Yilmaz, Mehmet; Suleyman, Bahadir; Mammadov, Renad; Altuner, Durdu; Bulut, Seval; Suleyman, Halis

Background and Objectives: The purpose of the study was to investigate the role of adrenaline (ADR), noradrenaline (NDR), and cortisol in the pathogenesis of the analgesic potency, duration, and epilepsy-like toxic effect of meperidine. Materials and Methods: The experimental animals were separated into 11 groups of six rats. In the meperidine (MPD) and metyrosine + meperidine (MMPD) groups, paw pain thresholds were measured before and after the treatment between the first and sixth hours (one hour apart). In addition, ADR and NDR analyses were performed before and after the treatment, between the first and fourth hours (one hour apart). For the epilepsy experiment, caffeine, caffeine + meperidine, and caffeine + meperidine + metyrapone groups were created, and the treatment was applied for 1 day or 7 days. Groups were created in which caffeine was used at both 150 mg/kg and 300 mg/kg. Epileptic seizures were observed in epilepsy groups, latent periods were determined, and serum cortisol levels were measured. Results: In the MPD group, pain thresholds increased only at the first and second hours compared to pre-treatment, while ADR increased at the third hour, leading to a decrease in pain thresholds. In the MMPD group, the increase in paw pain thresholds at 1 and 6 h was accompanied by a decrease in ADR and NDR. In the caffeine (150 mg/kg) + meperidine group, 1-day treatment did not cause epileptic seizures, while seizures were observed and cortisol levels increased in the group in which treatment continued for 7 days. When cortisol levels were compared between the group in which caffeine (300 mg/kg) + meperidine + metyrapone was used for 7 days and the animals receiving caffeine (300 mg/kg) + metyrapone for 7 days, it was found that cortisol levels decreased and the latent period decreased. Conclusions: The current study showed that if serum ADR and cortisol levels are kept at normal levels, a longer-lasting and stronger analgesic effect can be achieved with meperidine, and epileptic seizures can be prevented.

Dinc, Kemal; Kiremitli, Sevil; Suleyman, Bahadir; Mammadov, Renad; Yavuzer, Bulent; Bulut, Seval; Yazici, Gulce N.; Coban, Taha A.; Altuner, Durdu; Suleyman, Halis

Purpose: To investigate the protective effect of pycnogenol (PYC) against ischemia/reperfusion (I/R) ovarian damage induced by experimental ovarian torsion in rats using biochemical and histopathological methods. Methods: Four groups of rats (n = 6) were randomly assigned and designated as follows - SG: sham group, PCG: pycnogenol (40 mg/kg) group, IRG: ovarian ischemia-reperfusion group, and PIR: pycnogenol (40 mg/kg) + ovarian ischemia-reperfusion group. In IRG and PYC treatment groups, ischemia was induced in the right ovary for two hours with vascular clips. The ovary was reperfused for two hours after ischemia was induced. Then, the levels of malondialdehyde (MDA), total glutathione (tGSH), nuclear factor-kappa B (NF-κB), tumor necrosis factor-alpha (TNF-α) and interleukin-1β (IL-1β) in the ovarian tissues of rats were examined. Ovarian tissues were also examined histopathologically and follicle counts were performed. Results: Histopathological examination revealed that the developing follicles in ovary of I/R-induced group showed both morphological and cellular degeneration and vascular pathology. The PYC treatment group showed less ovarian damage, edema and vascular pathology than I/R-induced group (p < 0.05). The MDA, NF-κB, TNF-α and IL-1β levels in I/R-induced group were significantly higher than in SG and tGSH levels were significantly reduced by I/R damage (p < 0.05). Pycnogenol treatment reversed these biochemical indices as well as I/R-induced histopathological changes. Conclusion: Pycnogenol has a protective effect in rat ovaries by alleviating I/R-induced changes in biochemical parameters and histological disruptions. Further studies, including those in humans, to determine the effect of PYC on ovarian injuries would be required.

Effect of Thymoquinone on Diclofenac-Induced Liver Injury

Ekinci, Bilge; Altuner, Durdu; Suleyman, Bahadir; Mammadov, Renad; Bulut, Seval; Suleyman, Zeynep; Gul, Mehmet A.; Ergul, Cetin; Suleyman, Halis

Background and Objective: Thymoquinone (TQ) is an active phenolic compound obtained from Nigella sativa L. It has anti-inflammatory and antioxidant activity by inhibiting the overproduction of certain inflammatory molecules and lipid peroxidation. TQ also has a hepatoprotective effect. This study, it was aimed to biochemically investigate the effect of TQ on diclofenac (DC)-induced liver damage in rats. Materials and Methods: The animals were divided into healthy control (HG), only diclofenac (DG), only thymoquinone (TQG) and diclofenac+thymoquinone (DTQG) groups. The DC was injected intraperitoneally at a dose of 25 mg kg(-1). The TQ was administered orally to the stomach at a dose of 20 mg kg(-1). This procedure was repeated once a day for 7 days. Results: Biochemical test results showed that TQ significantly prevented the increase in oxidant parameters and the decrease in antioxidants in liver tissue, which is formed by DC. Also, DC and TQ inhibited the increase of proinflammatory cytokines in liver tissue. The TQ also prevented the decrease of COX-1 activity by DC and increased the inhibition of COX-2. The TQ also significantly suppressed the elevation of liver function markers such as Alanine Aminotransferase (ALT) and Aspartate Aminotransferase (AST) with DC. Conclusion: These findings indicated that the hepatotoxic effect of DC was due to oxidative stress and inhibition of cytoprotective structural COX-1 enzyme. It suggested that TQ may be useful in the treatment of DC-related liver injury.

Is Benidipine Effective in Preventing Gastric Ischemia/Reperfusion Injury?

Bilici, Sami; Tas, Hakan Gokalp; Mamedov, Renad; Suleyman, Bahadir; Kuyrukluyi, Ufuk; Yavuzer, Bulent; Bulut, Seval; Coban, Abdulkadir; Suleyman, Halis

Background and Objective: The preventive effect of benidipine on organs and tissues against oxidative and proinflammatory cytokine damage, whose production is elevated by I/R, has been demonstrated. The current study noted that benidipine affects the biochemical and histopathological effects of I/R therapy on gastric injury in rats. Materials and Methods: A total of 18 rats were divided into either a Sham operation group (SG), gastric ischemia/reperfusion (GIR) group or benidipine+gastric ischemia/reperfusion (BGIR) groups. Biochemical measures and histological exams were done in all three groups after I/R intervention. Results: The MDA, TNF-alpha, IL-1 beta and COX2 levels were significantly higher (p<0.001) in GIR compared to SG and BGIR, although tGSH and COX1 were significantly lower (p<0.001). The SG group was histopathologically normal. The surface epithelium broke off in spots, causing mild degeneration, the gland recesses were decreased, the gland neck areas were wide, the base areas were significantly oedematous and the blood capillaries showed moderate dilatation and congestion in the GIR group. Infiltration of polymorphonuclear cells was seen in the connective tissue surrounding the arteries and close to the gland bases. The BGIR group's histopathological results were not normal, although they were milder than the GIR group's. Conclusion: The results discovered that benidipine shifted the oxidant/antioxidant balance in I/R damage in favour of antioxidants, as well as the COX-1/COX-2 equilibrium in favour of COX-2, which is cytoprotective. It appears that administering benidipine for antioxidant and cytoprotective purposes before surgical operations that are likely to cause I/R damage may be advantageous.

Sahin, Nurullah; Delibas, Ilhan Bahri; Isaoglu, Unal; Suleyman, Bahadir; Yazici, Gulce Naz; Coban, Taha Abdulkadir; Uzel, Kemine; Suleyman, Halis; Arslan, Veysel

Background: Increased oxidants and proinflammatory cytokines play a role in the pathogenesis of ovarian ischemia-reperfusion (OIR) injury and related infertility. Carvacrol has antioxidant, antibacterial and anti-inflammatory properties. Methods: Protective effect of carvacrol against ischemia-reperfusion (IR)-related ovarian damage and infertility was investigated. IR process were applied to the ovaries of rats, which were divided into the following groups (n = 12): OIR, IR +50 mg/kg carvacrol (IRC-50), IR +100 mg/kg carvacrol (IRC-100) and sham group (SG). After the reperfusion process, six rats from each group were killed and the removed ovaries were examined biochemically and histopathologically. The remaining animals were kept two months with mature male rats to reproduce. Results: At a dose of 50 mg/kg, carvacrol suppressed the oxidant parameter increase and antioxidant decrease caused by IR in ovarian tissue. At a dose of 100 mg/kg, carvacrol antagonized both oxidant and proinflammatory cytokine increase and antioxidant decrease. Histopathologically, severe degeneration of follicles in the ovaries of the OIR group, necrotic cell accumulations, hemorrhage in the corpus luteum, edema in the interstitial tissue, polymorphous nuclear leukocyte (PNL) infiltration, and congestion and dilation of blood vessels were detected. Inflammatory symptoms such as edema in the ovarian tissue, congested dilated blood vessels and PNL infiltration were observed at a dose of 50 mg/kg of carvacrol, but these histopathological findings were not observed at a dose of 100 mg/kg. Conclusion: A dose of 100 mg/kg carvacrol, which eliminated inflammatory damage, significantly prevented the development of IR-induced infertility. Carvacrol may be beneficial in the treatment of IR-related ovarian damage and infertility. Copyright:

EFFECT OF NIMESULIDE AGAINST INDOMETHACININDUCED OXIDATIVE LIVER DAMAGE IN RATS

Yilmaz, Canan T.U.Z.; Suleyman, Bahadir; Mammadov, Renad; Ozcicek, Adalet; Bulut, Seval; Yazici, Gulce N.A.Z.; Gulapoglu, Mine; Suleyman, Halis

Indomethacin is used in the treatment of ankylosing spondylitis, osteoarthritis, tendinitis, and other inflammations. However, it has been reported indomethacin has nephrotoxic, gastrotoxic, and hepatotoxic effects. The aim of this study is to investigate the effect of nimesulide on indomethacin-induced liver damage in rats biochemically and histopathologically. Experimental animals (21 rats) were divided into 3 groups: the healthy (HG) group receiving distilled water as a solvent, the 25 mg/kg indomethacin administered (IDO) group, and the 50 mg/kg nimesulide + 25 mg/kg indomethacin administered (NIDO) group. Malondialdehyde (MDA), total glutathione (GSH) levels and cyclooxygenase-1 (COX-1), cyclooxygenase (COX-2) enzyme activities were determined from liver tissues. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels were also measured in serum. The data obtained in the IDO group were compared with the data obtained from the NIDO and MDA and decreased tGSH levels with indomethacin HG groups. Increased application in liver tissue approached the HG group with nimesulide application, and the difference was found to be insignificant. Nimesulide prevented the decrease in COX-1 and COX-2 levels observed with indomethacin administration. Again, the increase in ALT and AST levels, which increased with indomethacin administration, was prevented by nimesulide. As a result of the histopathological evaluation, it was observed that nimesulide reduced the damage caused by indomethacin. Nimesulide at 50 mg/kg prevented oxidative liver damage induced with indomethacin at 25 mg/kg. Our results suggest that nimesulide may be useful in the treatment of hepatotoxicity and COX-1 inhibition side effects without suppressing analgesic and anti-inflammatory activity due to indomethacin use.

The aim of the study to determine the effects of thiamine pyrophosphate (TPP) and tocilizumab (TCZ) either alone or in combination on ethanol-induced neuropathic pain and peripheral neuropathy in rat nerve tissues, biochemically and histopathologically. Albino Wistar male rats were divided into five groups as the healthy (HG), ethanol alone (EtOH), TPP + ethanol (TPE), TCZ + ethanol (TCE), and TPP + TCZ + ethanol (TPTE) groups. TPE (n = 6), TCE (n = 6) and TPTE (n = 6) groups were injected with 20 mg/kg TPP, 8 mg/kg TCZ, and 20 mg/kg TPP+8 mg/kg TCZ intraperitoneally (respectively). Distilled water were given the HG (n = 6) and EtOH (n = 6) groups. One hour after the drugs and distilled water were administered, 35% ethanol was administered orally by lavage at a 5 g/kg dose to the TPE, TCE, TPTE, and EtOH groups. Paw pain thresholds of animals were measured using Basile Algesimeter. TPTE, which suppressed the decrease of the pain threshold with ethanol more significantly than TPP and TCZ. The best inhibitor of MDA increase and inhibit the reduction of tGSH by etanol was listed as TPTE > TPE > TCZ. The potency of suppressing the increase in cytokine production with ethanol was listed as TPTE > TCE > TPE. In conclusion, the results indicate that TPP+TCZ may be more beneficial than TPP and TCZ alone in treating ethanol-related neuro-pathic pain.

Ekinci, Bilge; Suleyman, Bahadir; Mammadov, Renad; Bulut, Seval; Ozcicek, Adalet; Ergul, Cetin; Gulaboglu, Mine; Hayme, Serhat; Suleyman, Halis

Cyclooxygenase 2 (COX-2) is responsible for the therapeutic effects of indomethacin, while inhibition of the COX-1 enzyme and oxidative stress are responsible for its gastro-toxic effects. It has been reported that pycnogenol increases the expression of COX-1, suppresses the expression rate of COX-2 and oxidative stress. Our aim in this study is to investigate the antiinflammatory activities of indomethacin, pycnogenol, and their combination (PI) in rats and to examine their effects on stomach tissue. In the study, anti-inflammatory activity was investigated in carrageenan-induced inflammatory paw edema in albino Wistar male rats. Effects on stomach tissue were performed by applying the previous method. PI, indomethacin and pycnogenol were the best suppressors of carrageenan inflammation and oxidative stress in paw tissue, respectively. While the groups with the lowest COX-1 activity in paw tissue were IC, PIC and PC, respectively, PIC, IC and PC were the ones that best inhibited the increase in COX-2 activity. Pycnogenol inhibited the increase of malondialdehyde, the decrease of total glutathione and COX-1 in the stomach, and significantly suppressed the formation of indomethacin ulcers. Our experimental results showed that pycnogenol reduced the toxic effect of indomethacin on the stomach and increased anti-inflammatory activity. This beneficial interaction of pycnogenol and indomethacin suggests that PI will provide superior success in the treatment of inflammatory diseases.

The Effect of Cerebrolysin on Ischemia Reperfusion-Induced Oxidative and Inflammatory Ovarian Damage in Rats

Ozer, Mehmet; Ince, Sefa; Suleyman, Bahadir; Mammadov, Renad; Yavuzer, Bulent; Bulut, Seval; Akbas, Nergis; Mokhtare, Behzad; Mendil, Ali S.; Dinc, Kemal

SUMMARY. Ovarian ischemia-reperfusion (I/R) injury develops due to oxidative stress and inflammation that occurs when tissue is deprived of oxygen. Ovarian I/R damage also causes infertility. The aim of this study is to see the effect of ovarian I/R damage on reproductive functions and to determine the protection of antioxidant and anti-inflammatory cerebrolysin in possible damage. Albino Wistar-type female rats were divided into sham operation (SG), ischemia-reperfusion (IR) and cerebrolysin+ischemia-reperfusion (CIR) groups. After I/R application with cerebrolysin, the ovaries of six animals from each group were taken and biochemical and histopathological examinations were performed. Six animals from each remaining group were left to breed. Our test results showed that cerebrolysin decreased I/R-related oxidant, proinflammatory cytokine and tissue damage and increased antioxidant levels. In addition, cerebrolysis significantly inhibited the delay of the maternity period. These findings indicate that cerebrolysin may be helpful in the treatment of I/R-related ovarian damage and infertility.

Emir, Izzet; Altuner, Durdu; Gulaboglu, Mine; Coban, Taha Abdulkadir; Suleyman, Bahadir; Mammadov, Renad; Ozkaraca, Mustafa; Suleyman, Zeynep; Karakurt, Tulay Ceren Olmezturk; Suleyman, Halis

The aim of our study was to biochemically and histopathologically investigate the protective effect of adenosine triphosphate (ATP), benidipine, sugammadex and (ATP + benidipine + sugammadex) ABS against cardiac damage induced by abdominal clamping and unclamping (ACU) procedure in rats. Rats utilized were divided into six groups as Healthy (HG), Abdominal ACU (AACC), ATP + AACU (ATPG), Benidipine + AACU (BNDG), Sugammadex + AACU (SDXG), ABS + AACU (ABSG). One hour before anesthesia, ATP (25 mg/kg; ip) was administered to the ATPG group, Benidipine (4 mg/kg orally) was administered to the BNDG group, Sugammadex (4 mg/kg ip) was administered to the SDXG and ABS was administered with this specified dose and method to the ABSG group. Distilled water as a solvent was injected to AACC and HG groups. One hour after, laparotomy was performed to the rats, and the abdominal cavity was reached. Ischemia was provided for one hour and then reperfusion for 2 h placing an atraumatic clamp on the suprarenal abdominal aorta of the animals in all groups (except from the HG). The heart tissues extracted from the killed animals were examined biochemically and histopathologically. ATP and benidipine suppressed the increase of oxidant and proinflammatory cytokines better than sugammadex. The ABSG group was found to be almost the same as the healthy group biochemically and histopathologically (p > 0.05). ABS drug form alone was possible to be much more beneficial rather than ATP, benidipine and sugammedex in the treatment of cardiac damage induced by AACU procedure.

THE EFFECT OF CARVACROL UPON EXPERIMENTALLY INDUCED DIABETIC NEUROPATHY AND NEUROPATHIC PAIN IN RATS

Ekinci, Bilge; Suleyman, Bahadir; Mammadov, Renad; Gezer, Arzu; Mendil, Ali Sefa; Akbas, Nergis; Bulut, Seval; Dal, Cagatay Nusret; Suleyman, Halis

Diabetic neuropathies are the most frequent complication of diabetes. While numerous metabolic pathways are disrupted in diabetic neuropathy, oxidative stress has been indicated as a significant reason for this condition. In this study, the effect of carvacrol, which has antioxidant effects, on experimental diabetic neuropathy and neuropathic pain was investigated. Alloxan was used to induce diabetes in the experiment. Diabetes was created by administering 120 mg/kg of alloxan intraperitoneally (i.p) once a day for 3 days. Rats with a blood glucose concentration above 250 mg/kg in the blood taken from the tail veins at the end of three days were considered diabetic. Rats were categorized under healthy control (HG), alloxan-induced hyperglycemia (AG), and alloxan-induced hyperglycemia + carvacrol-treated (ACG) groups. Carvacrol was i.p injected at 50 mg/kg dose to the ACG (n = 6) group of rats with hyperglycemia. The same volume of distilled water as the solvent was applied in the same way to AG (n = 6) and HG (n = 6) rat groups. This procedure was repeated once a day for three months. Carvacrol showed an anti-hyperglycemic effect in diabetic rats, a protective effect against lowering pain threshold, and analgesic activity in rat paws. Carvacrol prevented the oxidant/antioxidant balance from changing in favor of oxidants. The results supported that carvacrol is an agent against alloxan-induced peripheral diabetic neuropathic pain.

The effects of taxifolin on neuropathy related with hyperglycemia and neuropathic pain in rats: A biochemical and histopathological evaluation

Alay, Murat; Sonmez, Miyase Gulcin; Sakin, Aysegul; Atmaca, Murat; Suleyman, Halis; Yazici, Gulce Naz; Coban, Abdulkadir; Suleyman, Bahadir; Bulut, Seval; Altuner, Durdu

Background. Hyperglycemia can be considered a determining factor in the development of diabetic neuropathy as well as neuropathic pain. There is a relationship between the excessive production of reactive oxygen species (ROS) and the pathogenesis of diabetic neuropathic pain. Taxifolin, on the other hand, is a flavonoid that has been documented to inhibit ROS production. Objectives. To investigate the effects of taxifolin, which has antioxidant and neuroprotective effects, on alloxan-induced hyperglycemia-induced neuropathy and neuropathic pain, biochemically and histopathologically. Materials and methods. The albino Wistar male rats were divided into 3 groups: Healthy group (HG), only alloxan group (AXG) and alloxan+taxifolin group (ATG). Hyperglycemia in animals was caused through intraperitoneal injection of alloxan at a dose of 120 mg/kg. Paw pain thresholds of animals were measured using Basile algesimeter. Sciatic nerve tissues were examined biochemically and histopathologically in order to evaluate neuropathy. Results. Our experimental results revealed that taxifolin significantly prevented the increase of plasma glucose concentration level with alloxan administration, the decrease of the paw pain threshold related to hyperglycemia, the change of oxidant-antioxidant balance in the sciatic nerve tissue in favor of oxidants, and the deterioration of tissue morphology in animals. Conclusions. Our experimental results indicate that taxifolin alleviates alloxan-induced hyperglycemia-related neuropathy and neuropathic pain.

Effect of taxifolin on acrylamide-induced oxidative and proinflammatory lung injury in rats: Biochemical and histopathological studies

Demir, Omer Faruk; Elma, Bekir; Suleyman, Bahadir; Ozkaraca, Mustafa; Mammadov, Renad; Suleyman, Halis; Coban, Taha Abdulkadir; Ozcicek, Fatih

Purpose: To examine the probable beneficial effects of taxifolin against acrylamide damage in lung tissue. Methods: 18 male albino Wistar rats were divided into healthy (HG), acrylamide (AG) and taxifolin + acrylamide (TAG) groups. Once a day for 30 days, acrylamide was orally administered to the AG group (50 mg/kg), while ACL (50 mg/kg) and TAX (20 mg/kg) were orally administered to TAG group. Protein concentration, malondialdehyde (MDA), and total glutathione (tGSH) levels as well as oxidant and antioxidant molecules concentrations of the rat lung tissues were measured. In addition, degree of mononuclear (MN) cell infiltration and bronchial-associated lymphoid tissue (BALT) hyperplasia was evaluated by the degree of hyperplasia (absent, mild, moderate, severe). The histopathological and biochemical data the groups were compared. Results: When compared in terms of MDA levels, it was found that the AG group had high MDA levels, and the TAG group had low MDA levels. (p < 0.001). TAG group was found to have a higher tGSH level than the AG group (p < 0.001). Compared to the AG group, lower TOS and higher TAS levels were obtained in the TAG group (p < 0.001). In addition, when TOS levels of TAG and HG groups were compared, the TOS levels between the two groups were statistically insignificant (p = 0.213). It has been observed that TAX administration prevents the increase in NF-ƘB level. When the NF-ƘB levels of the AG and TAG groups were compared with each other, there was a statistically significant difference (p = 0.001). In the AG group, severe MN cell hyperplasia and BALT hyperplasia were observed histopathologically. It was determined that these findings were alleviated in the TAG group. A histopathologically significant difference was found between AG and TAG groups (p < 0.05). Conclusion: Taxifolin has beneficial effects against lung injury caused by acrylamide, a health-damaging environmental factor. Regular use of taxifolin can be recommended, especially in people who are known to have intense contact with acrylamide. There is a need for research studies on this subject.

Objectives: Oral mucositis is a significant toxicity related to the mammalian target of rapamycin inhibitor everolimus. Oxidative stress and pro-inflammatory cytokines, which contribute to treatment-related mucositis, can betargeted with Hippophae rhamnoides extract (HRE). Herein, we assessed the effects of HRE on everolimus-inducedmucositis in rats. Methods: Eighteen rats were equally divided into healthy, everolimus, and everolimus plus HRE groups. Malondialdehyde (MDA) and total glutathione (tGSH) levels along with interleukin-1 beta (IL-1β) and tumor necrosis factor-alpha(TNF-α) gene expression levels were measured in the tongue and buccal mucosa tissues of all groups, histopathologicalchanges were also evaluated. We tested the significance of variations with one-way variance analysis. We also analyzedthe differences between groups with Kruskal–Wallis test and Mann–Whitney U-test.Results: HRE significantly decreased MDA and increased tGSH levels and reduced IL-1β and TNF-α gene expression inboth tissues administered everolimus (p<0.001 for each). Histological examination revealed that HRE improved epithelial formation and keratinization, disrupted by everolimus, and alleviated everolimus-related mononuclear cell infiltration (p<0.05 for each). Conclusion: In light of these results, HRE may be a promising agent to manage oral mucositis caused by everolimus, given the lack of effective therapeutic options for this type of adverse event.

Protective effect of nimesulide on the external ear damage induced by staphylococcusaureus inoculation in rats

Mammadov, Renad; Suleyman, Bahadir; Çoban, T. Abdulkadir; Salcan, Ismail; Akyüz, Sümeyye; Suleyman, Halis; Erhan, Ertugrul; Dilber, Muhammet

Since the external ear is covered with skin, infections that cause otitis externa are often produced by distinct skin flora in several different areas. Staphylococcus aureus (S. aureus) reproduces the most out of all bacteria isolated from external auditory canal skin culture samples. Proinflammatory cytokines are the main components responsible for tissue damage pathogenesis due to S. aureus. Nimesulide is a nonsteroidal anti-inflammatory drug that selectively inhibits cyclooxygenase-2 and also demonstrates antioxidant properties. The present study aimed to examine the antibacterial activity of nimesulide against S. aureus and to compare its effectiveness on otitis externa induced by S. aureus in male albino Wistar rats with that of cefazolin. The antimicrobial activity testing was conducted using the Kirby-Bauer disk diffusion method as described by the European Committee on Antimicrobial Susceptibility Testing. To induce otitis externa, we applied 0.5 ml of S. aureus to the ear skin using a hypodermic syringe (S. aureus strain TACK 25923 was dispersed on the 900x10-6 concentration of colony forming units per ml). Nimesulide and cefazolin were administered orally at a dose of 50 mg/kg. The antibacterial activity of cefazolin when used in equal doses (50 µg/ml) was more powerful against S. aureus than nimesulide. However, nimesulide reduced the macroscopic findings (such as oedema and redness) induced by S. aureus better than cefazolin. Additionally, nimesulide inhibited the increase of oxidant and proinflammatory indicators related to S. aureus, while cefazolin was able to inhibit only the increase in proinflammatory indicators. Our results showed that nimesulide was superior to cefazolin in terms of reducing the external ear damage caused by S. aureus. Therefore, nimesulide monotherapy or a combination of nimesulide and cefazolin therapy may be beneficial in the treatment of bacterial otitis externa.

Objective: Psychological stress is an important factor triggering depression and anxiety. Infertility is known to cause stress; however, it is not clearly known whether stress causes infertility as well. In addition, there are different opinions accounting for the relation of stress-induced oxidative stress to infertility and intrauterine growth restriction. The aim of the study is to examine the effect of sertraline, diazepam and melatonin on the infertility, intrauterine growth restriction and oxidative stress that can be caused by forced immobilization stress management (FISM) in female rats. Materials and methods: Wistar rats were grouped as healthy rats (HG) applied distilled water, stress treated control group (SC), and 20 mg/kg sertraline + stress (SS), 2 mg/kg diazepam + stress (DS) and 10 mg/kg melatonin + stress (MS) treated rats. The medicines were administered orally once a day for 30 days. At the end of this period, oxidant/antioxidant parameters were measured through the blood samples collected from the tail veins of all rats. Then the rats were kept in a suitable environment for 2 months for breeding. Results: FISM caused oxidative stress in blood serum of animals, infertility and intrauterine growth restriction (decrease in birth weight of the baby). Best medicines to suppress FISM-related oxidative stress are melatonin > diazepam > sertraline respectively, while sertraline > diazepam > melatonin were most successful in terms of preventing infertility. The best medicines preventing the FISM-caused intrauterine growth restriction were found to be melatonin > diazepam > sertraline, respectively. Conclusion: FISM causes oxidative stress in animals. Oxidative stress is understood to affect the intrauterine growth negatively although it is not a major component in the pathogenesis of infertility. While melatonin is only effective in preventing the oxidative stress-related intrauterine growth restriction, antidepressants and anxiolytic treatment were found to be helpful in preventing both infertility and intrauterine growth restriction.

Background: Acrolein is a reactive aldehyde that forms during burning of wood and other fuels. It is also a product of lipid peroxidation (LPO) reactions and is present in cigarette smoke. Acrolein is known to cause oxidative stress and inflammatory nerve tissue damage. Lutein is a tetraterpenoid molecule with antioxidant and anti-inflammatory properties. There appear to be no studies on the effect of lutein on vestibulocochlear nerve damage induced by acrolein. The aim of this study was to investigate the effect of lutein on vestibulocochlear nerve damage induced by acrolein in rats using biochemical and histopathological methods. Methods: The rats were divided into three groups (n = 6, for each group) a healthy control group (HG), an acrolein (ACR) group and a lutein and acrolein (LACR) group. In the LACR group, lutein was administered (1 mg/kg) via oral gavage. The ACR and HG groups received saline via oral gavage. Then, 1 h after the administration of lutein and saline, the LACR and ACR groups were treated with 3 mg/kg of acrolein via oral gavage. This procedure was repeated once a day for 30 days. Results: The results of biochemical experiments showed that in the vestibulocochlear nerve tissues of the animals treated with acrolein, the levels of malondialdehyde, total oxidants, nuclear factor kappa b, tumor necrosis factor alpha and interleukin 1 beta significantly increased, whereas the levels of total glutathione and total antioxidants decreased as compared to those in the HG and LACR groups. In addition, severe histopathological damage was observed in vestibulocochlear nerve tissue of the acrolein group, whereas this damage was alleviated in the lutein group. Conclusion: Lutein protected vestibulocochlear nerve tissue from acrolein-associated oxidative and proinflammatory damage. This suggests that lutein might be useful in preventing or treating acrolein-induced ototoxicity.

The effect of taxifolin on high-dose-cisplatin-induced oxidative liver injury in rats

Kurt, Nezahat; Türkeri, Ozge Nur; Suleyman, Bahadir; Bakan, Nuri

Background. Cisplatin is a non-specific platinum-based (derivative) chemotherapeutic agent that causes an increase in free radicals activity in the liver. Antioxidant activity of taxifolin has been demonstrated previously, and it has been reported that taxifolin inhibits the hydroxyl, radical in experimental studies. Objectives. No studies were found in the current literature examining the protective effect of taxifolin on cisplatin-induced oxidative liver damage. We aimed to determine the protective effect of taxifolin on cisplatin-induced hepatotoxicity in an experimental study. Materials and methods. In total, 18 albino Wistar male rats were assigned into 3 groups: healthy controls (HC group), 5 mg/kg of cisplatin administered for 8 days (CIS group) and 50 mg/kg of taxifolin + 5 mg/kg of cisplatin administered for 8 days (TCG group). Malondialdehyde (MDA), total glutathione (tGSH), total oxidant (TOS), and total antioxidant (TAS) capacity levels were measured in the extracted liver tissue. Results. Liver tissue MDA and TOS levels were significantly higher in the CIS group. In contrast, tGSH and TAS levels were significantly lower in the CIS group, administered cisplatin alone (p < 0.001), compared to other groups. In the TCG group, administered cisplatin + taxifolin, MDA and TOS levels were significantly lower, whereas tGSH and TAS levels were significantly higher than in the CIS group (p < 0.001). Conclusions. These results suggest that taxifolin may be useful in preventing cisplatin-related liver injury.
