Başaran, Eyüp; Çakmak, Reşit; Türkmenoğlu, Burçin; Akkoc, Senem; Köprü, Semiha

The current study focuses on the synthesis and characterization of six benzenesulfonamide-based Schiff base derivatives (7–12) with various electron-withdrawing and electron-donating substituents (−F, -CI, -Br, −CH3, and −OCH3) and the assessment of their antiproliferative activities against human lung (A549) and liver (HepG2) cancer cell lines using in vitro and in silico approaches. The structures of the synthesized compounds (7–12) were elucidated by elemental analysis and FT-IR, 1D (1H, 13C, APT, and DEPT-135), and 2D (HMQC and HMBC) NMR spectroscopies. The cytotoxic activities of the targeted compounds were determined at various concentrations against these cancer cell lines for 72 h, using the MTT method. The targeted molecules (7–12) demonstrated remarkable antiproliferative activities, with IC50 values ranging from 6.032–9.533 μM against the A549 cell line and 5.244–9.629 μM against the HepG2 cell line. These compounds showed activities at lower or very similar concentrations to cisplatin against the A549 cell line and at much lower concentrations than cisplatin against the HepG2 cell line. Among them, compounds 10 and 12 were found to be more effective against A549 and HepG2 cells, respectively, than cisplatin. These compounds were analyzed by interacting with the 1BNA, 4HJO, and 4ASD crystal structures in molecular docking studies. The docking score of 4ASD-compound 12 interaction was calculated as −4.045 kcal/mol, 4HJO-compound 10 interaction was calculated as −5.179 kcal/mol and 1BNA-compound 10 interaction was calculated as −8.571 kcal/mol and it was determined that these compounds were theoretically better than Cisplatin. In the present study, ADME data were estimated using the web tool SwissADME. With ADME, it was calculated that the logP value of compounds 7–12 was less than 5, the HBD number was 1, the HBA number was 7 or 8, and the molecular weight was less than 500. Properties such as the electrophilic index and chemical hardness of the designed compounds were examined by density functional theory (DFT) using B3LYP/6-311G**. In conclusion, these compounds have emerged as promising new anti-cancer drug candidates.

Ergül, Emine Esra; Özdemir, Elif Can; Ayçiçek, Hilal Büşra; Hayme, Serhat; Kemer, Aysel Özge; Sağlam Akkaya, Gonca

Introduction: Grip strength represents a critical, objective indicator of functional capacity in daily activities. This study sought to elucidate the impact of breast cancer-related lymphedema on grip strength and to explore the underlying factors contributing to its decline. Materials and method: This is a retrospective study including patients over the age of 65 who presented to our outpatient clinic after developing lymphedema following breast cancer treatment. Hand grip strength was measured using a hand dynamometer in patients with lymphedema, and data on lymphedema stage and clinical parameters were collected. Results: In older adults with lymphedema, grip strength was significantly reduced on the affected side (median: 8.0 kg [3–21]) compared to the non-affected side (median: 16.5 kg [10–29]; p < 0.001). Patients with more advanced stages of lymphedema were found to have lower grip strength on the affected side compared to those with earlier stages. In older adults with lymphedema, grip strength was also correlated with the subscales of quality of life and with fatigue, both considered important clinical outcomes. Conclusion: Our findings highlight that lymphedema in older adults is associated with clear functional impairment, as evidenced by reduced grip strength on the affected side and a progressive decline across disease stages, underscoring the importance of early intervention to preserve hand function.

Monosodium glutamate (MSG), a widely used food additive, has been associated with various health concerns, including potential reproductive toxicity. This study investigated the protective effects of black garlic (BG) ethanol extract against MSG-induced ovarian damage in rats. Thirty-two female rats in estrus were randomly divided into four groups (n = 8 per group): control (saline), BG (250 mg/kg BW), MSG (4 mg/g BW), and BG+MSG (combined treatment). Treatments were administered daily for 14 days. Ovarian tissues were collected for histopathological, immunohistochemical (IHC), and biochemical analyses. Histopathological examination revealed a significant reduction in cystic follicles in the BG+MSG group compared to the MSG group (p < 0.0001). IHC analysis showed decreased immunoreactivity of endothelin-1 and endothelin-2 in the BG+MSG group compared to the MSG group (both p < 0.01). Biochemical assays demonstrated significantly increased follicle-stimulating hormone (FSH), luteinizing hormone (LH), and estradiol levels in the BG+MSG group compared to the MSG group (all p < 0.05), while progesterone levels were significantly lower in the MSG group compared to the BG+MSG group (p < 0.05). These findings suggest that BG ethanol extract may mitigate MSG-induced ovarian dysfunction in rats by alleviating degenerative changes in follicles and modulating hormonal levels. This study provides insights into potential natural interventions for MSG-related reproductive toxicity.

Sahin, Dicle; Kepekci, Remziye Aysun; Türkmenoğlu, Burçin; Akkoc, Senem

Three new thymol-based molecules were synthesized and evaluated as anticancer, antimicrobial and antioxidant agents. Liver, colon, lung and prostate cancer cell lines were utilized in cytotoxicity tests. The results demonstrated that synthesized molecules had a cytotoxic effect against the screened cell lines. One of the molecules (4a) was found to have a higher efficacy towards the colon cancer cell line (DLD-1) with an IC50 value of 12.39 µM and the other (4c) towards the prostate cancer cell line (PC3) with an IC50 value of 7.67 µM than the positive control drug cisplatin. To assess the antimicrobial activity of molecules (4a–c), Gram-positive bacteria, Gram-negative bacteria and yeast were subjected to agar disc diffusion and broth microdilution assays. The investigation of antioxidant potential was conducted using the DPPH radical scavenging activity assay. While all compounds displayed strong cytotoxic and antioxidant properties, they exhibited only moderate antimicrobial activity. Molecular docking studies were performed on epidermal growth factor receptor (EGFR), vascular endothelial growth factor receptor 2 (VEGFR-2), focal adhesion kinase (FAK), B-Raf and phosphoinositide 3-kinase (PI3K). The binding energies and interactions obtained from the docking results of compounds (4a–c) supported the experimental results. Drug similarity rates and pharmacokinetic properties were analyzed with the absorption, distribution, metabolism and excretion (ADME) method. Geometric parameters such as chemical potential (µ), electrophilicity index (ω) and chemical softness (σ) of compounds (4a–c) were calculated using the 6-31*G basis set B3LYP method.

The present investigation focused on the comprehensive analysis of the phenolic profile of Centaurea glastifolia L. (Asteraceae) and the assessment of its diverse biological activities. Utilising LC-MS/MS, the phytochemical composition of the 70% methanol extract of Centaurea glastifolia (CG-ME) was thoroughly elucidated, revealing the presence of 30 distinct phytochemical compounds. Notably, major phenolic constituents identified in the extract included quinic acid, chlorogenic acid, luteolin-7-O-glucoside, kaempferol-3-O-glucoside, luteolin, and apigenin-7-O-glucoside. The antioxidant, antibacterial, antiproliferative, and cytotoxic activities of CG-ME were investigated. The CG-ME exhibited a moderate capacity for scavenging DPPH radicals (IC50: 50.05 ± 1.58 µg/mL) and FRAP (63.96 ± 0.39 mg TE/g extract), indicating a moderate level of antioxidant activity. Moreover, CG-ME demonstrated significant antiproliferative effects (GI50: 1.10 and 1.30 µg/mL) on cancer cells (C6 and HTC cancer cell lines, respectively) while displaying low cytotoxicity towards normal cells (LC50: >1000 µg/mL). In terms of antibacterial activity, CG-ME was found to be inactive against tested both Gram-positive and Gram-negative bacterial strains (MIC > 500 µg/mL). The extracts had a promising antiproliferative effect on C6, HeLa, and HT29 cancer cell lines with a less cytotoxic effect (10.5–14.2%) against normal cells.

Baykal, Saliha; Bozkurt, Abdullah; Çobanoğlu Osmanlı, Cansu; Önal, Bedia Sultan; Şahin, Berkan; Karadoğan, Zeynep Nur; Karadağ, Mehmet; Hangül, Zehra; Kılıçaslan, Fethiye; Ayaydın, Hamza

Aim: The aim of this study was to compare the clinical characteristics of childhood-onset schizophrenia (COS) and early-onset schizophrenia (EOS) during the first- episode psychosis and the stable period, to examine psychopharmacological treatment approaches, and to investigate potential predictive factors for prognosis. Methods: Demographic, clinical, and psychopharmacological therapy data for 31 patients diagnosed with COS and 66 with EOS were retrieved from the file records in this multicenter study. Symptom distribution and disease severity and course were evaluated twice, in the acute psychotic stage and in the latest stable phase, during follow-up using the positive and negative syndrome scale (PANSS) and clinical global impression (CGI) scales. Results: A statistically significant difference was observed between the groups' CGI improvement rates and median last stable stage PANSS positive, negative, and general psychopathology symptom scores (p =.005, p =.031, p =.005, and p =.012, respectively). Premorbid neurodevelopmental disorder and obsessive-compulsive disorder and comorbidities were more common in the COS group (p =.025 and p =.030, respectively), and treatment required greater multiple antipsychotic use in that group (p =.013). When the independent variables affecting the difference between pre- and post-treatment PANSS scores were examined using linear regression analysis, the model established was found to be statistically significant (F = 5.393; p =.001), and the group variable (p =.024), initial disease severity (p =.001), and socioeconomic level (p =.022; p =.007) emerged as predictive factors for the disease course. Conclusion: Although early diagnosis and treatment is an important factor in improving prognosis in schizophrenia, more specific predictors for schizophrenia need to be identified. Additionally, preventive programs and pharmacological methods need to be developed in children with neurodevelopmental problems, particularly those from low socioeconomic status families.

Novel therapeutic approaches targeting 5-HT7 receptors outside the central nervous system

Kilic, Melis; Karakoy, Zeynep; Halici, Hamza; Cadirci, Elif; Halici, Zekai

Serotonin (5-HT) is a neurotransmitter found throughout the human body that regulates many physiological events arising from the brain and central nervous system (CNS), such as sleep and appetite. However, it has many other functions in systems outside. In addition to the routine expression of 5-HT7 receptors in CNS regions, such as the pituitary gland, spinal cord, and hippocampus, many studies have reported the expression of these receptors in pathological conditions outside. The role of 5-HT7 receptors outside the CNS has been attracting increased attention in recent years. This review highlights the fact that 5-HT7 receptors are associated with diseases and systems beyond the CNS increasing or decreasing in response to cellular changes. Clinical, basic, in vivo and in vitro studies to date are described, but more research is needed to better understand the role of 5-HT7 receptors outside the CNS.

The effect of physical activity on life satisfaction among sports science students: a moderated mediation model of resilience and mindfulness

Secer, Emrah; Kacay, Zulbiye; Yildiz, Nuh Osman; Talaghir, Laurentiu-Gabriel; Cosoreanu, Dumitru Marius; Bentea, Cristina Corina

Background: This study examines the effect of physical activity (PA) on life satisfaction (LS), focusing on the mediating role of psychological resilience (PR) and the moderating role of mindfulness (MF). Method: Participants were 363 sports science students (240 male and 123 female). Data were collected through International Physical Activity Questionnaire-Short Form, the Life Satisfaction Scale, the Psychological Resilience Scale-Short Form and the Mindfulness Scale. The measurement model was tested using confirmatory factor analysis (CFA) in AMOS 21 and hypothesis were analyzed using Hayes’ (2022) Process Macro models (4, 1, and 7). Results: The mediation effect (Model 4) indicated that the indirect effect of PA on LS through PR was significant. The moderation analysis (Model 1) showed that mindfulness significantly moderated the relationship between PA and PR. Finally, the moderated mediation model (Model 7) confirmed that the indirect effect of PA on LS via PR was stronger at higher levels of mindfulness. Although the effect size was small, it highlights the practical importance of integrating mindfulness in resilience pathways. Conclusion: These findings suggest that physical activity contributes to life satisfaction by enhancing resilience, and that this indirect effect is conditioned by mindfulness. These findings point to the potential value of resilience-building and mindfulness-based programs in educational and psychological practice. However, as the study sample consisted of sports science students from a single faculty, the results should be interpreted with caution regarding generalizability.

    Yangılar, Filiz; Gerek, Merve Dilara

The study aimed to evaluate the potential of creating a functional beverage by enriching Anthriscus sylvestris (L.) kombucha production with guava peel and pulp, showcasing the variety and richness of the product formulations. This was done due to kombucha being a fermented beverage rich in various bioactive compounds that have significant effects on health and the possibility of enhancing product diversity through different formulations. Thus, the kombucha tea was physicochemical, phytochemical, antioxidant activity, and sensory evaluation during the 15-day fermentation period. The increased acetic acid bacteria and yeast count indicates that Anthriscus sylvestris is a viable substrate for the proliferation of Kombucha symbiotic microbes. Adding guava peel and rosemary oil to kombucha significantly increased its total phenolic, flavonoid, and vitamin C content (P<0.05). Anthriscus sylvestris-guava kombucha has antibacterial properties (zones of inhibition of 18.32 and 17.31 mm against Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922, respectively). Moreover, it also provided an inhibitory effect on α-glucosidase and α-amylase, enzymes associated with diabetes. The DPPH, ABTS*+, FRAP, CUPRAC, and ORAC capacities were found to be 66.89-78.90%, 70.83-97.25%, 367-723.15 µmol Trolox/mL, 381.40-460.45 µmol Trolox/mL, and 486.50-737.50 µmol Trolox/mL, respectively. Anthriscus sylvestris kombucha has well-balanced and pleasant sensory properties. These findings indicate that preparing Anthriscus sylvestris kombucha tea with guava may have health effects and that further research is needed to determine potential health benefits.

The Significance of Mono- and Dual-Effective Agents in the Development of New Antifungal Strategies

    Zobi, Cengiz; Algul, Oztekin

Invasive fungal infections (IFIs) pose significant challenges in clinical settings, particularly due to their high morbidity and mortality rates. The rising incidence of these infections, coupled with increasing antifungal resistance, underscores the urgent need for novel therapeutic strategies. Current antifungal drugs target the fungal cell membrane, cell wall, or intracellular components, but resistance mechanisms such as altered drug-target interactions, enhanced efflux, and adaptive cellular responses have diminished their efficacy. Recent research has highlighted the potential of dual inhibitors that simultaneously target multiple pathways or enzymes involved in fungal growth and survival. Combining pharmacophores, such as lanosterol 14α-demethylase (CYP51), heat shock protein 90 (HSP90), histone deacetylase (HDAC), and squalene epoxidase (SE) inhibitors, has led to the development of compounds with enhanced antifungal activity and reduced resistance. This dual-target approach, along with novel chemical scaffolds, not only represents a promising strategy for combating antifungal resistance but is also being utilized in the development of anticancer agents. This review explores the development of new antifungal agents that employ mono-, dual-, or multi-target strategies to combat IFIs. We discuss emerging antifungal targets, resistance mechanisms, and innovative therapeutic approaches that offer hope in managing these challenging infections.

Intrastromal Suturing Technique Compared With Interrupted Corneal Suturing Technique, Loose Suture and Knot Exposure: A Comparative Rabbit Study

Ozkan, Kaan; Aydın, Bahri; Ucgul, Ahmet Yucel; Bayrakceken, Kemal; Ozmen, Mehmet Cuneyt; Akata, Rustu Fikret

Purpose: This study aimed to assess the effectiveness of an innovative intrastromal suturing technique in an experimental rabbit model, comparing it to standard interrupted suturing, loose suture, and suturing with knot exposure. Methods: Fourteen adult male New Zealand White rabbits were included in this study. Each rabbit underwent suturing in both eyes, divided into four groups based on suturing techniques. The novel intrastromal suturing technique involved burying the entire suture material within the corneal stroma. Corneal neovascularisation (CoNV) areas were evaluated by image analysis and immune cell densities by in vivo confocal microscopy (IVCM). Results: The intrastromal suturing group demonstrated significantly smaller CoNV areas at both 1 week and 1 month post-suturing compared to other interventional groups, indicating effective mitigation of CoNV development and progression. Moreover, this group exhibited lower immune cell densities in the superficial stromal layer and endothelial layer, suggesting a reduced inflammatory response. Both the loose suture and the knot exposure groups exhibited significant levels of CoNV and heightened immune cell densities. Conclusion: This experimental study demonstrated effectiveness of intrastromal suturing technique in limiting CoNV and immune cell infiltration, common contributors to graft rejection and complications. Furthermore, the study revealed that loose sutures and those with exposed knots are likely to cause more severe CoNV and inflammation, compared to the traditional interrupted suturing technique and intrastromal suturing.

    • AIM: To investigate the effects of adenosine triphosphate (ATP) and melatonin, which have antioxidant and anti-inflammatory activities, on potential 5-fluorouracil (5-FU)-induced optic nerve damage in rats. • METHODS: Twenty-four rats were categorized into four groups of six rats: healthy (HG), 5-FU (FUG), ATP+5-FU (AFU), and melatonin+5-FU (MFU). ATP (4 mg/kg) and melatonin (10 mg/kg) were administered intraperitoneally and orally, respectively. One hour after ATP and melatonin administration, rats in the AFU, MFU, and FUG were intraperitoneally injected with 5-FU (100 mg/kg). ATP and melatonin were administered once daily for 10d. 5-FU was administered at a single dose on days 1, 3, and 5 of the experiment. After 10d, the rats were euthanized and optic nerve tissues were extracted. Optic nerve tissues were biochemically and histopathologically examined. • RESULTS: ATP and melatonin treatments inhibited the increase in malondialdehyde (MDA) and interleukin-6 (IL-6) levels, which were elevated in the FUG. The treatments also prevented the decrease in total glutathione (tGSH) levels and the superoxide dismutase (SOD) and catalase (CAT) activities (P<0.001). This inhibition was higher in the ATP group than in the melatonin group (P<0.001). ATP prevented histopathological damage better than melatonin (P<0.05). • CONCLUSION: ATP and melatonin have the potential to be used in alleviating 5-FU-induced optic nerve damage. In addition, ATP treatment shows better protective effects than melatonin.

Background: Functional strength training (FST) has gained considerable attention due to its potential in enhancing muscle strength, endurance, and body composition, especially among athletes. The purpose of this study was to assess the effects of a 12-week FST program on performance outcomes such as muscle strength, muscular endurance, and body composition in elite male field hockey players. Methods: The study involved 28 male athletes from the Türkiye Hockey Federation Super League, with a mean age of 27.54 ± 1.34 years and an average athletic experience of 7.62 ± 0.48 years. The mean height of participants was 180.28 ± 4.54 cm, and their mean weight was 75.59 ± 3.08 kg. Participants were randomly divided into two groups: the experimental group (n = 14) and the control group (n = 14). Pre-intervention assessments were conducted to evaluate muscle strength (Leg Extension, Leg Curl, Bench Press, Pushdown), muscular endurance (30-s Sit-up and Push-up tests), and body composition [Body Mass Index (BMI) and Body Fat Percentage (BFP)]. The experimental group participated in the FST program three times per week for 12 weeks, while the control group maintained their usual training regimen. Post-test evaluations were performed using the same testing protocols. Statistical Analysis: Data were analyzed using two-way repeated measures ANOVA to assess the interaction effects of group and time (pre-test vs post-test). Partial eta squared (η2) values were used to report effect sizes, and statistical significance was set at p < 0.05. Data analysis was carried out using SPSS 22.0 software. Normality was assessed through the Shapiro-Wilk test, and data distribution was further examined through skewness-kurtosis values, histograms, box plots, and Q-Q plots. Paired sample t-tests were performed for pairwise comparisons, with Cohen’s d used to determine the effect sizes. The classification for effect sizes followed Hopkins’ (2002) guidelines: small (≥0.01), moderate (≥0.06), and large (≥0.14). Results: Significant group × time interaction effects were found for all measured variables (p < 0.05). The experimental group showed greater improvements in body composition, muscle strength, and muscular endurance compared to the control group. Specifically, the experimental group experienced a significant reduction in BMI (pre-test: 21.17 ± 0.64 kg/m2, post-test: 19.84 ± 0.85 kg/m2, p < 0.001) and BFP (pre-test: 15.36% ± 0.62%, post-test: 12.13% ± 0.47%, p < 0.001), while the control group showed minimal changes in these variables. Muscle strength improvements in the experimental group were significant for Leg Extension (pre-test: 83.93 ± 4.87 kg, post-test: 66.07 ± 4.01 kg, p = 0.003), Leg Curl (pre-test: 99.29 ± 7.81 kg, post-test: 118.21 ± 5.04 kg, p < 0.001), and Bench Press (pre-test: 66.43 ± 6.91 kg, post-test: 87.14 ± 4.69 kg, p < 0.001). In contrast, the control group did not show significant improvements in these tests. Muscular endurance was also superior in the experimental group for both the 30-s Sit-up (pre-test: 19.21 ± 0.97, post-test: 23.36 ± 1.28, p < 0.001) and Push-up (pre-test: 24.66 ± 2.53, post-test: 27.04 ± 0.81, p < 0.001) tests. Effect sizes (Cohen’s d) indicated moderate to large effects for all measured variables, with d values ranging from 0.46 to 1.14 for strength and endurance improvements. Conclusion: The findings of this study demonstrate that a 12-week FST program significantly improves muscle strength, endurance, and body composition in elite male field hockey players. These results suggest that incorporating FST into athletic training regimens may optimize performance and enhance overall physical fitness in athletes.

Purpose: This study aimed to biochemically and histopathologically evaluate the protective and therapeutic effects of elamipretide and methylprednisolone on methanol poisoning-induced brain, optic nerve, and retinal toxicity. Method: In this study, 40 male Wistar Albino rats were divided into six groups: healthy control (HC), methotrexate (MTX, 0.3 mg/kg/d for 7 d), methotrexate + methanol (MTX-M, 0.3 mg/kg/d for 7 d + methanol 3 g/kg on Day 8), methotrexate + methanol + methylprednisolone (MTX-M-MPZ, 0.3 mg/kg/d for 7 d + methanol 3 g/kg on Day 8 + MPZ 1 mg/kg/d for 3 d), methotrexate + methanol + elamipretide (MTX-M-E, 0.3 mg/kg/d for 7 d + methanol 3 g/kg on Day 8 + elamipretide 5 mg/kg/d for 3 d), and methotrexate + methanol + methylprednisolone + elamipretide (MTX-M-MPZ-E, 0.3 mg/kg/d for 7 d + methanol 3 g/kg on Day 8 + MPZ 1 mg/kg/d + Elamipretide 5 mg/kg/d for 3 d). The rats were euthanized 8 h after the last drug administration. Histopathological and biochemical evaluations were performed on serum, caudatoputamen, and ocular tissues. Retinal degeneration was assessed using a scoring system where higher scores indicate less degeneration, with a score of 5 representing normal structure and 1 reflecting severe degeneration. Results: In the MTX-M-MPZ-E group, the retinal degeneration score was higher than in MTX-M group (p = 0.002). The apoptosis index in the retina was highest in MTX-M group, while it was lower in MTX-M-MPZ-E group compared to MTX-M group (p = 0.018). In addition, the apoptosis index in the caudatoputamen was lower in MTX-M-MPZ-E group compared to MTX-M group (p = 0.009). Conclusion: Combined elamipretide and methylprednisolone treatment improved optic nerve and retinal degeneration, reduced neuronal degeneration in the caudatoputamen, decreased oxidative stress and lipid peroxidation, and reduced apoptosis in the retina and caudatoputamen.

Albumin-based nanocarriers loaded with novel Zn(II)-thiosemicarbazone compounds chart a new path for precision breast cancer therapy

Danişman-Kalindemirtaş, Ferdane; Özerkan, Dilşad; Kariper, Ishak Afşin; Erdemir Cilasun, Gökçe; Ülküseven, Bahri; Erdem-Kuruca, Serap

This study explores the therapeutic potential of albumin-bound Zn(II)-thiosemicarbazone compounds (Alb-ZnTcA, Alb-ZnTcB) against breast cancer cells. Previous research indicates that these compounds hinder cancer cell proliferation by blocking DNA synthesis, promoting oxidative stress to induce apoptosis, and disrupting the cell cycle to inhibit cellular division. This study focuses on the loading and characterization of these potentially chemically unstable compounds on bovine serum albumin-based nanocarriers. Accordingly, unlike previous studies using albumin nanoparticles, in this study, ultraviolet light was used to precisely bind the therapeutic agent to albumin during the integration of thiosemicarbazones, achieving controlled nanoparticle size to control nanoparticle size. The mean diameter of Alb-ZnTcA nanoparticles was 32 nm, while Alb-ZnTcB exhibited an average diameter of 43 nm. Notably, Alb-ZnTcA displayed the highest cytotoxicity toward breast cancer cells, suggesting an optimal size for cellular uptake. Additionally, albumin-bound compounds showed enhanced cytotoxicity at lower concentrations, potentially minimizing adverse side effects. Apoptosis analysis indicated that both Alb-ZnTcA and Alb-ZnTcB induce cell death predominantly through apoptosis, effectively preventing the uncontrolled proliferation of cancer cells. These findings demonstrate the potential of Zn(II)-thiosemicarbazone compounds loaded on albumin-based nanocarriers for breast cancer treatment. The increased potency of Alb-ZnTcA and Alb-ZnTcB compared to free compounds, along with their ability to activate apoptotic signaling pathways in MCF-7 breast cancer cells, highlights a promising approach for future cancer therapies. This study suggests that albumin-bound Zn(II)-thiosemicarbazone compounds could offer a targeted and effective strategy in breast cancer treatment, leveraging the advantages of nanocarrier-based delivery systems.

The Key to Stability is Love? The Mediating Role of Work-Family Conflict in the Impact of Job Stress on Work Alienation and the Moderating Effect of Affective Commitment

    Yücel, İlhami; Torun, Nida Bengisu

The main aim of this research is to examine the mediating role of work-family conflict and the moderating effect of affective commitment in the relationship of job stress and work alienation to work. For this purpose, surveys were applied to health workers in the field such as midwives, nurses, emergency medical technicians and doctors in public hospitals. The universe of the study was composed of 844 employees of a public hospital in Türkiye. Surveys were distributed to 680 employees selected through simple incidental sampling from the universe through human resources department representation, and 427 accurately filled surveys were returned. The analysis of the data obtained was carried out through SPSS 22 and Amos 22 programs. Analyses were conducted using PROCESS Macro, developed by Hayes (2013), to determine the conditionally indirect effects. The findings revealed that work-family conflict has a mediating role in the relationship between job stress and work alienation and that employees’ level of affective commitment has a moderating effect. Thanks to this study, it was concluded that there is a meaningful and positive relationship between work-family conflict and alienation from work and that affective commitment has a moderating effect in this relationship. In addition, employees with low affective commitment compared to employees with high affective commitment to the organization were found to be stronger in the positive relationship. Theoretical and application implications are discussed in relation to “social change theory” and “social gender roles theory”. As a result of the analysis, the majority of the participants were male (58.8%), married (57%), 28-40 age group (46.5%) and undergraduate (49.5%).

Akkuş Uçar, Mine; Yılmaz, Coşkun; Soylu, Hakan Hüseyin; Sarıakçalı, Barış; Budak, Cemalettin; Kavuran, Korhan; Durmuşoğlu, Mehmet Vakif; Ceylan, Levent

Background: The impact of resistance training extends beyond the enhancement of muscle strength, encompassing improvements in physical performance, postural stability, and overall functional capacity. This study aimed to investigate the effects of a 4-week resistance training program on functional capacity, respiratory muscle strength, diaphragm thickness, and liver density in healthy older women. Methods: The study included 30 healthy women aged 60–80 years and was designed as a randomized controlled experimental trial. Participants were randomly assigned to a resistance training (RT) group or a control (CON) group. Each participant attended the laboratory on three occasions: during the initial visit, the study procedures were explained; the second visit involved baseline assessments (6MWT, diaphragm thickness and liver fat via ultrasound, and respiratory muscle strength); and final measurements were conducted after the 4-week training programme during the third visit. Results: When pre- and post-training measurements were compared, the resistance training (RT) group demonstrated a significantly greater improvement (8.02%) in the Six-Minute Walk Test (6MWT) distance compared to the control group (p < 0.001). In terms of diaphragm thickness (DT), the RT group showed a 16.66% increase, whereas the control group exhibited a 1.13% decrease (p < 0.001). Maximal inspiratory pressure (MIP) increased by 12.30% in the RT group, while it decreased by 7.25% in the control group, indicating a 19.55% greater improvement in the RT group (p < 0.001). Regarding maximal expiratory pressure (MEP), a significant improvement of 21.48% was observed in the RT group, whereas a 0.75% decrease was recorded in the control group, resulting in a 22.23% greater enhancement in the RT group (p < 0.001). Finally, liver density (LD) increased by 12.30% in the RT group, while it decreased by 7.25% in the control group (p < 0.001). Conclusion: As a result of resistance training, greater improvement was observed in functional capacity, respiratory muscle strength, diaphragm thickness and liver density in the training group compared to the control group.
