Possible Neuroprotective Effect of tDCS on Doxorubicin-Induced Neurotoxicity in Rats

Danisman, Betul; Gökçe, Yasin; Çiçek, Betul; Gül, Deniz Kantar; Derin, Narin

Doxorubicin (DOX), a chemotherapeutic medicine, are frequently utilized to treat various malignancies, even though it has well-documented negative side effects. Neurotoxic effect is one of the best known and occurs through several mechanisms including oxidative stress and neuro-inflammation. Transcranial direct current stimulation (tDCS), a non-invasive brain stimulation technique, is used in several neurodegenerative disease model because of its neuroprotective effects. The aim of this study was to investigate the effects of DOX on antioxidant and inflammatory markers, behavioral parameters and the possible neuroprotective effect of tDCS stimulation on these effects. To induce neurotoxicity, DOX was administered intraperitoneally (i.p.) at 2.15 mg/kg/day, 1 dose in 3 days, 7 times in total for 3 weeks. tDCS treatment was then applied for 5 consecutive days. Catalase (CAT), interleukin-1 beta (IL-1 beta), tumor necrosis factor-alpha (TNF-alpha), and interleukin 6 (IL-6) levels were evaluated. Open field test was used to analyzed to locomotor activity and behavioral changes. The DOX group had neuronal alteration as indicated by the elevation of neuro-inflammation markers (IL-1 beta, TNF-alpha, and IL-6) with reduced antioxidant marker (CAT). The tDCS-treated group showed a significant reduction in the inflammation biomarkers, with a significant improvement in the antioxidant marker. DOX administration impaired locomotor activity as well as anxiety behaviors of rats. tDCS have also been shown to be effective in preventing behavioral and locomotor changes; therefore, they may be considered as candidates to reduce toxicity effects in DOX treatments. Our findings reveal the importance of tDCS treatment to attenuate DOX-associated neurotoxicity. Through the modification and decrease of indicators of cellular toxicity and the increase of natural antioxidant enzymes, tDCS demonstrated considerable promise for avoiding DOX-induced neurotoxicity.

Are increased symptoms of orthorexia nervosa linked to self-perceived body image and nutritional adequacy of Turkish adults?

Icer, Mehmet Arif; Yesildemir, Ozge; Çelik, Mensure Nur; Gezmen-Karadag, Makbule

This cross-sectional study, conducted on 4003 participants aged between 18 and 45, aims to contribute to orthorexia nervosa (ON) research by examining the relationship between ON, body image and nutritional adequacy. The Orthorexia Nervosa-11 scale (ORTO-11) and body perception scale were applied to individuals, and food consumption records were taken with a 24-hour dietary recall method. The study found that more individuals with orthorexia nervosa (ON) considered themselves overweight compared to non-ON individuals. Evaluating diet quality, the mean adequacy ratio was significantly higher in ON individuals. Nutrient adequacy ratios for total fiber, magnesium, zinc, thiamine, folic acid, and vitamin C were higher, while those for carbohydrate and iron were lower in ON individuals. Additionally, increasing the number of snacks, protein, and total fiber adequacy decreased the ORTO-11 score. Distorted body image perception may be a risk factor for ON, but healthy eating fixation does not lead to nutrient inadequacy unless it involves progressive food restriction.

Akyüz, Sumeyye; Sümer, Betul; Günay, Murat; Tanriverdi, Elif Seren; Gülhan, Baris

Background Haematobacter massiliensis is a Gram-negative, non-motile, non-spore-forming, non-fermenting, pleomorphic, and aerobic bacillus. It is rarely isolated from bloodstream infections such as bacteremia, particularly in immunosuppressed patients.Case presentation We report the case of a 67-year-old female patient with a recent history of coronary angiography who presented to the Infectious Diseases outpatient clinic in February 2023 with complaints of fever, chills, rigors, and sweating. Blood cultures obtained for fever etiology identified Haematobacter massiliensis as the causative agent using MALDI-TOF MS and 16S rRNA gene sequencing. The patient was successfully treated and discharged in good health. Conclusions The paper reports the first case of bacteremia caused by Haematobacter massiliensis in Turkey. Haematobacter species are clinically significant due to their potential to cause bloodstream infections, particularly in immunosuppressed patients, and should be considered in the differential diagnosis. In patients presenting with fever of unknown origin, bacteremia, particularly when atypical growth patterns are observed in cultures, Haematobacter massiliensis should be considered a rare but potential pathogen.

Prevalence and clinical correlates of mesenteric panniculitis in urinary stone disease

Öztürk, Mehmet Eren; Kahraman, Sevket; Yalçin, Ahmet; Sunar, Mukadder

Purpose: The aim of this study is to determine the prevalence of mesenteric panniculitis in patients with urinary system calculi and identify factors, such as hydronephrosis and calculus location, that might be associated with the presence of mesenteric panniculitis. Materials and Methods: In this retrospective observational study, the prevalence of mesenteric panniculitis was determined in 692 patients with urolithiasis using non-contrast-enhanced computed tomography imaging between 2017 and 2020. The grade of hydronephrosis and the level of urinary calculi were recorded for each patient, and statistical analysis was performed to assess the link between these features and mesenteric panniculitis. Results: The prevalence of mesenteric panniculitis in patients with urolithiasis was 6.35% and it was associated with the presence of hydronephrosis. It was more common in patients who had both ureterolithiasis and nephrolithiasis with no statistically significant difference regarding in age, gender, or the presence of either only nephrolithiasis or only ureterolithiasis. Conclusion: Mesenteric panniculitis is more common in patients with both ureterolithiasis and nephrolithiasis and can be easily diagnosed with non-contrast computed tomography imaging. Because the mesentery and urinary system share a common lymphatic drainage system, increased lymphatic drainage due to hydronephrosis has been shown to contribute to the development of mesenteric panniculitis.

Kizilgöz, Volkan; Aydin, Sonay; Kantarci, Mecit; Kockara, Nizamettin; Gülmez, Ali Osman; Gür, Volkan

Aim: There are many studies in the literature presenting morphological predisposing factors of anterior cruciate ligament (ACL) tears. Insall-Salvati index (ISI), which is widely used to measure patellar height, was investigated as a predisposing risk factor for the ruptures of ACL. Material and Methods: Patellar tendon length (PTL), Patellar length (PL), and anterior tibial translation (ATT) were measured, and ISI values were calculated for the study and control groups. Knee magnetic resonance (MR) images of 33 patients who had undergone ACL reconstruction due to ruptured ACL, which is surgically confirmed (study group), and 90 patients who had undergone meniscus operation with surgically confirmed intact ACL (control group) were reinterpreted by two reviewers in terms of ISI and ATT values. Independent samples T-test was used to compare the results of the study and control groups. Additionally, intraclass correlation coefficient (ICC) values were presented to verify the interobserver difference between reviewers. Results: PTL, ATT, and ISI values showed a statistically significant difference, and higher values were obtained in the study group than in the control group (p<0,001). There was excellent agreement between reviewers regarding PTL, PL, ISI, and ATT measurements. Discussion: ISI had been used as an indicator of patellar dislocation. In this research, ISI was investigated to be in a relationship with ACL injuries in operated patients. The results of this study indicated higher ISI values in patients with ACL rupture. High ISI values might be a predisposing factor for ACL rupture and may be used in preventive medicine in terms of ACL injuries.

Effect of Hippophae Rhamnoides Fruit Extract on Doxorubicin-Induced Oxidative Brain Damage and Behavioral Impairment in Rats

Borekci, Taha Berkay; Altuner, Durdu; Cicek, Betul; Bulut, Seval; Coban, Abdulkadir Taha; Suleyman, Halis

Background and Objective: Doxorubicin (DOX) use can promote neurobehavioral changes and neurodegeneration, which have been attributed to oxidative stress. Thus, this study aimed to examine the effect of Hippophae rhamnoides L., fruit extract (HRe), against possible oxidative brain damage and behavioral disorders in rats caused by DOX.Materials and Methods: A total of 24 male Sprague-Dawley rats were utilized in this study and were divided randomly into four groups (n = 6 in each groups): CG, healthy control; HRe, 50 mg/kg HRe; DOX, 5 mg/kg i.p., in a single intraperitoneal dose of DOX; Hre + DOX, 50 mg/kg HRe + 5 mg/kg DOX. HRe was administered orally once a day for two weeks, while DOX was administered intraperitoneally twice a week for two weeks. Subsequently, behavioral tests were performed-the sucrose preference test (SPT) and pole test-to assess depression-like behaviors and motor function, respectively. Then, the level of oxidative stress was biochemically evaluated in the brain tissues of the rats. One-way analysis of variance (ANOVA) was conducted, followed by a post hoc Tukey's test for the statistical analysis. A p-value < 0.05 was considered statistically significant. Results: The HRe treatment markedly reduced DOX-induced depression-like behaviors and improved motor dysfunction. The HRe treatment also restored the impaired antioxidant response by inhibiting the DOX-related malondialdehyde increase and reducing the decrease in total glutathione levels, as well as superoxide dismutase and catalase activities. Conclusion: The present study indicates that HRe treatment has beneficial effects on motor dysfunction as well as depression-like behavior associated with neurodegeneration following DOX-induced brain damage. Possible mechanisms underlying these beneficial effects include lipid peroxidation inhibition and restoration of antioxidant defense mechanisms by HRe.

Effects of a semester-long massage practice on low back pain and physical fitness in university students

Erzeybek, Mustafa Said; Ka, Fatih; Eynur, Baybars Recep; Enur, Atul; Onen, Mert Eray

This quasi-experimental study aimed to investigate the effects of a semester-long massage practice on low back pain and physical fitness among university students. A total of 124 students (87 male, 37 female) enrolled in a sports science program participated in structured massage training using the Albert Hoffa technique. Over a 14-week period, students practiced three times per week under supervision. Physical activity was assessed using the short-form International Physical Activity Questionnaire (IPAQ), and back pain was measured using the Oswestry Disability Index (ODI). Findings indicated a significant moderate negative correlation between physical activity and low back pain levels (r =-.62, p < .001), with moderate to large effect sizes observed across outcomes. These results demonstrate that semester-long educational massage interventions can enhance physical activity, reduce musculoskeletal pain, and may represent a promising, non-pharmacological strategy for managing musculoskeletal complaints in young adults. Future research should extend to clinical populations, employ longer follow-up periods, and incorporate standardized protocols to strengthen generalizability.

Dose-Dependent Pulmonotoxic Effects of Favipiravir in Rats Biochemical and Histopathological Evaluation

Suleyman, Zeynep; Altuner, Durdu; Suleyman, Halis; Coban, Taha Abdulkadir; Ozkaraca, Mustafa

Background and Objective: Favipiravir is associated with more serious side effects at higher doses. This experimental study proposed to investigate the effect of favipiravir on dose-dependent lung toxicity in rats biochemically and histopathologically. Materials and Methods: The rats were divided into four groups as healthy (HG), 100 mg/kg favipiravir (FAV-100), 200 mg/kg favipiravir (FAV-200) and 400 mg/kg favipiravir (FAV-400). Favipiravir 100, 200 and 400 mg/kg doses were administered by oral gavage to the other groups except HG. To the HG group, only distilled water (0.5 mL) was applied in the same way. This procedure was repeated twice a day for a week. Then, the rats were euthanised with high-dose anaesthesia and lung tissues were removed. Oxidative stress parameters such as malondialdehyde (MDA), Total Glutathione (tGSH), superoxide dismutase (SOD), total oxidant status (TOS) and total antioxidant status (TAS) were examined. After the one-way ANOVA, the Tukey's post hoc test was performed. Results: Favipiravir dose-dependently increased MDA and TOS also decreased tGSH, SOD and TAS in rat lung tissue. As favipiravir was given in increasing doses, it was easier to observe the changes between the different groups. This was also supported by the histopathological data. Histopathologically, interstitial pneumonia and lymphoid hyperplasia were mild in the 100 mg/kg favipiravir group, severe at high doses. Conclusion: As the dose of favipiravir increased, oxidant levels increased and antioxidant levels decreased in the lung tissue. In line with these results, it was observed that favipiravir caused a dose-dependent pulmonotoxic effect in rats.

Combination of Silver Nanoparticles Synthesized Using Karaerik Extract and Cisplatin: Effects on Breast Cancer Cells

    Atici, Tugba; Altun Çolak, Deniz

Nanoparticle-based therapies provide targeted drug delivery with reduced side effects, making them promising options for cancer treatment. This study examines the anticancer and oxidative effects of green-synthesized silver nanoparticles (AgNPs) derived from the Karaerik grape (Vitis vinifera L., Vitaceae), a variety known for its high antioxidant capacity, both alone and in combination with cisplatin (CP) on MDA-MB-231 breast cancer cells. AgNPs were synthesized using the plant leaf extract and characterized through UV-Vis, SEM-EDX, and FT-IR techniques. MDA-MB-231 cells were exposed to 10 and 20 jig mL-1 AgNPs, 25 jiM CP, and their combinations for 24, 48, and 72 hours. Cytotoxicity was analyzed using the MTT assay, and IC50 values were determined. Parameters of oxidative stress, including total oxidant status (TOS), total antioxidant status (TAS), and oxidative stress index (OSI), were evaluated to assess oxidative balance disruption. The combination of AgNPs and CP led to a significantly greater reduction in cell viability compared to individual treatments (p<.05). The IC50 value of CP decreased in the presence of AgNPs, indicating an enhanced cytotoxic effect. Additionally, a significant increase in TOS and OSI levels, along with a decrease in TAS, suggested that the combination therapy induced oxidative stress-mediated cell death. Green-synthesized AgNPs from Karaerik grape leaves enhance CP-induced cytotoxicity by modulating oxidative stress pathways, indicating their potential role as adjuvants in breast cancer therapy. These findings underscore the importance of green nanotechnology-based strategies in oncology.

Background and Aims: Myrtle (Myrtus communis L.) has been used as a food and folk medicine since ancient times. The aim of this study was to determine the antioxidant capacity, inhibition of tyrosinase enzyme, and cytotoxicity of 12 extracts from the fruit, seed, leaves, and twigs of Myrtus communis L. subsp. communis. Methods: The cytotoxic effects on human lung (A549), liver (Hep3B), prostate (PC-3) and breast (MCF7) cancer cells and healthy L929 cells were determined using the MTT method. Tyrosinase enzyme inhibition was investigated using L-tyrosine as the substrate. The antioxidant capacity was determined using the DPPH, TEAC and CUPRAC methods. Results: It was found that the extracts did not affect the viability of healthy cells but showed concentration-dependent toxic effects on cancer cells. The ethyl acetate sub-extracts of the plant parts showed a higher cytotoxic effect on all cell lines, and the ethyl acetate sub-extract of the seed was the most effective with 72.6% proliferation inhibition against the Hep3B. The methanol extract and ethyl acetate sub-extract of the seed showed the highest inhibition against tyrosinase and the values were 388.6 and 412.8 mg KAE/ g extract, respectively. While all extracts showed significant antioxidant activities in all methods, the ethyl acetate sub-extract of seeds showed the highest. Conclusion: This is the first comparative study to investigate the antiproliferative effects of extracts from various parts of Myrtus communis L. subsp. communis. The results show that seeds in particular can be a promising, effective and recyclable resource that can be safely used in the pharmaceutical and cosmetic industries.

Regulatory activity of nobiletin on MAPK/NF-κB signaling pathway in indomethacin-induced gastric damage

Uslu, Gozde Atila; Uslu, Hamit; Çoban, Taha Abdulkadir; Özkan, Ebru Bardas

Nobiletin (NOB) is an important flavonoid obtained from citrus fruits such as Citrus depressa, Citrus reticulata, Citrus sinensis and Citrus limon. Studies have demonstrated that effects are anti-inflammatory, antioxidant, immunomodulatory, anticancer, neuroprotective, anti-atherosclerosis, antiapoptotic, and antidiabetic. There are still limited studies on the efficacy of nobiletin on the pathways underlying gastric damage. Therefore, in the present study, we investigated both the effects of nobiletin on the mitogen-activated protein kinase (MAPK)/nuclear factor kappa B (NF-kappa B) signaling pathway, one of the pathways in indomethacin (IND)-induced inflammation and whether it has gastroprotective activity. NOB+IND and PAN+IND groups were treated with substances for 7 days (10 mg/kg NOB, 5 mg/kg PAN). On the 8th day, gastric damage model was created with a single dose of 100 mg/kg indomethacin. In indomethacin-induced gastric injury, nobiletin significantly decreased NF-kappa B-p65, MAPK levels and significantly increased prostaglandin E2 (PGE2) production in the stomach. In addition, nobiletin administration caused a decrease in interleukin 6 (IL-6), tumor necrosis factor alpha (TNF-alpha) and interleukin 1 alpha (IL-1 alpha) levels and an increase in interleukin 10 (IL-10) levels against indomethacin-induced inflammation. When the ulcerative areas were evaluated, it was found that ulcerative areas were significantly reduced in the nobiletin group compared to the indomethacin only group, and when the ulcer inhibition levels of nobiletin and pantoprazole administration were examined, both substances showed similar results. When these results were evaluated as a whole, it was determined that nobiletin had a candidate anti-inflammatory potential for the prevention of inflammation in the stomach and showed a strong gastroprotective effect.

Role of imaging in chronic otitis media and its complications

    Memis, Kemal Bugra; Aydin, Sonay

Chronic otitis media (COM) is a long-standing inflammatory condition affecting the middle ear and mastoid cavity, often resulting in progressive structural damage and functional deficits. Radiological imaging is fundamental in diagnosing the disease, assessing its severity, and identifying possible complications. The literature indicates that the prevalence rates of extracranial and intracranial complications range from 0.69% to 5%, while the mortality rate for intracranial complications is 26%. While magnetic resonance imaging is particularly useful in distinguishing soft tissue abnormalities and detecting intracranial extensions like meningitis, brain abscess, and sigmoid sinus thrombosis, high-resolution computed tomography remains the preferred modality for evaluating bony erosion, cholesteatoma, and mastoid involvement. Key complications such as ossicular chain destruction, facial nerve damage, and labyrinthine fistulae can be precisely identified using advanced imaging modalities, allowing for timely and effective surgical intervention. This minireview underscores the essential role of radiology in both diagnosing and managing COM, highlighting critical imaging findings that facilitate early detection and inform treatment decisions. A collaborative approach among radiologists, otolaryngologists, and infectious disease specialists is crucial for improving clinical outcomes in affected patients.
