Removal of the toxic effects of chlormadinon acetate on the development of Drosophila melanogaster via the use of nordihydroguaiaretic acid

    Altun, D.; Uysal, H.; Ayar, A.; Aşkın, H.

In this study, the effects of chlormadinon acetate (CMA) and CMA + nordihydroguaiaretic acid (NDGA) on various developmental stages of Drosophila melanogaster were investigated. Different concentrations of CMA (1.0; 3.0; 5.0 and 10.0 μM/100 mL medium) and CMA + NDGA as the concentrations of CMA (1.0 + 1.0; 3.0 + 3.0; 5.0 + 5.0 and 10.0 + 10.0 μM/100 mL medium) were carried out during the developmental periods of the flies. When F1progeny of control and application groups were compared, CMA was found to extend the process of metamorphosis and decrease the total offspring numbers. However, these negative effects were inhibited by NDGA treatment at different concentrations. These results suggest that NDGA could effectively inhibit CMA-induced abnormalities in developmental stages of D. melanogaster. It was found that the difference between the groups was significantly important (p < 0.05).

Antioxidant and antimicrobial activity of Turkish endemic Sonchus erzincanicus extracts; [Türkiyede yetişen endemik Sonchus erzincanicus'un ekstraktlarının antioksidan ve antimikrobiyal aktiviteleri]

Mavi, Ahmet; Yiǧit, Nimet; Yiǧit, Demet; Kandemir, Ali

Sonchus erzincanicus (Asteraceae) is a plant species that is endemic to Turkey and grows in Erzincan. Lipid peroxidation inhibition, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and total phenolic content of the methanol and water extracts of the plant were determined. Antimicrobial activity of the extracts was also tested against 107 clinical isolates of human pathogenic microorganism strains belonging to Enterobacter aerogenes, Escherichia coli, Proteus mirabilis, Pseudomonas aeruginosa, Staphylococcus aureus, and Candida albicans using the disc-diffusion method, and the minimal inhibition concentration (MIC) value of each active extract was determined. The extracts were observed to have antioxidant potential, although no better than that of BHT (butylated hydroxytoluene), which is a wellknown commercial antioxidant. The extracts exhibited antimicrobial effects on all the tested microorganism species, except for P. aeruginosa and E. aerogenes. In addition, antifungal activity of the methanol extract against C. albicans was better than that of the water extract (inhibition zone: 25 mm; MIC: 0.156 mg/mL). As a consequence, it was observed that the methanol and water extracts of Sonchus erzincanicus had in vitro lipid peroxidation inhibition and antimicrobial effects against some microorganisms.

Diet-induced obesity suppresses ghrelin in rat gastrointestinal tract and serum

Sahin, Ibrahim; Aydin, Suleyman; Ozkan, Yusuf; Dagli, Adile Ferda; Akin, Kadir Okhan; Guzel, Saadet Pilten; Catak, Zekiye; Ozercan, Mehmet Resat

The aims of the present study were to examine ghrelin expression in serum and gastrointestinal tract (GIT) tissues, and to measure tissue ghrelin levels and obesity-related alterations in some serum biochemical variables in rats with diet-induced obesity (DIO). The study included 12 male rats, 60 days old. The rats were randomly allocated to two groups (n = 6). Rats in the DIO group were fed a cafeteria-style diet to induce obesity, while those in the control group were fed on standard rat pellets. After a 12 week diet program including an adaptation period all rats were decapitated, tissues were individually fixed, ghrelin expression was examined by immunohistochemistry, and tissue and serum ghrelin levels were measured by radioimmunoassay. Serum biochemical variables were measured using an autoanalyzer. When the baseline and week 12 body mass index and GIT ghrelin expression were compared between DIO and control rats, BMI had increased and ghrelin expression decreased due to obesity. The RIA results were consistent with these findings. Serum glucose, LDL cholesterol, and total cholesterol levels were elevated and HDL cholesterol significantly decreased in the DIO group. A comparison of GIT tissues between the control and obese groups demonstrated that ghrelin was decreased in all tissues of the latter. This decrease was brought about a decline in the circulating ghrelin pool. This suggests that rather than being associated with a change in a single tissue, obesity is a pathological condition in which ghrelin expression is changed in all tissues.

The effect of patient education and home monitoring on medication compliance, hypertension management, healthy lifestyle behaviours and BMI in a primary health care setting

    Hacihasanoǧlu, Rabia; Gözüm, Sebahat

Aims: The aim of this study was to determine the effect of anti-hypertensive patient-oriented education and in-home monitoring for medication adherence and management of hypertension in a primary care setting, by providing education on healthy lifestyle behaviours and medication adherence. Background: Hypertension is the third most common cause of death worldwide. Prevalence of hypertension in Turkey is approximately 30% in the general population and 45-50% in population over 50. Design: Randomised controlled study. Methods: This study conducted in Turkey used a study group comprised of 120 subjects (40 Group A, 40 Group B, 40 controls), all previously diagnosed with hypertension and who started medication therapy at least oneyear prior to start of study. The study was conducted between February-November 2006 at public primary health care facilities and homes of the study participants. Participants in Group A and B received a total of six monthly education sessions, four times during clinic visits and two home visits. Medication adherence education for Groups A and B and education about healthy lifestyle behaviours for Group B were administered in a structured and individualised format. The control group was routinely monitored in health care facilities. Results: Healthy lifestyle behaviours and perception of self-efficacy regarding medication adherence showed improvement after education sessions in Groups A and B. Systolic and diastolic blood pressures of subjects in Group A and B showed a significant decrease compared with those of the control group; the blood pressure decrease in Group B was greater than in Group A. Conclusion. Nurses play an important role in uncontrolled hypertension detection and can improve medication adherence and healthy lifestyle behaviours. Relevance to clinical practice. Patient education medication adherence alone and in combination with healthy lifestyle behaviour teaching is an effective tool for blood pressure reduction in the hypertensive population in primary health care settings.

Investigation of the protective effects of vitamin C, Iron and desferrioxamine on heat stress resistance in the model organism Caenorhabditis elegans

Kiliçgün, Hasan; Olgun, Abdullah; Köksal, Zeynep

The aim of this study was to determine the effects of Vitamin C (Vit. C), iron, the iron chelating agent desferrioxamine (DF) and their most effective dose combinations on thermotolerance of Caenorhabditis elegans (C. elegans). Due to the fact that studies on aging and lifespan take long time, thermotolerance was used as a first step analysis for using the time more effectively. The data of this study showed that when compared with the control group, the use of vitamin C, DF and iron alone or their combinations were associated with an increase in the survival of C. elegans under heat shock. However, the treatment with DF and its combinations resulted in a survival rate significantly higher than the other combinations. The results of our study suggested that under conditions of heat stress, treatment of DF has the most significant beneficial effect on survival of C. elegans, suggesting some possible pleiotropic mechanisms other than iron chelation.

Determination of the effects of genistein on the longevity of Drosophila melanogaster Meigen (Diptera; Drosophilidae)

Altun, Deniz; Uysal, Handan; AşkIn, Hakan; Ayar, Arif

In this study, the effects of genistein on the longevity of Drosophila melanogaster were investigated. The effects of different concentrations of genistein (1, 3, 5 and 10 μM/100 mL medium) were separately administered one by one to female and male populations of D. melanogaster for application groups. In the control group, the maximum life span was determined to be 57 days for ♀♀, 46 for ♂♂. The maximum life span for the lowest (1.0 μL) and highest (10.0 μL) application groups among the adult populations of D. melanogaster subjected to genistein were observed to be 54, 50, 40 and 36 days for ♀♀ and 51, 48, 40 and 33 days for ♂♂. These values indicate a negative correlation (R = 0.513 for ♂♂ and R = 0.509 for ♀♀) between the maximum life span of the application groups and changing genistein concentrations.

A novel and one-pot synthesis of new 1-tosyl pyrrol-2-one derivatives and analysis of carbonic anhydrase inhibitory potencies

Alp, Cemalettin; Ekinci, Deniz; Gültekin, Mehmet Serdar; Şentürk, Murat; Şahin, Ertan; Küfrevioǧlu, Ömer Irfan

Here we propose a novel one-pot synthesis of new tosyl-pyrrole derivatives. By means of the new developed method, pyrrole derivatives were synthesized at room temperature in a single step, and a useful method is proposed for the synthesis of similar compounds. Moreover, inhibitions of two human cytosolic carbonic anhydrase (hCA, EC 4.2.1.1) isozymes I and II by 1-tosyl-pyrrole and 1-tosyl-pyrrol-2-on derivatives were investigated. 1-Tosyl-pyrrole, 1-tosyl-1H-pyrrol-2(5H)-one, 5-hydroxy-1-tosyl-1H-pyrrol-2(5H)-one and 5-oxo-1-tosyl-2,5-dihydro-1H-pyrrol-2-yl acetate showed inhibitory action with Ki values in the range of 14.6-42.4 μM for hCA I and 0.53-37.5 μM for hCA II, respectively. All pyrrole derivatives were competitive inhibitors with 4-nitrophenylacetate as substrate. Some new synthesized pyrrole derivatives showed very effective hCA II inhibitory effects, in the same range as the clinically used sulfonamide acetazolamide, and might be used as leads for generating enzyme inhibitors targeting other CA isoforms.

Correlation between antioxidant effect mechanisms and polyphenol content of Rosa canina

    Kilicgun, Hasan; Altiner, Dehen

Rosa canina L. is a member of Rosaceae family, which is well-known for its high phenolic contents. These compounds are known to possess antioxidant, antimutagenic and anticarcinogenic effects. However, they have yet to pass controlled clinical trials for efficacy, and their potential for prooxidant activity is an understudied field of research. In order to estimate the correlations between phenolic contents and antioxidant/proxidant effect mechanisms, different concentrations of R. canina fruit extracts were examined in this study. R. canina showed antioxidant activities at all concentrations with respect to the reducing power, hydrogen peroxide scavenging activity and superoxide anion radical scavenging (O2-) activity assays, whereas a negative correlation was observed with the metal ion chelating activity and free radical scavenging activity [1,1-diphenyl-2-picryl-hydrazil (DPPH) % inhibition] assays at higher concentrations with the phenolic content of R. canina. These results suggest that R. canina may act not only as an antioxidant, but also as a prooxidant with the effects depending on its concentrations.

Ghrelin, nitrite and paraoxonase/arylesterase concentrations in cement plant workers

Aydin, Suleyman; Aydin, Suna; Croteau, Gerry A.; Sahin, Íbrahim; Citil, Cihan

Occupational cement dust exposure has been associated with an increased risk of liver abnormalities, pulmonary disorders, and carcinogenesis. Decreased antioxidant capacity and increased plasma lipid peroxidation have been posed as possible causal mechanisms of disease. Accordingly, this study examined the serum paraoxonase (PON1) arylesterase (AE), ghrelin, HDL-C, LDL-C and serum nitrite (NOx) levels in cement dust exposed workers. Twenty-eight volunteer male cement plant workers and 30 volunteer control male workers, aged 29-54 years, participated. The concentrations of serum PON1, AE, NOx, ghrelin, and HDL-cholesterol and LDL-cholesterol were measured in both groups. PON-1, AE, ghrelin and HDL-cholesterol were lower in the cement plant workers than in controls. Serum nitrite (NOx), and LDL-C levels in cement plant workers were higher (p<0.05) than in the control group workers. No correlation was observed between the serum levels of HDL-cholesterol and PON1 and between HDL-cholesterol and ghrelin. A weak negative correlation was detected between the serum ghrelin and NOx. The study results strongly suggest that HDL-paraoxonase, AE, ghrelin, HDL-C, and high NOx, and LDL-C levels may have a role in disease involving oxidative damage. However, some studies are necessary to address the association between occupational dust exposure and respiratory symptoms.

Altun, Deniz; Ayar, Arif; Uysal, Handan; Kara, Aye Aydan; Nal, Elif Leman

Context:Stachyss species have been used as a medicine for centuries throughout the world. Stachys lavandulifolia Vahl. var. lavandulifolia (Lamiaceae) is of interest to researchers because the constituents such as betulinic acid, oleanolic acid, rosmarinic acid, and ursolic acid are similar to other Stachys species commonly used as an alterative in medicinal preparations. Objective:The present study investigated the effects of water extract (SLE w) and ethanol extract (SLEe) obtained from S. lavandulifolia (SLE) on the longevity of Drosophila melanogaster Meigen. Materials and methods:The effects of different concentrations of SLE (ControlDMSO; 4.0; 12.0 and 20.0 μL/100mL medium) were administered separately to female and male populations of D. melanogaster for control and SLE groups. Results:In all application groups, each population's longevity increased, depending on the concentration of SLE. The mean life-span of the extract groups which are applied with SLE(w) was determined to be shorter than the extract groups which are applied with SLE(e). For example, the maximum mean life-span applied with SLE(w) increased from 31.86±0.92 days to 43.21±1.33 days and the maximum mean life-span applied with SLE(e) increased from 31.86±0.92 days to 49.62±1.62 days in females. Conclusion:These findings demonstrate that the constituents of S. lavandulifolia have great potential as a source for natural health products for D. melanogaster management.

Measurement of secondary school student's test-anxiety levels and investigation of their causes

Gürses, Ahmet; Kaya, Özgür; Doǧar, Çetin; Günes, Kübra; Yolcu, H. Hasan; Keser, H; Ozcinar, Z; Kanbul, S

Test anxiety is concerned about a large portion of our society (students, family environment). The portion affected from the test anxiety is increasing year by year. Test anxiety is not related with studying or studying much. The idea that studying much and getting new knowledge cause anxiety and stress is common. A student being tired by studying much can rest after a brief relaxation and exercise. What creates the anxiety is the fear of being unsuccessful and the thought that is the end of some values because of the failure. Determination of students' test anxiety is very important for the success in applied teaching activities. In this study intending to determine secondary school students' test anxiety, test anxiety scale and data collected from interviews were evaluated from the view of gender, class grades and enrolled educational institutions. After the analyses gained by descriptive statistical techniques and One-Way ANOVA, it was found that the students had mid-level test anxiety, and girls had higher test anxiety level than boys.

Sildenafil is a strong activator of mammalian carbonic anhydrase isoforms I-XIV

Abdülkadir Coban, T.; Beydemir, Şükrü; Gücin, Ilhami; Ekinci, Deniz; Innocenti, Alessio; Vullo, Daniela; Supuran, Claudiu T.

Sildenafil citrate, a phosphodiesterase-5 (PDE5) inhibitor widely used for the treatment of erectile dysfunction was investigated for its interaction with the zinc-enzyme carbonic anhydrase (CA, EC 4.2.1.1), as it has in its molecule a piperazine moiety also found in some CA activators (CAAs). Sildenafil was a potent, low micromolar activator of several CA isozymes, such as CA I, VA and VI (KAs in the range of 1.08-6.54 μM), and activated slightly less the isoforms CA III, IV and VA (KAs of 13.4-16.8 μM). CA isozymes II, IX, XIII and XIV showed activation constants in the range of 27.5-34.0 μM, whereas the least activated isoforms were CA VII and XII (KAs of 72.9-73.0 μM). Sildenafil citrate was also given orally to Sprague-Dawley rats at 1 mg/kg body weight. Red blood cell CA activity was inhibited in the treated animals at 3-5 h post-administration (in the range of 60-85%), probably due to NO/nitrite formed by PDE5 inhibition or by another, unknown mechanism. Whether CA activation by sildenafil has clinical consequences in humans is beyond the scope of the present work and warrants further studies.
