Büyüklü, Mutlu; Kürüm, Akif Turhan; Tatlís, Ersan; Set, Turan

Background: Levosimendan is known with its two-sided effects of strengthening myofibril contraction without increasing myocardial oxygen demand. Anemia is a deteriorating situation that causes increase of drug dosing in patients with heart failure. Objectives: In this study, we compared the effectiveness of levosimendan treatment in decompensated heart failure patients with or without anemia. Methods: Twenty-three anemic patients having class 3 or 4 heart failure according to New York Heart Association (NYHA) and an ejection fraction of below 35% were included to the study. Another 23 patients with the same cardiac diagnosis but without anemia served as control group. Twenty-four hours levosimendan treatment was added to the traditional heart failure treatment of these patients. Samples were taken to measure serum tumor necrotizing factor alpha (TNF-alpha), aminoterminal pro-brain natriuretic peptide (NT-proBNP) and matrix metalloproteinase-1 (MMP-1) levels before and after the administration. Results: There was no significant difference between serum TNF-alpha and MMP-1 levels before and after the treatment (p>0.05). Although NT-proBNP level decreased in both groups after the treatment this was not statistically significant (p=0.531 and p=0.913 for anemia and control groups respectively). Significant restoration of functional capacity was seen in both groups assessed according to NYHA (p<0.001 and p=0.001 for anemia and control groups respectively). Conclusion: Levosimendan treatment shows similar effects in heart failure patients with anemia to that of patients without anemia. However, the early effect of this treatment on TNF-alpha, NT-proBNP and MMP-1 levels is not evident. It provides significant improvement in functional capacity without influence from anemia.

The inhibitory effects of L-adrenaline on lactoperoxidase enzyme purified from bovine milk

Şisecioglu, Melda; Gülçin, Ilhami; Çankaya, Murat; Ozdemir, Hasan

L-Adrenaline belongs to a group of compounds known as catecholamines, which play an important role in the regulation of the physiological process in living organisms. In the present study, the inhibitory effect of L-adrenaline on lactoperoxidase was examined. Lactoperoxidase (E.C.1.11.1.7) was purified from bovine milk with three consecutive steps: Amberlite CG-50 resin, CM-Sephadex C-50 ion-exchange, and Sephadex G-100 gel filtration chromatography. Lactoperoxidase was purified with a yield of 42.18%, a specific activity of 30.33 EU/mg proteins, and 20.77 purification fold. Enzyme purity was determined with SDS-PAGE, where a single band was observed. The Rz (A 412/A280) value for lactoperoxidase was 0.9. The effect of L-adrenaline on lactoperoxidase was determined using ABTS as a chromogenic substrate. The half maximal inhibitory concentration (IC50) value and an inhibition constant (Ki) values for L-adrenaline were 34.5 and 2.26 μM, respectively. L-Adrenaline was found to be a non-competitive inhibitor.

Examination of the tissue ghrelin expression of rats with diet-induced obesity using radioimmunoassay and immunohistochemical methods

Aydin, Suleyman; Sahin, Ibrahim; Ozkan, Yusuf; Dag, Ersel; Gunay, Ahmet; Guzel, Saadet Pilten; Catak, Zekiye; Ozercan, Mehmet Resat

Currently, obesity is an important health problem in all countries, both developed and developing. Dietary habits and neurohormonal imbalances play a critical role in obesity. Circulating amounts of ghrelin, which is a neurohormonal hormone, decrease with obesity and increase with weight loss. Although it is known that both mRNA and peptide version of the ghrelin hormone are expressed in almost all tissues of both humans and animals, it is not known how obesity changes the expression of this hormone in the tissues, with the exception of the gastrointestinal system tissues. Therefore, the objective of the present study is to show how diet-induced obesity in rats changes ghrelin expression in all system tissues, and thus, to shed light on the etiopathology of obesity. The study included 12 male and 12 female 2-month-old Wistar albino species rats. The animals in the control group were fed on standard rat pellet, while those in the experiment group were fed ad libitum on a cafeteria-style diet for 2 months. When their body mass index reached 1 g/cm 2, diet-induced obese (DIO) rats were sacrificed in a sterile environment after one night fasting. Ghrelin localizations in the tissues were studied immunohistochemically using avidin-biotin-peroxidase complex (ABC) method, while tissue ghrelin amounts were analyzed using radioimmunoassay (RIA) method. When the ghrelin amounts in the urogenital system (with the exception of kidney tissues), sensory organs, respiratory system, immune system, skeletal muscle system, cardiovascular system, nervous system, and adipose tissue of rats analyzed by RIA method were compared to those in the control group, tissue ghrelin amounts in the DIO group were found lower. Immunohistochemical findings which showed a similar fall in ghrelin concentrations in the tissues were parallel to RIA results. In addition, ghrelin was shown to be synthesized in the cardiovascular system, heart muscle cells, tails of the sperms, hair follicles, lacrimal glands, tongue, and teeth of rats for the first time in this study and ghrelin syntheses in these tissues were found to decrease in obesity. Nutritional obesity is among the most common causes of obesity and the findings we have obtained through diet-induced obesity will contribute to the illumination of the etiopathology of obesity.

The past and present of paraoxonase enzyme: Its role in the cardiovascular system and some diseases

Aydin, Suleyman; Sahin, Ibrahim; Aydin, Suna; Aksoy, Aziz; Citil, Cihan

Although paraoxonase is synthesized in many tissues including the heart, colon, kidneys, lungs, small intestines and brain, its major locus of synthesis is the liver. PON1 is in close association with apolipoproteins and protects LDL against oxidation. It was reported that PON1 quantities dropped to 40 times lower than normal in cardiovascular diseases and diseases like diabetes, ulcerative colitis, Crohn's disease, chronic renal failure, SLE, Behcet's disease, cancer, hepatitis B, obesity, metabolic syndrome, Alzheimer's and dementia. It is speculated that the concerning decline in serum PON1 amount results from single nucleotide polymorphism in the coding (Q192R, L55M) and promoter (T-108C) sites of the PON1 gene. Additionally, circulating amounts of PON1 are affected by vitamins, antioxidants, fatty acids, dietary factors, drugs, age and lifestyle. This collection attempts to review and examine the past and present studies of paraoxonase and its relation with the cardiovascular system and some relevant diseases.

Life span effects of Hypericum perforatum extracts on Caenorhabditis elegans under heat stress

    Kiliçgün, Hasan; Göksen, Gülden

Background: The beneficial effects of antioxidants in plants are mainly extrapolated from in vitro studies or short-term dietary supplementation studies. Due to cost and duration, relatively little is known about whether dietary antioxidants are beneficial in whole animals′ life span or not. Materials and Methods: To address this question, under heat stress (35°C), Hypericum perforatum was extracted with petroleum ether and the nematodes Caenorhabditis elegans exposed to three different extract concentrations (1mg/mL, 0.1mg/mL, 0.01mg/mL) of H. perforatum. Results: We report that Hypericum perforatum extracts did not increase life span and slow aging related increase in C. elegans. Moreover, one fraction (1mg/mL) increased declines of C. elegans life span and thermotolerance. Conclusion: Given this mounting evidence for life span role of H. perforatum in the presence of heat stress in vivo, the question whether H. perforatum acts as a prooxidant or an antioxidant in vivo under heat stress arises.

Nesfatin-1 and other hormone alterations in polycystic ovary syndrome

Deniz, Rulin; Gurates, Bilgin; Aydin, Suleyman; Celik, Husnu; Sahin, Ibrahim; Baykus, Yakup; Catak, Zekiye; Aksoy, Aziz; Citil, Cihan; Gungor, Sami

Polycystic ovary syndrome (PCOS) is commonly characterised by obesity, insulin resistance (IR), hyperandrogenemia and hirsutism. Nesfatin-1 a recently discovered hormone, acts upon energy balance, glucose metabolism, obesity and probably gonadal functions. This study was to evaluate the circulating levels of nesfatin-1 in patients with PCOS (n = 30) and in age and body mass index (BMI)-matched controls (n = 30). PCOS patients had significantly lower levels of nesfatin-1 (0.88 ± 0.36 ng/mL) than healthy controls (2.22 ± 1.14 ng/mL). PCOS patients also had higher gonadotropin and androgen plasma concentrations, Ferriman-Gallwey scores, blood glucose levels and a homeostasis model of assessment-IR index (HOMA-IR) index than in healthy women. Correlation tests in PCOS subjects detected a negative correlation between nesfatin-1 levels and BMI, fasting blood glucose, insulin levels and a HOMA-IR index. Lower nesfatin-1 concentration may plays a very important role in the development of PCOS.

The inhibition of two human cytosolic carbonic anhydrase (hCA, EC 4.2.1.1) isozymes I, II and human serum isozyme VI, with a series of tosylited aromatic amine derivatives was investigated. The KI ranges of compounds 1-14 and acetazolamide against hCA I ranged between 1.130 and- 448.2 μM, against hCA II between 0.103 and- 14.3 μM, and against hCA VI ranged between 0.340 and- 42.39 μM. Tosylited aromatic amine derivatives are thus interesting hCA I, II and VI inhibitors, and might be used as leads for generating enzyme inhibitors eventually targeting other isoforms which have not been assayed yet for their interactions with such agents.

Genotoxic evaluation of selective serotonin-reuptake inhibitors by use of the somatic mutation and recombination test in Drosophila melanogaster

Gürbüzel, Mehmet; Oral, Elif; Kizilet, Halit; Halici, Zekai; Gulec, Mustafa

This study evaluated different concentrations of selective serotonin-reuptake inhibitors (citalopram and sertraline) for genotoxicity by use of the somatic mutation and recombination test (SMART) in Drosophila melanogaster. Three-day-old larvae, trans-heterozygous for the multiple wing hairs (mwh) and flare (flr3) genes were treated with these two compounds. Two recessive markers were located on the left arm of chromosome 3, i.e. 'multiple wing hairs' (mwh) in map position 0.3 and 'flare-3' (flr3) at 38.8, while the centromere was located in position 47.7. SMART is based on the loss of heterozygosity, which may occur through various mechanisms, such as mitotic recombination, mutation, deletion, half-translocation, chromosome loss, and non-disjunction. Genetic changes occurring in somatic cells of the wing's imaginal discs, cause the formation of mutant clones on the wing blade. The results of this study show that citalopram had a genotoxic effect in the Drosophila SMART. Sertraline, however, did not show any genotoxic effect in balancer heterozygous wings. This study concluded that more information is needed to be certain regarding the mutagenic effects of sertraline.

Hierarchical factor structure of the Turkish version of the posttraumatic growth inventory in a normal population; [Travma sonrası büyüme envanteri'nin Türkçe versiyonunun normal toplumda hiyerarşik faktör yapısı]

Kaǧan, Mücahit; Güleç, Mustafa; Boysan, Murat; Çavuş, Hayati

AIM: Negative consequences of traumatic experiences, such as depression, anxiety, or dissociative symptoms, etc. have been reported by many researchers. However, it is proposed that stressful events not only lead to poor psychological outcomes but also may trigger positive changes. Several instruments gauging posttraumatic benefits have been developed to examine the effects of factors that may promote positive psychological outcomes in the aftermath of stressful events. The Posttraumatic Growth Inventory (PTGI) is one of the prominent instruments that assess posttraumatic positive changes. In this study we aimed to assess psychometric properties of the Turkish version of the PTGI. METHOD: This study differed to an extent from previous studies concerning the features of the PTGI. We used a dispositional form of the instrument in a sample recruited from high school and university students. Our data were collected from 723 volunteers. 367 subjects were males (50.76%) and 356 subjects were females (49.24%). Also we did not specify any selection criteria in recruiting subjects owing to their adverse life experiences with a presumption that stressful life events are not uncommon in normal population. We administered to participants a dispositional form of the Posttraumatic Growth Inventory and the Personal View Survey-III. Item statistics for the measure were computed. We performed an explanatory factor analysis by using principal components with promax rotation and a confirmatory factor analysis by using structural equation modeling. Since the factor inter-correlations were higher than.40 we computed Schmid-Leiman transformation to obtain second-order general factor loadings. Inner consistencies and 15-day test-retest intracorrelations were calculated. RESULTS: Item discrimination indexes ranged from.28 to.72. Promax rotated principal components analysis pointed out a three-factor structure. It was found in model testing with structural equation modeling that three-factor structure was valid for the Turkish version of the PTGI. After computing second-order factor loadings, we detected that general factor accounted for 64 percentage of the total variance. Three subscales of the measure were the Changes in Self-Perception, Changes in Philosophy of Life, and Changes in Relationship. Internal consistency for the Changes in Self-Perception subscale was 0.88, for the Changes in Philosophy of Life subscale was 0.78, for the Changes in Relationship was 0.77, and for the overall items was 0.92. 15-day test-retest intra-correlation for the composite scores was 0.83 and intra-correlations for the subscale scores ranged from 0.70 to 0.85. CONCLUSION: Turkish version of the measure revealed a three-factor first-order structure. However, it seems that the concept of posttraumatic benefits measured by the PTGI has a tendency to represent a unidimensional psychological construct in Turkish sample. Since the three-factor structure was validated, three-subscales may also be used to make an extensive assessment instead of composite scores. The PTGI is a valid and reliable measure to be used in research purposes among Turkish individuals.

Investigation of radioactivity level in soil and drinking water samples collected from the city of Erzincan, Turkey

    Yalcin, P.; Terzi, M.; Bastug, A.; Taskin, H.; Varinlioglu, A.; Tasdelen, B.; Kam, E.; Bozkurt, A.

This study is part of an effort to assess the level of background radiation for Erzincan Province of eastern Turkey. Radionuclide activity concentrations in soil samples were measured through gamma-ray spectrometry and the average activities were determined as 8.93, 11.39, 281.94, and 9.52 Bq/kg for the radionuclides 238U, 232Th, 40K, and 137Cs, respectively. The average annual effective dose from these natural radioactivity sources ( 238U series, 232Th series and 40K,) was calculated to be 27.9 μSv. Radioactivity levels in drinking and potable water samples were studied using a multi-channel low level proportional counter. The average gross alpha activity concentration was found to be 0.0477 Bq/L (min. 0.007 Bq/L; max. 0.421 Bq/L) and the average gross beta activity was measured as 0.104 Bq/L (min. 0.008 Bq/L; max. 1.806 Bq/L). These values lead to an average annual effective dose of 9.75 μSv from the alpha emitters and 56.34 μSv from the beta emitting radionuclides in water. The radioactivity levels in the water samples investigated were found to comply with the reference levels recommended by WHO and the regulations set forth by the Turkish Health Ministry.

Comparison of central corneal thickness measurements by Galilei Dual-Scheimpflug Analyzer® and ultrasound pachymeter in myopic eyes

Yeter, Volkan; Sönmez, Baris; Beden, Ümit

    ■ BACKGROUND AND OBJECTIVE: To compare the central corneal thickness (CCT) measurements by Galilei Dual-Scheimpflug analyzer (GSA) (Ziemer Group, Port, Switzerland) and ultrasound pachymeter (UP) in myopic eyes. ■ PATIENTS AND METHODS: In this prospective study, CCT of 161 myopic eyes of 81 refractive surgery candidates (35 female, 46 male; mean age: 23.18 ± 4.08 years) were measured by GSA and UP consecutively. The data were analyzed statistically by paired t test, Bland-Altman plot, and Pearson correlation test to assess the agreement of the measurements. ■ RESULTS: The mean CCTs obtained by GSA and UP were 559.85 ± 30.87 and 560.41 ± 34.45 μm, respectively. No significant difference was found between the measurements (P = .684) and regression analysis showed a high correlation between the measurements obtained with both devices (r = 0.86; P < .001). A strong agreement between the two methods was also found by Bland-Altman plot. ■ CONCLUSION: In myopic eyes, the CCT measurements of GSA and UP are similar and highly correlated. Because of high agreement between these devices, the GSA is a non-contact method that may be an alternative to UP for measurement of CCT.

Radiological characterisation of Artvin and Ardahan provinces of Turkey

    Kucukomeroglu, B.; Yesilbag, Y.O.; Kurnaz, A.; Celik, N.; Cevik, U.; Celebi, N.

Indoor radon concentration measurements were carried out and corresponding annual effective doses due to exposure to indoor radon were determined in Artvin and Ardahan provinces located in the eastern part of Turkey. The measurements were performed for four seasons in order to determine the seasonal fluctuations mostly observed in indoor environments. Indoor radon concentration values were observed to range from 21 to 321 Bq m-3 for the Artvin province and from 53 to 736 Bq m-3 for the Ardahan province. It was observed that minimum indoor radon concentration values were obtained in summer, while the highest ones were observed in winter. Indoor radon concentration values of the current study were compared with those of other provinces in Turkey. As elevated indoor radon concentrations are mostly correlated with high 238U activity concentrations in soil, a total of 57 and 33 soil samples were collected from the Artvin and Ardahan provinces, respectively, to determine 238U activity concentration as well as the concentration of 232TH and 40K-naturally occurring radionuclides. It was also observed that soil samples collected from the study areas contained 137Cs as an artificial radionuclide.

Ghrelin expression of endometrium hyperplasia and endometrioid carcinoma

Dagli, Adile F.; Aydin, Suleyman; Kocdor, Hilal; Gurates, Bilgin; Sahin, Ibrahim; Catak, Zekiye; Ozercan, Mehmet R.; Ozercan, Ibrahim. H.

Background. Endometrium carcinoma ranks fourth among female carcinomas. Therefore, early diagnosis of endometrium pre-malignant lesions is emphasised, and attempts are made to identify the risk factors. Since hyperplasias, particularly those with atypia, are held responsible for the development of the most common endometrium carcinomas, it is important to definitely distinguish between well-differentiated carcinomas and hyperplasia with atypia. In the present study, we aimed to explore whether ghrelin expression had a role in distinguishing between benign, pre-malignant and malignant lesions of endometrium. Methods. Tissue ghrelin expressions of a total of 60 cases, who were diagnosed in the Pathology Department Laboratory of Fiirat University Medical School, and of whom 10 were in the proliferation phase, 10 had simple hyperplasia without atypia, 10 had simple hyperplasia with atypia, 10 had complex hyperplasia without atypia, 10 had complex hyperplasia with atypia and 10 had endometrioid carcinoma cases, were examined using immunohistochemical method. Additionally, tissue samples were homogenised to analyse tissue ghrelin levels in the supernatants according to RIA method. Samples from the parotid glands were used as positive control for ghrelin. Cells that exhibited cytoplasmic staining with ghrelin antibody were evaluated as positive. Results. Immunohistochemical examination showed that ghrelin expression increased markedly in the proliferation phase, relative to hyperplasias and carcinoma. These results were parallel to ghrelin levels in tissue supernatants. Immunohistochemical and RIA analysis results indicate that ghrelin expression either markedly decreases or is entirely depleted in endometrial carcinomas. Conclusions. Therefore, we think that ghrelin expression can be useful in differentiating not only endometrium carcinomas from benign lesions but also complex hyperplasias with atypia, which pose diagnostic difficulties.

Materials and Methods: The study sample included patients with physical illnesses (n = 124) and healthy controls (n = 209). Symptoms of stress, self-perception, interpersonal style, and anger were evaluated using the Stress Symptoms Scale, Social Comparison Scale, Interpersonal Style Scale, and Multidimensional Anger Questionnaire, respectively. The role of self-perception, interpersonal style, and anger in stress experienced in the patients and controls was compared. Results: The patients had higher stress symptoms, perceived themselves more negatively, had more problematic interpersonal styles, and more intense anger than the controls. The higher stress symptoms in the patients and the lower symptoms in the controls was predicted by 4 common variables3/4dissatisfaction with life, dissatisfaction with interpersonal relationships, negative self-perception, and aggressive expression of anger. Another predictive variable in the psychosomatic group was age, whereas in the control group gender and internalized anger were predictive variables. Conclusion: As the physical illnesses had by those in the patient group are stress-related, inclusion of psychosocial interventions in treatment protocols, such as communication skills, and stress and anger management training, and interventions aimed at increasing a positive self-perception might improve their QoL.
