Impact of chitosan–thyme and chitosan–rosemary essential oil coatings on biochemical, oxidative, and volatile profiles of Turkish fermented sausage ‘sucuk’ during ripening

Demirok Soncu, Eda; Özdemir Orhan, Necla; Küçükkaya, Seren; Arslan Solmaz, Betül; Soyer, Ayla

In the current study, the effects of chitosan coatings combined with thyme or rosemary essential oils (EOs), applied as natural antifungal agents to prevent superficial fungal growth on the surface of sucuk, were investigated throughout the ripening period in terms of lipolytic, proteolytic, and oxidative changes, as well as volatile composition and sensory properties. Six sucuk groups were produced by covering the casing surface on day 4 of production with distilled water (DW), acetic acid (AA), potassium sorbate (PS), chitosan (C), chitosan+thyme EO (CT), or chitosan+rosemary EO (CR). Superficial fungal growth was reduced approximately 4-log units in chitosan+EO treatments compared to the control. The lowest fungal count (log CFU/cm2) was observed in PS-treated sucuk (0.60), followed by CT (2.06) and CR (2.09), whereas this value was 5.95 in the DW group (p<0.05). Coatings containing EOs retarded lipid oxidation as well as free fatty acid (FFA) formation, as the lowest values were recorded in the CT and CR groups, with FFA contents of 4.75% and 4.96% and thiobarbituric acid reactive substances values of 0.60 and 0.60 mg MA/kg, respectively. The lowest sulfhydryl reduction (32%) was also observed in the CT and CR groups, compared to the PS, in which the reduction was 39% (p<0.05). The fatty acid profile and sensory properties of the sausages were not adversely affected by the superficial chitosan–EO coatings. A total of 46 and 67 volatile compounds were identified in sausages and casings, respectively. The major volatile constituents of sucuk in the CT and CR groups were acids, terpenes, sulfur compounds, and aldehydes, accounting for 10.41–17.07%, 58.89–66.88%, 5.36–6.70%, and 12.68–12.85% of the total volatile composition, respectively. In conclusion, CT or CR coating can be considered an alternative antifungal approach to synthetic treatments for preventing superficial fungal growth without adversely affecting the characteristic quality properties of sucuk.

Ertugay, Mustafa Fatih; Ozdokur, Kemal Volkan; Ertugay, Neşe

In this study, it was aimed to determine the traffic and agricultural activity-induced heavy metal including arsenic (As), cadmium (Cd), cobalt (Co), chromium (Cr) and lead (Pb) pollution of sugar beet plants (Beta Vulgaris) grown along the Erzincan-Erzurum D100-25 highway and to reveal the possible change in the amount of heavy metals in sugar beet plants depending on distance to the highway. For this purpose, sugar beet plants were collected from 5 points of 10, 20, 30, 40 and 50 meters away from the highway and heavy metal levels of the samples were determined by inductively coupled plasma mass spectrometry (ICP-MS). The results showed that sugar beet plants were contaminated with various heavy metals such as Cr (585.5-2687.8 µg kg-1) > Co (143.4-500.7 µg kg-1) > Pb (52.1-476.7 µg kg-1) > As (9.1-112.4 µg kg-1) > Cd (4.4-65.2 µg kg-1) depending on the distance from the highway. It was determined that heavy metal pollution levels in sugar beet plants decreased with distance from the motorway and there was a high correlation (As: 0.977; Cd: 0.944; Co: 0.995; Cr: 0.989 and Pb: 0.999) between distance and heavy metal content.

The assessment of rice cultivars widely produced in Turkey in terms of arsenic, cadmium, aluminum and copper: effect of soaking methods and household cooking

Özdokur, Kemal Volkan; Özdokur, Seyhan; Güneş, Abdulaziz; Ertugay, Neşe; Ertugay, Mustafa Fatih

Industrialization-driven heavy metal contamination is a growing concern, as residues are increasingly found in foods and tap water due to soil absorption by plants. This study aimed to assess the amount of heavy metals including arsenic, cadmium, aluminum, and copper in widely produced domestic rice cultivars in Türkiye, and to evaluate the effect of pretreatments and domestic cooking on the concentration of heavy metals. Six different rice cultivars were analyzed using inductively coupled plasma-mass spectrometry to determine the levels of heavy metals. Among these six cultivars, the highest heavy metal contamination and also the most consumed was selected to evaluate the effect of soaking processes accompanied by acetic acid (0.25, 0.50, 0.75, and 1%) and salt (1%) at different soaking times (2, 4, 8, and 12 h) and domestic cooking. The concentrations of heavy metals in the primary rice cultivar with the highest contamination were 64.9, 1.3, 2476.7 and 3185.2 μgkg−1 dry weight, respectively. The results showed that soaking in salt (1%) and acetic acid (1%) before cooking was the most effective treatment for removing heavy metals (As = 57.8%, Cd = 100%, Al = 98%, and Cu = 85.7%) at a soaking time of 12 h.

Effect of supercritical melocan (Smilax excelsa L.) leaf extract on quality attributes of beef meatballs during cold storage

Selbeş, Esra Bostancı; Şahin, Özlem; Vural, Halil

This study investigated the effects of melocan leaf extract (ME), obtained by supercritical carbon dioxide (Sc-CO2) extraction, on the quality characteristics of beef meatballs during cold storage (0, 3, 6, and 9 days). ME was incorporated at three levels (0.1 %, 0.5 %, 1 % – marked as ME0.1, ME0.5 and ME1). The results showed that the use of ME significantly delayed lipid oxidation in beef meatballs during cold storage (p < 0.05). While the thiobarbituric acid reactive substances (TBARS) value reached 2.46 mg·kg-1 malonaldehyde (MA) in the control group, the meatballs with no ME content, the TBARS values of ME0.1, ME0.5 and ME1 reached 1.39, 0.92, and 1.00 mg·kg-1 MA, respectively. As the extract content increased, the lightness L* and redness a* values of the meatballs decreased (p < 0.05). ME effectively inhibited lipid oxidation and increased the antioxidant capacity in beef meatballs, without significantly affecting total mesophilic aerobic bacteria and total mould and yeast counts (p > 0.05). In terms of sensory evaluation scores, the group containing 0.1 % ME was the closest to the control group, and since it considerably prevented lipid oxidation even at this level, the use of 0.1 % ME in beef meatballs is recommended.

Isolation and Characterization of Lytic Bacteriophages Specific to Foodborne Pathogenic Pseudomonas aeruginosa and Use as a Biopreservative in Chicken Meat

Yilmaz, Nazife; Yildirim, Zeliha; Sahin, Tuba Sakin; Karaoğlan, Mert

In this study, bacteriophages specific to Pseudomonas aeruginosa strains were isolated from wastewaters, and their potential to be used as a bioprotective in chicken meat was investigated. Then, among the 35 isolated bacteriophages, two phages (vB_PaeS-K10 and vB_PaeS-K13) with a wide host cell range and strong clear plaque formation were selected and their morphological, physical–chemical, and genetic properties and their inhibitory effects against the host cell in chicken meat were determined. As a consequence of the examination by TEM (transmission electron microscopy), vB_PaeS-K10 and vB_PaeS-K13 phages belong to the family Siphoviridae of the order Caudovirales. The genome size, multiplicity of infection value, latent period, burst time, and burst size of vB_PaeS-K10 phage were 54.20 kb, 10, 75 min, 30 min, and 34.86, respectively, whereas those of vB_PaeS-K13 phage were 44.06 kb, 0.01, 20 min, 35 min, and 35.95. Both of them were resistant to high temperature and entirely active between pH 3 and 12, and the rate of absorption of the phages against their host cells increased with Ca2+ ions. Lytic activity test revealed that both phages reduced host cell density in LB broth at 4 °C and 37 °C. Moreover, vB_PaeS-K10 and vB_PaeS-K13 phages lethally infect P. aeruginosa at 4 °C in chicken meat, and this situation is the evidence of their potential to be utilized as bioprotective in chicken meat.

Effects of tumbling and ultrasound-assisted curing on quality of smoked beef tongue

    Şahin, Özlem; Vural, Halil

Smoked beef tongue (SBT) is a meat product obtained by cooking and smoking brined beef tongue. This study aimed to reduce the curing time of SBT and evaluate the effects of different curing methods on product quality during 90- day storage period. Ultrasound-assisted (UAC), intermittent ultrasound-assisted (IUC), tumbling (TAC), and vacuum tumbling (VTC) methods were applied, with traditional curing (TC) used as the control. According to ANOVA results, the highest redness (a*) value was observed in the TC group (14.98), but there was no significant difference compared to UAC, IUC, and VTC (p > 0.05). Curing methods had no significant effect on sensory properties (p > 0.05). The highest salt content was found in TC (4.23%), while the lowest was in IUC (1.61%), resulting in a 41–62% reduction depending on the method (p < 0.05). The highest water activity (0.960) and water-holding capacity (0.46) were observed in IUC. Weight yield ranged from 60.73% (VTC) to 65.77% (TC). The new methods also resulted in higher levels of sulfur compounds and aromatic hydrocarbons, while TC had higher levels of Maillard reaction products such as furans, pyrazines, pyridines, phenolics, ketones, and acids. Curing time was reduced from 8 days to 6–10 h. Overall, the novel curing methods effectively reduced curing time while maintaining the technological, chemical, and sensory quality of SBT. Implementing these methods in industrial production could enhance both product quality and production efficiency.

Co-Expression of Chaperones for Improvement of Recombinant Geobacillus stearothermophilus Lipase Production in Pichia pastoris

    Erden-Karaoglan, Fidan

Pichia pastoris is among the most popular expression systems for recombinant protein production. The ability to secrete high titers of recombinant proteins is one of its primary advantages. Co-expression of folding-assisting factors is considered one of the strategies to improve protein production with this expression system. In this study, the effect of PDI, KAR2, HAC1, and ERO1 co-expression on Geobacillus stearothermophilus lipase production in P. pastoris was investigated. The chaperones were co-expressed under the regulation of the GAP promoter in clones with different lipase production levels (LPS#2 and LPS#8). Results showed that lipase-PDI co-expression clones have the highest activity. The effect of the other chaperones had varying effects for LPS#2 and LPS#8 clones. LPS#2-PDI and LPS#8-PDI were further analyzed to determine the effect of dimethyl sulphoxide (DMSO). Different concentrations (0.5%, 1%, and 2%) of DMSO were tested. The highest activity was obtained with approximately 1.5-fold activity in the LPS#8-PDI clone with 0.5% and 1% DMSO concentration levels. Comparison of the fermentation kinetic parameters revealed that the activity level of 56.54 U/mL provided with LPS#8 increased to 73.84 U/mL in the LPS#8-PDI clone with PDI co-expression, and when PDI co-expression was combined with DMSO conditions (0.5%), it reached 114.46 U/mL. It was observed that the productivity (U/g wet cell/g substrate/h) of the LPS#8 clone was increased 1.27-fold with LPS#8-PDI and 2.05-fold with LPS#8-PDI-DMSO conditions. In future studies, it is possible to achieve higher protein production levels by optimization at the fermenter scale.

Antioxidant properties of bovine liver protein hydrolysates and their practical application in biphasic systems

Arslan, Betul; Xiong, Youling L; Soyer, Ayla

BACKGROUND: The influence of protein hydrolysate produced from bovine liver protein hydrolysate (LPH) by enzymatic hydrolysis, using Alcalase/Protamex (1:1), on lipid dispersions was investigated. LPH production was optimized to maximize the antioxidant activity (at 45, 50, and 55 °C for 12, 18, and 24 h). Different concentrations of LPHs (1, 3, and 5 mg/g) were added to emulsions and to liposomes. Lipid oxidation level and particle size of the lipid dispersions were monitored for 14 days of storage at 25 °C. RESULTS: Radical scavenging activity and reducing power were the highest at 45 °C after 24 h of hydrolysis. Electrophoresis pattern showed that the antioxidant activity was arising from the peptides with molecular weight around 10 kDa. Lipid oxidation occurred more rapidly in samples without LPH during storage. In emulsions, lower thiobarbituric acid-reactive substance and conjugated diene values were measured with increasing concentrations of LPH at day 14. Accordingly, particle size of the samples containing 5 mg/g of LPH was smaller than those of other groups. Phase separation was observed only in lecithin emulsion without LPH at day 14. The use of LPH in liposome limited the lipid oxidation and maintained the size of the particles independently from the concentration. CONCLUSION: This study highlights the potential applications of animal by-products as natural antioxidants in complex food systems. The results demonstrate that LPH, particularly when hydrolyzed at optimized conditions, can effectively inhibit lipid oxidation. The findings suggest that biphasic systems incorporating LPH have promising prospects for enhancing the stability and quality of food products.
