Corneal neovascularization (CV) is a vision-threatening condition characterized by the pathological ingrowth of blood vessels. While corticosteroids and anti-VEGF agents are standard treatments, their prolonged use is associated with significant ocular side effects. This study aimed to develop and optimize nanosuspensions of Apigenin (APG) and Apigenin-7-O-glucoside (APG 7) to enhance their ocular bioavailability and evaluate their efficacy in the treatment of CV. APG and APG 7 were isolated from Petroselinum crispum . Nanosuspensions were formulated using an anti-solvent precipitation method, optimized via a 23 factorial design considering PVA concentration, Span 80 concentration, and homogenization speed. The physical properties, including particle size, PDI, and zeta potential, were characterized. In vivo efficacy was assessed over a 7-day treatment period in an alkali-burn induced rat CV model through clinical scoring, histopathology, and immunohistochemical analysis of VEGFA and IL-6 expression. The optimized APG nanosuspensions exhibited a mean particle size of ∼166.9 nm with high stability. In vivo studies demonstrated that 0.05% APG nanosuspension significantly inhibited corneal angiogenesis, reduced stromal edema, and downregulated VEGFA and IL-6 expression compared to the untreated injury group (p < 0.05). Conversely, APG 7 formulations failed to show significant therapeutic efficacy, likely due to limited corneal permeability attributed to the glycoside moiety. The 0.05% APG nanosuspension presents a promising, non-invasive therapeutic alternative for CV management, offering effective anti-angiogenic activity with improved ocular delivery characteristics.

Gözcü, Sefa; Akşit, Zeynep; Dursun, Ahmet
Objective: The purpose of this study was to characterize the chemical composition of the essential oil (EO) derived from the cones of Cupressus arizonica Greene collected from Erzincan and to evaluate its antibacterial potential against a range of pathogenic microorganisms, including strains not previously investigated for this species. Methods: The essential oil was obtained from fresh cones via hydrodistillation for 4 hours. Chemical profiling was performed using Gas Chromatography-Mass Spectrometry (GC-MS). The antibacterial activity was assessed against six pathogenic bacteria using the disc diffusion method, with imipenem serving as a positive control. Results: GC-MS analysis identified 35 compounds, representing 93.83% of the total oil. The major components were α -pinene (53.02%), 3-carene (13.31%), α -myrcene (4.54%), sabinene (4.24%), and limonene (3.47%). The oil exhibited selective antibacterial activity, with inhibition zones recorded for Salmonella enterica (10.0 ± 0.3 mm), Clostridium perfringens (8.1 ± 0.5 mm), and Bacillus cereus (8.0 ± 0.6 mm). However, no inhibitory effect was observed against Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli. Conclusion: This study identifies a distinct α-pinene/3-carene chemotype of C. arizonica influenced by the regional flora of Erzincan. Notably, this research represents the first report of the inhibitory efficacy of C. arizonica cone essential oil against the anaerobic pathogen Clostridium perfringens. The findings suggest that this essential oil could be a promising natural candidate for targeted applications against certain foodborne and anaerobic pathogens.
- ArticleAçık Erişim2026Comparative evaluation of the cytotoxic and antioxidant effects of apricot (Prunus armeniaca L.) fruit and traditionally prepared apricot jam: Differential impacts on healthy and cancer cell lines
Güven, Zühal Bayrakçeken; Gülmüş, Cennet
Cancer is a complex, multifactorial disease in which the toxic side effects of drugs hinder patients' quality of life and adherence to treatment. During this period, people try to cope with the negative effects of treatment and disease by consuming natural foods. This study compared the antiproliferative and antioxidant effects of apricot (Prunus armeniaca L.) fruit and traditionally prepared jam. Cytotoxic effects were investigated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method in L929, A549, CaCo-2, Hep3B, MCF-7 and PC-3 cell lines. Antioxidant capacity was determined by DPPH, CUPRAC, and TEAC methods. While jam exhibited cytotoxic effects in healthy cells at concentrations exceeding 400 µg/mL, the fruit aqueous extract did not reduce cell viability. In cancer cells, the fruit aqueous extract showed a concentration-dependent cytotoxic effect, with 64.60% and 58.8% cytotoxicity observed against the Hep-3B cell line at 800 and 1600 µg/mL, respectively. Notably, jam was found to promote cancer cell proliferation at higher concentrations, a critical observation that contrasts sharply with the fruit extract's antiproliferative effects. In all experiments, the aqueous extract showed a higher antioxidant effect than the jam. The results indicate that the antiproliferative and antioxidant effects of jam compared to fruit were reduced due to added sugar and high temperature.

Koç, Alparslan; Polat, Eyup Can
Objective: Clinical pharmacy is a health field that is spreading all over the world. Numerous studies have demonstrated that clinical pharmacy services offered to healthcare providers lead to improved therapeutic outcomes. Intensive care units are the primary clinics where clinical pharmacists work. Due to factors such as variable clinical conditions of patients and the use of multiple medications, drug-related problems are frequently encountered in intensive care units. This study aims to conduct a comprehensive bibliometric analysis of research on clinical pharmacy in intensive care units by examining productivity trends, citation impact, collaboration networks, and thematic evolution using VOSviewer software. Methods: We used all relevant studies from the Web of Science database published between 1981 and 2024 that included the keywords "intensive care unit" and "clinical pharmacy" for this analysis. The analyses were performed using the VOSviewer (v1.6.20) software. Results: A total of 866 publications were analyzed. The country with the most publications was the United States. Original research articles were the most common publication type, and the most cited article was by Tsuji et al. (n=535). The most frequently used keywords were "critical care", "intensive care unit", "clinical pharmacy", and "clinical pharmacist". Conclusion: Our results show that research on clinical pharmacy in intensive care units is increasing steadily every year and that the services provided in these settings have a beneficial effect on treatment outcomes. Therefore, our study will be an important guide for researchers investigating clinical pharmacy services in intensive care units.

This study comparatively evaluated the phytochemical and biological profiles of Origanum leptocladum and O. vulgare subsp. hirtum (Lamiaceae). Aqueous extracts yielded more material than methanolic ones, with O. leptocladum showing superior extractability. GC-MS analysis revealed distinct chemotypes: O. leptocladum was rich in cis-sabinene hydrate and p-cymene, whereas O. vulgare was dominated by carvacrol. Methanol extracts of O. leptocladum exhibited the highest alpha-glucosidase inhibition (90.10%) and antioxidant capacity, correlating with elevated phenolic content. Molecular docking indicated strong binding affinities of carvacrol, cis-sabinene hydrate, and p-cymene to alpha-glucosidase, alpha-amylase, AChE, and BChE, implying multitarget inhibitory potential. ADME predictions confirmed favourable pharmacokinetic properties and BBB permeability, especially for carvacrol and cis-sabinene hydrate. This first report on alpha-glucosidase and alpha-amylase inhibition by O. leptocladum underscores its promising antidiabetic and neuroprotective potential, supporting its value for pharmaceutical and nutraceutical development.

Myriophyllum spicatum L. methanolic extract was examined through integrated biochemical, phytochemical, computational analyses. The extract exhibited strong alpha-glucosidase inhibitory activity with an IC50 value of 198.54 & micro;g/mL, demonstrating markedly higher potency than reference inhibitor acarbose (IC50= 4738.12 & micro;g/mL), while no inhibitory activity was detected against alpha-amylase under tested conditions. In anticholinesterase assays, extract showed acetylcholinesterase inhibition with an IC50 value of 9.70 & micro;g/mL, whereas standard inhibitor donepezil exhibited a significantly stronger activity (IC50= 0.30 & micro;g/mL). No inhibitory effect was observed against butyrylcholinesterase for the extract, while donepezil displayed potent inhibition (IC50= 15.40 & micro;g/mL). Extract showed strong ABTS center dot+ scavenging activity (85.7% at 40 & micro;g/mL) but weak DPPH center dot activity (25.6% at 25 & micro;g/mL), suggesting higher affinity for hydrophilic radicals. High total phenolic, flavonoid and tannin contents correlated with antioxidant potential. LC-MS/MS revealed thirteen phenolics, mainly vanillic acid, gallic acid and cyanidin-3-O-glucoside. Docking studies supported selective alpha-glucosidase binding of vanillic acid.
- ArticleAçık Erişim2026Antioxidant and Antimicrobial Activities and Phenolic Composition of Vaucheria aversa Isolated from the Tigris River (Diyarbakır, Türkiye)
Uç, Osman; Pabuçcu, Koksal
This study aims to evaluate the pharmaceutical potential of Vaucheria aversa isolated from the Tigris River through the assessment of its antioxidant capacity, antimicrobial activities, and phenolic compound composition. In this study, samples collected from benthic habitats were purified using a mechanical isolation method. The phenolic and flavonoid contents of the prepared extracts were measured by spectrophotometric methods, and their phenolic profiles were determined using LC-MS/MS analysis. Antioxidant activities were assessed using FRAP, DPPH, and ABTS assays, while antimicrobial activities were tested on E. coli, S. aureus, and C. albicans using the agar well-diffusion method. The total phenolic content of V. aversa was determined to be 64.28 f 2.11 mg GAE/g, while the flavonoid content was 15.21 f 0.89 mg QE/g. Regarding antioxidant activities, the FRAP value was 65.55 f 3.18 mg TE/g, the DPPH IC 50 value was 38.25 f 1.41 & micro;g/ml, and the ABTS IC50 value was 16.39 f 1.02 & micro;g/ml. LC-MS/MS analysis revealed that cinnamic acid and gallic acid were present at higher levels compared to other phenolic compounds. In antimicrobial assays, ethanol and methanol extracts exhibited strong inhibition zones against E. coli and C. albicans. The activity of V. aversa against Gram (-) negative bacteria and fungi was found to be particularly noteworthy compared to other Vaucheria species. Its antioxidant and antimicrobial capacities are presumed to be directly associated with its phenolic compound content. The findings of this study indicate that in order to evaluate V. aversa as a natural antioxidant and antimicrobial agent in pharmaceutical research, further compound isolation and in vivo investigations are required to elucidate its effective pharmaceutical potential in detail.

Objective: Allergic conjunctivitis is one of the most prevalent conditions affecting the eyes, and various medications are used in its treatment. In this study, olopatadine and verbascoside were used in combination. Methods: A factorial design was employed to evaluate the effects of Poloxamer 407 and sodium alginate. The impact of polymer concentration on parameters such as viscosity, gelation time, mucoadhesion, pH, and gelation temperature–considered indicators of ideal formulations–was assessed. Results: The optimal formulation, determined through the factorial design, included 0.5% sodium alginate and 20% poloxamer 407. The obtained values were viscosity at 25 °C = 7451.8 cP, viscosity at 35 °C = 11367.3 cP, pH = 7.12, gelation temperature = 30 °C, gelation time = 52 s, and mucoadhesion = 0.301 N. Cytotoxicity tests on ARPE-19 and L929 cells, along with irritation studies, indicated acceptable safety profiles. In vitro release tests demonstrated an initial burst release of verbascoside and olopatadine during the first 2 h, followed by a controlled release over the subsequent 8 h. Mathematical analysis of the release kinetics revealed a good fit with the Weibull model. No adverse effects were observed in rats during Draize’s in vivo test. Conclusions: Consequently, the drug-loaded in situ gel formulation may be a promising option for treating allergic conjunctivitis.

Ozdemir Sarikaya, Bengul; Gul, Huseyin Fatih; Gozcu, Sefa; Yayla, Muhammed
The present study aimed to investigate the therapeutic effects of Malus floribunda fruit ethanol extract on metabolic parameters and insulin signalling in a letrozole-induced PCOS-associated insulin resistance model. 60 female Sprague-Dawley rats were randomly allocated into five groups: Healthy Control, IR, IR + Metformin (300 mg/kg), IR + MF250, and IR + MF500. Treatments were administered orally for 15 days. Fasting insulin, HOMA-IR, OGTT, lipid profile, and reproductive hormones were assessed. Histological and IHC analyses of InR, IRS-1, and IRS-2 were performed on muscle and adipose tissues. The IR group exhibited significantly elevated insulin (p<0.001), HOMA-IR (p<0.001), testosterone, triglycerides, LDL, and total cholesterol levels, with reduced HDL and oestrogen levels compared to controls. Treatment with M. floribunda, particularly at 500 mg/kg, significantly improved insulin levels (p<0.001), HOMA-IR (p<0.001), lipid profile, and hormonal parameters. IHC analysis revealed suppressed InR, IRS-1, and IRS-2 expression in muscle and adipose tissues of the IR group, which was restored following M. floribunda treatment. M. floribunda extract administration, particularly at 500 mg/kg, significantly improved glucose homeostasis, lipid metabolism, hormonal balance, and insulin signalling pathway components (InR, IRS-1, IRS-2) in a letrozole-induced PCOS-associated insulin resistance model. These findings suggest that M. floribunda may serve as a promising alternative therapeutic agent for insulin resistance management.
