Introduction and objectives: Testicular ischemia/reperfusion (I/R) injury develops after torsion and following detorsion of the testis. Reactive oxygen species were produced and oxidative damage begins to occur due to I/R process. Nimesulide, which is a specific cyclooxygenase-2 inhibitor drug, have antioxidant, antiinflammatory, analgesics and antipyretic effects. We aimed to investigate biochemically and histopathologically effect of nimesulide on testis I/R injury in rats induced by the testicular torsion–detorsion. Material and methods: In this study, 24 albino Wistar male rats were divided into four groups (6 rats in each group): ischemia/reperfusion applied + 50 mg/kg nimesulide administrated (NIM-50), ischemia/reperfusion applied + 100 mg/kg nimesulide administrated (NIM-100), ischemia/reperfusion applied (IR) and Sham surgery (SS) groups. Nimesulide was administered to NIM-50 and NIM-100 groups at the 50 mg/kg and 100 mg/kg doses before 2 h applied I/R procedures. The IR group were applied only I/R procedures, no drug treatment was applied. Animals were sacrificed under high dose anesthesia and left testes were extracted. Testes were examined biochemically and histopathologically. Results: Total glutathione (tGSH) and cyclooxygenase-1 (COX-1) levels were increased in the NIM-50 and NIM-100 groups compared to IR group. The levels of COX-2, malondialdehyde (MDA), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) were lower in the NIM-50 and NIM-100 groups than in the IR group. Some histopathological changes seen in IR group. This findings were decreased in NIM-50 group and prevented in NIM-100 group. Conclusion: Nimesulide prevented inflammation and oxidative stress. Our results suggest that nimesulide may be have a protective effect on testicular I/R injury.

Nonsteroidal anti-inflammatory drugs (NSAID) are among the aggressive factors causing gastric ulcer. They cause oxidative damage in the gastric tissue and lead to intracellular calcium deposition. Lercanidipine is a calcium channel blocker derived from the third generation dihydropyridine. The aim of this study is to analyse the effect of lercanidipine on indomethacin-induced gastric ulcers. A total of 24 albino Wistar male rats were divided into four groups; those who received indomethacin 25 mg/kg (IND), 5 mg mg/kg lercanidipine +25 mg/kg indomethacin (LC-5), 10 mg/kg lercanidipine+25mg/kg indomethacin (LC-10) and healthy rats who received 0.5 mL distilled water. Six hours after the application of indomethacin, the animals were sacrificed by high dose thiopental sodium. The stomachs of the animals were excised to perform a macroscopic analysis and the ulcerous region was measured on millimeter paper. All the stomachs were subjected to a biochemical analysis. Macroscopic analysis revealed hyperaemia on the gastric surface of the indomethacin group. Ulcerous tissues formed by oval, circular or irregular mucosal defects in varying diameters and depths were observed on the whole surface of the stomach. Hyperaemia was lower and ulcerous region was smaller in groups LC-5 and LC-10 compared to IND group. Malondialdehyde and myeloperoxidase levels were significantly lower and total glutathione and cyclooxygenase-1 activity were higher in groups LC-5 and LC-10. Lercanidipine did not change the cyclooxygenase-2 activity. Lercanidipine in doses 10 mg/kg is more effective compared to 5 mg/kg. Lercanidipinine can be useful in the treatment of NSAID-induced gastric damage.

Peptic ulcer has been reported to increase acid secretion and lead to oxidative stress. Benidipine is an antihypertensive drug with antioxidant properties. This study aims to investigate the effects of benidipine on indomethacin-induced gastric ulcers in rats. Four groups were designed in this study. Gastric ulcer was occurred with indomethacin in the first three groups. These groups were treated with famotidine, benidipine and distilled water respectively 5 minutes before indomethacin administration. The fourth group rats didn’t have gastric ulcer and they received only distilled water. Six hours after drug administration gastric tissue was extracted and macroscopic and biochemical examinations were performed. Damaged areas in the stomach of benidipine and famotidine receiving animal groups were smaller than the indomethacin received animals. Indomethacin elevated the levels of malondialdehyde and myeloperoxidase in the stomach tissue (p<0.0001), and also decreased glutathione, glutathione related enzymes and superoxide dismutase (p<0.0001). Benidipine may be useful in preventing the toxic effect of indomethacin on the stomach.

No previous studies have examined the effect of Hippophae rhamnoides fruit extract (HRE), which contains both water and fat soluble vitamins, on cisplatin-induced ototoxicity. Therefore, the aim of the study was to investigate the effects of FIRE on cisplatin-induced ototoxicity in Guinea pigs and to evaluate the biochemical, gene expression and histopathological changes associated with cisplatin use. Experimental animals were divided into three groups: healthy (HG), HRE+cisplatin (HRC) and cisplatin control (CCG) groups. Biochemical, gene expression and histopathological examinations were carried on the removed inner ear tissues. In the HRC group, the oxidant parameter was lower and the anti-oxidant parameter was higher than in the CCG. These results are supported by gene expression levels and histopathological results. The use of FIRE against cisplatin-induced oxidative ototoxicity may be easier, more cost effective and more beneficial than the use of vitamins alone or in combination with each other.

Oxidative stress is considered responsible for cisplatin autotoxicity. The aim of this study was to examine the protective effect of rutin on cisplatin-induced oxidative vestibular nerve damage in rats. Animals were divided into four groups (n = 6): healthy group (HG), 50 mg/kg rutin + cisplatin group (RG-50), 100 mg/kg rutin + cisplatin group (RG-100), and cisplatin group (CG). Rutin was administered orally for eight days, and 5 mg/kg cisplatin was injected intraperitoneally once every two days. Malondialdehyde, myeloperoxidase, and total glutathione levels were measured in serum and vestibular nerve tissue and examined histopathologically. Malondialdehyde and myeloperoxidase levels were lower in the serum and vestibular nerve tissue of RG-50 and RG-100 groups compared to the CG group; total glutathione was higher. Also, several histopathological changes were seen in the vestibular nerve tissue of the CG group. Rutin inhibited nerve tissue damage and may protect vestibular nerve tissue from cisplatin-induced oxidative damage.

Purpose: To investigate the effects of the EtOAc extract of U. longissima which is uninvestigated previously on esophagogastric cancer induced in rats with N-methyl-N-nitro-N-nitrosoguanidin (MNNG). Methods: The anticancer activity of EtOAc extract of U. longissima was examined in the esophagogastric adenocarcinoma models induced in rats with MNNG. EtOAc extract of U. longissima, 50 and 100 mg/kg oral doses were administered once daily for six months. MNNG induced differentiated and undifferentiated type adenocarcinomas in the esophageal and gastric tissues of rats. Results: EtOAc extract of U. longissima obtained from U. longissima prevented gastric and esophageal cancerogenesis induced in rats with MNNG. EtOAc extract of U. longissima did not have a lethal effect at doses of 500, 1000 and 2000 mg/kg. The prominent anticarcinogenic activity of EtOAc extract of U. longissima 50 and 100 mg/kg suggests that it is not toxic and it is selective to the cancer tissue. Conclusion: This information may shed light on clinical implementation of EtOAc extract of U. longissima in future.

The aim of this study was to examine the effects of Hippophae rhamnoides extract (HRE), known to have antioxidant properties, on ischemia reperfusion-induced brain injury in rats. Eighteen albino Wistar male rats were divided into three equal groups: 15 min of brain ischemia-3 h of reperfusion (BIR) group, 50 mg/kg HRE + brain ischemia-reperfusion (HRE + BIR) group, and control group for sham operation (SHAM). Malondialdehyde (MDA) level, myeloperoxidase (MPO) activity and 8-OHdG levels significantly increased, whereas total glutathione (tGSH) level, glutathione reductase (GSHRd), glutathione s-transferase (GST) activities decreased in the brain tissues of BIR group compared to SHAM group. HR application reversed these changes and brought the levels closer to the level of SHAM group. This study has revealed that HR inhibits IR-induced oxidative damage and inflammation in rat’s brain biochemically and it is effective in maintaining antioxidant activity and protecting against DNA damage in the tissue.

Ototoxicity of cisplatin is one of the major dose-limiting side effects. Cisplatin-dependent ototoxic-ity is known to be associated with reactive oxygen species. Protective effect of taxifolin, being a flavanone found in onions, milk thistle, French maritime, and Douglas fir bark, against cisplatin-associated ototoxicity will be examined in this study. There are no studies in the literature examining the protective effect of taxifolin against cisplatin-induced ototoxicity. 50 mg/kg taxifolin was orally administered to TXC group rats (n-6), and distilled water was orally administered as a solvent to CG (n-6) and HG (n-6) groups. One hour later, 5 mg/kg cisplatin was administered intraperitoneally (i.p) to TXC and CG groups. This procedure was repeated once a day for 7 days. At the end of this period, all animals were sacrificed by high-dose anesthesia (50 mg/kg thiopen-tal sodium) and biochemical and histopathological examinations were performed on the dissected cochlear tissue. Our biochemical test results showed that oxidant parameters were significantly increased whereas antiox-idant parameters were significantly decreased in the cisplatin group (CG) compared to taxifolin (TXC) and healthy (HG) groups (p < 0.0001 for both parameters). Histopathological results showed that severe cochlear vestibular membrane degeneration, dilated conjunctival blood vessels, edema and destruction developed in the cisplatin group. However, no pathological findings were found in the taxifolin-treated group except for mild degeneration and edema. This information suggests that taxifolin may be useful in the treatment of cisplatin-associated oxidative cochlear damage.

Purpose: Diabetic retinopathy (DR) is one of the leading causes of blindness. In DR patients, antioxidant defence is disrupted, and production of reactive oxygen species and pro-inflammatory cytokines such as interleukin 1β (IL-1β) and tumour necrosis factor alpha (TNF-α) increases. Taxifolin has been reported to suppress reactive oxygen species, IL-1β and TNF-α production. The aim of this study is to biochemically and histopathologically examine the protective effect of taxifolin against DR damage induced by alloxan. Materials and methods: Alloxan received rats with a blood glucose level of ≥250 mg/dL were divided into taxifolin-treated (TAX) (n = 6), diabetic control (DC) (n = 6) groups. There were rats received only saline in non-diabetic control (NC) group (n = 6). Taxifolin (50 mg/kg) was orally administered to the TAX group rats. DC and NC rats received the same volume of saline as a solvent. This procedure was repeated once a day for 3 months. At the end of this period, animals were killed by high dose thiopental sodium anaesthesia. Histopathological examinations were then performed on excised rat eyes. Malondialdehyde (MDA), total glutathione (tGSH), IL-1β and TNF-α levels were measured in obtained blood samples. Results: MDA, IL-1β and TNF-α levels were significantly increased in blood samples of DC group rats with hyperglycemia induced by alloxan compared with NC group (p < 0.0001), and decreased in the TAX group compared with the DC group (p < 0.0001). The levels of tGSH were significantly decreased in blood samples of DC group rats compared with NC group (p < 0.0001), and increased in the TAX group compared with the DC group (p < 0.0001). Histopathologically, retinal ganglion cells of the TAX group had a slightly dilated and congested blood vessel, and severe damage was inflicted to the retinal ganglion cell layer of the DC group. Conclusions: Experimental results suggest that taxifolin may be beneficial in the treatment of DR.

An ischaemia/reperfusion (I/R) injury in an organ can also cause damage to other organs. In this study, the effects of nizatidine were investigated on ovarian I/R injury and subsequent heart damage in rats. Rats were divided into groups; ovarian I/R (IRG), nizatidine 25 mg/kg+ ovarian I/R (NIR), and control (CG) groups. The NIR and IRG groups were given 2 hours of ischaemia followed by 2 hours of reperfusion by applying, and subsequently releasing, a vascular clip to the right ovarian artery. While oxidant levels in the IRG group were higher than NIR and CG groups, antioxidant levels were lower. Necrosis, congested blood vessels, haemorrhage, leukocyte infiltration and oedema were observed in the ovarian and heart tissues of the IRG group. These findings were reduced in the NIR group. Nizatidine suppressed oxidative stress in ovarian and heart tissues and may be useful in prevention of I/R–induced ovarian and cardiac injury.

Postoperative pain begins with tissue incision, continues with the production of free oxygen radicals, and is intensified by the inflammatory response. We investigated the effect of esomeprazole on postoperative pain and lung and heart complications of laparotomy. Albino Wistar-type rats were grouped into the paw scalpel incision (PSI), applied control (SIC), PSI + 20 mg/kg esomeprazole (SIE), PSI + 20 mg/kg meperidine (SIM), and healthy group (HG). For the laparotomy test, the rats were grouped as follows: laparotomy applied control (LTC), laparotomy + 20 mg/kg esomeprazole (LTE), laparotomy + 20 mg/kg meperidine (LTM), and healthy group (HG). Meperidine suppressed the PSI-induced pain. Esomeprazole decreased the level of oxidant and proinflammatory cytokines in the serum and lung and heart tissues of the LTE group and in the paw tissues of the SIE group, but meperidine did not. There were no histopathological findings in the lung and heart tissues of the LTE group, but tissue damage was found in the LTC and LTM groups. Esomeprazole may help reduce major surgery complications.

Thiamine pyrophosphate (TPP) is the active metabolite of thiamine. This study aimed to investigate the effects of thiamine and TPP on cisplatin-induced peripheral neuropathic pain (PNP). Male albino Wistar type Rattus norvegicus were divided into six groups (n=6) that received 2 mg/kg cisplatin (CIS), 25 mg/kg thiamine (TM), 2 mg/kg cisplatin+25 mg/kg thiamine (CTM), 25 mg/kg TPP (TPP), 2 mg/kg cisplatin+25 mg/kg TPP (CTPP), or distilled water (healthy group; HG) for 8 days intraperitoneally. Analgesic effect was measured with a Basile Algesimeter. IL-1β, malondialdehyde (MDA), total glutathione (tGSH), thiamine, and TPP were determined in blood samples. Histopathological examinations were performed on removed sciatic nerves. The percent analgesic effects of the CTM and CTPP groups were calculated to be 21.3% and 82.9%, respectively. Increased production of IL-1β and MDA by cisplatin was inhibited by TPP, while it was not inhibited by thiamine. Conversion of thiamine to TPP significantly decreased in the CIS group. Histopathological and biochemical investigations demonstrated that hyperalgesia and sciatic nerve damage developed in the CIS and CTM groups with low TPP levels. These results indicate that cisplatin inhibits the formation of TPP from thiamine, leading to severe PNP. This finding suggests that TPP may be more beneficial than thiamine for the treatment of cisplatin-induced PNP.

The aim of this study was to investigate the effect of thiamine pyrophosphate (TPP), administered via sugar water, on retinal neovascularisation in rats. Animals were assigned to three groups, namely the TPP sugar-water group (TPSWG, n = 12), the control group (CG, n = 12) and the healthy group (HG, n = 12). The TPSWG was injected intraperitoneally with TPP once a day for 6 months. CG and HG rats were given distilled water in the same way. TPSWG and CG rats were left free to access an additional 0.292 mmol /ml of sugar water for 6 months. The fasting blood glucose (FBG) levels of the animals were measured monthly. After 6 months, biochemical, gene expression and histopathologic analyses were carried out in the retinal tissues removed from the animals after they were killed. The measured FBG levels were 6.96 ± 0.09 mmol/ml (p < 0.0001 vs. HG), 6.95 ± 0.06 mmol/ml (p < 0.0001 vs. HG) and 3.94 ± 0.10 mmol/ml in the CG, TPSWG and HG groups, respectively. The malondialdehyde (MDA) levels were found to be 2.82 ± 0.23 (p < 0.0001 vs. HG), 1.40 ± 0.32 (p < 0.0001 vs. HG) and 1.66 ± 0.17 in the CG, TPSWG and HG, respectively. Interleukin 1 beta (IL-1β) gene expression was increased (3.78 ± 0.29, p < 0.0001) and total glutathione (tGSH) was decreased (1.32 ± 0.25, p < 0.0001) in the retinal tissue of CG compared with TPSWG (1.92 ± 0.29 and 3.18 ± 0.46, respectively). Increased vascularisation and oedema were observed in the retinal tissue of CG, while the retinal tissues of TPSWG and HG rats had a normal histopathological appearance. A carbohydrate-rich diet may lead to pathological changes in the retina even in nondiabetics, but this may be overcome by TPP administration.

Objective: Cisplatin is an anticancer drug used for treating childhood solid tumors. Symptoms related to cisplatin-induced cardiovascular adverse effects may be mild or severe. Rutin (vitamin P1) has many properties, including as antioxidant, anticancer, antidiabetic, antimicrobial, antiulcer, and tissue renewal properties. Therefore, we aimed to biochemically, histopathologically, and immunohistochemically demonstrate the effect of rutin on cisplatin-induced cardiotoxicity in rats. Methods: The rats included in our study were divided into four groups: Healthy group (HE), 5-mg/kg cisplatin group (CP), 50 mg/kg rutin+5-mg/kg cisplatin (CR-50), 100-mg/kg rutin+5-mg/kg cisplatin (CR-100) group. Results: CP group administered cisplatin had significantly increased blood, serum, and cardiac tissue malondialdehyde (MDA), interleukin 1 beta (IL-1β), tumor necrosis factor alpha (TNF-α), troponin I, creatine kinase (CK), and CK-MB levels compared to the HE group, whereas there was a significant decrease in the total glutathione (tGSH) levels. Rutin was observed to prevent the increase in MDA, IL-1β, TNF-α, troponin I, CK, and CK-MB levels as well as prevent the decrease in tGSH levels more significantly when administered at a 100-mg/kg dose than at a 50-mg/kg dose. Histopathologically, cardiac necrosis, dilated/congested blood vessels, hemorrhage, polymorphonuclear leukocyte, edema, and cells with pyknotic nuclei were observed in the CP group. Rutin was shown to prevent cisplatin-induced cardiac damage more effectively when used at a100-mg/kg dose than at a 50-mg/kg dose. Conclusion: These results suggest that rutin is useful for preventing cisplatin-related cardiovascular damage.

Introduction: We compared the side effects of ketamine and thiopental used alone and of a ketamine/thiopental combination dose on the brain, heart, and bronchial tissues of rats. Material and methods: Three groups received intraperitoneal injections of 30 mg/kg ketamine (K-30); 15 mg/kg thiopental (T-15); or of both in combination (KTSA). These doses were doubled in another set of study groups (K-60, T-30, and KTA groups, respectively). Optimal anesthesia duration was examined in all groups. Results: Anesthesia did not occur with 30 mg/kg ketamine or 15 mg/kg thiopental. However, when used alone ketamine and thiopental led to oxidative stress in the striatum, heart, and bronchial tissues. Conversely, combined administration of anesthetics and subanesthetic doses were found not to create oxidative stress in any of these areas. The highest level of adrenaline in blood samples collected from the tail veins was measured in the KTA-60, and the lowest amount in the T-30. Creatine kinase activity was highest in the KTA-60 group (p < 0.001). When we compared for all 5 groups to untreated control group; the creatine kinase-MB activities were significiantly different in K-30, T-15 and T-30 (p < 0.001). Conclusions: The studied doses of ketamine led to oxidative stress by increasing the amount of adrenaline. Thiopental increased oxidative stress with decreases in adrenaline. A longer anesthetic effect with minimal adverse events may be achieved by ketamine and thiopental in combination.

Background. Oxidative stress and interleukin-1 beta (IL-1β) have been reported to play a role in the pathogenesis of nephrotoxicity induced by cisplatin. Objectives. The objective of this study was to investigate the effect of anakinra, which is an IL-1β receptor antagonist, on cisplatin-induced nephrotoxicity in rats, through biochemical, gene expression and histopathological analyses. Material and methods. The study was designed with 4 groups. For 1 week, the control group (C) and the cisplatin (Cis) group received distilled water, while the cisplatin + anakinra 50 (Cis + ANA50) group and the cisplatin + anakinra 100 (Cis + ANA100) group were intraperitoneally administered 50 mg/kg and 100 mg/kg of anakinra, respectively. The Cis, Cis + ANA50 and Cis + ANA100 groups were intraperitoneally injected with a 2.5 mg/kg dose of cisplatin for 7 days. After sacrifice, the kidney tissue of each rat was extracted for the assessment of the malondialdehyde (MDA) and total glutathione (tGSH) levels, and for gene expression analyses of IL-1β. The kidney tissues were histopathologically evaluated. Statistical analyses of the data were performed using one-way analysis of variance (ANOVA). Results. The administration of cisplatin (the Cis group) yielded a higher level of MDA (4.75 ±0.25 nmol/mL; p < 0.001) and lower levels of tGSH (1.80 ±0.35 mg/L; p < 0.001) compared to other groups. Cisplatin also increased IL-1β gene expression (6.33 ±0.27 gene expression levels; p < 0.001) compared to other groups. The impact of anakinra on the MDA and tGSH levels, and on IL-1β gene expression induced by cisplatin was observed as a reversal of these findings (p < 0.05). Anakinra better prevented an increase of the levels of MDA and IL-1β at a dose of 100 mg/kg compared to a 50 mg/kg dose. Conclusions. Anakinra prevents oxidative kidney damage induced by cisplatin in a dose-dependent manner. This result suggests that anakinra may be useful in the treatment of cisplatin-induced kidney damage.

Etoricoxib features antioxidant and anti-inflammatory properties concomitantly, suggesting that it may be beneficial in testicular ischemia reperfusion (I/R) damage. Our aim is to investigate the effects of etoricoxib on testicular I/R damage induced with torsion-detorsion (TD). The etoricoxib + torsion-detorsion (ETD) groups of animals were given etoricoxib in 50 and 100 mg/kg of body weight (ETD- 50 and ETD-100), while the testes torsion-detorsion (TTD) and sham operation rat group (SOG) animals were given single oral doses of distilled water as a solvent. TTD, ETD-50 and ETD-100 groups were subjected to 720o degrees torsion for four hours, and detorsion for four hours. The SOG group was not subjected to this procedure. Biochemical, gene expression and histopathological analyses were carried out on the testicular tissues. The levels of malondialdehyde (MDA), myeloperoxidase (MPO), interleukin-1 beta (IL-1β) and tumor necrosis factor alpha (TNF-α) were significantly higher, and the levels of total glutathione (tGSH) and glutathione reductase (GSHRd) were significantly lower in the TTD group, compared to the ETD-50, ETD-100 and SOG groups. Etoricoxib at a dose of 100 mg/kg better prevented I/R damage than the 50 mg/kg dose. Etoricoxib may be useful in clinical practice in the reduction of I/ R damage on testes caused by torsion-detorsion.
