Background and Objectives: Post-menopausal estrogen decline is considered a contributing factor to sarcopenia, and inspiratory muscle training (IMT) may provide benefits in this demographic. This study examined the impact of a four-week IMT program on diaphragm thickness, abdominal wall muscle thickness (AWMT; transversus abdominis, internal oblique, and external oblique), and liver fat percentage in healthy elderly women. Materials and Methods: Twenty-six women aged 60–80 years were randomly assigned to an IMT group (n = 13) or a control group (n = 13). The IMT group used the PowerBreathe® Classic device at 40% of maximal inspiratory pressure (MIP), with weekly increments of 10%. Training was performed twice daily, five days per week, with 30 breathing cycles per session (60 per day). The control group maintained their usual routines. AWMT, diaphragm thickness (DT), and fatty liver density (FLD) were measured by a radiologist before and after the intervention. Results: After four weeks, the IMT group showed significant improvements in all parameters compared to controls. Mid-diaphragm thickness (MDT) increased by 11.44% (effect size (ES) = 0.358, p < 0.001) versus 0.76% in controls (p = 0.271). Posterior diaphragm thickness (PDT) improved by 7.48% (ES = 0.282, p < 0.001) versus 0.38% (p = 0.564). Right AWMT increased by 12.7% (ES = 0.492, p < 0.001) compared to 0.10% (p = 0.872), and left AWMT increased by 9.93% (ES = 0.395, p < 0.001) versus 2.64% (p = 0.014). FLD improved by 11.79% (ES = 0.959, p < 0.001) in the IMT group, while the control group showed no meaningful change (−0.13%, p = 0.847). Conclusions: A short-term IMT protocol significantly enhanced diaphragm and AWMT and reduced liver fat in elderly women. These findings support the use of IMT as a simple, non-invasive intervention to preserve musculoskeletal and metabolic health in aging populations.

Objective: In this study, novel 5-fluoroindolylmethylene hydrazone derivatives were synthesized and assessed for their potential inhibitory effects on human carbonic anhydrase I and II (hCA I and II) enzymes through both in vitro and in silico approaches. Material and Method: Accordingly, starting from 5-fluoroindole, 5-fluoroindole-3-carboxaldehyde was synthesized. The target final compounds were then obtained by condensing substituted phenylhydrazine with 5-fluoroindole-3-carboxaldehyde, resulting in three Schiff base derivatives (2a, 2b, and 2c). Result and Discussion: The synthesized compounds demonstrated effective inhibition of cytosolic carbonic anhydrase isoforms hCA I and II, with Ki values ranging from 32.28±7.09 to 74.86±10.90 nM for hCA I and 8.79±1.92 to 60.40±14.64 nM for hCA II. Among them, compound 2a exhibited the most potent inhibitory effect on both isoenzymes. In vitro results were verified with the results obtained by docking studies and interactions with enzymes were demonstrated. These novel 5-fluoroindolylmethylene hydrazone derivatives show promise as potent inhibitors of cytosolic CA isoenzymes.

Vaginal microbiota is essential for mucosal immunity, pathogen defense, and homeostasis. Disruption of this balance can promote opportunistic infections, notably by Candida albicans. This study investigates the therapeutic potential of Morus alba (MA) extract and Lactobacillus rhamnosus (LR) in a rat model exposed to Candida albicans exosomes (CAE). CAE induced a 1.3–5.1-fold upregulation in SAP1–10 gene expression, with SAP4 showing the highest increase (p ≤ 0.05). MA and LR monotherapies selectively suppressed SAP6 (41%) and SAP4 (23%), respectively. Notably, combination therapy (CAE+MA+LR) synergistically inhibited all SAP genes (0.8–1.1-fold, p ≤ 0.05). 16S rRNA analysis showed that LR-containing groups maintained Lactobacillales abundance (37.77%–38.7%), while MA reduced Mycobacteriales by 68.5% (p = 0.004). Microbial diversity was lower in the MA group (H = 3.242) but higher in LR groups (H = 5.493–5.598). Histopathology revealed severe ovarian inflammation in the CAE group (87.4%, p = 0.0022) with a 4.2-fold increase in IL-6, while combination therapy reduced inflammation and IL-6 levels by 60%–72% (p ≤ 0.05). Immunofluorescence confirmed downregulation of TLR4 and Caspase-3 with treatment. FIB-SEM and NTA analyses showed that CAE exosomes had a heterogeneous morphology (99.3 ± 31.1 nm) and high polydispersity (PDI = 0.31). These results suggest that MA and LR synergistically reduce fungal virulence, restore microbial balance, and offer a promising adjuvant strategy against vaginal candidiasis.
- MakaleAçık Erişim2025Development of Potent Type V MAPK Inhibitors: Design, Synthesis, and Biological Evaluation of Benzothiazole Derivatives Targeting p38α MAPK in Breast Cancer Cells
Zoatier, Bayan; Gizem Yildiztekin, K.; Abdullah Alagoz, M.; Hepokur, Ceylan; Burmaoglu, Serdar; Algul, Oztekin
Type V MAPK inhibitors are distinguished by their capacity to target both the ATP binding site and a specific allosteric site on the enzyme. The present work utilized in silico analysis with Maestro 13.8.135 (Schrodinger) software in conjunction with experimental investigations to enhance the antiproliferative efficacy and forecast the likely mechanism of action of benzothiazole derivatives. Approximately 28 compounds were developed, produced, and assessed for their antiproliferative properties against two breast cancer cell lines: ER+ (MCF7) and ER- (MDA-MB-231), in addition to one normal mouse fibroblast cell line (L929). Their antiproliferative activities were evaluated via the MTT test, with doxorubicin and cisplatin serving as reference drugs for comparison. Consequently, the compounds with the greatest activity against the MCF7 cell line were chosen, and their inhibitory effects on the p38α MAPK enzyme were examined. The molecular docking studies of compounds 15 and 19 demonstrated significant binding affinities for p38α MAPK. Molecular dynamics simulations conducted over 100 ns revealed that compounds 15 and 19 exhibit stability inside both the ATP-binding domain and the lipid domain of p38α MAPK. The research focused on creating effective Type V MAPK inhibitors demonstrate that compounds 15 and 19 possess considerable ability to inhibit p38α MAPK, hence establishing them as promising anticancer agents.

- MakaleAçık Erişim2025Turkish men’s experiences of positive SMA carrier results: a qualitative study within a gendered screening framework
Gürsoy, Ersan; Yeğ, Mert
Background: Spinal muscular atrophy (SMA) carrier screening has been part of Türkiye’s premarital and prenatal program since 2021, using a stepwise algorithm that first tests men. Little is known about how male carriers interpret a positive result—particularly when their partners test negative—within the country’s sociocultural context. This study qualitatively explored Turkish men’s emotional, cognitive, and interpersonal responses to a positive SMA carrier result to illuminate needs that current screening practices may overlook. Methods: Eleven male participants who had received a positive SMA carrier result while their partners tested negative were recruited through purposive sampling from official health records in Erzincan Province. Data were collected via in-depth, semi-structured interviews conducted by telephone between November and December 2024. Interviews explored emotional responses, information-seeking behaviour, reproductive decision-making, and communication with healthcare professionals. All interviews were audio-recorded, transcribed verbatim, and analysed thematically using Braun and Clarke’s six-phase framework, supported by NVivo 12 software. Results: Participants described intense, sometimes overwhelming emotions; widespread confusion stemming from scarce or inconsistent genetic counselling; and a heavy reliance on internet and social-media sources that often intensified fear. Disclosure decisions were shaped by stigma concerns and extended-family expectations, while interactions with healthcare professionals ranged from reassuring to dismissive. Many men postponed or reconsidered childbearing, weighed assisted-reproduction options, and ultimately reported greater awareness of genetic risk—some becoming advocates for broader screening. Across accounts, carrier status was framed not as a purely biomedical label but as a socially embedded responsibility compounded by the gendered design of the screening algorithm. Conclusion: Findings highlight the psychosocial burden placed on male carriers and point to gaps in counselling and follow-up care. Integrating accessible, culturally sensitive, and gender-aware genetic counselling—through tele-genetics, community outreach, and primary-care training—could mitigate distress and support informed reproductive choices. Addressing these needs would strengthen the ethical and practical impact of Türkiye’s SMA screening program and offer guidance for similar initiatives elsewhere.
- MakaleAçık Erişim2025New 1,2,3-triazole derivatives as acetylcholinesterase and carbonic anhydrase inhibitors: Synthesis, molecular docking, and solubility
Efeoglu, Cagla; Demir, Yeliz; Türkeş, Cüneyt; Yabalak, Erdal; Seferoglu, Zeynel; Nural, Yahya
In this study, a series of new 1,2,3-triazole derivatives were synthesized in 84–93 % yield using copper-catalyzed azide-alkyne cycloaddition (CuAAC) click chemistry and characterized by 1H/13C NMR, FT-IR, and HRMS analyses. The synthesized compounds (4a–h) were evaluated for their inhibitory activities against human carbonic anhydrase isoforms I and II (hCA I and hCA II) and acetylcholinesterase (AChE), which are clinically relevant targets in neurological and metabolic disorders. Among them, compounds 4f and 4g exhibited the most potent dual inhibitory activities. Compound 4f showed KI values of 144.30 nM for hCA II and 205.10 nM for AChE, while compound 4g exhibited KI values of 239.10 nM for hCA II and 125.90 nM for AChE. These values demonstrate that 4f and 4g are more effective than the reference drugs acetazolamide (hCA II, KI = 381.44 nM) and tacrine (AChE, KI = 255.44 nM). Structure-activity relationship (SAR) analysis revealed that hydrophobicity, steric bulk, and aromaticity significantly influenced enzyme affinity. In silico docking confirmed strong interactions with key active site residues. Furthermore, ethanol solubility profiling revealed that polar and hydrogen-bonding groups significantly improved solubility, while bulky or aromatic hydrophobic substituents reduced it. The combined biological activity and solubility data emphasize the potential of these triazole derivatives particularly 4f and 4g as promising candidates for multitarget drug design and further preclinical development.
- MakaleAçık Erişim2025Social Isolation in Turkish Adolescents: Translation, Cross-Cultural Adaptation, and Validation of the Social Isolation Questionnaire
Çevik Özdemir, Hamide Nur; Ayran, Gülsün
Objectives: Social isolation is an important public health issue that is becoming increasingly prevalent among adolescents today. Recognizing the risk of social isolation in children and adolescents during their developmental years can contribute to the prevention of depression and anxiety disorders, as well as risky behaviors such as suicide and substance abuse. Valid and reliable measurement tools are needed to assess social isolation in adolescents. There is a lack of surveys to identify gaps in social isolation among Turkish adolescents. This study aims to adapt the Social Isolation Questionnaire (QIS) scale, which was developed for adolescents, to Turkish culture and to evaluate its psychometric properties. Methods: This descriptive and methodological study was conducted between July and November 2024 with 1922 adolescents. Data were collected using an introductory information form, the QIS, and the UCLA Loneliness Scale. In the evaluation of the data, explanatory and confirmatory factor analysis, Cronbach’s alpha, split-half, item–total score correlation, and test–retest analysis were used. Results: The average age of the adolescents was 13.97 ± 1.67 years (min = 12, max = 17). A total of 50.6% of the adolescents were male and 49.4% were female. A total of 81.2% of the adolescents had a nuclear family structure. Exploratory factor analysis revealed a three-factor structure explaining 55.97% of the variance. The factor loadings were greater than 0.30, and all fit indices were greater than 0.90. The total Cronbach’s α value of the scale was 0.83, while the values for the subdimensions ranged from 0.73 to 0.75. Conclusions: The Turkish version of the QIS is a valid and reliable measurement tool for assessing social isolation in adolescents. This questionnaire can be used by health professionals and researchers to identify adolescents at risk of social isolation and plan appropriate mental health interventions. This questionnaire can be used in studies focusing on adolescent mental health and well-being, contributing to the development and implementation of strategies in line with the Sustainable Development Goals.

This study aimed to investigate biochemically and histopathologically the protective effect of adenosine triphosphate (ATP) and coenzyme Q10 (CoQ10) against potential hydroxychloroquine (HCQ)-induced retinal damage in rats. Twenty-four male albino Wistar-type rats were randomly separated into four groups: healthy (HG), receiving HCQ (HQG), receiving ATP + HCQ (AHQ), and receiving CoQ10 + HCQ (CoQHQ). ATP (4 mg/kg, intraperitoneal) was given to the AHQ, and CoQ10 (10 mg/kg, oral) to the CoQHQ. Rats in the HQG, AHQ, and CoQHQ were given HCQ (120 mg/kg, oral) 1 h after administering ATP and CoQ10. Treatments continued once a day for seven days. On the 8th day, the rats were sacrificed with 50 mg/kg sodium thiopental, and the eyes were removed. Malondialdehyde (MDA), total glutathione (tGSH), superoxide dismutase (SOD), and catalase (CAT), and interleukin-6 (IL-6) levels were measured in the retrieved eye tissues and retinal tissues were assessed histopathologically. An increase in MDA and IL-6 levels and a decrease in tGSH, SOD, and CAT levels were detected in the eye tissues of the HQGcompared to the HG. HCQ-induced changes in oxidant and antioxidant levels were significantly suppressed by ATP and CoQ10 treatment. ATP was more successful than CoQ10 in this inhibition. Severe damage was observed in the eye tissues of the HQG group, whereas the damage was mild in the AHQ and moderate in the CoQHQ. Although both ATP and CoQ10 have the potential to be effective in the prevention of HCQ-induced retinal damage through antioxidative activity, ATP appears to be the more preferable treatment approach.

Akşit, Hüseyin; Gergin, Ömer
This study aimed to synthesize and characterize silver nanoparticles using Alchemilla erzincanensis extract and to evaluate their antimicrobial potential against selected bacterial strains. Silver nanoparticles (AgNPs) were successfully synthesized using an environmentally friendly green synthesis approach based on the extract of A. erzincanensis, an endemic plant species in Turkiye. The synthesized nanoparticles were characterized using Scanning Electron Microscopy (SEM), X-ray Diffraction (XRD) and Energy-Dispersive X-ray Spectroscopy (EDX) techniques. SEM analysis revealed that the AgNPs were spherical in shape with sizes ranging from 34.24 to 48.43 nm. XRD results confirmed the face-centered cubic (FCC) crystalline structure of the particles. EDX spectra indicated a high silver content. Antibacterial activity tests showed that the AgNPs exhibited notable inhibitory effects particularly against Gram-positive bacteria such as Bacillus cereus and Staphylococcus aureus and also demonstrated activity against resistant Gram-negative bacteria like Pseudomonas aeruginosa. This study is the first to report the use of A. erzincanensis for AgNP synthesis and highlights its potential contribution to the development of novel antimicrobial agents against antibiotic-resistant pathogens.

Background and Objectives: Chronic mucosal infection with Helicobacter pylori (H. pylori) plays a key role in the development of gastroduodenal disorders such as chronic gastritis, peptic ulcers, gastric lymphoma, and gastric cancer by triggering local immune responses and inducing hypoxic and inflammatory conditions in the gastric mucosa. This study aims to evaluate the potential diagnostic value of hypoxia-inducible factors HIF-1α and HIF-2α, along with transmembrane prolyl 4-hydroxylase (P4H-TM), as biomarkers in H. pylori-positive patients. Additionally, the study investigates the association between these markers and alterations in lipid profiles, as well as their involvement in the molecular mechanisms underlying gastric conditions like gastritis, particularly in the context of H. pylori infection. Materials and Methods: This study was conducted at Istanbul Avcılar Murat Kölük State Hospital’s General Surgery Outpatient Clinic. A total of 60 participants were included: 40 patients diagnosed with chronic gastritis (20 H. pylori-positive and 20 H. pylori-negative) and 20 healthy controls confirmed negative by 13C-urea breath test. Blood samples were collected for ELISA analysis of HIF-1α, HIF-2α, and P4H-TM levels. Additionally, lipid profiles were measured and compared among the groups. Results: No significant differences were found among the groups in terms of demographic factors such as age, sex, or body mass index (BMI). However, significant variations were observed in the levels of HIF-1α, HIF-2α, and P4H-TM across all groups (p < 0.001 for each marker). These markers were substantially elevated in the H. pylori-positive gastritis group compared to both the H. pylori-negative and healthy control groups. Receiver Operating Characteristic (ROC) curve analysis revealed that all evaluated markers exhibited strong diagnostic accuracy in differentiating H. pylori-positive individuals from other groups. HIF-1α (AUC: 0.983) and HIF-2α (AUC: 0.981) both achieved 100% sensitivity with specificities of 93.3% and 91.1%, respectively. P4H-TM showed an AUC of 0.927, with 85% sensitivity and 95.6% specificity. Conclusions: These findings indicate that HIF-1α, HIF-2α, and P4H-TM may serve as effective biomarkers for diagnosing H. pylori-positive patients and may be linked to changes in lipid metabolism. The elevated expression of these markers in response to H. pylori infection highlights their potential roles in the inflammatory and hypoxic pathways that contribute to the pathogenesis of gastric diseases such as gastritis.

Fifty male rats were randomly divided into five groups. The control group received distilled water orally for 11 days, while the acrylamide group received acrylamide (50 mg/kg, i.g.) for 11 days. The MEL10 + ACR and MEL20 + ACR groups received intraperitoneal injections of melatonin at doses of 10 mg/kg and 20 mg/kg, respectively, followed by acrylamide (50 mg/kg, i.g.) administered 1 h after melatonin injection. The MEL20 group received melatonin injections (20 mg/kg) for 11 days. Lung tissues collected at the end of the study underwent biochemical, histopathological, immunohistochemical, immunofluorescence, and in silico analyses. Acrylamide caused oxidative stress, inflammation, apoptosis, and tissue damage in the lungs. Melatonin treatment alleviated acrylamide-induced lung damage by exhibiting antioxidant, anti-inflammatory, and anti-apoptotic effects. Melatonin significantly improved the histopathological changes caused by acrylamide in lung tissue. Melatonin may have protective effects on health by regulating cellular processes such as oxidative stress, antioxidant enzyme activity, inflammation, and apoptosis through MT1 and MT2 receptors. Melatonin mitigates oxidative stress, inflammation, apoptosis, and tissue damage in acrylamide-induced lung injury in rats.

Colorectal adenocarcinoma, the second deadliest malignancy, is characterized by chromosomal instability and mismatch repair deficiencies, leading to apoptosis resistance. Previously, we developed Au(I) complexes [Ni(hydeten)Au2(CN)4] (C1), [Ni(bishydeten)Au2(CN)4] (C2), and [Cd(edbea)2][Au(CN)2]2.H2O (C3) that induce apoptosis in cancer cells without toxicity to normal cells in vitro. In this study, we evaluate the in vivo antitumor efficacy of these Au(I) complexes, assessing tumor volume, survival rate, and protein markers related to cell cycle, apoptosis, and oxidative stress through immunohistochemistry, gene expression, and ELISA assays. Molecular docking studies of the complexes with proteins revealed the best binding interactions. In molecular docking analysis, compounds C1, C2, and C3 interacted with more than one target. The best interaction results were obtained with PI3K alpha target among these targets. ETotal energy values presenting the docking score value on PI3K alpha (PDB ID:5ITD) were calculated as -1653.5 kJ/mol for C1, -1863.4 kJ/mol for C2, and -2420.4 kJ/mol for C3. In the HT29 xenograft tumor model, treatment with Au(I) complexes (0.5 mg/kg) resulted in a significant reduction in tumor volume and increased survival. Histopathological analysis confirmed minimal side effects on tissues, while assessment of peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT) levels indicated that the mechanism of action was not based on oxidative stress-induced cell death. Overall, Au(I) complexes demonstrated selective low toxicity and antitumor effects via apoptosis, suggesting their potential as therapeutic agents for colorectal adenocarcinoma.

Doxorubicin (DOX), although widely used as a potent chemotherapeutic agent, is limited by its dose-dependent cardiotoxicity. Esculetin (E), a naturally occurring coumarin derivative, has been reported to exert antioxidant and anti-apoptotic effects in various tissues. This study aimed to evaluate the cardioprotective effects of esculetin against DOX-induced cardiac injury in rats by examining changes in the expression of genes related to apoptosis, oxidative stress, and mitochondrial function. Male Wistar rats were randomly assigned into six groups (Control, DOX, E50, E100, DOX + E50, DOX + E100). Cardiotoxicity was induced by administering DOX was administered as six intraperitoneal injections of 5 mg/kg each over 14 days (cumulative dose = 30 mg/kg). Serum cardiac markers (CK-MB, LDH, and cTn-I) were measured, and cardiac tissues were subjected to histopathological examination and RT-qPCR analysis for target gene expression. In addition, hierarchical clustering heatmap was employed to evaluate multidimensional gene expression patterns across groups. DOX treatment significantly elevated serum cardiac injury markers and upregulated pro-apoptotic genes (Casp3, Casp9, Anf, Bnp, β-Mhc), while downregulating genes associated with mitochondrial biogenesis (Pgc1α) and antioxidant response (Foxo1, Cox2). These findings suggest esculetin as a potential adjunctive candidate for cardioprotection during DOX chemotherapy. However, given the absence of protein-level validation, further studies are warranted to confirm whether the observed gene expression changes translate into corresponding alterations at the protein level.
