhibited by TPP, while it was not inhibited by thiamine. Conversion of thiamine to TPP significantly decreased in the CIS group. Histopathological and biochemical investigations demonstrated that hyperalgesia and sciatic nerve damage developed in the CIS and CTM groups with low TPP levels. These results indicate that cisplatin inhibits the formation of TPP from thiamine, leading to severe PNP. This finding suggests that TPP may be more beneficial than thiamine for the treatment of cisplatin-induced PNP.

Suleyman, Bahadir; Bilgin, Asli O.; Mammadov, Renad; Cimen, Ferda K.; Gulaboglu, Mine

Amiodarone, an antiarrhythmic drug, is a benzofuran derivative. Oxidative stress plays a role in the mechanism of amiodarone-induced pulmonary toxicity. Taxifolin has antioxidant and anti-in-flammatory activities. The aim of the study was to investigate the effects of taxifolin on amiodarone-induced pulmonary toxicity in rats. In total 18 male albino Wistar rats were divided into three groups (n = 6 in each group): a taxifolin 50 mg/kg plus amiodarone 30 mg/kg (TAM) group, an amiodarone 30 mg/kg group (AMD) and a healthy group (HG). Malondialdehyde (MDA) levels, total glutathione (tGSH) levels, total oxidant status (TOS) and total antioxidant status (TAS) were measured in lung tissues, and histopathological examinations were performed. MDA and TOS levels in the AMD group were higher than those in the other two groups. In contrast, tGSH and TAS levels in the AMD group were lower than those in the TAM and HG groups. Alveolar destruction, marked oedema, alveolar septal fibrosis, leukocyte infiltration and haemorrhages were seen in the AMD group. Only slight oedema and congestion were observed in the TAM group. Taxifolin may be useful in preventing amiodarone-induced pulmonary toxicity.

Effect of anakinra, an interleukin one beta antagonist, on oxidative testicular damage induced in rats with ischemia reperfusion; [Efecto de la anakinra, que es un antagonista de la interleucina 1 beta en oxidativo daño testicular, inducida en ratas con isquemia reperfusión]

Hirik, Erkan; Suleyman, Bahadir; Mammadov, Renad; Yapanoglu, Turgut; Cimen, Ferda Keskin; Cetin, Nihal; Kurt, Nezahat

Background: It has been reported in the literature that proinflammatory interleukin-1 beta (IL-1β) is increased in cases of testicular ischemia reperfusion (I/R) damage. This information suggests that anakinra, an IL-1β antagonist, may be effective in testicular I/R damage. Objective: In our study, we investigated the effect of anakinra on testicular I/R damage induced in rats with torsion/detorsion. Methods: The 50 mg/kg anakinra + testicular torsion/detorsion (KTD-50) and 100 mg/kg anakinra + testicular torsion/detorsion (KTD-100) groups received an intraperitoneal (i.p.) injection of 50 mg/kg and 100 mg/kg of anakinra, respectively. In turn, the testicular torsion/detorsion (TTD) and sham operation (SOG) groups received a single dose of distilled water as a solvent 1 h before ketamine anaesthesia. After the testes of the TTD, KTD-50 and KTD-100 groups were subjected to torsion and detorsion for 4 h each, the rats were killed with a high-dose anaesthesia, and their testicles were removed and evaluated through biochemical, gene expression and histopathological examinations. The results were evaluated in comparison with those of the SOG group. Results: The levels of malondialdehyde (MDA), myeloperoxidase (MPO) and IL-1β showed significant increases in the TTD group, which underwent torsion/detorsion, compared to the KTD-50, KTD-100 and SOG groups. Conversely, the levels of glutathione (tGSH), glutathione peroxidase (GPO) and glutathione s-transferase (GST) were found to be significantly higher in the KTD-50, KTD-100 and SOG groups than in the TTD group. Conclusion: Anakinra at a 100 mg/kg dose histologically suppressed better oxidative stress and tunica albuginea, germ cell, seminiferous tubule and interstitial damage in the testicular tissue compared to a 50 mg/kg dose. Experimental results indicate that anakinra might be beneficial in the attenuation of testicular I/R damage.

The protective effect of thiamine pryophosphate against sugar-induced retinal neovascularisation in rats

Cinici, Emine; Mammadov, Renad; Findik, Huseyin; Suleyman, Bahadir; Cetin, Nihal; Calik, Ilknur; Balta, Hilal; Tas, Ismail Hakki; Sener, Ebru; Altuner, Durdu

The aim of this study was to investigate the effect of thiamine pyrophosphate (TPP), administered via sugar water, on retinal neovascularisation in rats. Animals were assigned to three groups, namely the TPP sugar-water group (TPSWG, n = 12), the control group (CG, n = 12) and the healthy group (HG, n = 12). The TPSWG was injected intraperitoneally with TPP once a day for 6 months. CG and HG rats were given distilled water in the same way. TPSWG and CG rats were left free to access an additional 0.292 mmol /ml of sugar water for 6 months. The fasting blood glucose (FBG) levels of the animals were measured monthly. After 6 months, biochemical, gene expression and histopathologic analyses were carried out in the retinal tissues removed from the animals after they were killed. The measured FBG levels were 6.96 ± 0.09 mmol/ml (p < 0.0001 vs. HG), 6.95 ± 0.06 mmol/ml (p < 0.0001 vs. HG) and 3.94 ± 0.10 mmol/ml in the CG, TPSWG and HG groups, respectively. The malondialdehyde (MDA) levels were found to be 2.82 ± 0.23 (p < 0.0001 vs. HG), 1.40 ± 0.32 (p < 0.0001 vs. HG) and 1.66 ± 0.17 in the CG, TPSWG and HG, respectively. Interleukin 1 beta (IL-1β) gene expression was increased (3.78 ± 0.29, p < 0.0001) and total glutathione (tGSH) was decreased (1.32 ± 0.25, p < 0.0001) in the retinal tissue of CG compared with TPSWG (1.92 ± 0.29 and 3.18 ± 0.46, respectively). Increased vascularisation and oedema were observed in the retinal tissue of CG, while the retinal tissues of TPSWG and HG rats had a normal histopathological appearance. A carbohydrate-rich diet may lead to pathological changes in the retina even in nondiabetics, but this may be overcome by TPP administration.

Ozcicek, Adalet; Ozcicek, Fatih; Cimen, Ferda Keskin; Mammadov, Renad; Cankaya, Murat; Ezmeci, Talat; Altuner, Durdu

Background. Oxidative stress and interleukin-1 beta (IL-1β) have been reported to play a role in the pathogenesis of nephrotoxicity induced by cisplatin. Objectives. The objective of this study was to investigate the effect of anakinra, which is an IL-1β receptor antagonist, on cisplatin-induced nephrotoxicity in rats, through biochemical, gene expression and histopathological analyses. Material and methods. The study was designed with 4 groups. For 1 week, the control group (C) and the cisplatin (Cis) group received distilled water, while the cisplatin + anakinra 50 (Cis + ANA50) group and the cisplatin + anakinra 100 (Cis + ANA100) group were intraperitoneally administered 50 mg/kg and 100 mg/kg of anakinra, respectively. The Cis, Cis + ANA50 and Cis + ANA100 groups were intraperitoneally injected with a 2.5 mg/kg dose of cisplatin for 7 days. After sacrifice, the kidney tissue of each rat was extracted for the assessment of the malondialdehyde (MDA) and total glutathione (tGSH) levels, and for gene expression analyses of IL-1β. The kidney tissues were histopathologically evaluated. Statistical analyses of the data were performed using one-way analysis of variance (ANOVA). Results. The administration of cisplatin (the Cis group) yielded a higher level of MDA (4.75 ±0.25 nmol/mL; p < 0.001) and lower levels of tGSH (1.80 ±0.35 mg/L; p < 0.001) compared to other groups. Cisplatin also increased IL-1β gene expression (6.33 ±0.27 gene expression levels; p < 0.001) compared to other groups. The impact of anakinra on the MDA and tGSH levels, and on IL-1β gene expression induced by cisplatin was observed as a reversal of these findings (p < 0.05). Anakinra better prevented an increase of the levels of MDA and IL-1β at a dose of 100 mg/kg compared to a 50 mg/kg dose. Conclusions. Anakinra prevents oxidative kidney damage induced by cisplatin in a dose-dependent manner. This result suggests that anakinra may be useful in the treatment of cisplatin-induced kidney damage.

Yapanoglu, Turgut; Ozkaya, Fatih; Yilmaz, Ali Haydar; Mammadov, Renad; Cimen, Ferda Keskin; Hirik, Erkan; Altuner, Durdu

Etoricoxib features antioxidant and anti-inflammatory properties concomitantly, suggesting that it may be beneficial in testicular ischemia reperfusion (I/R) damage. Our aim is to investigate the effects of etoricoxib on testicular I/R damage induced with torsion-detorsion (TD). The etoricoxib + torsion-detorsion (ETD) groups of animals were given etoricoxib in 50 and 100 mg/kg of body weight (ETD- 50 and ETD-100), while the testes torsion-detorsion (TTD) and sham operation rat group (SOG) animals were given single oral doses of distilled water as a solvent. TTD, ETD-50 and ETD-100 groups were subjected to 720o degrees torsion for four hours, and detorsion for four hours. The SOG group was not subjected to this procedure. Biochemical, gene expression and histopathological analyses were carried out on the testicular tissues. The levels of malondialdehyde (MDA), myeloperoxidase (MPO), interleukin-1 beta (IL-1β) and tumor necrosis factor alpha (TNF-α) were significantly higher, and the levels of total glutathione (tGSH) and glutathione reductase (GSHRd) were significantly lower in the TTD group, compared to the ETD-50, ETD-100 and SOG groups. Etoricoxib at a dose of 100 mg/kg better prevented I/R damage than the 50 mg/kg dose. Etoricoxib may be useful in clinical practice in the reduction of I/ R damage on testes caused by torsion-detorsion.

Doxorubicin (DOX) inhibits the enzyme tyaminpyrophosphokinase (TPK). Hence the synthesis of thiamine pyrophosphate (TPP) which is the active metabolite of thiamine stops. This leads to oxidative damage. In the literature, no studies on the protective effect of TPP against doxorubicin-induced oropharyngeal, nose and cochlear oxidative damage were found. In this study was investigated whether DOX produces oxidative stress in the oropharyngeal, nose and cochlea of animals and it examines the protective effect of TPP against DOX toxicity on these tissues. Guinea pig experimental animals were divided into groups as the controls. One group was given DOX, another group was given TPP + doxorubicin (TDOX) and the final group was the healthy group (HG). The TDOX group (n = 6) received an intraperi-toneal (ip) injection of 25 mg/kg TPP. The DOX (n = 6) and HG (n = 6) animals were given distilled water in the same way. TDOX and DOX animals were administered ip 5 mg/kg DOX one hour after the administration of TPP and distilled water once a day for seven days. At the end of this period, animals were sacri-fied with a high dose of anesthesia and the levels of malondialdehyde (MDA), myeloperoxidase (MPO), ni-tric oxide (NO), total glutathione (tGSH) and glutathione reductase (GSHRd) were determined in the removed oropharyngeal, nasal, and cochlear tissues. In addition, the TNF-α gene expression was measured. DOX was demonstrated to significantly increase the levels of MDA, MPO and NO and to reduce the levels of tGSH and GSHRd in the oropharyngeal, nasal and cochlear tissues of animals. TPP prevented the increase of the levels of MDA, MPO, NO and TNF-α with doxorubicin. TPP supressed the oxidative stress induced by DOXin the oropharyngeal, nasal and cochlear tissues. It can be suggested that TPP can be used against DOX toxicity

Nayki, Umit Arslan; Nayki, Cenk; Cetin, Nihal; Cimen, Ferda Keskin; Coban, Abdulkadir; Mammadov, Renad; Tas, Ismail Hakkı; Malkoc, Ismail

Aim: To investigate the effect of Kineret® on ischemia reperfusion (IR) injury in rat ovaries. Methods: Rats were divided into four groups: ovarian IR (IRG); 50 mg/kg Kineret® + ovarian IR (KIR-50); 100 mg/kg Kineret® + ovarian IR (KIR-100); and sham operation (SOC). KIR-50 (n = 10) and KIR-100 (n = 10) groups received an intraperitoneal injection of Kineret® at doses of 50 and 100 mg/kg, respectively. IRG and SOC (n = 10) rat groups were given distilled water as solvent using the same method. The results were compared between the groups. Results: In rats in which IR occurred, oxidant parameters, such as malondialdehyde (MDA) and myeloperoxidase (MPO), were increased, the level of proinflammatory interleukin 1 beta (IL-1β) was elevated and total glutathione (tGSH) as an antioxidant was decreased in the ovarian tissues. Administration of Kineret® at a dose of 100 mg/kg inhibited the increase of MDA, MOP and IL-1β and a decrease in tGSH caused by IR more significantly than administration of Kineret® at a dose of 50 mg/kg. In addition, 100 mg/kg Kineret® significantly decreased severe hemorrhage, degeneration and inflammatory signs in the follicular cells, caused by IR. Kineret® at 100 mg/kg markedly ameliorated increased apoptosis in ovarian tissue with IR more significantly than 50 mg/kg kineret. Conclusion: Our findings indicate that Kineret® might be useful in clinical practice for the treatment of damage that may occur as a result of ovarian torsion.

Kuyrukluyıldız, Ufuk; Küpeli, İlke; Bedir, Zehra; Özmen, Özgür; Onk, Didem; Süleyman, Bahadır; Mammadov, Renad; Süleyman, Halis

Objective: Paclitaxel is used in the treatment of cancer, and it may cause interleukin-1 beta (IL-1β)-related peripheral neuropathic pain. While our primary aim was to investigate the analgesic efficacy of an IL-1β antagonist, a secondary outcome was to assess whether a correlation exists between analgesic effects and antioxidant activity. Methods: A total of 24 albino Wistar male rats were divided into the following groups: paclitaxel-control, paclitaxel+50 mg kg−1 anakinra, paclitaxel+100 mg kg−1 anakinra and healthy group (HG). After the normal paw pain threshold in all animal groups was measured using a Basile algesimeter, a single dose of 2 mg kg−1 paclitaxel was intraperitoneally administered on the 1st, 3rd, 5th and 7th days. Anakinra was intraperitoneally administered following the final paclitaxel administration. The paw pain thresholds in the groups were measured before and seven days after paclitaxel administration and at the 1st and 3rd hours after anakinra administration. After the third hour of measurement, the rats were killed with high doses of ketamine, and the paw tissues were removed. Malondialdehyde, myeloperoxidase and total glutathione levels were measured in claw tissues, and IL-1β gene expression was determined. The biochemical results were compared with the results of the HG; in the meanwhile the claw pain threshold results were compared with the results obtained after the last paclitaxel and the results obtained from the 1st and 3rd hours after the anakinra application. Results: The claw paw pain threshold of the rats decreased one and three hours after anakinra administration. Further, 100 mg kg−1 anakinra had greater analgesic activity than 50 mg kg−1 anakinra. A correlation was found between the antioxidant and analgesic activities of 100 mg kg−1 anakinra. Conclusion: Anakinra may be useful to reduce paclitaxel-induced neuropathic pain; further, 100 mg kg−1 anakinra may have greater analgesic and antioxidant activities.

Ince, Ilker; Aksoy, Mehmet; Ahiskalioglu, Ali; Comez, Mehmet; Dostbil, Aysenur; Celik, Mine; Yilmaz, Ismayil; Mammadov, Renad; Dogan, Hasan; Boztok Ozgermen, Basak

Background: This study investigated the effects of metamizole and paracetamol on pain and oxidative stress induced by scalpel incision and carrageenan in rats. Materials and methods: Total of 144 rats were divided into groups of 12 animals. Six groups each were used for scalpel incision and carrageenan tests. Pain was inflicted by applying a scalpel incision or carrageenan. Pain-created groups by scalpel incision received metamizole (SIM) or paracetamol (SIP) at doses of 250 or 500 mg/kg. Pain-created groups by carrageenan received metamizole (CAM) or paracetamol (CAP) at doses of 250 or 500 mg/kg. Analgesic activity was determined by Basile Algesimeter. The COX-2 and MPO gene expressions were determined, and malondialdehyde and tGSH were measured in rat paws. Results: In the scalpel incision test, pain was reduced in groups of SIM-250 and SIM-500 in the first hour by 65.2% and 91.3%, respectively, and in the third hour by 51.9% and 77.8%, respectively, compared with the SIC group. In SIP-250 and SIP-500 groups, pain was reduced in the first hour by 43% and 74%, respectively, and by 33.4% and 59.3%, respectively, in the third hour compared with the SIC group. In the carrageenan test, in groups CAM-250 and CAM-500, pain was reduced in the first hour by 72.3% and 86.1%, respectively, and by 65.8% and 71.4%, respectively, in the third hour compared with the CCG group. In groups CAP-250 and CAP-500, pain was reduced in the first hour by 52.8% and 69.4%, respectively, and by 28.6% and 25.8%, respectively, in the third hour compared with the CCG group. Metamizole inhibited COX-2 gene expression at a dose of 500 mg/kg in the carrageenan test. At doses of 250 and 500 mg/kg, metamizole reduced COX-2 and MPO gene expressions and oxidative stress induced by scalpel incision or carrageenan. But both doses of paracetamol were unable to suppress that parameters. Conclusions: Our results show that metamizole is more effective than paracetamol for treating surgical trauma-related pain, inflammation, and oxidative stress and hence may be a preferential drug to paracetamol.

Controlled reperfusion for different durations in the treatment of ischemia-reperfusion injury of the rat ovary: Evaluation of biochemical features, molecular gene expression, and histopathology

Yapca, Omer Erkan; Kumbasar, Serkan; Salman, Suleyman; Yaral, Oguzhan; Sener, Ebru; Mammadov, Renad; Tekin, Yesim Bayoglu; Aksoy, Aysenur; Albayrak, Abdulmecit; Cetin, Nihal

High numbers of proinflammatory cells (PMNLs), which are carried by the blood to ischemic tissue during reperfusion, are considered responsible for inducing the inflammatory response that occurs in ischemia-reperfusion (I/R) injury. Our objective was to determine the controlled reperfusion (CR) interval duration (CRID) that would minimize the injury caused by the PMNLs that infiltrate ischemic tissue. Animal groups were divided into the following groups: Sham group, ovarian I/R group (OIR), and ovarian ischemia controlled-reperfusion groups OICR-1, OICR-2, OICR-3, OICR-4, OICR-5, OICR-6, which had their ovarian artery opened and then closed for 10, 8, 6, 4, 2, or 1 s, respectively. The results show that the COX-2 activity and the gene expression decreased while the COX-1 activity and the gene expression were found to be increased in parallel to the shortening of the period in CRID. From the histopathological examinations, the findings of hemorrhage, edema, congested vascular structures, degenerated cells, and migration and adhesion of PMNLs were scaled as follows: Sham group < OICR-6 < OICR-5 < OICR-4 < OICR-3 < OICR-2 < OICR-1. The results from the histopathological assessments were consistent with the molecular and biochemical findings. In conclusion, our findings suggest that increased COX-2 activity plays a role in I/R injury of the rat ovary, and that controlled reperfusion for 3, 2, or 1 s following 2 h of ischemia may attenuate the effects of I/R injury.
