Marine algae-derived polyamines and phlorotannins as cholinesterase inhibitors: In vitro and in silico analysis

Necip, Adem; Işık, Mesut; Türkeş, Cüneyt; Ünal, Dilek; Kırboğa, Kevser Kübra; Beydemir, Şükrü; Rudrapal, Mithun

The limitations of conventional cholinesterase inhibitors in Alzheimer's disease (AD) therapy namely, their short half-lives and adverse side effects—necessitate the exploration of natural alternatives. This investigation examined the inhibitory potential of selected polyamines (putrescine and spermidine) and a phlorotannin derivative (phloroglucinol), naturally present in the macroalgae species Cystoseira crinita and Stypopodium schimperi, against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) using in vitro and in silico approaches. Additionally, radical scavenging potentials of each sample were determined. Enzyme activity assays employing the Ellman method revealed that putrescine and spermidine reached similar inhibition levels (approximately 32 %), while Cystoseira crinita acetone extract demonstrated superior potency with an IC50 of 0.085 mg/mL, indicating robust enzyme modulation. Complementary molecular docking studies conducted with Schrödinger Maestro 2020 elucidated the formation of stable enzyme-ligand complexes through hydrogen bonding and π-π interactions with critical residues including HIS447, TYR337, and TRP86 within the AChE active site. Moreover, crude macroalgal extracts, particularly those derived from C. crinita and S. schimperi, exhibited marked AChE inhibition, further substantiating the therapeutic potential of these marine-derived compounds. Collectively, these findings demonstrate that the polyamines and phlorotannin derivative studied not only effectively regulate cholinesterase activity but also offer naturally derived structural frameworks for the development of novel AD treatments. Phloroglucinol showed a high scavenging of DPPH (85%) and ABTS (98%) radicals, close to the activity of the reference compound trolox (96% for DPPH and 94% for ABTS). Further pharmacokinetic and toxicological evaluations are warranted to ascertain their clinical viability in AD management.

Examining the Mediating Role of Entrepreneurial Intention in the Relationship Between General Self-Efficacy and Career Stress Among Pharmacy Students; [Eczacılık Öğrencilerinde Genel Öz Yeterlilik ve Kariyer Stresi Arasındaki İlişkide Girişimcilik Niyetinin Aracı Rolünün İncelenmesi]

    Özkaya, Yağmur; Hakverdi̇, Gülden

This study examined the relationships among general self-efficacy, entrepreneurial intention, and career stress among pharmacy students and tested whether entrepreneurial intention mediated the relationship between general self-efficacy and career stress. The study was conducted with 282 students enrolled in the Faculty of Pharmacy at Erzincan Binali Yıldırım University. The data were analyzed using Pearson correlation analysis and Hayes' PROCESS macro (Model 4). The results showed that general self-efficacy was positively and significantly associated with entrepreneurial intention (r = .46, p < .001) and negatively and significantly associated with career stress (r = -.39, p < .001). Entrepreneurial intention was also negatively and significantly associated with career stress (r = -.41, p < .001). According to the mediation analysis, the total effect of general self-efficacy on career stress was significant and negative (B = -.3868, p < .001). When entrepreneurial intention was included in the model, the direct effect of general self-efficacy on career stress decreased but remained significant (B = -.2567, p = .001). In addition, the indirect effect of general self-efficacy on career stress through entrepreneurial intention was significant (B = -.1301, 95% bootstrap CI [-.2175, -.0664]). The findings suggest that entrepreneurial intention partially mediates the relationship between general self-efficacy and career stress. In this context, students with higher levels of general self-efficacy appear to have stronger entrepreneurial intentions, which may, in turn, be associated with lower levels of career stress.

Synthesis of Novel 4,6-Dimethoxyindole-Based Hydrazones Derived From 2,4-Dimethoxybenzaldehyde: Design, Cytotoxic Activity, Molecular Docking, and ADME-Tox Studies

Çakmak, Reşit; Başaran, Eyüp; Akkoç, Senem; Türkmenoğlu, Burçin; Köprü, Semiha; Bingül, Murat

In this study, a series of indolyl hydrazone derivatives (5–10) were designed and synthesized from 2,4-dimethoxybenzaldehyde (DMBA) (1) as potential chemotherapeutic agents. The structures of the synthesized compounds were confirmed by elemental analysis, FT-IR, 1D, and 2D NMR spectroscopy, and high-resolution mass spectrometry (HRMS). Their cytotoxic activities were evaluated against human breast (MCF-7), liver (HepG2), and colorectal (DLD-1) cancer cell lines. All compounds exhibited cytotoxic effects at micromolar concentrations, with IC50 values ranging from 8.97–26.07 µM for MCF-7, 22.50–70.99 µM for HepG2, and 5.93–66.31 µM for DLD-1 cells. Compound 10 demonstrated the strongest activity against MCF-7 and DLD-1 cells, with IC50 values of 8.97 ± 0.31 and 5.93 ± 0.11 µM, respectively. Compound 7 showed the highest antiproliferative activity against HepG2 cells (22.50 ± 1.23 µM), exceeding the activity of cisplatin (37.27 ± 2.42 µM). Molecular docking studies revealed favorable binding affinities of the most active compounds (7 and 10) toward selected targets. In addition, in silico ADME-toxicity analyses predicted acceptable physicochemical and pharmacokinetic properties, supporting the potential of these derivatives as promising anticancer candidates.

Patient vs. physician narratives on refractive surgery in Turkish YouTube videos: a comparative reflexive thematic analysis

    Gürsoy, Nurcan; Gürsoy, Ersan

Correctable refractive errors are a major, preventable cause of visual impairment. Refractive surgery is widely promoted online, yet how Turkish-language YouTube videos frame benefits, risks, recovery, and long-term outcomes—and how this framing differs between patient and physician narrators—remains underexplored. We aimed to qualitatively compare patient- and physician-generated Turkish-language YouTube videos on refractive surgery and to describe audience engagement. We conducted a reflexive thematic analysis (Braun & Clarke) of 64 publicly available videos (29 patient, 35 physician) meeting predefined criteria (Turkish; primarily refractive surgery; ≥1 min; ≥1,000 views; ≥240p). Searches were performed on 15 July 2025 using predefined search strings. Videos were transcribed verbatim and inductively coded in NVivo by two researchers. Between-group differences in engagement metrics were assessed with Mann–Whitney U tests, with a one-video-per-channel sensitivity analysis to address potential clustering. Patient narratives foregrounded lived experience (decision-making, perioperative discomfort, postoperative visual fluctuations, and symptoms such as dry eye and glare/halos) and often raised concerns about commercialization. Physician narratives emphasized candidacy assessment, procedure selection, recovery timelines, and risk mitigation. In the one-video-per-channel sensitivity analysis (patient n = 27; physician n = 23), patient videos received more likes (median 300 [IQR 1,639] vs. 59 [269], p = 0.009) and showed a higher like-to-view ratio (0.013 [0.01] vs. 0.006 [0.01], p < 0.001), whereas view counts were not significantly different (24,000 [98,900] vs. 18,000 [48,500], p = 0.224). Turkish-language YouTube narratives share experiential touchpoints but diverge systematically in how risks, commercialization, and expectations are framed by patients versus physicians. Findings support the need for balanced, accurate, and discoverable patient-facing materials tailored to platform dynamics.

Machine learning-guided repurposing of FDA-approved quinolones as dual cholinesterase inhibitors: A multi-level docking, molecular dynamics, DFT, and SHAP-based analysis

    Türkeş, Cüneyt

Alzheimer's disease (AD) involves progressive cholinergic degeneration, with acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) playing key enzymatic roles in its pathology. In this study, we computationally repurposed four FDA-approved quinolone antibiotics, Norfloxacin, Sparfloxacin, Gatifloxacin, and Nalidixic acid, as potential dual-site cholinesterase (ChE) inhibitors using a hybrid in vitro/in silico workflow. Enzyme inhibition assays identified Norfloxacin as the most potent AChE inhibitor (KI = 1.08 μM), while all compounds displayed non-competitive inhibition toward BChE. Molecular docking and MM-GBSA binding free energy analyses revealed key interactions within the catalytic gorge of AChE, supported by hydrogen bonding with Phe295 and Arg296, as well as π–π contacts with Tyr124. Density functional theory computations highlighted the influence of frontier orbital distribution on binding affinity, particularly for Norfloxacin and Sparfloxacin. An explicit-solvent molecular dynamics simulation of the AChE–Norfloxacin complex further confirmed the stability of the docking-derived binding mode over 100 ns. In an exploratory fashion, SHAP-based machine learning models were applied to a descriptor set derived from QikProp, SwissADME, and Jaguar outputs, suggesting that BBB-related indices and HOMO energy contribute to AChE inhibition, whereas the energy gap is more relevant for BChE; these trends, however, are constrained by the small four-compound dataset and should be regarded as hypothesis-generating. In silico ADME/Tox profiling indicated favorable oral drug-like properties, low predicted CYP450 inhibition liabilities, and physicochemical profiles compatible with CNS-oriented optimization, although passive BBB permeability was not predicted to be high. Finally, systems-level enrichment (STRING, GeneCards) provided a qualitative network context linking ACHE and BCHE to neurodegeneration. Together, these data position Norfloxacin and Sparfloxacin as computationally prioritised candidates whose ChE-related repurposing potential warrants further validation in dedicated cellular and in vivo models.

Antioxidant, Antidiabetic, Antiproliferative Activities, and Verbascoside Content of Four Endemic Verbascum Species: V. leiocarpum, V. ozturkii, V. alyssifolium, and V. tuna-ekimii

Şimşek, Samed; Akşit, Hüseyin; Köksal, Ekrem; Altay, Ahmet

This study, for the first time, investigated the antioxidant, antidiabetic, and antiproliferative properties of methanolic extracts from the aerial and root parts of four endemic Verbascum species—Verbascum leiocarpum Murb., Verbascum ozturkii Karavel., Uzunh. & S.Çelik, Verbascum alyssifolium Boiss., and Verbascum tuna-ekimii Karavel., A.Duran & Hamzaoğlu—and quantitatively determined their verbascoside content using HPLC-DAD. Antioxidant capacity was evaluated by DPPH and FRAP assays, and total phenolic content (TPC) and total flavonoid content (TFC) were also determined. Antidiabetic activity was assessed through α-glucosidase inhibition, and antiproliferative effects were determined in A549, MDA-MB-453, and HT-29 human cancer cell lines. Among the tested samples, the root of V. leiocarpum exhibited the highest TPC (50.89 mg GAE/g extract), FRAP value (124.16 mg TE/g extract), DPPH scavenging ability (IC50: 58.82 μg/mL), and verbascoside content (129.06 mg verbascoside/g extract). This extract also exhibited significant antiproliferative effects against A549 and MDA-MB-453 cells, with inhibition rates of 85% and 69%, respectively. In contrast, both aerial and root extracts of V. tuna-ekimii exhibited low biological activity and a lower concentration of verbascoside. Pearson correlation analysis demonstrated strong positive associations between verbascoside content and both antioxidant and antiproliferative parameters. However, no significant correlation was found between verbascoside content and α-glucosidase inhibition, suggesting that other phytochemicals may be responsible for the antidiabetic activity. These findings indicate that V. leiocarpum, especially its root extract, may be considered a valuable source of natural antioxidants and antiproliferative agents. The study highlights the pharmacological potential of endemic Verbascum species that have not been comprehensively investigated phytochemically or pharmacologically and supports their further evaluation in the context of phytopharmaceutical research.

Lessons learned from the 2023 Türkiye twin earthquakes: how radiology facilities can respond to disasters

    Aydın, S.; Yurttutan, N.; Kızıldağ, B.; Korkmaz, İnan; Peköz, B.Ç.; Karazincir, S.; Onur, M.R.

On 6 February 2023, back-to-back earthquakes (Mw 7.7 and 7.6) in southeastern Türkiye produced a prolonged mass-casualty incident, placing heavy demands on radiology services nationwide. Operational and clinical lessons were derived from frontline experience to guide emergency imaging under disaster conditions. Surge rostering and rapid staff redistribution were implemented; point-of-care ultrasound was employed for damage-control triage; and “CT-first” pathways were adopted for polytrauma. Workflow continuity during PACS/network outages was maintained with paper logs, downtime order sets, and standardized critical findings phrasing. Disaster teleradiology was activated through streamlined credentialing and ID-agnostic reporting, enabling rapid capacity expansion and support for hospitals beyond the epicentral region. Dominant injury patterns observed were crush injury with acute compartment syndrome; traumatic brain injury; chest trauma with sequential rib fractures, pulmonary contusion and pneumothorax; lumbar-predominant spinal fractures; and pelvic and long-bone injuries. Imaging correlates of rhabdomyolysis and non-lethal pneumomediastinum compatible with the Macklin effect are summarized. Protocol adaptations that preserved throughput and safety included abbreviated, low-dose trauma CT; cautious contrast use in renally vulnerable patients; and corridor-based scanner prioritisation during overflow. On the basis of these observations, a radiology preparedness bundle is presented: redundant power and IT failover; predefined modality-prioritisation matrices; caches of portable US/X-ray for secondary triage; streamlined trauma CT checklists; templated reports that foreground immediate actions; and interoperable teleradiology hooks that can be switched on rapidly. These measures are transferable to earthquakes and other mass-casualty events, sustaining diagnostic capacity, communication, and team alignment under extreme constraints.

Experimental and theoretical investigation of a benzazole-2-carboxamide derivative: Crystal structure, spectroscopic characterization, antiproliferative evaluation, and molecular docking studies

Kılıç, Selçuk; Aktekin, Mine Buga; Turkmenoglu, Burcin; Sert, Yusuf; Caglar, Bulent; Çırak, Çağrı; Algul, Oztekin

In this study, the structural, spectroscopic, and biological properties of a benzazole-2-carboxamide derivatives, N-(1H-benzo[d]imidazole-2-yl)-4-chlorobenzamide, were comprehensively investigated through both experimental and theoretical approaches. First, the crystal structure of the compound was revealed with the help of single-crystal X-ray diffraction (XRD). Subsequently, density functional theory (DFT) calculations were performed using Gaussian 09 W to optimize the molecular geometry, simulate vibrational spectra, predict nuclear magnetic resonance (NMR) parameters, analyze frontier molecular orbitals HOMO (Highest Occupied Molecular Orbital) - LUMO (Lowest Unoccupied Molecular Orbital), and generate molecular electrostatic potential (MEP) surface maps. To enhance the accuracy of vibrational assignments, potential energy distribution (PED) analysis was conducted via the VEDA4 software. Furthermore, theoretical infrared (IR) and NMR data were compared with experimental results, with results from FT-IR, ¹H-NMR, and ¹³CNMR spectra demonstrating strong agreement and validating the computational models. In addition to spectroscopic characterization, the antiproliferative activity of the synthesized compound against human breast cancer cell line MCF7 and a normal mouse fibroblast cell line (L929) was evaluated. Among the tested compounds, those exhibiting the highest inhibitory activity against MCF7 cells were selected for further analysis. In order to explain the underlying molecular interactions, molecular placement studies were carried out with the help of the Schrödinger Maestro software program, which targets the basic protein binding sites to predict binding affinity and interaction patterns. Collectively, this integrated study provides valuable insights into the structural, spectroscopic, and biological behavior of benzazole-based derivatives, highlighting the synergy between experimental techniques and computational modeling in modern drug discovery research.

A Comparative Evaluation of the Therapeutic Effects of Adenosine Triphosphate, Coenzyme Q10, Pyridoxine, and Thiamine Pyrophosphate in a Linezolid-Induced Peripheral Neuropathic Pain Model in Rats

Ozgodek, Habip Burak; Ince, Ramazan; Kahramanlar, Agah Abdullah; Yavuzer, Bulent; Sezgin, Esra Tuba; Mammadov, Renad; Bakan, Nuri; Suleyman, Halis

Background/Objectives: Linezolid is an oxazolidinone antibiotic whose prolonged use is associated with peripheral neuropathy, hyperlactatemia, and metabolic acidosis. These adverse effects are primarily linked to the inhibition of mitochondrial protein synthesis, respiratory chain dysfunction, and oxidative stress. Given the central role of impaired energy metabolism and redox imbalance in drug-induced peripheral neuropathy, therapeutic strategies targeting mitochondrial function are of particular interest. Accordingly, this study aimed to comparatively evaluate the effects of adenosine triphosphate (ATP), coenzyme Q10 (CoQ10), pyridoxine, and thiamine pyrophosphate (TPP) on linezolid-induced peripheral neuropathic pain in rats. Methods: Sixty male albino Wistar rats were assigned to ten groups: healthy (HG); ATP-only (ATPG, 5 mg/kg, intraperitoneally); CoQ10-only (CQ10G, 10 mg/kg, orally); pyridoxine-only (PDXG, 50 mg/kg, orally); TPP-only (TPPG, 20 mg/kg, intraperitoneally); linezolid-only (LZDG, 125 mg/kg, orally); linezolid+ATP (ATLG); linezolid+CoQ10 (CQLG); linezolid+pyridoxine (PXLG); and linezolid+TPP (TPLG). Treatments were administered once daily for ATP, CoQ10, and TPP, and twice daily for linezolid and pyridoxine for 14 days. Oxidative stress indices (MDA, tGSH, SOD, CAT) were quantified in the sciatic nerve using ELISA. Serum lactate dehydrogenase (LDH) activity and blood lactate levels were determined to evaluate metabolic disturbances. Mechanical paw withdrawal thresholds were measured using the Randall–Selitto test both before and after treatment. Results: Linezolid significantly reduced paw withdrawal thresholds and induced oxidative stress, antioxidant depletion, increased LDH activity, and hyperlactatemia. Co-treatment with ATP and CoQ10 attenuated oxidative stress but did not significantly improve linezolid-induced reductions in nociceptive thresholds. In contrast, pyridoxine partially alleviated linezolid-induced neuropathic pain and improved biochemical parameters. Notably, TPP exerted the most robust protective effect, preserving nociceptive thresholds and effectively normalizing oxidative stress and metabolic indices. Conclusions: These findings identify TPP as a promising therapeutic strategy for mitigating linezolid-induced peripheral neuropathic pain by targeting mitochondrial energy metabolism and pyruvate–lactate homeostasis.

Game-based and individualized movement training improves physiological and motor outcomes in young adults with autism spectrum disorder: an experimental study from Türkiye

Aydın, Ahmet Serhat; Söyler, Mehmet; Zileli, Raif; Halmatov, Medera; Gürkan, Alper Cenk; Akcan, İbrahim Orkun; Yılmaz, Coşkun

Background: This study aims to examine the effects of game-based and individualized movement training (GBIMT) program on physiological (body weight, body fat percentage, BMI) and motor outcomes (balance, reaction time, motor speed) in young adults diagnosed with autism spectrum disorder (ASD). Methods: An experimental design with pre-test–post-test control groups was used in the study. A total of 20 participants (10 in the experimental group, 10 in the control group) with a mean age of 22.2 ± 4.8 years were monitored for eight weeks. The experimental group participated in GBIMT sessions for 60 min per day, two days per week, while the control group continued their usual educational routines. Body composition (InBody 270 Body Composition Analyzer, model Plus 270, South Korea), balance (TechnoGym, Cesena, Italy), reaction time (BlazePod Ltd., Israel), and motor speed (Computerized Tapping Test, Neurosoft Inc., USA) parameters were used as measurements. Results: Repeated measures ANOVA results indicated a significant reduction in body weight in terms of the experimental group (p = 0.014, η² = 0.289). Large effect sizes were observed for body fat percentage and BMI; however, the group × time interactions were not found to be statistically significant. A significant improvement was observed in the motor speed parameter within the experimental group (p = 0.032, η² = 0.231). Reaction time decreased by 9.8%, although the group × time interaction stayed within the significance limit (p = 0.063). A trend toward improvement was observed in balance parameters; however, no statistically significant difference was found. Conclusion: The findings indicate that GBIMT produced significant improvements in body composition and motor speed after implementing for eight weeks, and also showed a strong tendency to affect reaction time. As a controlled experimental study conducted in Türkiye, this research offers a unique contribution to the literature by demonstrating the effects of game-based and individualized movement programs on both physiological and motor outcomes in individuals with ASD. UV-synthesized albumin-based doxorubicin nanoparticles induce apoptosis and modulate leukemia surface markers while overcoming drug resistance

Danışman-Kalındemirtaş, Ferdane; Sert, Esra; Kariper, İ. Afşin; Korkut, Işık Neslişah; Erdem-Kuruca, Serap

Although chemotherapeutic agents such as doxorubicin (DOX) are used to treat leukemia, their application is often limited by systemic toxicity, low specificity, and the development of drug resistance. The development of drug delivery systems can overcome the problems associated with conventional delivery methods. In this study, innovative albumin-based UV-DOX-NPs were synthesized by UV irradiation and evaluated for their leukemia inhibitory activity; they represent a novel formulation strategy that has seen limited application to date. Their effects on leukemic Jurkat and THP-1 cells were extensively studied. Advanced techniques such as dynamic light scattering (DLS), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and energy dispersive X-ray analysis (EDX) were used for characterization. UV-DOX-NPs exhibited increased cytotoxicity and significantly lower IC50 values compared to free DOX. Apoptosis rates were also higher in both cell lines following DOX-NP treatment. Flow cytometry revealed a significant downregulation of leukemia-associated surface markers, including CD5, CD7, and CD3 in Jurkat cells and CD7, CD14, and CD33 in THP-1 cells. In addition, UV-DOX-NPs significantly reduced the expression of ABCG2, an important MDR-associated transporter. Our results demonstrate that UV-synthesized, albumin-based UV-DOX-NPs not only enhance antileukemic efficacy but also modulate the leukemic immunophenotype and overcome multidrug resistance. These results provide a promising platform for a more targeted and safer leukemia therapy.

Lower Trochlear Facet Asymmetry Ratio Is Associated with Objective Patellar Instability in a Retrospective MRI Case-Control Study with Patella Alta-Positive Controls

Kuru, Muhammed; Koçkara, Nizamettin; Gökgöz, Mehmet Burak; Gür, Volkan; Yapıcı, Furkan

Background/Objectives: Because patella alta frequently accompanies objective patellar instability, separating trochlear morphology from patellar height is difficult. We evaluated whether the trochlear facet asymmetry ratio was associated with instability among knees with patella alta. Methods: This retrospective case-control MRI study included 123 knees: 43 with objective patellar instability, 43 normal-height controls, and 37 patella alta-positive controls without instability identified by re-screening the eligible control pool. Nine morphologic parameters were measured. The primary analysis compared the instability group with patella alta-positive controls; a supportive analysis used the full cohort. Results: The facet asymmetry ratio showed a graded pattern across groups (instability, 42.7 ± 12.4%; patella alta-positive controls, 59.2 ± 10.7%; normal-height controls, 70.3 ± 11.4%; p < 0.001). In the primary comparison, it had the highest observed apparent area under the curve (0.855), although its confidence interval overlapped with those of quantitative trochlear depth and tibial tubercle-trochlear groove (TT-TG) distance; the Insall–Salvati ratio showed weaker discrimination (0.679). After adjustment for the Insall–Salvati ratio and age, a lower facet asymmetry ratio remained associated with instability (odds ratio per 1-percentage-point increase, 0.903; 95% confidence interval, 0.854–0.956; p < 0.001). This association was attenuated after additional adjustment for quantitative trochlear depth or for tibial tubercle-trochlear groove distance together with Wiberg type. Interobserver reliability was excellent (intraclass correlation coefficient, 0.994). Conclusions: In this selected cohort, a lower trochlear facet asymmetry ratio was associated with objective patellar instability after adjustment for patellar height and age. The internally derived 49% threshold is exploratory and requires external validation before clinical use.

Relactation Success: A Randomized Controlled Trial of Health Promotion Model-Based Education and Follow-Up

Daştan Yilmaz, Ayşe; Yeşilçiçek Çalik, Kıymet

Background: Early breastfeeding cessation due to physiological and psychosocial factors limits long-term maternal and infant health benefits. Relactation, the reinitiation of breastfeeding, is a promising strategy to restore lactation and improve well-being. This study aimed to evaluate the effect of education and follow-up based on Pender’s Health Promotion Model (HPM) on relactation success among mothers who had discontinued breastfeeding or experienced low milk supply. Methods: This randomized controlled trial was conducted between July and December 2023 in a hospital and five family health centers in eastern Türkiye. Seventy-three mothers (38 intervention, 35 control) with infants aged 15 days to 4 months who had ceased breastfeeding or used formula for 2–8 weeks were included. The intervention group received 8 weeks of relactation education and follow-up based on the HPM, whereas the control group received routine postnatal care. Data were collected using a personal information form, the Breastfeeding Self-Efficacy Scale-Short Form (BSES-SF), the Breastfeeding Motivation Scale (BMS), the LATCH tool, and a tool for assessing the level of maternal milk intake (IMDAT). Results: At the final follow-up, 65.8% of mothers in the intervention group achieved complete relactation, compared to 8.6% in the control group. The intervention group also had significantly higher BSES-SF, LATCH, and IMDAT scores. However, no significant differences were found in the BMS intrinsic motivation and identified regulation subscales. Conclusion: HPM-based education and follow-up significantly enhanced relactation success, self-efficacy, and breastfeeding skills.

Pilot Study on the Effects of First-Line Antituberculosis Drugs and Their Combinations on Selected Reproductive Endpoints in Female Rats

Uyar, Elif Esra; Yavuzer, Bulent; Babayeva, Mansura; Yucel, Nurinisa; Gunay, Murat; Suleyman, Halis

Background: The reproductive toxicity of first-line antituberculosis drugs remains poorly understood, particularly when used in combination. Rifampicin, isoniazid, pyrazinamide, and ethambutol are essential in tuberculosis therapy, but their potential influence on female fertility is uncertain. This pilot study evaluated their effects, given alone or in dual, triple, and quadruple combinations, on oxidative stress, endocrine markers, and reproductive outcomes in healthy female rats. Materials and Methods: Ninety-six albino Wistar-type female rats were divided into sixteen groups of six animals each and treated with single, dual, triple, or quadruple regimens of first-line antituberculosis drugs for 28 days. After treatment, two sexually mature males were introduced per group, and therapy continued for seven additional days. Serum malondialdehyde (MDA), total glutathione (tGSH), prolactin, and anti-Mullerian hormone (AMH) levels were measured, and fertility outcomes were evaluated. Results: In single-drug groups, MDA increased and tGSH decreased, but detectable infertility was not recorded. Prolactin remained stable except in the pyrazinamide group, where it declined. Dual-drug regimens increased oxidative imbalance; fertility failure occurred only in pyrazinamide-lacking groups and was accompanied by higher prolactin and lower AMH. Triple and quadruple combinations produced prominent oxidative imbalance. In triple-drug regimens, infertility was lower in pyrazinamide-containing groups than in the pyrazinamide-free group, but this pattern was not maintained in the quadruple regimen. Fertility impairment was not consistently aligned with the degree of oxidative stress and may involve prolactin and AMH alterations. Conclusions: These findings suggest that reproductive impairment under these experimental conditions may involve endocrine alterations and cannot be explained solely by serum oxidative imbalance. Pyrazinamide-associated fertility preservation appeared context-dependent and requires further confirmation in larger mechanistic studies with broader reproductive and endocrine assessment.

Biotechnological application of Escherichia coli outer membrane vesicles for the delivery of Myristica fragrans phytochemicals in pancreatic cancer therapy

Celebi, Ozgur; Baser, Sumeyye; Ari, Betul; Celebi, Demet; Dabanlioglu, Bulent; Genc, Sidika; Taghizadehghalehjoughi, Ali

Bacterial outer membrane vesicles (OMVs) are emerging as natural nanocarriers for cancer therapy due to their ability to deliver bioactive molecules and to penetrate the tumor microenvironment. This study investigated the combined effects of Escherichia coli-derived OMVs and Myristica fragrans extract on pancreatic ductal adenocarcinoma (PDAC) cells. OMVs (50 µg/mL) and M. fragrans extract (IC₅₀: 25 µg/mL) significantly reduced PANC-1 cell viability and increased LDH release after 24 h of treatment (p < 0.01). The combination treatment induced oxidative stress, evidenced by increased total oxidant status and decreased antioxidant capacity. Molecular analysis showed upregulation of PTEN expression, reduced phosphorylated AKT levels, and an increased IL-1β/IL-10 ratio. These changes elevated the Bax/Bcl-2 ratio and activated caspase-3, indicating induction of apoptosis (p < 0.01). Our findings suggest that OMVs can effectively deliver M. fragrans phytochemicals to cancer cells, modulating oxidative balance and triggering apoptosis through the PTEN/AKT signaling pathway. This study provides preclinical evidence that combinations of microbial vesicles and plant extracts may enhance therapeutic efficacy against PDAC.

Background: Hydroxychloroquine (HCQ), widely used in autoimmune and inflammatory diseases, has been associated with cardiotoxicity driven by oxidative, mitochondrial, and metabolic disturbances. However, no comparative evidence exists regarding whether thiamine, thiamine pyrophosphate (TPP), or their combination (TTPC) can mitigate HCQ-induced myocardial injury. Objective: This study examined the biochemical and histopathological effects of thiamine, TPP, and their combination in a rat model of HCQ-induced cardiomyopathy. Methods: Thirty male Wistar rats were assigned to five groups: healthy control, HCQ, thiamine + HCQ, TPP + HCQ, and TTPC + HCQ. Thiamine (20 mg/kg, intraperitoneal), TPP (20 mg/kg, intraperitoneal), or TTPC (20 mg/kg each, intraperitoneal) was administered once daily, followed by HCQ (120 mg/kg, oral, twice daily). After seven days, cardiac tissue was analyzed for MDA, tGSH, SOD, and CAT, while serum TnI, lactate, and LDH were measured from tail-vein blood samples. Cardiac samples underwent histopathological examination. Results: HCQ exposure markedly increased MDA, TnI, LDH, and lactate levels while reducing tGSH, SOD, and CAT, indicating severe oxidative and metabolic insult. Thiamine co-treatment failed to ameliorate these disturbances. Conversely, TPP restored redox balance, attenuated biomarker elevations, and improved cardiac biochemical profiles. TTPC produced comparable improvements but did not exceed those of TPP alone. Histopathologically, HCQ caused pronounced myocyte degeneration and mononuclear infiltration, whereas TPP and TTPC groups showed only mild inflammatory changes with preserved myocardial architecture. Conclusions: HCQ induces a marked redox imbalance accompanied by well-defined histopathological myocardial degeneration. TPP afforded robust cardio-protection, whereas thiamine offered no meaningful benefit. Collectively, these findings position TPP as a biologically plausible, clinically relevant candidate for mitigating HCQ-induced cardiomyopathy.

Aldose reductase (ALR2), a key enzyme in the polyol pathway, plays a significant role in the onset and progression of diabetic complications, rendering it a critical pharmacological target. In this study, a novel series of twenty-four sulfonate ester-functionalized rhodanine derivatives (compounds 1–24) were rationally designed, synthesized via Knoevenagel condensation, and comprehensively evaluated for their inhibitory activity against ALR2. Spectroscopic and spectrometric methods confirmed the structural integrity of the synthesized compounds. In vitro enzyme inhibition assays revealed that all compounds acted as competitive inhibitors, with several analogues, particularly compounds 6 and 8, exhibiting stronger ALR2 inhibition (Ki = 0.43 µM and 0.48 µM, respectively) than the reference drug epalrestat (Ki = 0.98 µM). Structure–activity relationship (SAR) analysis highlighted the critical influence of para-substituted electron-donating (e.g., methyl) and electron-withdrawing (e.g., nitro, halogen) groups on binding potency. Molecular docking of the most potent inhibitor (compound 6) demonstrated a stable binding pose supported by key interactions, including hydrogen bonding with His110 and π–π stacking with Phe122. In silico ADME profiling confirmed favorable drug-likeness properties for all derivatives. Cytotoxicity studies on L929, A549, and RG-2 cell lines revealed that most compounds were less toxic than the reference drug at lower concentrations, with compound 8 showing a promising cytotoxic profile. These findings position rhodanine–sulfonate hybrids as promising scaffolds for the development of next-generation ALR2 inhibitors for the treatment of diabetic complications.

Objective: This study was conducted to examine the effect of smombie (attention distraction and alienation from the environment due to smartphone use) and phubbing (ignoring others by being preoccupied with one’s phone during face-to-face interactions) levels on adolescents’ psychological well-being. Method: This descriptive cross-sectional study was conducted with 626 adolescents living in Turkey between June–September 2025. Data was collected via an online survey administered through Google Forms. The data collection tools used were the Smombie Scale for Adolescents, the General Phubbing Scale, and the Psychological Well-Being Scale. R programming language 4.1.3, G*Power 3.1, and SPSS-22 programs were used in the analysis of the study. Results: According to hierarchical regression analysis results, smombie level explained 15.7% of the variance in psychological well-being (R2 = 0.157; F(1,624) = 117.64, p < 0.001) and was found to be negatively associated with psychological well-being (β = − 0.398; 95% CI [− 0.475, − 0.329]). With the addition of phubbing level, the total variance explained by the model increased to 22.3% (R2 = 0.223; F(2,623) = 90.70, p < 0.001). In this model, both smombie (β = − 0.171; 95% CI [− 0.266, − 0.079]) and phubbing (β = − 0.344; 95% CI [− 0.224, − 0.130]) levels showed statistically significant and negative relationships with psychological well-being; with phubbing being a stronger predictor. Conclusion: This study demonstrates that smombie and phubbing behaviors in adolescents are negatively associated with psychological well-being. The cross-sectional design of this study limits causal inferences. Therefore, future research should employ a longitudinal design to determine the direction of these relationships and their effects over time.

Introduction: Cystic fibrosis (CF) is a life-limiting autosomal recessive disorder caused by mutations in the CFTR gene. In Turkiye, high genetic diversity and consanguinity contribute to a heterogeneous CFTR variant spectrum; however, genotype–phenotype data remain limited. Methods: This retrospective study analyzed 92 Turkish CF patients (36 homozygous, 56 compound heterozygous) to characterize CFTR variant diversity and clinical outcomes. Next-generation sequencing (NGS) was performed and confirmed by Sanger sequencing. Clinical data were analyzed using chi-square and nonparametric tests (p < 0.05). Results: A total of 46 distinct CFTR variants were identified. The most frequent were F508del (24%), followed by Y515X (7.33%), G542X (6%), N1303K (5.33%) and D110H (4%). Meconium ileus (p = 0.017) and CF-related diabetes mellitus (p = 0.013) were significantly more frequent among homozygous patients. Pseudomonas aeruginosa and Staphylococcus aureus were the most prevalent pathogens. CFTR modulator therapy eligibility was significantly higher among compound heterozygous patients (p = 0.007). Non-missense variants were associated with higher sweat chloride levels and more frequent severe pancreatic insufficiency and abnormal fecal fat quantification compared with missense variants (p = 0.037; p = 0.001; p = 0.033). Conclusion: This study highlights the heterogeneous CFTR variants in Turkiye and suggests genotype–phenotype correlations. Homozygous patients tended to show more severe gastrointestinal and endocrine involvement, while compound heterozygotes may exhibit comparatively milder disease features and higher treatment eligibility.
