- ArticleAçık Erişim2026Effect of adding a superficial parasternal intercostal plane block to the serratus anterior plane block on postoperative analgesic outcomes in breast cancer surgery: a prospective, randomized, double-blind trial
Akbulut, Fazil Ahmet; Kürşad, Hüsnü; Sözüak, Muhammed Emin; Ahiskalioglu, Ali
Background: Breast cancer surgery is frequently associated with moderate to severe postoperative pain. While the serratus anterior plane block (SAPB) provides effective anterolateral chest wall analgesia, it may inadequately cover the anterior intercostal nerve branches. This study aimed to determine whether adding a superficial parasternal intercostal plane block to SAPB improves postoperative opioid consumption, pain scores, and dermatomal sensory coverage. Methods: In this prospective, randomized, double-blind trial, 64 patients aged 18–65 years undergoing elective breast surgery were randomly allocated into two groups. Group S (n = 32) received a preoperative SAPB with 30 mL of 0·25% bupivacaine combined with a parasternal injection of 10 mL saline. Group S + P (n = 32) received a SAPB with 30 mL of 0·25% bupivacaine plus a parasternal block with 10 mL of 0·25% bupivacaine. All patients were managed with an identical multimodal postoperative analgesia protocol. The primary outcome was postoperative opioid consumption. Secondary outcomes included visual analogue scale (VAS) pain scores, dermatomal sensory analysis and side effects. Results: Baseline demographic characteristics and surgical variables were comparable between groups (p > 0·05). Sensory blockade of the T3, T4, and T5 dermatomes was significantly more frequent in the S + P group than in the S group (p < 0·05). However, postoperative VAS pain scores at rest and during movement, as well as total opioid consumption, were comparable between groups (p > 0·05). Conclusions: Although the addition of a superficial parasternal intercostal plane block to SAPB resulted in wider anterior chest wall dermatomal coverage, it did not confer additional benefit in terms of postoperative pain scores or opioid consumption under a multimodal analgesia regimen. These findings suggest that SAPB-based strategies, when combined with systemic analgesics, may provide sufficient postoperative analgesia in patients undergoing mastectomy.

Vatansever, Mehmet; Gürsoy, Ersan; Hayme, Serhat
Background: This study aimed to evaluate the hemoglobin, albumin, lymphocyte, and platelet (HALP) score as an exploratory adjunctive marker associated with reduced bone density, particularly in resource-limited settings. Methods: This cross-sectional study was conducted between October 2022 and October 2023 at the Physical Therapy and Rehabilitation Hospital in Trabzon, Türkiye. This study included 267 patients who had not been previously diagnosed with osteopenia or osteoporosis and had undergone a dual-energy X-ray absorptiometry (DEXA) scan within the previous year. The participants were categorized into the normal, osteopenic, and osteoporotic groups according to their DEXA results. Statistical analysis included receiver operating characteristic (ROC) curve assessment, sensitivity and specificity calculations, and multiple logistic regression analysis to evaluate the discriminative performance of the HALP score and its adjusted association with reduced bone density. Results: Patients with reduced bone density (osteopenia or osteoporosis) had substantially lower HALP scores than those with a normal bone density. The area under the curve (AUC) for the HALP score was 0.625, indicating a limited discriminative ability. The HALP score exhibited a sensitivity of 65.36% and a specificity of 55.26% for identifying reduced bone density. In multivariable analysis, higher HALP remained associated with lower odds of osteopenia/osteoporosis after adjustment (OR = 0.757, 95% CI 0.624–0.917, p = 0.004). Conclusion: Given its limited discriminative ability, HALP should not be interpreted as a stand-alone screening test. At most, it may represent a low-cost exploratory adjunctive signal that could help support consideration of confirmatory DEXA in resource-limited settings, pending external validation. Trial registration: Not applicable.

Crimean-Congo hemorrhagic fever (CCHF) during pregnancy is associated with severe maternal and fetal complications, but evidence remains limited to heterogeneous case reports. We synthesized patient-level data from published and unpublished cases identified through systematic database searches and direct collaboration with centers in CCHF-endemic areas. We harmonized patient-level clinical, laboratory, obstetric, treatment, and outcome data for 55 pregnancies. The maternal mortality rate was 28.8% (15/52), and fetal loss occurred in 57.4% (31/54) of cases. Maternal mortality rates increased with advancing gestation, reaching 31.3% in the third trimester, but fetal loss was highest (85.7%) in the first trimester. Ribavirin treatment during the third trimester was associated with lower observed maternal mortality rates (odds ratio 0.33, 95% CI 0.13–0.84; p = 0.031). Our findings highlight gestational age–specific risks of CCHF and support the need for standardized management protocols and strengthened CCHF surveillance during pregnancy in endemic regions.
- ArticleYalnız künye2026Synthesis of new halo-substituted quinolinecarboxaldehyde-hydrazone derivatives, structural illuminations, investigation of effects on acetylcholinesterase activity and molecular modeling studies
Sirinzade, Hanif; Faydali, Nagihan; Muhammed, Muhammed Tilahun; Dilek, Esra
Acetylcholinesterase (AChE) is a key serum esterase with antiatherosclerotic effects by preventing lipid peroxide oxidation. AChE inhibitors also serve as therapeutic targets for Alzheimer's disease. Newly developed halo-substituted quinolinecarboxaldehyde-hydrazone derivatives (1a–1 s) show significant pharmacological potential due to their broad biological activities. This study evaluated the inhibitory effects of halo-substituted quinolinecarboxaldehyde-hydrazone derivatives on AChE. Acetylcholine esterase IC₅₀ values ranged from 0.143 to 2.015 µM, and Kᵢ values from 0.078 ± 0.030 to 1.074 ± 0.105 µM. Tacrine served as the positive control. Molecular docking studies revealed that compounds 1b and 1k displayed pi-pi interactions (pi-alkyl, pi-pi stacking) and conventional hydrogen bonding. The molecular dynamics simulation study disclosed that compounds 1b and 1k formed stable complexes with the enzyme structure. Moreover, the density functional theory study demonstrated that the active compounds had similar electrical property and reactivity. Together with this, compound 1e was anticipated to exhibit the highest chemical stability among the compounds investigated. All of the compounds were shown to have a high absorption from the gastrointestinal tract and to meet Lipinski's criterion in SwissADME. As a result, it was determined that new halo-substituted quinoline carboxaldehyde-hydrazone derivatives showed strong inhibition effect on this enzyme. In comparison to the reference chemical Tacrine, 1k had the most potent inhibitory activity against AChE. In this context, we anticipate that the findings of this study will aid in identifying the adverse effects of both existing and novel quinolinecarboxaldehyde-hydrazone-based pharmaceutical drugs under development. Additionally, it should be effective in the production of novel AChE inhibitors.

Although chemotherapy is still a widely used primary treatment modality for many cancers, it is an accepted fact that it causes many undesirable side effects and toxicity. Until a new, side-effect-free treatment modality is developed, new resources will continue to be sought to prevent and reduce the adverse effects of chemotherapy. In this study, we investigated the potential of apigenin, selenium, and their combination to prevent cardiovascular toxicity induced by 5-fluorouracil (5FU), an important chemotherapeutic agent. The study consisted of control (C), 5FU, apigenin + 5FU (Api + 5FU), selenium + 5FU (Se + 5FU), and apigenin + selenium + 5FU (Api + Se + 5FU) groups. Api (50 mg/kg) and Se (0.5 mg/kg) were administered orally to rats for 7 days. On Day 5, 5FU was administered i.p. at a dose of 150 mg/kg. Administration of 5FU significantly reduced GSH levels and CAT and TrxR activities in both cardiac and thoracic aortic tissues. It also decreased Bcl-2 expression and increased Bax and Caspase-3 expression in cardiac tissue, while reducing Bcl-2 and increasing Bax expression in the thoracic aorta. Thoracic aortic MDA levels were also significantly elevated. On the contrary, oxidative stress and apoptosis were significantly decreased, and cardiovascular damage was attenuated in apigenin and selenium treated groups. In this study, it was determined that 5FU caused a higher level of toxicity to the heart compared to the thoracic aorta. In addition, both apigenin and selenium were shown to have significant protective effects on the cardiovascular system against 5FU toxicity. However, the cardiovascular protection against 5FU toxicity was found to be higher when apigenin and selenium were used together.

BACKGROUND This study aimed to establish normative values for the femoral inclination angle (FIA) and medial proximal femoral angle (MPFA) using magnetic resonance imaging (MRI), which minimizes projectional and rotational errors, in a large cohort. Sex-based and side-to-side comparisons, as well as correlations with age, were also investigated for these proximal femoral angle measurements. MATERIAL AND METHODS After exclusion of patients with trauma, infection, prior surgery, advanced osteoarthritis, femoroacetabular impingement, avascular necrosis, coxa vara, coxa valga, and low-quality magnetic resonance images, 631 pairs of proximal femurs were retrospectively evaluated for FIA and MPFA measurements. All measurements were performed by consensus between 2 radiologists, then analyzed regarding sex and side differences and correlations with age. RESULTS The mean FIA was 128.53±4.62°, whereas the mean MPFA was 86.62±3.40°. No significant differences were observed between men and women (FIA: P=0.057, MPFA: P=0.050) or between right and left sides (FIA: P=0.178, MPFA: P=0.853). Age was not significantly correlated with either proximal femoral angle measurement. No significant association was found between age and FIA (right: P=0.554, left: P=0.321) or MPFA (right: P=0.954, left: P=0.935). CONCLUSIONS This large MRI-based study provides normative reference values for FIA and MPFA. Neither angle was significantly influenced by sex, side, or age, suggesting that these parameters remain relatively stable across adult populations. These findings may serve as useful reference values for assessment of proximal femoral morphology and preoperative orthopedic planning.

Ocular ischemic syndrome (OIS) is a vision-threatening disorder characterized by retinal ischemia, oxidative stress, and inflammation. In this study, the protective effects of anakinra, an interleukin-1 receptor antagonist, were investigated and compared with those of tocilizumab, an interleukin-6 receptor antagonist. Twenty-four male Wistar rats were randomly assigned to four groups (n = 6 per group): sham-operated (SG), carotid artery clamping/unclamping (CUG), anakinra-treated carotid artery clamping/unclamping (ACUG), and tocilizumab-treated carotid artery clamping/unclamping (TCUG). An experimental ocular ischemic injury model induced by transient bilateral common carotid artery occlusion was established by transient bilateral common carotid artery clamping followed by reperfusion. Retinal tissues were analyzed for malondialdehyde (MDA), total glutathione (tGSH), superoxide dismutase (SOD), catalase (CAT), tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and interleukin-6 (IL-6) levels, and histopathological examinations were performed. OIS markedly increased MDA, TNF-α, IL-1β, and IL-6 levels while significantly decreasing tGSH, SOD, and CAT activities. Both anakinra and tocilizumab attenuated oxidative stress and inflammatory responses; however, anakinra produced notably greater improvements in MDA, tGSH, SOD, and CAT levels. Histopathological evaluation demonstrated that both treatments reduced retinal injury, edema, vascular congestion, and inflammatory cell infiltration. Although retinal architecture appeared slightly better preserved in the anakinra-treated group, histopathological scores were comparable between the treatment groups. These findings suggest that anakinra may attenuate oxidative and inflammatory retinal damage in this experimental ocular ischemic injury model and may provide greater biochemical protection than tocilizumab, whereas histopathological protection was comparable between treatments. Nevertheless, further experimental and clinical studies are required to confirm these findings.

Polystyrene nanoplastics (PsNPs) represent an emerging environmental threat with potential adverse effects on pancreatic tissue. This study investigated whether probiotic supplementation (PB) could attenuate PsNPs-induced pancreatic injury by modulating oxidative stress, inflammation, and P2X7R/Caspase-1 signaling. Forty male Sprague–Dawley rats were divided into five groups: control, probiotic (PB 1010), PsNPs (100 nm, 10 mg/kg/day), PsNPs + PB 109, and PsNPs + PB 1010. After 35 days of oral administration, oxidative stress markers, inflammatory cytokines, P2X7R/Caspase-1 gene expression, signaling proteins (NF-κB, PI3K, p-PI3K, p38 MAPK, TGF-β), histopathology, and immunohistochemistry (Bcl-2, Caspase-3, Insulin, P2X7 and Caspase-1) were evaluated. PsNPs significantly upregulated P2X7R and Caspase-1 expression (Sp < 0.0001), elevated NF-κB levels (p < 0.0001), increased IL-1β and TNF-α, and induced oxidative stress with elevated MDA, depleted SOD (p < 0.001), and reduced GSH (p < 0.01). Histopathology revealed inflammatory infiltration and cellular degeneration. Insulin immunoreactivity decreased while Caspase-3 increased. Probiotic co-treatment at 101⁰ CFU/day significantly attenuated P2X7R/Caspase-1 upregulation, reduced NF-κB expression, restored SOD activity and GSH levels, normalized cytokines, and preserved Insulin immunoreactivity. PsNPs induce pancreatic injury through P2X7R/Caspase-1/NF-κB signaling activation. Multi-strain probiotic supplementation exerts dose-dependent protective effects by modulating purinergic signaling and restoring antioxidant capacity.
- ArticleYalnız künye2026Impact of CT-based 3D modelling on hip joint anatomical measurements: a comparative study with 2D methods
Otugüzel, Mahmut; Subaşı, İzzet Özay; Tanoğlu, Oğuzhan
Introduction: The functionality of the hip joint is primarily determined by the anatomical relationship between acetabulum and femur. Impairment of this relationship can lead to diseases such as femoroacetabular impingement and hip osteoarthritis. In patients undergoing total hip arthroplasty, accurate placement of the acetabular and femoral components in accordance with native 3-dimensional anatomy (3D) is crucial for obtaining prosthesis stability and survival, also for preventing postoperative complications such as wear, dislocation and osteolysis. Therefore, 3D determination of native anatomical features of the hip joint is highly important. Methods: A total of 100 adults were randomly selected. Right femur and acetabulum were segmented using MIMICS program. 2D and 3D measurements of femoral anteversion according to posterior condylar and transepicondylar axis, acetabular anteversion, acetabular inclination and combined anteversion were determined. The differences between gender groups and the correlations of two- and three-dimensional measurement results were evaluated. Results: According to 2D and 3D measurements, statistically significant differences were observed between gender groups in all parameters except for 2D acetabular inclination. A very strong correlation was observed between 3D femoral anteversion measurements performed using the posterior condylar axis and transepicondylar axis. Our findings revealed statistically significant differences between 2D and 3D measurements in most parameters, which have clinical relevance. Conclusions: A very strong correlation between femoral anteversion measurements obtained using the transepicondylar and posterior condylar axes supports the intraoperative use of the transepicondylar axis reliably. We believe the data obtained from our study will contribute to understand the 3-dimensional native anatomy of the hip joint.

Yalınkılıç, Abdulaziz; Huyut, Zübeyir; Akyol, Mehmet Edip; Gündemir, Damla Ayan; Huyut, Mehmet Tahir; Çetin, Yaser Said
Objective: Prestin, expressed in cochlear outer hair cells, is essential for auditory signal amplification and may serve as a biomarker for cochlear injury related to trauma. This study aimed to investigate the potential role of prestin in the diagnosis and prognosis of cranial trauma. Methods: A total of 150 participants were included, comprising 110 patients aged 18–70 who were treated for head trauma and 40 age-matched healthy controls. The patients were divided into two groups of 55 each, with Glasgow coma scale (GCS) ≥13 and GCS ≤8, according to the GCS. Prestin was measured by enzyme-linked immunosorbent assay in serum on days 1, 3, and 7 in the patient and also the control groups. In the diagnosis of trauma and determining trauma severity, important risk levels (cut-off values) according to prestin's measurement days were calculated by receiver operating characteristic (ROC) analysis. Results: Prestin values in both the GCS ≥13 and GCS ≤8 groups were dramatically lower than those in the control on all measurement days. Additionally, prestin values in the GCS ≤8 group on days 3 and 7 tended to be lower than those in the GCS ≥13 group. Furthermore, there was a tendency for serum prestin values to increase as time progressed especially in the GCS ≥13 group. ROC analysis showed that prestin values on the first, third, and seventh days showed high accuracy in detecting traumatized individuals with area under the curve (AUC) values of 85%, 81%, and 75%, respectively (sensitivity: 84%, 79%, and 83%, respectively). Also, prestin data on the first day accurately detected the severity of trauma at a high rate (sensitivity, 82%; AUC, 90%). Conclusion: Our findings suggest that serum prestin levels on the first day after trauma may provide supportive information regarding trauma severity. However, further controlled studies are needed before definitive diagnostic or prognostic use.

Vescalagin have been isolated from Camu-camu (Myrciaria dubia). Vescalagin compound was a found to be the main C-glycosidic ellagitannin with a bridging open ring d -glucose. A variety of techniques were used to assess vescalagin's antioxidant capacity, including its capacity to reduce potassium ferric cyanide, scavenge N,N-dimethyl-p-phenylenediamine dihydrochloride radicals (DMPD•+), scavenge 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulphonate) radicals (ABTS•+), scavenge 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH·), and reduce cupric ions (Cu2+). Likewise, butylated hydroxyanisole (BHA), Trolox, α-tocopherol, and butylated hydroxytoluene (BHT) were employed as reference antioxidants for assessment. For DPPH radical scavenging, vescalagin had an IC50 value of 9.65 μg/mL (r2: 0.9225), while the IC50 values for BHA, BHT, α-Tocopherol, and Trolox were 10.10, 25.95, 11.31, and 7.05 μg/mL properly. The resarch revealed that vescalagin reacted better than BHA, α-tocopherol, and BHT with regard to DPPH· scavenging, but nearly similiar to Trolox. Vescalagin was also shown to be a polyphenolic secondary metabolite that inhibited a number of metabolic enzymes, particularly α-glycosidase, carbonic anhydrases I and II (CA I and II), butyrylcholinesterase (BChE), and acetylcholinesterase (AChE). The Ki values of vescalagin showed 5.87, 3.89, 11.75, 16.23, and 16.08 nM towards AChE, BChE, hCA I, hCA II, and α-glycosidase, respectively. These enzymes are linked to several ilness such as diabetes, Alzheimer disease (AD), epilepsy and glaucoma. The obtained findings clearly indicate that vescalagin can be used as a reference polyphenolic antioxidant or enzyme inhibitor in analytical and screening assays.

Karadogan, Muhammed Talha; Yeni, Yesim; Yeter, Bahtinur; Gungor, Ali; Altuner, Durdu; Azade, Faraz; Gursul, Cebrail; Suleyman, Halis
Although doxorubicin (DOX) is a first-line antineoplastic drug, its clinical use is limited due to its widespread side effects. Hepatic and renal metabolic clearance causes DOX accumulation in these organ systems, ultimately leading to oxidative-inflammatory damage. This study investigated the potential impact of syringic acid (SA), a natural antioxidant phenolic compound, against in vivo DOX-evoked hepatorenal damage. SA was orally administered to rats at 100 mg/kg body weight for 10 consecutive days. On the 8th day of the study, DOX was administered at 20 mg/kg by single-dose intraperitoneal injection to prompt hepatorenal toxicity. Hepatorenal toxicity was assessed by measuring serum alanine transferase (ALT), aspartate transferase (AST), blood urea nitrogen (BUN), and creatinine (Cre). Histopathological evaluation of kidney and liver tissues was also performed to observe DOX-related damage. The concentrations of malondialdehyde (MDA) and glutathione (GSH), along with the activities of catalase (CAT) and superoxide dismutase (SOD), were measured in the hepatorenal tissues, in addition to the concentration of 8-OHdG in serum, to determine the protective effect of SA against oxidative stress. Tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) levels were investigated with molecular and immunohistochemical analysis in hepatorenal tissues, evaluating the inflammation. SA treatment prevented DOX-aberrant elevation of hepatorenal biomarkers in serum, besides histopathological alterations in rats’ liver and kidney. Additionally, SA markedly counteracted DOX-provoked oxidative damage as evidenced by outstanding reduction in lipid peroxidation and 8-OHdG, conjointly promoted GSH contents, and amelioration of activities of SOD and CAT. Similarly, SA-treated rats exhibited attenuation of hepatorenal inflammation via diminished TNF-α and IL-6 levels. SA exerted a hepatorenal protective effect in rats by exhibiting antioxidant and anti-inflammatory activities.

5-fluorouracil (5-FU) is an important chemotherapeutic agent that has been used alone or in combination with other chemotherapeutics for years in the treatment of various malignancies. However, as with other chemotherapeutic agents, the disadvantage of 5-FU is that it damages healthy cells as well as cancer cells. Natural compounds are the first option that comes to mind to reduce and/or prevent undesirable side effects of chemotherapeutics. In this study, we aimed to determine the effects of Juglans regia extract (JR), whose antioxidant, anti-inflammatory, anti-diabetic, and antimicrobial effects were previously determined, on 5-FU-induced autophagy, oxidative stress, and apoptosis after its constituents were determined by LC-MS/MS. Experimental groups were formed as Control (C), JR, 5-FU, and JR + 5-FU. Rats were pretreated with JR at a dose of 200 mg/kg for seven days, and on the eighth day, a single dose of 100 mg/kg 5-FU was administered intraperitoneally to the JR + 5-FU and 5-FU groups. 5-FU disturbed the oxidant-antioxidant balance by increasing Keap1, TOS, and 8-OHdG levels and decreasing TAS levels and triggered cellular autophagy and apoptosis by increasing LC3B, Bax, and Caspase-3 levels and decreasing Bcl-2. JR pretreatment was shown to have modulating effects on the Keap1/Nrf2 pathway in the reorganization of the oxidant-antioxidant balance, to show anti-apoptotic activity, and to partially alleviate autophagy. JR’s bioactive components were shown to alleviate oxidative stress and associated DNA damage via Keap1/Nrf2 and also attenuate fluorouracil-induced kidney damage by reducing autophagy and apoptosis, two important forms of programmed cell death, and have a promising potential against nephrotoxicity.

Onopordum acanthium L. (Asteraceae), a traditionally used medicinal plant, was investigated for its morphological, phytochemical, and biological properties in this comprehensive study. Methanolic and aqueous extracts were prepared from different parts of the plant (root, stem, leaf, flower, and fruit) and analyzed for their phenolic composition, antioxidant, antidiabetic, anticholinesterase, antimicrobial, and genotoxic activities. LC-MS/MS analysis revealed that chlorogenic acid was most abundant in the flower (3045.38 ng/mL) and root (728.53 ng/mL) aqueous extracts, while quinic acid reached 856.27 ng/mL in the root. Quercetin, apigenin, and luteolin were also detected at significant levels. The fruit methanol extract showed the highest total phenolic (100.78 ± 0.0015 µg GAE/mg), flavonoid (603.67 ± 0.0015 µg RE/mg), and tannin (186.22 ± 0.0015 µg TAE/mg) contents. Antioxidant assays demonstrated notable ABTS•⁺ (38.38 ± 0.0042%) and DPPH• (28.43 ± 0.0252%) scavenging capacities in the same extract. Regarding enzyme inhibition, the flower aqueous extract showed the strongest α-glucosidase inhibition (45.58%), while the fruit aqueous extract exhibited moderate α-amylase inhibition (26.33%). The stem methanol extract displayed the highest acetylcholinesterase inhibition (19.02%), whereas the root aqueous extract showed the highest butyrylcholinesterase inhibition (10.76%). Antimicrobial assays revealed moderate antifungal activity, particularly against Candida tropicalis (MIC = 312.5 µg/mL), with the flower and fruit methanol extracts being the most effective. Genotoxicity assessment using Ames and Allium tests confirmed biosafety up to 5 mg/plate and 250 mg/L, respectively, except for flower extracts, which showed slight cytogenotoxicity. Overall, this study highlights O. acanthium as a promising natural source of phenolic compounds with therapeutic potential, particularly in managing oxidative stress, diabetes, and neurodegenerative disorders.

This study reports the synthesis and characterization of mixed-ligand Cu(II), Co(II), and Mn(II) complexes derived from 2-(((2-hydroxyphenyl)imino)methyl)-4-methoxy-6-nitrophenol (L1) and 5-nitro-1,10-phenanthroline (L2). The ligand and complexes were characterized by 1H/13C NMR (L1 only), FT-IR, UV–vis, LC-MS/MS, TGA, and elemental analysis. Cytotoxicity studies against A549 and HTB-54 lung cancer cells identified the Cu(II) complex as the most active compound, with IC50 values of 24.05 and 22.08 µM, respectively, while showing lower toxicity toward BEAS-2B normal cells (IC50 = 39.08 µM). The Cu(II) complex significantly inhibited cell migration and induced apoptosis, supported by increased Bax expression in qPCR analysis. Molecular docking with selected cancer-related protein targets further supported its proposed anticancer mechanism. The catalytic activity of the complexes was investigated for the solvent-free cycloaddition of CO2 with epoxides. The Co(II) complex exhibited the highest catalytic performance, affording 86.5% yield and 97.0% selectivity in the presence of DMAP under optimized conditions. Density functional theory (DFT) calculations provided the EHOMO, ELUMO, chemical potential, hardness, softness, electrophilicity index, and MEP profile of the ligand and complexes, demonstrating their multifunctional potential for anticancer and sustainable catalytic applications.

Introduction: The genus Tanacetum has a long history of use and is well known for its traditional applications in food, medicine, cosmetics, and agriculture. The biological effects of Tanacetum armenum were investigated, and its phytochemical profiles were analysed. Methods: Phytochemical analysis was conducted using liquid chromatography-mass spectrometry/mass spectrometry, and antioxidant, target enzyme inhibition, and antigenotoxic effects were evaluated. The antiproliferative effects and mechanisms of the extracts were investigated in vitro, and the possible mechanisms of the main compounds were analysed in silico. Results: Phytochemical profile analysis revealed significantly higher levels of chlorogenic acid (64.894 mg/g extract), quinic acid (45.377 mg/g extract), and 1,5-dicaffeoylquinic acid (18.150 mg/g extract). The methanol extract (IC50:116.56 ± 2.84 µg/mL) exhibited strong tyrosinase enzyme inhibition than ethylacetate (IC50:184.04 ± 3.02 µg/mL) and aqueous extracts (IC50:340.84 ± 3.46 µg/mL). The aqueous extract (IC50:328.23 ± 4.22 µg/mL) showed the highest inhibition of acetylcholinesterase, while the methanol extract (IC50:504.87 ± 4.78 µg/mL) demonstrated the highest inhibition of butyrylcholinesterase. The ethylacetate extract exhibited the strongest antimicrobial effect against Staphylococcus aureus, with a MIC value of 62.5 µg/mL. The extracts did not show any genotoxic effects. The ethyl acetate extract exhibited significant cytotoxic effects (IC50:25.00–108.16 µg/mL) against cancer cell lines (p < 0.001). To investigate the mechanism by which the extracts inhibit MCF-7 cell viability, the expression levels of the cyclin E1 and caspase-8 were evaluated using western blot analysis. In addition, a colony formation assay was performed, and migration potential was analysed using a scratch wound healing assay. Using in silico approaches, the molecular docking and binding parameter values of cyclin E1 and caspase-8 were calculated. Chlorogenic acid was found to have the best binding value and mode of in silico interaction with the target crystal structures. The docking scores calculated from these binding parameter values were -7.824 kcal/mol for the cyclin E1 (8H6P) target, -7.126 kcal/mol for the caspase-8 (3KJQ) target, and -8.182 kcal/mol for the human oestrogen receptor (3ERT) target. Conclusion: T. armenum, which has demonstrated a strong cytotoxic effect through in vitro and in silico mechanism studies, can be considered a promising potential natural resource for cancer treatment.
