Effects of tDCS on glutamatergic pathways in epilepsy: neuroprotective and therapeutic potential

    Demirdogen, Filiz; Akcay, Guven

Epilepsy is a chronic neurological disease characterized by recurrent seizures caused by abnormal electrical activity in the brain. The aim of our study was to investigate the effect of tDCS on oxidative stress, Ca2+, glutamate, GABA, AMPAR1, and NMDAR1 levels in kindling-induced epilepsy model. Behavioral tests evaluated motor and cognitive functions, while assessing oxidative stress, Ca2+, glutamate, GABA, AMPAR1, and NMDAR1 levels in hippocampal tissue. tDCS stimulation therapy demonstrates a neuroprotective effect on motor and cognitive function postepilepsy. Our study reveals an increase in TOC, Ca2+, glutamate, GABA, AMPAR1, and NMDAR1 levels and a decline in total antioxidant capacity (TAC) following PTZ-induced seizures. However, tDCS treatment led to a significant decrease of Ca2+, total oxidant capacity (TOC), glutamate, GABA, AMPAR1, and NMDAR1 levels in the epilepsy cohorts, while simultaneously causing a spike in TAC levels. The study’s results showed that tDCS treatment could have a therapeutic effect on oxidative stress, Ca2+, TOC, glutamate, GABA, AMPAR1, NMDAR1, and TAC in both acute and chronic kindling epilepsy models.

The use of highly sensitive and reliable analytical methods is essential for Extractables & Leachables studies. Especially the determination of N-Nitrosamines in drugs, which have carcinogenic properties and may contaminate drugs at trace levels, is quite important. In this study, a new, sensitive, short-time and reliable liquid chromatography with tandem mass spectrometry method was developed for the analysis of 15 N-Nitrosamines defined in the European Pharmacopoeia within the scope of Extractables & Leachables studies and validated according to the International Council for Harmonization (ICH Q2 (R2)). The analysis of N-Nitrosamines was carried out in positive mode using an Atmospheric Pressure Chemical Ionization source in the dynamic multiple reaction monitoring scanning mode. In the chromatographic separation, gradient elution was applied using a reverse phase Phenyl column and the mobile phase (A: 0.1 % formic acid in ultrapure water, B: 0.1 % formic acid in methanol); total analysis time was 16 mins and the flow rate was optimized as 0.6 mL/min. N-Nitroso-dimethylamine-d6 was used as an internal standard. The developed method was used in extractables studies to control the potential presence of N-Nitrosamines that may be caused by interactions between the product and primary packaging materials (e.g. polypropylene bag, LDPE container, disposable eye drop packaging and bromobutyl stopper). It was also successfully applied to pharmaceutical preparations containing sugammadex, metformin, gliclazide and paracetamol in the leachables studies.

In Vivo Evidence for the Preventive Role of Vaccinium macrocarpon Aiton in Indomethacin-Induced Gastric Ulcer: Focusing on Antioxidant, Anti-Inflammatory and Anti-Apoptotic Mechanisms

Aydin, Ismail Cagri; Ferah Okkay, Irmak; Okkay, Ufuk; Ozkaraca, Mustafa; Yesilyurt, Fatma; Yilmaz, Aysegul; Cicek, Betul; Karatas, Ozhan; Kalkandelen, Hilal Kadioglu; Aydin, Bilge

The present study aimed to unveil the gastroprotective potential of Vaccinium macrocarpon (VM) extract and its mechanism of action against indomethacin (INDO)-induced gastric ulcers in rats. To achieve this goal, rats were pretreated with either omeprazole (20 mg/kg) or VM (100 mg/kg) orally for 14 consecutive days. Gastric tissue samples were collected and various parameters were evaluated to understand the mechanism of VM's action, including the levels of superoxide dismutase, malondialdehyde, glutathione, CAT and transforming growth factor beta (TGF-β), as well as the mRNA expression levels of tumour necrosis factor alpha, interleukin 1 beta, nuclear factor kappa B (NF-κB) and inhibitor kappa B (IκB). Additionally, the immunopositivity of cyclooxygenase (COX)-1, COX-2, PGE2, proliferating cell nuclear antigen (PCNA) and caspase-3 was assessed. The total amount of phenolic compounds present in the VM extract was high (58.08 µg/mL gallic acid equivalent/mg extract). The healing effect of VM was demonstrated by an increase in the expression of PCNA. Furthermore, the level of TGF-β was found to increase upon treatment with VM. Analyses of COX-1, COX-2 and PGE2 expression in gastric tissue confirmed the gastroprotective effect of VM. Notably, the expression of NF-κB was markedly reduced, whereas that of IκB was substantially increased. Overall, the findings of this study demonstrate that VM extract has gastroprotective and curative effects against INDO-induced ulcers through its antioxidant, anti-inflammatory, mucosal regenerative and anti-apoptotic activities. Therefore, VM may serve as a useful adjuvant treatment for nonsteroidal anti-inflammatory drugs–induced gastric ulcer disease.

Cephalometric Examination of Soft Palate, Hyoid and Upper Airway Anatomy in Obstructive Sleep Apnea Syndrome Patients

Celik, Elif; Celik, Fatih; Unver, Edhem; Karavas, Erdal; Diyarbakır, Samih

Objectives: In this study, the dimensions of the soft palate, the position of the hyoid bone in relation to the mandibular plane and C3 vertebrae, and the width of the upper airway were measured on lateral cephalograms of patients with obstructive sleep apnea syndrome of different severities diagnosed by polysomnography. This study aimed to investigate the association between increased apnea severity and anatomical formations relevant to dental practice. Materials and Methods: Our study was designed as a retrospective data analysis of 182 patients, 78 females and 104 males. In our study, patients were divided into four groups: patients with simple snoring with an Apnea-Hipopnea index (AHI) value below 5, patients with mild OSAS with an AHI value of 5–15, patients with moderate OSAS with an AHI value of 15–30 and patients with severe OSAS with an AHI value above 30. Measurements obtained from lateral cephalograms were compared between these groups. Results: Statistically significant narrowing was observed in all other groups compared to the control group in oropharyngeal width values in upper airway measurements (P<0.05). The distance between the hyoid and mandibular plane was significantly lower in the control group than in the severe OSAS group (P=0.001). A statistically significant elongation of the soft palate was also observed in patients with OSAS compared to the control group (P<0.05). Conclusions: Our findings support that the position of the hyoid bone is important in the diagnosis of OSAS. Furthermore, a significant correlation was found between soft palate length and OSAS. These results suggest that surgical techniques such as maxillary advancement or mandibular advancement, or both, to increase airway width may be effective in the treatment of OSAS. SYNTHESIS, CHARACTERISATION AND INVESTIGATION OF THE ANTICANCER POTENTIAL OF CARMOFUR-LOADED SILVER NANOPARTICLES; [KARMOFUR YÜKLÜ GÜMÜŞ NANOPARTİKÜLLERİNİN SENTEZİ, KARAKTERİZASYONU VE ANTİKANSER POTANSİYELİNİN ARAŞTIRILMASI]

DanışmanKalindemi̇rtaş, Ferdane; Kari̇per, İshak Afşin; Özerkan, Dilşad; Kuruca, Dürdane Serap

Objective: The term "triple-negative breast cancer" (TNBC) is used to describe tumours that do not express oestrogen receptor (ER), progesterone receptor (PR), or human epidermal growth factor receptor 2 (HER2). TNBC tends to be more aggressive than other types of breast cancer. Current antineoplastic drugs have limited treatment options for malignant breast cancer owing to their narrow therapeutic index, toxicity, resistance, and nonselectivity. Therefore, there is a need for the prompt development of new medicinal drugs for TNBC. Here, we investigated the growth inhibition potential of carmofur-bonded silver nanoparticles (AgNPs-Car) on two TNBC cell lines, MDA-MB-231 and 4T1, and compared the effects with non-cancerous Human umbilical vein endothelial cells (HUVECs). Material and Methods: AgNPs-Car were synthesised and characterised by FTIR, DLS, SEM, and EDX. The anticancer effect was evaluated using a 3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide (MTT) assay. Results: AgNPs-Car was determined to be predominantly more effective than Car alone. Mainly, 4T1 cells were 5.7-fold more sensitive to AgNPs-Car than Car alone. While AgNPs showed no considerable toxicity on HUVECs, they significantly induced the cytotoxicity of MDA-MB-231 and 4T1 cells. Conclusion: Our results showed that AgNPs-Car is a promising anticancer agent due to its highly potent and selective growth inhibitory effect on TNBC cells.

Background: Tramadol, a weak opioid analgesic agent, is known to induce ovarian damage. Previous studies have held oxidative stress responsible for the adverse effects of tramadol on female reproduction. This study examined the protective effects of ATP against tramadol-induced ovarian damage and reproductive dysfunction in rats. Methods: Rats were divided into four groups (n = 12); healthy (HG), only ATP (ATPG), only tramadol (TMDG), and ATP + tramadol (ATMG). ATP was injected intraperitoneally at 25 mg/kg. Tramadol at 50 mg/kg was initiated one hour after ATP. The treatment was administered once a day for 14 days. Six rats from each group were euthanized. For two months, the remaining rats were paired with male rats. Rats that failed to give birth during this period were considered infertile. A maternity period was calculated for the rats that were delivered. Results: Tramadol caused an increase in malondialdehyde and interleukin-6, and decreased total glutathione, superoxide dismutase, and catalase levels in the ovarian tissue. Furthermore, tramadol disrupted the histological structure of the ovaries, and immunohistochemical staining revealed severe immunopositivity. Tramadol again caused infertility and delayed pregnancy in fertile women. By suppressing biochemical changes, ATP significantly reduced tramadol-induced ovarian damage. Both histopathologically and immunohistochemically, ATP treatment regressed ovarian damage. Additionally, ATP significantly reduced tramadol-induced infertility and maternal delay. Conclusions: The results indicate that tramadol-induced oxidative and inflammatory ovarian injury, infertility, and caspase 3 were suppressed by ATP, as demonstrated by our experimental findings.

    Akşit, Zeynep

In this study, the essential oils of the basal leaves (blEOs) and peduncles (pEOs) -stalks bearing flowers- of Scrophularia catariifolia were separately analyzed using GC-MS for the first time. The main components of two distinctive parts were determined as 4-hexen-1-ol (8.03/6.34%), 1-hexanol (5.48/9.15%), 3-octenol (16.94/6.83%), benzene ethanol (17.21/9.82%), and phytol (15.18/28.68%) for basal leaves and peduncles, respectively. The essential oils showed remarkable antimicrobial activity against the four food-borne pathogens tested. Pseudomonas aeruginosa, Clostridium perfringens, and Bacillus cereus were found to be more susceptible to peduncle essential oil. Two essential oils showed similar activity against Staphylococcus aureus. The essential oils were found to be inactive against Escherichia coli. These findings highlight the potential of S. catariifolia essential oils as natural antimicrobial agents for food safety and preservation. Further investigations are needed for their key applications.

Determination of the effect of pyocyanin and Saccharomyces boulardii on gut microbiota and TLR4/MyD88/NF-κB and NLRP3 signaling pathways in sepsis induced by cecal ligation and puncture in rats

Ucar, Mahmut; Celebi, Ozgur; Celebi, Demet; Baser, Sumeyye; Guler, Mustafa Can; Tanyeli, Ayhan; Kılıclıoglu, Metin; Yılmaz, Ahmet; Yıldırım, Serkan

Sepsis is a life-threatening systemic inflammatory condition marked by a dysregulated host response to infection. The intestinal microbiota plays a pivotal role in maintaining immune homeostasis and epithelial barrier integrity, whereas dysbiosis significantly contributes to the pathogenesis of sepsis. This study investigates the effects of the Pseudomonas aeruginosa-derived metabolite Pyocyanin and the probiotic Saccharomyces boulardii (S. boulardii) on microbial composition and the TLR4/MyD88/NF-κB/NLRP3 signaling axis in a cecal ligation and puncture (CLP)-induced rat model of sepsis. The experimental design assessed the synergistic or antagonistic effects of single and combined treatments using molecular, microbiome, and immunohistochemical parameters to evaluate histopathological damage and microbial ecological dynamics. Seven experimental groups were established following CLP. Intra-abdominal Pyocyanin (10 nmol/g) and oral probiotic (10⁶ CFU/kg) treatments were administered either individually or in combination. Focused Ion Beam - Scanning Electron Microscopy (FIB-SEM) analyses revealed that the amorphous structure of Pyocyanin interacted with the surface of S. boulardii. Western blot analysis showed a 2.3-fold increase in TLR4/NF-κB expression in the CLP group (p ≤ 0.05), which synergistically rose to 4.5-fold with Pyocyanin (p ≤ 0.001), whereas probiotic treatment decreased expression levels by 35%. According to 16 S rRNA sequencing, Pyocyanin reduced α-diversity by increasing Lactobacillaceae abundance to 32.66% (Shannon index: 3.598 vs. 4.433 in control), while S. boulardii enhanced β-diversity by elevating Coriobacteriaceae (5.85%) and Prevotellaceae (10.63%) levels (Tables 2, 3 and 4). PCoA confirmed 41.7% Bray-Curtis dissimilarity between groups at the species level (PERMANOVA R²=0.38, p = 0.002). Histopathologically, severe hepatocyte necrosis (73.2 ± 6.1%, p = 0.0022) and a 4.2-fold increase in hepatic TGF-β expression were observed in the CLP group, whereas epithelial barrier damage was significantly attenuated in the probiotic groups. Immunofluorescence analysis revealed that combined treatment reduced Caspase-8 and TLR4 expression by 28% compared to Pyocyanin alone (p ≤ 0.05). In conclusion, S. boulardii supported microbiota homeostasis by suppressing TLR4/NF-κB signaling, whereas Pyocyanin exacerbated the inflammatory response via NLRP3 activation. These findings provide molecular evidence supporting probiotic-assisted immunomodulatory strategies in sepsis therapy.

Targeting oxidative stress and metal imbalance in cerebral ischemia: the neuroprotective role of sodium valproate

Ulaş, Mustafa; Gökay Argadal, Ömer; Bardaş Özkan, Ebru

In brain ischaemia (BI), the oxidative and ionic stresses lead slowly but definitely to the injury of neurones. The present investigation is looking to check the status of certain oxidative stress markers and trace elements which are induced in response to brain ischaemia with sodium valproate (VPA), a neuroprotective agent, in the rats exposed to the model of BI. Although VPA is already used for clinical purposes, it deserves careful examination of the potentially detrimental effects of VPA in conditions of ischaemia closely linked to hepatotoxicity and thrombocytopenia. Forty rats were randomly assigned into five groups (n = 8 per group): Control, Sham, Brain Ischaemia (BI), BI + Valproic Acid (VPA), and VPA only. Ischaemia was induced in the BI and BI + VPA groups by bilateral occlusion of the common carotid arteries for 30 min. VPA was administered orally at a dose of 100 mg/kg daily for seven days in the BI + VPA and VPA-only groups. This dose was based on previous preclinical safety and efficacy data. Control, Sham, and BI groups received saline. The Sham group underwent surgical procedures without carotid occlusion. The biochemical investigations pertaining to cortical tissue demonstrated the fact that BI has decreased all antioxidant enzyme levels of SOD, CAT, GPx, and those of trace elements such as Mg, Zn, and Se. At the same time, malondialdehyde (MDA) and neurotoxic metals (Cr, Fe, Cu) increased. These changes brought by BI were restored through treatment with VPA on oxidative stress and the restoration of trace element balance along with reductions in those associated with oxidative damage. The results thus reinforce the capacity of VPA for neuroprotective benefits against ischaemic damage inflicted within brain tissue and hence merit the further exploration of the mechanistic action of this therapeutic A.

Background: Artificial intelligence can be an important tool in developing creative thinking while supporting individualized learning and problem-solving skills in educational processes. However, the effect of artificial intelligence on creative thinking may differ depending on factors such as students’ digital literacy level and the academic support they receive. In this context, how artificial intelligence can encourage creative thinking in educational processes and how this process is shaped by digital literacy and academic support stands out as an issue to be investigated. Purpose: This study artificial intelligence to determine the effect of artificial intelligence use on creative thinking skills in nursing students, the mediating role of digital literacy in this effect, and the situational mediating effect of academic support. Method: This cross-sectional and descriptive correlational study was conducted with 426 nursing students from three universities in different regions of Turkey during the fall semester of 2024–2025. Simple random sampling method was used. Data were collected through demographic information form and four valid and reliable scales. Result: Research findings show that using artificial intelligence substantially and significantly affects Creative Thinking (β = 0.70, p < 0.001). Digital literacy plays mediating role in this relationship, indirectly strengthening the effect of artificial intelligence use on creative thinking (β = 0.422, LLCI = 0.357, ULCI = 0.490). As a moderating factor of this relationship, the academic support creates a more limited effect at low levels of academic support (Effect = 0.224). At the same time, it maximizes this effect at high levels of support (Effect = 0.298). Conclusion: The effect of artificial intelligence use on Creative Thinking is significantly strengthened by the mediating role of digital literacy and the moderating role of academic support. These findings suggest that developing digital skills and strengthening academic support mechanisms are critical to integrating artificial intelligence technologies into educational processes. © The Author(s) 2025.; Artificial intelligence significantly increases the creative thinking tendencies of nursing students both directly and through digital literacy. Salvia pachystachya is a medicinally promising plant species, yet its pharmacological properties remain underexplored. This study aimed to investigate its anatomical, phytochemical and bioactive characteristics to assess its therapeutic potential comprehensively. Microscopic analysis revealed distinctive anatomical features, including quadrangular stems and glandular trichomes on leaves, providing valuable insights into the plant's structural properties. Phytochemical profiling of methanol and aqueous extracts identified chlorogenic acid as the major component (11,052.88–25,718.77 ng/mL), while essential oils were predominantly composed of phytol (48.4%) and caryophyllene oxide (26.2%). The total phenolic content and antioxidant capacity analyses showed that the methanol extract exhibited significant ABTS•+ scavenging activity (90.06%). Bioactivity assays demonstrated the plant's antidiabetic, anticholinesterase and antimicrobial properties. The LC–MS/MS analysis of extracts revealed that chlorogenic acid was the most abundant secondary metabolite among the analysed compounds, with significant variations across different plant parts and extraction solvents. The highest concentration was detected in the aerial part methanol extract (34,885.06 ng/mL), followed by the flower methanol extract (25,718.77 ng/mL). Genotoxicity assessments, including Ames, Escherichia coli WP2, and Allium cepa tests, confirmed the nongenotoxic nature of the extracts. Furthermore, molecular docking indicated strong binding affinities of chlorogenic and quinic acids to target enzymes, supporting their therapeutic relevance. These findings highlighted S. pachystachya as a promising candidate for further pharmacological research, particularly for its potential in combating oxidative stress-related conditions such as diabetes and neurodegenerative diseases.

Segmental Non-Mass Enhancement Features in Breast Magnetic Resonance Imaging: A Multicenter Retrospective Study of Histopathologic Correlations

Aydin, Hale; Bozkurt, Cansu; Hayme, Serhat; Bilge, Almila Coskun; Oztekin, Pelin Seher; Avdan Aslan, Aydan; Ozcan, Irem; Gultekin, Serap; Eren, Abdulkadir; Durur Subası, Irmak

Background/Objectives: Segmental non-mass enhancement (NME) is the breast MRI distribution pattern with the highest positive predictive value (PPV) for malignancy. Despite its diagnostic relevance, its imaging characteristics have rarely been examined in isolation, leaving uncertainty in clinical practice. This multicenter retrospective cohort study aimed to evaluate multiparametric MRI features—including internal enhancement pattern, dynamic contrast-enhanced (DCE) kinetics, and diffusion restriction—in segmental NME to identify malignancy predictors. Methods: This retrospective cohort review included 14,834 breast MRI reports from five institutions (September 2017–February 2024), identifying 103 women (mean age, 44.4 ± 9.9 years) with segmental NME (70 malignant, 33 benign). MRI was performed at 1.5 T or 3 T using standardized protocols. Two breast radiologists, blinded to pathology, assessed internal enhancement, DCE kinetics, diffusion restriction, and short tau inversion recovery (STIR) features according to BI-RADS. Statistical analyses included chi-square/Fisher’s tests and logistic regression. Results: Clustered ring enhancement (CRE) was significantly associated with malignancy (p = 0.004). Fast initial-phase enhancement (p < 0.001) and delayed-phase washout (p = 0.011) also correlated with malignancy. On multivariate analysis, fast initial-phase enhancement remained an independent predictor (odds ratio [OR] = 5.133, p = 0.031), whereas slow enhancement predicted benignity (OR = 0.194, p = 0.020). Histologies included ductal carcinoma in situ, invasive ductal carcinoma, granulomatous mastitis, and benign hyperplastic lesions. Conclusions: This study, focusing exclusively on segmental NME, identifies CRE, fast initial-phase enhancement, and washout kinetics as reliable imaging biomarkers. Incorporating these features into breast MRI interpretation may improve diagnostic accuracy, risk stratification, and management decisions.

Increasing antifungal resistance and side effects of existing drugs demand alternative approaches for treating Candida (C.) infections. This study aimed to comprehensively evaluate the antifungal efficacy of myricetin (MYR), a natural flavonoid, against both fluconazole (FLC)-resistant and susceptible clinical Candida strains, with a particular focus on its inhibitory effects on C. albicans biofilms. Antifungal susceptibility was evaluated on Candida spp. by the broth microdilution method, and the impact of myricetin on C. albicans biofilms was determined using the Cell Counting Kit-8 (CCK-8) assay. To understand the molecular mechanisms underlying the antibiofilm properties of myricetin, expression analysis of genes in the RAS1/cAMP/EFG1 pathway (ALS3, HWP1, ECE1, UME6, HGC1) and cAMP-dependent protein kinase regulation (RAS1, CYR1, EFG1) involved in the transition from yeast to hyphae was performed. Field emission scanning electron microscopy (FESEM) was used to study the ultrastructural changes and morphological dynamics of Candida biofilms after exposure to MYR and FLC. The in vivo toxicity of myricetin was evaluated by survival analysis using the Galleria mellonella model. Myricetin significantly suppressed key genes related to hyphae development (RAS1, CYR1, EFG1, UME6, and HGC1) and adhesion (ALS3 and HWP1) in both clinical and reference Candida strains at a concentration of 640 µg/mL. FESEM analysis revealed that myricetin inhibited hyphae growth and elongation in C. albicans. This study highlights the promising antibiofilm potential of myricetin through a significant inhibition of biofilm formation and hyphal morphogenesis.

Since the beginning of the pandemic, few studies have explored the histopathological and molecular characteristics of placentas from SARS-CoV-2-positive pregnant women. This study aimed to investigate changes in the expression of angiogenic growth factors in the placentas of SARS-CoV-2-infected pregnant women (COVID-19 group) and compare them with placentas from non-infected women (control group). Placentas collected at birth from 20 healthy women (control group) and 20 women who tested positive for COVID-19 were prepared for histopathological analysis. Immunohistochemical staining was performed on placental tissues using antibodies against Angiotensin-Converting Enzyme 2 (ACE2), Vascular Endothelial Growth Factor (VEGF), Vascular Endothelial Growth Factor Receptor 1 (VEGFR-1), and Platelet-Derived Growth Factor (PDGF). Initially, the presence of SARS-CoV-2 in placental tissue was evaluated through ACE2 immunohistochemical staining. ACE2 expression was weak in the placental syncytiotrophoblast cells of non-infected women, whereas it was markedly increased in the syncytiotrophoblasts of placentas from women infected with COVID-19. VEGF expression was strong in the endothelial and stromal cells of villi in the control group, but significantly reduced in the villi of infected placentas. VEGFR-1 was strongly expressed in the endothelial membranes of villi in the control group, but expression was notably weaker in infected placentas. PDGF expression was strong in the syncytiotrophoblast and stromal cells of the control group but was significantly weaker in COVID-19-positive placentas. VEGF, VEGFR-1, and PDGF were strongly expressed in the placental villi of the control group, and their marked reduction in COVID-19-positive placentas suggests that SARS-CoV-2 infection may adversely affect the development and differentiation of the placental villous tree.

Introduction: The impact of laparoscopic surgery on homeostatic systems necessitates careful consideration of intra-abdominal pressure (IAP) management. This study investigated the effects of low-pressure pneumoperitoneum with deep neuromuscular blockade (NMB) on surgeon satisfaction, haemodynamics and post-operative outcomes in laparoscopic cholecystectomy patients. Patients and Methods: The study design involves prospective randomised control. Ninety patients were assigned to low (7-10 mmHg, n = 45) or normal (12-16 mmHg, n = 45) IAP groups. Deep NMB, guided by train-of-four monitoring, was administered. This study evaluated surgical rating scale scores, haemodynamics and post-operative outcomes through a literature review. A computer programme (IBM, SPSS) was used for statistical analysis. Chi-square and Mann-Whitney U tests were used to analyse patients' IAP levels, additional NMB requirements, surgical rating scale scores and numerical rating scales. Patient demographics and other intraoperative and post-operative variables were analysed with Student's t-test and the Mann-Whitney U test. Values of P < 0.05 were considered to indicate statistical significance. Results: No significant demographic differences were observed. The low-pressure group exhibited lower post-operative pain (P < 0.01) and reduced analgesia requirements (P = 0.00). On analysis of the surgeon rating scale, no disparities were evident between the groups. NMB usage correlated with height and weight (P < 0.01). Heart rate showed no intergroup differences. The MAP measured after 15 min was lower in Group L, and the difference was significant (P = 0.023). The SAP measured after 30 min was lower in Group L, and the difference was significant (P = 0.017). Blood gas values and surgical field visibility were unaffected by the IAP. The positive correlations between NMB, height and weight aligned with previous research. Conclusion: This study highlights successful laparoscopic cholecystectomy under low IAP, deep NMB and favourable post-operative outcomes. Despite these limitations, the findings contribute to optimising laparoscopic surgical approaches.
