Background Olaparib, a poly (ADP-ribose) polymerase (PARP) inhibitor widely used in cancer therapy, has been associated with gastrointestinal adverse effects; however, the underlying mechanisms remain unclear. Previous studies have suggested that oxidative stress may contribute to olaparib-induced tissue injury. Objective This study aimed to investigate the protective effects of adenosine triphosphate (ATP) against olaparib-induced gastric damage in rats. Methods Animals were divided into healthy control (HG), olaparib alone (OLPG), ATP alone (ATPG), and ATP + olaparib (ATOLG) groups. Gastric injury was evaluated by macroscopic assessment of ulcer area and ulcer number. Histopathological alterations were examined using hematoxylin-eosin (H&E) staining, and oxidative stress status was assessed by measuring lipid peroxidation and antioxidant defense parameters. Results Olaparib administration induced marked gastric mucosal injury, characterized by increased ulcer area and ulcer number, severe histopathological damage, and pronounced oxidative stress. In contrast, ATP co-administration significantly attenuated gastric injury, as evidenced by reduced ulcer area and ulcer number, improved histopathological architecture, and restoration of oxidative balance. Compared with the OLPG group, the ATOLG group showed significant improvement in macroscopic, histopathological, and oxidative stress parameters. Conclusion ATP administration attenuated olaparib-induced gastric damage and improved oxidative stress parameters in rats. Although the underlying mechanisms were not directly investigated in the present study, the obtained findings provide preliminary experimental evidence suggesting that ATP may have protective potential against olaparib-induced gastric injury.

Potential Hepatoprotective Effects of Adenosine Triphosphate Against Olaparib-Induced Oxidative Liver Injury: An Experimental Rat Model

Baykan, Ahmed Ramiz; Cerrah, Serkan; Kartal Baykan, Emine; Tanas, Serdar; Yavuzer, Bulent; Yazici, Gulce Naz; Kuzucu, Mehmet; Altuner, Durdu; Suleyman, Halis

Background: Olaparib, a clinically established poly (ADP-ribose) polymerase (PARP) inhibitor widely used in oncology, has significantly improved outcomes in several malignancies. However, increasing evidence indicates that olaparib may induce hepatocellular injury through mechanisms involving oxidative stress, inflammation, and mitochondrial dysfunction. Adenosine triphosphate (ATP), a key regulator of cellular bioenergetics and redox homeostasis, may confer protection against oxidative tissue injury. This study investigated the hepatoprotective effects of ATP against olaparib-induced oxidative liver damage in rats and compared its efficacy with melatonin. Methods: Twenty-four male albino Wistar rats were randomly assigned to four experimental groups (n = 6): healthy control (HG), olaparib-treated (OLP), ATP plus olaparib (ATOL), and melatonin plus olaparib (MLOL). Olaparib was administered at a dose of 100 mg/kg orally twice daily, whereas ATP (5 mg/kg, intraperitoneally) and melatonin (10 mg/kg, orally) were administered once daily. The treatments were administered for 14 consecutive days. Hepatic oxidative and inflammatory status was assessed by measuring malondialdehyde (MDA), total glutathione (tGSH), superoxide dismutase (SOD), catalase (CAT), interleukin-6 (IL-6) and ATP levels in liver tissue. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were measured as biochemical indicators of hepatocellular injury. Liver tissues were also examined histopathologically. Results: Olaparib administration significantly increased hepatic MDA and IL-6 levels together with serum ALT and AST activities, while significantly reducing hepatic tGSH and ATP levels and SOD and CAT activities compared with the healthy group (p < 0.001). Both ATP and melatonin significantly attenuated these alterations (p < 0.001). Notably, ATP demonstrated a more pronounced protective effect than melatonin, particularly in suppressing aminotransferase elevations and improving histopathological liver architecture. Conclusion: ATP significantly alleviates olaparib-induced hepatotoxicity by attenuating oxidative stress, suppressing inflammatory responses, restoring antioxidant defense mechanisms and significantly replenishing depleted hepatic ATP levels. These findings suggest that ATP may represent a promising therapeutic strategy for preventing PARP inhibitor-associated drug-induced liver injury.

Comparative Protective Effects of Anakinra and Tocilizumab in Experimental Ocular Ischemic Syndrome

Karimov, Shavkatbek; Sezgin, Esra Tuba; Mammadov, Renad; Suleyman, Bahadir; Yaqudov, Kamandar; Yazici, Gulce Naz; Coban, Taha Abdulkadir; Suleyman, Halis; Bayrakceken, Kemal

Ocular ischemic syndrome (OIS) is a vision-threatening disorder characterized by retinal ischemia, oxidative stress, and inflammation. In this study, the protective effects of anakinra, an interleukin-1 receptor antagonist, were investigated and compared with those of tocilizumab, an interleukin-6 receptor antagonist. Twenty-four male Wistar rats were randomly assigned to four groups (n = 6 per group): sham-operated (SG), carotid artery clamping/unclamping (CUG), anakinra-treated carotid artery clamping/unclamping (ACUG), and tocilizumab-treated carotid artery clamping/unclamping (TCUG). An experimental ocular ischemic injury model induced by transient bilateral common carotid artery occlusion was established by transient bilateral common carotid artery clamping followed by reperfusion. Retinal tissues were analyzed for malondialdehyde (MDA), total glutathione (tGSH), superoxide dismutase (SOD), catalase (CAT), tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and interleukin-6 (IL-6) levels, and histopathological examinations were performed. OIS markedly increased MDA, TNF-α, IL-1β, and IL-6 levels while significantly decreasing tGSH, SOD, and CAT activities. Both anakinra and tocilizumab attenuated oxidative stress and inflammatory responses; however, anakinra produced notably greater improvements in MDA, tGSH, SOD, and CAT levels. Histopathological evaluation demonstrated that both treatments reduced retinal injury, edema, vascular congestion, and inflammatory cell infiltration. Although retinal architecture appeared slightly better preserved in the anakinra-treated group, histopathological scores were comparable between the treatment groups. These findings suggest that anakinra may attenuate oxidative and inflammatory retinal damage in this experimental ocular ischemic injury model and may provide greater biochemical protection than tocilizumab, whereas histopathological protection was comparable between treatments. Nevertheless, further experimental and clinical studies are required to confirm these findings.

Dilber, Muhammet; Suleyman, Zeynep; Yucel, Nurinisa; Yazici, Gulce Naz; Mammadov, Renad; Cakir, Osman Kagan; Celik, Fatih; Coban, Taha Abdulkadir; Salcan, Ismail; Baykal, Huseyin

Background: Stomatitis is the most common mucosal side effect associated with Hydroxychloroquine (HCQ). Hippophae Rhamnoides (HR) is known to contain several phytonutrients, including anti-inflammatory, and antioxidant properties. This study aimed whether HR can mitigate HCQ-induced oxidative stress and inflammation in rat cheek tissue and to corroborate biochemical findings with macroscopic and histopathological evaluations. Methods: Twenty four rats were used and split into four groups: healthy control group (HG), receiving HR alone, receiving HCQ alone, and receiving both HR and HCQ (HRHQ). The drugs were administered for 10 days, and at the end of the 10th day, the rats were euthanized with high dose of thiopental sodium anesthesia and the inner cheek tissues removed. Results: HR decreased malondialdehyde, and pro-inflammatory cytokine levels, which increased by HCQ, while it increased anti-oxidant levels, which decreased by HCQ. The inner cheek tissue was examined macroscopically for edema, hemorrhagic cracks, hyperemia and gray appearance that developed due to HCQ use, which were alleviated with HR. Histopathologically, epithelial necrosis, muscle degeneration, scar tissue, polymorphonuclear cell (PMNL) infiltration, haemorrhage and edema were observed in the HCQ group. In contrast, in the HRHQ group, epithelial necrosis, and muscle degeneration were normal, haemorrhage was absent, PMNL infiltration persisted in some-areas, and moderate capillary congestion and dilatation were observed. detected. Conclusion: The results indicate that HR may be a potential candidate for preventing and treating stomatitis associated with HCQ use. Translating these findings into clinical practice could support the safe and effective use of HR as a therapeutic agent.

Salcan, Ismail; Dilber, Muhammed; Suleyman, Zeynep; Yucel, Nurinisa; Salcan, Sara; Kesan, Sefa; Yazici, Gulce Naz; Celik, Fatih; Koseturk, Merve; Alp, Nurdan Alcan

Inflammation, oxidative damage, and adenosine triphosphate (ATP) depletion play a role in the7 pathogenesis of cisplatin (CIS)-induced oral mucositis. Objective: The purpose of this research is to examine8 the impact of ATP against potential oral mucositis development in cisplatin-treated rats. Methodology All rats9 were randomly assigned to four groups, namely healthy control group (HG), ATP group (ATPG), Cisplatin10 group (CISG), and ATP + Cisplatin group (ATCS). Firstly, ATP 4 mg/kg was administered via intraperitoneal11 injection (IP) to both ATPG and ATCS groups. The same volume of normal saline was injected into HG and12 CISG groups. After 1 h, cisplatin 5 mg/kg was administered via IP to CISG and ATCS groups. The drugs13 were taken 1x1 for 7 d. Later, tongue tissues were collected from all groups. Biochemical, macroscopic,14 and histopathological examinations were performed on all tissues. Results: ATP inhibited cisplatin-induced15 oxidative damage and pro-inflammatory cytokines levels in tongue tissue. In the CIS group, a significant16 number of distinct sulcus formations were found in the apex and corpus, as well as a few ulcer foci in the17 corpus, significant papilla loss, and bleeding. Meanwhile, in the ATP group, a similar appearance to healthy18 tissue was observed. Histopathologically, it was determined that in cisplatin-aggravated tongue tissue19 damage, filiform papillae decreased when ATP was administered, and the arrangement and structures of20 the epithelium, blood capillaries, muscle groups, and adipose cell groups were normal. Conclusions: Oral21 mucositis caused by cisplatin is alleviated by ATP. These findings may be useful for developing new therapeutic22 approaches to prevent or treat mucositis, a side effect so severe that can lead to treatment discontinuation.23 24.

Ates, Irem; Isik, Bahar; Gozen, Fusun; Yazici, Gulce Naz; Gulaboglu, Mine; Mammadov, Renad; Huseynova, Gulbeniz; Altuner, Durdu; Suleyman, Halis

Background: Tramadol binds to opioid receptors and inhibits norepinephrine and serotonin reuptake, causing serotonin syndrome. Tianeptine stimulates serotonin reuptake and reduces serotonin levels. The aim of this study was to investigate whether tianeptine is effective against serotonin syndrome that may occur with serotoninergic drugs such as tramadol and citalopram. Methods: Rats were divided into eight groups (n = 6) that received tramadol (50 mg/kg), citalopram (10 mg/kg), or tianeptine (5 mg/kg) alone or a combination of tramadol + citalopram, tramadol + tianeptine, citalopram + tianeptine or tramadol + citalopram + tianeptine at the same doses administered to the stomach by oral gavage for 3 weeks. The healthy control group was given saline. Malondialdehyde, total glutathione, superoxide dismutase, and catalase levels were measured in removed brain tissues. The tissues were also examined histopathologically. Results: In the tramadol, tramadol + citalopram, and tramadol + citalopram + tianeptine groups, malondialdehyde levels were found to be higher compared to the control group, while glutathione, superoxide dismutase, and catalase levels were found to be lower. In other groups, values close to the control group were measured. Morphological degeneration was observed in neurons in the tramadol + citalopram group. The swelling of astrocytes and pericellular edema in oligodendrocytes were also observed. A significant population increase was noted in microglial cells. Blood vessels belonging to the tissue were observed to be severely dilated and congested. Histopathological damage was partially resolved in the group given tramadol + citalopram + tianeptine. Conclusions: The tramadol + citalopram combination caused severe oxidative stress in brain tissue. Tramadol alone caused mild damage in brain tissue, whereas tianeptine prevented the brain damage caused by tramadol.

Effect of Sugammadex on Ischemia-Reperfusion-Induced Oxidative and Inflammatory Ovarian Damage in Rats: Biochemical and Histopathological Evaluation

Kadioglu, Berrin Goktug; Suleyman, Bahadir; Mammadov, Renad; Yavuzer, Bulent; Altuner, Durdu; Yazici, Gulce Naz; Gulaboglu, Mine; Suleyman, Halis

Background and Objective: Polymorphonuclear leukocytes (PMNLs) play an important role in the pathogenesis of oxidative ischemia-reperfusion (I/R) damage. There is information that sugammadex inhibits PMNL infiltration into organs and tissues undergoing I/R. Present study aimed to investigate the potential protective effect of sugammadex against I/R-induced ovarian damage in rats and to elucidate its mechanism of action. Materials and Methods: The 24 female albino Wistar-type rats were divided into four groups as follows: Sham operation (SG), ovarian I/R (IRG), sugammadex (4 mg/kg) +ovarian I/R (SIR-4) and sugammadex (8 mg/kg) +ovarian I/R (SIR-8). Sugammadex was administered intraperitoneally. The animals were subjected to 2 hrs of ischemia followed by 2 hrs of reperfusion. Results: Sugammadex significantly inhibited the increase in I/R-induced malondialdehyde and the decrease in total glutathione, superoxide dismutase and catalase levels in ovarian tissue at a dose of 8 mg/kg compared with the dose of 4 mg/kg. Moderate follicular degeneration, vascular congestion, necrosis, oedema and hemorrhage were observed in the sugammadex (4 mg/kg) group, which significantly reduced the increase in I/R-related PMNL infiltration in ovarian tissue. Only mild vascular congestion was observed in the ovarian tissue of the 8 mg/kg sugammadex group, which did not show PMNL infiltration. Conclusion: Histopathological findings show that sugammadex protects ovarian tissue from oxidative and inflammatory injury of the I/R by inhibiting PMNL infiltration. Results also suggest that an 8 mg/kg dose of sugammadex, which completely inhibits PMNL infiltration into ovarian tissue, may be more beneficial than the 4 mg/kg dose in the treatment of I/R-induced ovarian damage.

Aim: The current study aimed to investigate the protective effects of adenosine triphosphate (ATP), metyrosine, and melatonin on possible methylphenidate cardiotoxicity in rats using biochemical and histopathological methods. Methods: Thirty rats were separated into five groups: healthy (HG), methylphenidate (MP), ATP + methylphenidate (ATMP), metyrosine + methylphenidate (MSMP), and melatonin + methylphenidate (MLMP). ATP (5 mg/kg) was given intraperitoneally once daily, metyrosine (50 mg/kg) orally twice daily, and melatonin (10 mg/kg) orally once daily. Methylphenidate (10 mg/kg) was administered orally once daily for 1 h after ATP, metyrosine and melatonin. The protocol was repeated for 30 days. Subsequently, blood samples were taken from the tail veins of the animals to measure adrenaline, noradrenaline, dopamine, troponin I (TP I) and creatine kinase MB (CK-MB) levels; the animals were then euthanized and the heart tissues were extracted. Tissues were analyzed for malondialdehyde (MDA), total glutathione (tGSH), superoxide dismutase (SOD), and catalase (CAT) and histopathologically. Results: In MP group, MDA, adrenaline, noradrenaline, dopamine, TP I, and CK-MB levels increased (p < 0.001) and tGSH, SOD, and CAT levels decreased (p < 0.001) compared to HG, and histopathologic damage developed. Oxidant levels were lower and antioxidant levels were higher in ATMP, MSMP, and MLMP groups compared to MP group (p < 0.001). Catecholamine levels were measured lower in the MSMP group compared to the MP group (p < 0.001). TP I and CK-MB levels were lower in ATMP, MSMP and MLMP groups compared to MP (p < 0.05), with the lowest being in rats given ATP (p < 0.001). ATP, melatonin, and metirozin applications were effective to different degrees in preventing histopathological changes. Conclusion: This study may guide clinical trials using ATP and melatonin to prevent methylphenidate-induced myocardial injury.

Cim, Emine Fusun Akyuz; Suleyman, Zeynep; Suleyman, Halis; Yazici, Gulce Naz; Coban, Taha Abdulkadir

Objectives: Our aim was to evaluate the comparative effects of sertraline and vortioxetine against stress-induced brain injury in rats. Methods: The rats were assigned to a nonstress group (NSG), stress-treated control (StC), sertraline + stress (SSt), and vortioxetine + stress (VSt) groups. Sertraline and vortioxetine (10 mg/kg) were given orally by gavage to the SSt and VSt groups. One hour later, all animals (except NSG) underwent forced immobilization to establish a stress model (2 hours). The drugs were given once a day for 30 days. The animals were killed with ketamine 150 mg/kg, and tissues were removed from the cerebral cortex. One-way analysis of variance and Fisher post hoc least significant difference were conducted for the analysis. Results: The malondialdehyde (nmol/mL) level was 2.58 ± 0.48 in the NSG, 8.09 ± 0.57 in the StC, 3.84 ± 0.53 in the SSt, and 2.84 ± 0.20 in the VSt group (P < 0.0002). The total glutathione (mmol/g) was 7.15 ± 0.59 in the NSG, 2.41 ± 0.43 in the StC, 4.58 ± 0.26 in the SSt, and 5.98 ± 0.13 in the VSt (P < 0.0002). The total oxidant status (mmol H2O2Eq/L) level was 3.56 ± 0.20 in the NSG, 9.99 ± 0.74 in the StC, 4.97 ± 0.39 in the SSt, and 3.81 ± 0.31 in the VSt (P < 0.0002). The total antioxidant status (mmolTroloxEq/L) level was 8.65 ± 0.37 in the NSG, 3.04 ± 0.22 in the StC, 6.29 ± 0.34 in the SSt, and 7.61 ± 0.40 in the VSt (P < 0.0002). Sertraline reduced pericellular edema in astrocytes and oligodendrocytes and decreased perivascular edema, dilatation, and congestion of blood vessels, whereas these were not seen with vortioxetine. Conclusions: Compared with sertraline, vortioxetine is a neuroprotective antidepressant with higher antioxidant activity and can more effectively prevent stress-induced brain tissue injury.

Icel, Aykut; Suleyman, Bahadir; Mammadov, Renad; Bulut, Seval; Cicek, Betul; Yazici, Gulce Naz; Gulaboglu, Mine; Ozcicek, Fatih; Suleyman, Halis

Background and Objective: The toxic effect of atorvastatin on the liver has been associated with oxidative stress. The Liv-52 is a polyherbal formulation with known hepatoprotective properties. In this study, the preventive effect of Liv-52 against the potential hepatotoxicity of atorvastatin was investigated. Materials and Methods: Eighteen rats were categorized into three groups of six rats each: Healthy group (HG), atorvastatin-treated group (ATC) and Liv-52+atorvastatin-treated group (LAT). The Liv-52 was orally administered at 50 mg kg(-1) and atorvastatin was orally given at 20 mg kg(-1) after 1 hr of Liv-52 administration. The Liv-52 and atorvastatin were administered once daily for two months. At the end of two months, blood samples were collected from the rats. Subsequently, rats were sacrificed under high-dose (50 mg kg(-1)) thiopental anesthesia and liver tissues were extracted. Biochemical analysis of extracted liver tissues and serum was performed. Liver tissues were additionally analyzed for histopathology. Results: The Liv-52+atorvastatin administration inhibited the atorvastatin-induced increase in malondialdehyde and decreased total glutathione and superoxide dismutase activities in the liver tissues. In addition, the rats receiving Liv-52 had lower levels of serum alanine aminotransferase, aspartate aminotransferase and lactate dehydrogenase compared to the atorvastatin group. Histopathologic analysis demonstrated that Liv-52 protected the liver tissue against atorvastatin-induced injury. Conclusion: The Liv-52 therapy may be useful in preventing atorvastatin-induced liver injury.

The effect of carvacrol on the proinflammatory cytokines, histology, and fertility outcome of cisplatin-related ovarian change in a rat model

Dinc, Kemal; Ozyurt, Ramazan; Coban, Taha Abdulkadir; Yazici, Gulce Naz; Suleyman, Zeynep; Yavuzer, Bulent; Suleyman, Halis

Objective: In women, agents used in chemotherapy treatment have side effects such as accelerating follicular depletion and early menopause. Thus, cytotoxic treatments may cause various effects ranging from partial damage to the ovary to premature ovarian failure (POI) and infertility. This study aimed to investigate the protective effect of carvacrol on cisplatin (CIS)-induced reproductive toxicity in female rats. Materials and methods: The animals were divided to four groups; a healthy group (HG), administered only cisplatin 2.5 mg/kg (CIS); cisplatin 2.5 mg/kg + carvacrol mg/kg (CC-50), and cisplatin 2.5 mg/kg + carvacrol 100 mg/kg (CC-100). In this study, the CC-50 and CC-100 groups were injected with carvacrol at 50 and 100 mg/kg intraperitoneally (IP). The CIS and HG groupswere administered normal saline as a solvent in the same way. One hour afterwardthe CC-50 and CC-100 groups were injected with cisplatin at 2.5 mg/kg IP. This procedure was continued once a day for 14 days. At the end of this period, six rats from each group were euthanized with high-dose anaesthesia. Biochemical (oxidant-antioxidant and proinflammatory cytokines) and histopathological examinations were performed on the right ovarian tissue removed from the dead rats. The remaining (n = 6 in each group) animals were kept in the laboratory with mature male rats for two months for breeding. Rats that didn't give birth within two months were considered infertile. A one-way ANOVA test was used for the biochemical analysis, the a Kruskal Wallis test was used for the histopathological analysis. Results: It has been observed that cisplatine causes oxidative stress and inflammatory damage in the ovarian tissue of animals and ultimately causes infertility due to this oxidative stress. While carvacrol significantly suppressed cisplatin-related oxidative stress in ovarian tissue at the 50 and 100 mg/kg doses, it could suppress proinflammatory cytokine increase only at thecytokine increase only at the 100 mg/kg dose. In addition, carvacrol significantly reduced the development of cisplatin-related infertility (from 0 to 83.3%) at a dose of 100 mg/kg. Conclusion: These findings suggest that carvacrol at high doses can reduce the harmful effects of cisplatin on the ovary and improve ovarian reserve in rats.

Tocilizumab is effective in preventing ovarian injury induced by ischemia-reperfusion in rats

Uzel, Kemine; Lakhno, Igor; Turkler, Can; Kuzucu, Mehmet; Yazici, Gulce Naz; Mammadov, Renad; Suleyman, Bahadir; Kale, Ahmet; Suleyman, Halis

Ovarian torsion can be defined as the bending of the ovaries on the supporting ligament, disrupting both venous and arterial blood circulation. Insufficient blood flow causes ovarian tissue hypoxia and leads to ischemia. This study aimed to investigate whether tocilizumab has a protective effect on ischemia-reperfusion injury due to ovarian torsion in rats. Eighteen female Wistar albino rats were divided into three equal groups (Sham (SG), ischemia-reperfusion (OIR), and ischemia-reperfusion+tocilizumab (OIRT)). Degeneration, necrosis, vascular dilatation/congestion, interstitial edema, hemorrhage, and polymorphonuclear lymphocyte (PMNL) infiltration scores were significantly different between the groups (p=0.001 for all parameters). Moreover, the OIRT group had a significant improvement in these criteria compared to the OIR group (p<0.05). Additionally, there was a considerable difference between OIRT and OIR groups in the number of primordial, developing, and atretic follicles groups (p<0.05), while there was no difference in the number of corpus luteum (p=0.052). Stress markers or cytokines, such as MDA, tGSH, NF-κB, TNF-α, IL-1β, and IL-6, were significantly different between groups (p<0.05). Furthermore, a significant improvement was found in the measured variables when the OIRT group was compared with the OIR group (p<0.05). Tocilizumab may be an alternative option for treating ischemia-reperfusion injury due to ovarian torsion.

Protective Effect of Lycopene against Ethanol-induced Oxidative Kidney Damage in Rats

Yilmaz, Canan Tuz; Suleyman, Bahadir; Mammadov, Renad; Bulut, Seval; Gursul, Cebrail; Yazici, Gulce Naz; Gulaboglu, Mine; Suleyman, Halis

SUMMARY. Alcohol consumption can have dramatic effects on organs and tissue cells, with kidneys being some of the most affected organs. This study investigates the effect of lycopene on ethanol-induced oxidative kidney damage in rats. Albino Wistar rats were randomly divided into three groups. The first group served as the healthy group (HG), receiving distilled water as a solvent; the second group was the ethanol group (EtON), receiving only ethanol; the third group received both lycopene and ethanol (LEt). While ethanol ad-ministration increased malondialdehyde (MDA), total oxidant status (TOS), and oxidative stress index (OSI) levels, it caused a decrease in total glutathione (tGSH) and total antioxidant status (TAS) levels. Also, ethanol caused histopathological damage to the kidney. It has been observed that lycopene inhibits ethanol-induced biochemical and histopathological changes in kidney tissue. Lycopene inhibited the change in the oxidant-anti-oxidant balance towards oxidants under ethanol treatment and prevented ethanol-associated kidney oxidative damage.

The Effect of Carvacrol on Oxidative and Inflammatory Liver Damage due to Staphylococcus aureus Infection in Rats

Dabanlioglu, Bulent; Suleyman, Bahadir; Mammadov, Renad; Coban, Taha Abdulkadir; Yazici, Gulce Naz; Akyuz, Sumeyye; Yavuzer, Bulent; Bulut, Seval; Suleyman, Halis

SUMMARY. Staphylococcus aureus is a bacterial pathogen that can cause oxidative stress. Carvacrol has anti-microbial, antioxidant, anti-inflammatory and hepatoprotective effects. This study evaluated the antibacterial effect of carvacrol and its effect on liver damage by S. aureus. The rats were divided into four groups: con-trol(HG), S. aureus injected (SaG), S. aureus + carvacrol (SaC), and S. aureus + cefazolin (SaCF). S. aureus was injected intraperitoneally (ip) to the SaG, SaC and SaCF groups. 24 h after the injection, carvacrol was given to the SaC group and cefazolin to the SaCF group as ip. Carvacrol and cefazolin administration was continued for 7 days. According to our antibiogram results, although the antibacterial effect of carvacrol was weaker than cefazolin, it prevented the increase of malondialdehyde (MDA), aminotransferase (ALT), aspar-tate aminotransferase (AST) and proinflammatory cytokines and the decrease of total glutathione (tGSH) better (p < 0.001). It also managed histopathological damage better than cefazolin. The results indicate that carvacrol may be helpful in the treatment of liver damage from infection.

Cicek, Ibrahim; Altuner, Durdu; Suleyman, Zeynep; Mammadov, Renad; Coban, Taha Abdulkadir; Gulaboglu, Mine; Yazici, Gulce Naz; Bayrakceken, Kemal; Suleyman, Halis

5-Fluorouracil (5-FU) is also known to cause ocular toxicity. The purpose of this study was to investigate the effects of Carvacrol upon possible 5-FU-induced optic nerve damage in rats biochemically and histopathologically. In this study totally, eighteen albino Wistar male rats were used. The rats were grouped under three categories as the healthy (HG) (n=6), 100 mg/kg 5-FU treated alone (FUG) (n=6) and 50 mg/ kg Carvacrol+100 mg/kg 5-FU (CFU) groups (n=6). Carvacrol at 50 mg/kg was injected intraperitoneally (ip) to the CFU group. The same volume of 0.9% NaCl was administered as the solvent ip to HG and FUG groups. One hour after administrating Carvacrol and solvent, 100 mg/kg 5-FU was ip injected to CFU and FUG groups. Carvacrol was administered once daily for 10 days. Three-dose 5-FU was administered on the 1st, 3rd, and 5th days as one dose each. On the 10th day, the animals were euthanised with high-dose anesthesia and optic nerve tissues were removed. Malondialdehyde (MDA), reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), and Interleukin (IL-6) levels were measured in optic nerve tissues. The tissues were also examined histopathologically. The results obtained from HG and CFU groups were compared with the FUG group. Significantly higher MDA and IL-6 levels (p < 0.001) and significantly lower (p < 0.001) tGSH, SOD, and CAT levels were determined in optic nerve samples of the FUG group rather than HG and CFU groups. In histopathological analyses, determined that astrocyte and oligodendrocyte nuclei were mostly darkly stained, the majority of astrocytes were hypertrophied and degenerated, and some oligodendrocytes were degenerated in the FUG group unlike HG and CFU groups. Moreover, the optic nerve tissue was determined to be vacuolized and intensely edematous, and blood capillaries were intensely dilated and congested. In conclusion, 5-FU caused optic neuropathy due to oxidative and inflammatory damage, and Carvacrol reduced this oxidative and inflammatory damage and histopathological changes in tissues. This revealed that Carvacrol was possibly to be efficient upon 5-FU-induced optic nerve toxicity.

Otoprotective Effect of Nimesulide: Biochemical and Histopathologic Evaluation

Kuduban, Ozan; Suleyman, Zeynep; Gulaboglu, Mine; Yazici, Gulce Naz; Suleyman, Halis

Background and Objective: It is known that nimesulide reduces the toxic effects of Non-Steroidal Anti-Inflammatory Drugs (NSAID), eliminating their inhibitory effects on cyclooxygenase-1 (COX-1) enzyme. COX-1 enzyme inhibition is thought to be responsible for aspirin ototoxicity therefore current study aimed to investigate the effect of nimesulide on aspirin ototoxicity in rats. Materials and Methods: Nimesulide 100 mg kg(-1) orally was administered to the nimesulide+aspirin group (NASA) and the same volume of distilled water as solvent was administered to the aspirin group (ASA) and the healthy (HG) group. Aspirin at a dose of 1000 mg kg(-1) was administered orally to the NASA and ASA group one hour after the administration of nimesulide and solvent. This procedure was repeated once a day for 7 days. Then, the animals were euthanized with a high dose of anaesthesia (50 mg kg(-1) thiopental sodium) and their cochlea and cochlear nerve tissues were removed. Results: Malondialdehyde (MDA) level was significantly higher and total glutathione (tGSH) and cyclooxygenase-1 COX-1 levels were significantly lower in the ASA group in which prominent histopathological damage was found in the cochlea and cochlear nerve, compared to the HG and NASA group. The COX-2 level was found to be almost the same in all the g roups. This indicates that aspirin reduces the COX-1 and tGSH levels and increases the MDA level, causing ototoxicity. Conclusion: Aspirin significantly decreased the COX-1 activity in the cochlea and cochlear nerve tissue, compared to the healthy and nimesulide group. Current study concluded that the co-administration of nimesulide and aspirin will increase the therapeutic effect of aspirin and reduces its side-effects.

The Effect of Anakinra on Acrylamide-induced Peripheral Neuropathy and Neuropathic Pain in Rats

Ersoy, Alevtina; Tanoglu, Ceyda; Yazici, Gulce Naz; Coban, Taha Abdulkadir; Mammadov, Renad; Suleyman, Halis

Acrylamide is a neurotoxic compound. Moreover, anakinra is an interleukin-1 (IL-1) receptor antagonist used in rheumatoid arthritis treatment. This study investigated the effect of anakinra on acrylamide-related neuropathy and neuropathic pain. Acrylamide exposure caused a significant decrease in the pain threshold; an increase in malondialdehyde (MDA), tumor necrosis factor -alpha (TNF-alpha), and interleukin-1 beta (IL-113) levels; and a decrease in total glutathione (tGSH) values in the sciatic nerve. This indicates hyperalgesia presence, oxidative stress, and peripheral nerve tissue inflammation. Anakinra treatment significantly reduced the MDA, IL-113, and TNF-alpha levels, and increased the pain threshold and mean tGSH values. The analgesic effect of anakinra was 67.9% at the first hour, increasing to 74.9% and 76.7% at the second and third hours, respectively. The group receiving acrylamide exhibited histopathological changes (e.g., swollen and degenerated axons, hypertrophic and hyperplasic Schwann cells, and congested vessels). The use of anakinra significantly improved these morphological changes. Anakinra is concluded to reduce neuropathic pain and prevent neurotoxic effect of acrylamide on peripheral nerves due to its analgesic, antioxidant, and anti-inflammatory properties.

The effect of rutin on pulmonary contusion induced by blunt trauma in rats: Biochemical and histopathological evaluation

Elma, Bekir; Mammadov, Renad; Bilgin, Yasin; Yazici, Gulce Naz; Süleyman, Halis; Çoban, Taha Abdulkadir

Background: This study aims to investigate the possible protective effects of rutin, also called vitamin P1, against pulmonary contusion induced by blunt chest trauma in a rat model. Methods: Thirty male albino Wistar rats were separated into three equal groups as healthy group, trauma group, and trauma+rutin group. After anesthesia provided by intraperitoneal administration of 60 mg/kg ketamine and xylazine by inhalation at appropriate intervals, 200 g weight was dropped from 1 m height to the anterior chest wall of the animals in the trauma group (n=10) and trauma+rutin group (n=10) and pulmonary contusion was created. Thirty min after the trauma, 50 mg/kg of rutin was administered into the stomach of trauma+rutin group animals orally with gavage. The rats received rutin once daily for two days and were sacrificed 48 h later. Their lung tissues were removed and examined biochemically and histopathologically. Results: Nuclear factor-kappa B, cyclooxygenase-2, and malondialdehyde levels increased in the trauma group compared to the healthy group, and rutin administration prevented this increase. Total glutathione levels decreased in the trauma group, and rutin administration also prevented this decrease. The histopathological findings were compatible with the biochemical findings. Conclusion: Our study results suggest that rutin has a protective effect on contused lung tissue in rats.

The effect of carvacrol on oxido-inflammatory ovarian injury and infertility induced by ischemia-reperfusion in rats

Sahin, Nurullah; Delibas, Ilhan Bahri; Isaoglu, Unal; Suleyman, Bahadir; Yazici, Gulce Naz; Coban, Taha Abdulkadir; Uzel, Kemine; Suleyman, Halis; Arslan, Veysel

Background: Increased oxidants and proinflammatory cytokines play a role in the pathogenesis of ovarian ischemia-reperfusion (OIR) injury and related infertility. Carvacrol has antioxidant, antibacterial and anti-inflammatory properties. Methods: Protective effect of carvacrol against ischemia-reperfusion (IR)-related ovarian damage and infertility was investigated. IR process were applied to the ovaries of rats, which were divided into the following groups (n = 12): OIR, IR +50 mg/kg carvacrol (IRC-50), IR +100 mg/kg carvacrol (IRC-100) and sham group (SG). After the reperfusion process, six rats from each group were killed and the removed ovaries were examined biochemically and histopathologically. The remaining animals were kept two months with mature male rats to reproduce. Results: At a dose of 50 mg/kg, carvacrol suppressed the oxidant parameter increase and antioxidant decrease caused by IR in ovarian tissue. At a dose of 100 mg/kg, carvacrol antagonized both oxidant and proinflammatory cytokine increase and antioxidant decrease. Histopathologically, severe degeneration of follicles in the ovaries of the OIR group, necrotic cell accumulations, hemorrhage in the corpus luteum, edema in the interstitial tissue, polymorphous nuclear leukocyte (PNL) infiltration, and congestion and dilation of blood vessels were detected. Inflammatory symptoms such as edema in the ovarian tissue, congested dilated blood vessels and PNL infiltration were observed at a dose of 50 mg/kg of carvacrol, but these histopathological findings were not observed at a dose of 100 mg/kg. Conclusion: A dose of 100 mg/kg carvacrol, which eliminated inflammatory damage, significantly prevented the development of IR-induced infertility. Carvacrol may be beneficial in the treatment of IR-related ovarian damage and infertility. Copyright:

Efficiency of hypericum perforatum, povidone iodine, tincture benzoin and tretinoin on wound healing

Yalcınkaya, Esin; Basaran, Mustafa Mert; Tunckasık, Mehmet Emin; Yazici, Gulce Naz; Elmas, Çigdem; Kocaturk, Sinan

Different topical agents have been used to accelerate wound healing. The purpose of this study is to compare the safety and efficacy of topical application of the extract of Hypericum perforatum (HPE), povidone iodine (PI), tincture benzoin (TB) and tretinoin (T) on surgical wound healing. Ten adult female, Wistar albino rats were included in the study. HPE, PI, TB and T solutions were applied on the wounds. After seven days, tissue samples were collected and inflammatory cells, re-epithelialization, granulation tissue, angiogenesis, collagen accumulation, hemorrhage and lysis of cells were investigated histopathologically. No dermal toxicity was noted. HPE, TB, PI have all showed good epithelialization and granulation, but HPE showed the most advanced stage of healing within a short period of time. HPE had significantly higher values of re-epithelialization and collagen accumulation, but lower inflammatory cell count and granulation tissue. TB had the second best in re-epithelialization, collagen accumulation and the highest granulation tissue. PI induced better reepithelialization and granulation than the control group with remarkable cell lysis. As a result, HPE can be a safe, effective, and cheap agent that can be used for surgical wounds.
