In this study, the possible uses of glassworts as potential food ingredients and their antiproliferative activity against colorectal adenocarcinoma cells together with their antioxidant and phytochemical profiles were investigated for the first time. MeOH extracts of five different taxa collected from different localities were screened for their antioxidant capacities by DPPH (IC50 2.91-5.49mg/ml) and ABTS (24.4-38.5molTE/gextract) assays. Salicornia freitagii exhibited the highest DPPH radical scavenging activity. LC/MS/MS analysis displayed that vanillic acid and p-coumaric acid were two main phenolic compounds in the extract. Salicorniafreitagii extracts also exhibited high antiproliferative activity against HT-29 (IC50 1.67mg/ml) and Caco-2 (IC50 3.03mg/ml) cells for 72h. Mineral analysis indicated that all the species with different proportions of elemental components contained high amount of cations. These results indicate that investigated glassworts, with their high phenolic and mineral contents and also notable antioxidant and cytotoxic properties, may be utilized as a promising source of therapeutics.

Aim: Early stage breast cancer is being determined at increasing rates associated with an increase in the application of breast cancer screening programs. There is no consensus as to the surgical technique to be applied to these patients. In this study, the surgical techniques applied in our practice were examined together with the clinical characteristics of patients determined with early stage breast cancer. Patients and Methods: A retrospective examination was made of the records of 95 patients diagnosed with early stage breast cancer in our clinic at Suleyman Demirel University Medical Faculty Hospital in 2013-2015. Results: All the patients diagnosed and treated in our clinic for early stage invasive ductal carcinoma were female. Patients were diagnosed at a median age of 57 years. The treatment applied was breast-conserving surgery and sentinel lymph node sampling in 32,6%, breast-conserving surgery and axillary dissection in 8,4%, simple mastectomy and sentinel lymph node sampling in 4,2% and modified radical mastectomy in 54,7%. Within the surgical procedures applied for early stage breast cancer, breast-conserving surgery was applied at the rate of 41,4% and sentinel lymph node sampling at 36,8%. Conclusion: No difference was determined between the patients with early stage breast cancer who underwent breast-conserving surgery and those who underwent mastectomy in terms of general survival and disease-free survival periods. From a cosmetic aspect, the breast-conserving surgery was without doubt more successful and is being increasingly used in our clinical practice.
- MakaleAçık Erişim2017Modulation of mRNA expression and activities of xenobiotic metabolizing enzymes, CYP1A1, CYP1A2, CYP2E1, GPx and GSTP1 by the Salicornia freitagii extract in HT-29 human colon cancer cells

Phase I-II detoxification and antioxidant enzymes are responsible for the detoxification and elimination of activated carcinogens, acting as important biomarkers for chemoprevention. Among them, cytochrome P450s plays a prominent role in the metabolic activation of xenobiotics. The herb Salicornia freitagii (SF) (Amaranthaceae) is known for its anticancer, antioxidant, antidiabetic and antiinflammatory activities. In this study, we determined the bioactive phenolics in the SF methanol extract and investigated its antiproliferative potential in HT-29 human colon cancer cells. We also investigated the modulation of some phase I and II enzyme (CYP 1A1, 1A2, 2E1, GSTP1 and GPx) mRNA expression and enzymatic activities by the SF extract and its major bioactive phenolic compounds. LC/MS-MS analysis showed that the main phenolic compounds of the methanolic SF extract are vanillic acid (48 mu g/100g) and p-coumaric acid (10.8 mu g/100g). SF extract, vanillic acid and p-coumaric acid exhibited high antiproliferative activities in HT-29 cells, with IC50 values of 81.79 mu g/mL, 98.8 mu M and 221.6 mu M, respectively. The mRNA expression levels of CYP1A2 and CYP2E1 were decreased, while those of GSTP1 and GPx in HT-29 cells were increased after application of either the SF extract or vanillic acid. The SF extract by itself also increased the activities of GPx and GSTP1 enzymes 1.68- and 1.49-fold, respectively. Our data indicate that the SF extract and its major bioactive compound, vanillic acid, could exert a modulatory effect on the expression of enzymes that are involved in xenobiotic activation and detoxification pathways in the gastrointestinal tract. For this reason, SF can be considered as a natural source of chemopreventive agents.

The treatment modalities for odontomas are generally depend on the tumors size. Small and medium lesions can usually be removed easily allowing preservation of surrounding anatomical structures. In our study, we reported a conservative surgical treatment of a large complex odontoma. A 19-year-old woman was referred to our clinic after an incidentally observed lesion on her right mandibular angle. The patient was symptom-free at the time of visit. Computed tomography (CT) images showed a mass with a size of 3.5 cm x 3 cm x 2 cm. CT sections and tridimensional images showed partially eroded buccal and lingual cortex. Surgical treatment was indicated with an initial diagnosis of compound odontoma. The lesion removed after sectioning with bur and maxillo-mandibular fixation (MMF) were not thought to be necessary while the buccal and lingual cortexes were mostly reliable for preventing a fracture. In our case, the size of the odontoma was suitable for a conservative treatment method and with this modality we managed to prevent a possible fracture and eliminate the disadvantages of MMF.

In this study, effectiveness of 3 warm-up (WU) modes on 50m free style on child swimmers has been evaluated. In repeated-measures counterbalanced design, 10 male swimmers of a local sports club (10-12 ages) have tried out 50m freestyle after each WU on different days. Each WU took 15 minutes and the intensity of WU has been checked over by Borg's rating of perceived exertion (RPE) 15-point scale. a) no warm-up (NWU): the swimmers sat or laid down for 15 minutes, b) dryland warm-up (DWU): continuous rope-jumping at RPE 12 for 5 minutes, 2 sets 10x15s calisthenics with 15s breathing spaces and continuous rope jumping at RPE 14 for 5 minutes, c) in-water warm up (SWU): continuous freestyle swimming at RPE 12 for 5 minutes, freestyle swimming with 8x15m maximum performance with 30s breathing spaces and continuous freestyle swimming at RPE 14 for 5 minutes. RPE, stroke count and heart rate (HR) have been evaluated together with the best time in swimming performance. Compared with NWU swimming time post DWU was faster, however the fastest swimming time was achieved post SWU. After NWU swimming RPE was significantly higher. Significant difference could not be found in terms of HR and stroke count. Consequently, SWU is better for swimming performance and DWU can be used when swimming pool is not available.

In this study, the finger and hand force developments of the students who have taken selective/applied Tennis and Massage courses at the University have been examined. From the students of the Department of Physical Education and Sport, 19 healthy females and 73 healthy males (age = 21.25 +/- 1.55 years (average +/- SD) who have taken selective Tennis courses;51 healthy males (age = 22.00 +/- 1.04 years (average +/- SD) who have taken Massage courses; and as the control group,16 healthy womenand50 healthy males(age = 21.72 +/- 1.47 years (average +/- SD) have been participated to the study. The age, length, body weight, grip strength of both hands as well as the finger grip strength of the subjects have been recorded. The course schedule has been set as once a week four hours practice for both tennis and massage. Two weeks of the education and training program that takes twelve weeks in total were assigned for theoretical classes. The remaining period of ten weeks was for practice classes and the measurements were performed before and after this ten weeks period. The hand grip strength measurement has been carried out with a Takkei branded hand dynamometer whereas for the measurement of the finger grip strength, a Baseline branded pinch meter has been used. For both the pre-test and final test of the finger grip and hand grip strength measurements, the paired sample t test has been used in terms of in-group comparisons, whereas for the inter-group comparisons, one-way ANOVA has been used. For the significant F values, post hoc Tukey test has been used. The right hand and the left hand grip values of both test groups as well as the values of the control group have been significantly increased between the pre-test and final test. Particularly, preferring the exercises that improve the hand and finger grip strengths would enable a better racket handle grip as well as an improved shot efficiency for tennis. This would also enable masseurs/masseuses to apply an efficient massage therapy with higher pressure.
