Corneal neovascularization (CV) is a vision-threatening condition characterized by the pathological ingrowth of blood vessels. While corticosteroids and anti-VEGF agents are standard treatments, their prolonged use is associated with significant ocular side effects. This study aimed to develop and optimize nanosuspensions of Apigenin (APG) and Apigenin-7-O-glucoside (APG 7) to enhance their ocular bioavailability and evaluate their efficacy in the treatment of CV. APG and APG 7 were isolated from Petroselinum crispum . Nanosuspensions were formulated using an anti-solvent precipitation method, optimized via a 23 factorial design considering PVA concentration, Span 80 concentration, and homogenization speed. The physical properties, including particle size, PDI, and zeta potential, were characterized. In vivo efficacy was assessed over a 7-day treatment period in an alkali-burn induced rat CV model through clinical scoring, histopathology, and immunohistochemical analysis of VEGFA and IL-6 expression. The optimized APG nanosuspensions exhibited a mean particle size of ∼166.9 nm with high stability. In vivo studies demonstrated that 0.05% APG nanosuspension significantly inhibited corneal angiogenesis, reduced stromal edema, and downregulated VEGFA and IL-6 expression compared to the untreated injury group (p < 0.05). Conversely, APG 7 formulations failed to show significant therapeutic efficacy, likely due to limited corneal permeability attributed to the glycoside moiety. The 0.05% APG nanosuspension presents a promising, non-invasive therapeutic alternative for CV management, offering effective anti-angiogenic activity with improved ocular delivery characteristics.

Introduction: Insulin resistance (IR) is a central metabolic feature of polycystic ovary syndrome (PCOS). Althou gh metformin improves insulin sensitivity, its adverse effects limit long-term use, prompting interest in herbal alternatives. The present study aimed to investigate the effects of Cichorium intybus L. ethanolic extract on PCOS-associated IR in a letrozole-induced rat model, compared with metformin. Materials and Methods: A total of twenty-four female Sprague–Dawley rats were allocated into four groups: healthy control (H), PCOS-associated insulin resistance (IR), metformin-treated (MET), and Cichorium intybus ethanolic extract–treated (C.I). PCOS was induced by oral letrozole administration (1 mg/kg/day) for 21 days. Following confirmation of IR by estrous cycle disruption and oral gl ucose tolerance test (OGTT), treatments were administered orally for 15 da ys. Metabolic parameters, serum insulin, glucose, lipid profile, sex hormones, and HOMA-IR index were evaluated. Skeletal muscle tissues were examined histopathologically and by immunohistochemical analysis of insulin receptor (INSR), insulin receptor subsrate 1 (IRS1), and insulin receptor subsrate 2 (IRS2) expression. Results: PCOS-associated IR rats showed impaired glucose tolerance, hyperinsulinemia, increased HOMA-IR, dyslipidemia, and hormonal imbalance. C. intybus ethanolic extract treatment significantly improved glucose tolerance, insulin levels, HOMA-IR, and lipid profile, with effects comparable to metformin. While metformin markedly restored INSR, IRS1, and IRS2 expression in skeletal muscle, C. intybus produced only moderate improvements. Conclusion: C. intybus ethanolic extract improved metabolic, hormonal, and biochemical paramete rs in PCOS-associated IR. However, its effects on skeletal muscle insulin signaling markers were less pronounced than metformin, suggesting a complementary therapeu tic role mainly through systemic metabolic regulation.

From nature to nanoscale: advances, challenges, and preclinical translation of nanofiber drug delivery systems containing plant-derived active ingredients

Polat, Heybet Kerem; Turanlı, Yasin; Kurt, Nihat; Gözcü, Sefa; Tunçel, Emre; Bacanak, Hilal; Aytekin, Eren; Ünal, Sedat

Since the early 1960s, nanotechnology has been a critical area of science, allowing for the development of sophisticated nanomaterials. Nanofibers, one of the most widely used nanotechnological drug delivery systems, have emerged as a highly versatile platform within modern pharmaceutical sciences. By combining various polymers with active herbal ingredients, these systems mimic the natural extracellular matrix and provide improved functions such as a high surface area-to-volume ratio, drug targeting, and controlled drug release. Preclinical studies demonstrate that phytochemical-containing nanofibers have improved therapeutic profiles, including higher anti-inflammatory, antioxidant, antimicrobial, and antineoplastic effects. In various biomedical applications, such as promoting tissue engineering (such as bone and nerve regeneration), limiting tumor growth, and accelerating wound closure, it was shown that nanofibers may overcome the physicochemical limitations of herbal drugs such as low solubility and bioavailability. Despite promising preclinical study results, there are significant obstacles in the way of the commercialization of herbal drug-loaded nanofibers. The difficult standardization of multi-component herbal extracts, stabilization drawbacks, and the absence of scalable industrial manufacturing equipment that can maintain repeatable characterization of nanofibers are some of the major obstacles, in addition to a lack of clinical trials. To navigate the clinical translation of nanofibers, interdisciplinary collaboration regarding quality control, safety, and regulatory pathways is strictly necessary due to the case-by-case review approach utilized by regulatory bodies such as the US Food and Drug Administration (FDA) and European Medicines Agency (EMA). This review offers a comprehensive overview of research on herbal drug-loaded nanofibers and contributes a novel perspective with regulatory and clinical translational insights.

Material and Method: The medicinal plant species employed by the indigenous population for therapeutic purposes were systematically gathered and identified. Comprehensive data on traditionally utilized information were compiled, and herbarium materials were prepared. These materials have been deposited at the Iğdır National Wild Life Museum (INWM), affiliated with Iğdır University. Result and Discussion: This research identified a total of 58 taxa of medicinal plants belonging to 31 families. Among these, 50 species were found to grow naturally, while 8 species were cultivated. The predominant plant families included Asteraceae (8), Apiaceae (4), Lamiaceae (4), and Rosaceae (4). Infusion emerged as the most widely employed preparation method. The practice of traditional medicine remained prevalent among the population in Ağrı. Nevertheless, with the increasing availability of health services in the region, herbal medicine appeared to be more closely associated with healthcare and illness prevention than with curative purposes. The influx of new immigrants also contributed to the erosion of traditional knowledge. Notably, there is a discernible decline in traditional knowledge regarding the use of medicinal plants, both among younger generations and due to migration. Furthermore, this research serves as a foundational resource for prospective scientific inquiries aimed at the development of novel commercial drugs derived from plant sources.

BACKGROUND: Allergic conjunctivitis is one of the most common eye disorders. Different drugs are used for its treatment. Hesperidin is an active substance isolated from Citrus sinensis L. (Rutaceae) fruit peels, with known anti-inflammatory activity but low solubility. It was complexed with cyclodextrin and encapsulated in situ gel to extend its duration in the eye. RESULTS: The optimized formulation comprised 1% hesperidin, 1.5% hydroxyethyl cellulose, and 16% poloxamer 407. The viscosity at 25 °C was 492 ± 82 cP, and at 35 °C it was 8875 ± 248 cP, the pH was 7.01 ± 0.03, gelation temperature was 34 ± 1.3 °C, and gelation time was 33 ± 1.2 s. There was a 66% in vitro release in the initial 2 h, with a burst effect. A lipoxygenase (LOX) inhibition test determined that hesperidin was active at high doses on leukotyrens seen in the body in allergic diseases. In cell-culture studies, the hesperidin cyclodextrin complex loaded in situ gel, BRN9-CD (poloxamer 16%, hydroxy ethyl cellulose (HEC) 1.5%), enhanced cell viability in comparison with the hesperidin solution. It was determined that BRN9-CD did not cause any irritation in the ocular tissues in the Draize test. CONCLUSION: The findings of this study demonstrate the potential of the in situ gel formulation of hesperidin in terms of ease of application and residence time on the ocular surface. Due to its notable LOX inhibition activity and positive outcomes in the in vivo Draize test, it appears promising for incorporation into pharmaceutical formulations.
