Bingöl, Koray; Arslan, Mithat Kerim

Aim: Lumbosakral transitional vertebrae (LSTV) are anatomical variations where a vertebra exhibits characteristics of both lumbar and sacral vertebrae. This study aims to assess anatomical changes in LSTV patients by measuring intervertebral disc height (IVDh), vertebral foramen anteroposterior diameter (FVAPd), and intervertebral foramen height (FIVh), with consideration of age and gender differences. Material and Methods: This retrospective study included 274 patients diagnosed with LSTV. All MRI images were acquired with a 1.5T MRI machine and a 32-channel lumbar coil. The presence of LSTV, forming the morphological data, was identified from coronal and sagittal reformat images and was classified as either lumbarization or sacralization. Morphometric data (IVDh, FIVh and FVAPd) were acquired from axial, sagittal, and coronal planes using both soft tissue and bone window settings. Results: The mean patient age was 41.6±13.0 years, with a female majority (55.8%). LSTV was predominantly lumbarization (96.4%), with sacralization in the remaining cases. The overall mean FVAPd was 11.2±2.4 mm, mean IVDh was 9.1±1.8 mm, and mean FIVh was 18.8±2.4 mm on the right and 19.1±2.4 mm on the left. IVDh levels were consistent across age groups, while FVAPd values were similar between genders. Patients aged 40 and under had higher FVAPd and FIVh levels, and male patients had higher IVDh and FIVh levels compared to females. Conclusions: The morphometric characteristics of LSTV may vary according to age and gender. Considering these factors in accurately identifying the anatomical variations of LSTV may play a significant role in determining appropriate treatment and management strategies.

Does the incidence of post-COVID pulmonary complications in vaccinated individuals correlate with the types of vaccines they received?

Çınar, Hasibe Gökçe; Memis, Kemal Bugra

Aim: Although several research have been undertaken to investigate the impact of the vaccination on long Coronavirus Disease 2019 (COVID-19) syndrome or post-acute sequelae, there is a lack of published evidence on the long-term effects of vaccines on lung-sequelae-related disease. Considering the limited global COVID-19 vaccine distribution, it is essential to establish the impact of vaccination in reducing pulmonary complications. Turkey has been offering COVID-19 vaccines from two platforms, including BNT162b2 (Pfizer-BioNTech, mRNA vaccine) and CoronaVac (Sinovac, inactivated vaccine). This study aimed to evaluate the efficacy of BioNTech and Sinovac vaccines in reducing post-COVID-19 pulmonary complications in individuals. Material and Methods: A total of 94 patients COVID-19 pneumonia patients who were categorized based on the quantity of BioNTech or Sinovac vaccines they received before their first COVID-19 infection were included. The inclusion criteria consisted of a confirmed diagnosis of COVID-19 pneumonia through polymerase chain reaction testing, availability of the mentioned before and follow-up computed tomography scans, and administration of at least one dose of vaccine. Results: The number of complications in patients fully vaccinated with Sinovac and who experienced post-COVID lung complications was significantly greater than in those vaccinated with BioNTech. The C-reactive protein and D-Dimer measurements of individuals who experienced complications in the Sinovac vaccinated group were significantly elevated on the index date. Conclusion: The quantity of lung sequelae after COVID and laboratory parameters indicating this result were found to be higher in inactivated virus vaccines than in mRNA vaccines. This suggests that the protection of inactivated vaccines may be insufficient in severe cases.

Detection of Lung Cancer Cells Using Deep Learning Methods

    Akar, Funda; Genç, Muhittin

Lung cancer stands out as a high mortality, fatal disease worldwide. Early diagnosis is crucial for effective treatment of this disease; however, treatment options can be limited when it is often diagnosed in advanced stages. This study examines the role of artificial intelligence (AI) techniques in early diagnosis of lung cancer and emphasizes the advantages it provides. Particularly, the ability of deep learning algorithms to extract meaningful features from complex datasets indicates significant potential for detecting early stages of lung cancer. In this context, it is anticipated that AI-supported diagnostic systems have the potential to significantly improve lung cancer diagnostic methods by reducing the workload of radiologists and increasing accuracy rates. In this study, a total of 6 datasets were obtained by applying Gabor filter and Histogram Equalization+CLAHE filter to original datasets. The results obtained in the diagnosis of lung cancer using Convolutional Neural Networks (CNN) and YOLO algorithms are evaluated in two different categories. One of these categories is the investigation of the effect of image preprocessing methods. The other is the investigation of the effect of dataset partitioning into training, testing, and validation on success. According to the results obtained, the highest success rate in terms of F1 Score for the CNN model was achieved in both dataset partitioning (70%-20%-10% and 60%-20%-20%) with the datasets subjected to Histogram Equalization+CLAHE filter. It was obtained as 99%. For the YOLO model, the highest success rate was determined as 96% F1 Score with the same preprocessing technique and dataset partition. The effect of image preprocessing and dataset partitioning on success is not as high in the YOLO model as it is in the CNN model.

Quantum Chemical Computations, Molecular Docking, and ADMET Predictions of Cynarin

    Yavuz, Sevtap Çaglar

Cynarin (1,3-o-dicaffeoylquinic acid) is one of the biologically active functional food components which is the most well-known caffeoylquinic acid derivative found in artichoke. The structural and electronic features of cynarin compound were investigated theoretically using density functional theory (DFT). The highest occupied molecular orbital (HOMO) and the least occupied molecular orbital (LUMO) are the most significant orbitals in molecules, these orbitals are quite helpful to know several molecular features such as the chemical reactivity, kinetic stability, electronegativity, chemical potential, electrophilicity index, chemical hardness and softness and electronegativity. Molecular orbital analysis HOMO-LUMO was used to explore the stability of the molecule. Moreover, physicochemical properties, drug-likeness, and toxicity estimation of the cynarin compound were appraised owing to ADMET (including absorption, distribution, metabolism, excretion, and toxicology). Molecular docking was carried out to examine the biological activity of the cynarin compound. 5A19, a liver cancer biomarker, is human methionine adenosyl-transferase enzymes. Cynarin-methionine adenosyl-transferase enzyme binding energy value was calculated as -7.9 kcal/mol. As a result, this in silico study confirmed that cynarin has the potential to be a drug by revealing its protective effect against liver diseases.

Objective: This study aimed to investigate the gastroprotective effects of Laurus nobilis leaves essential oil (LANO) against indomethacin (INDO)-induced gastric ulcers in rats. Materials and Methods: In this study, an indomethacin-induced gastric ulcer model was employed. 30 Sprague-Dawley rats were divided into five groups (n=6): Control, LANO, INDO, INDO with famotidine (FAM), and INDO with LANO. Indomethacin (25 mg/kg) induced ulcers, while LANO and FAM were administered by oral gavage at 200 mg/kg and 40 mg/kg, respectively. Gastric tissues underwent histopathological examination for ulceration, and biochemical assays measured total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI), and nitric oxide (NO) levels. Results: Compared to the INDO group, treatment with LANO significantly decreased the number of gastric ulcer foci. Biochemically, LANO moderated TOS and OSI levels and preserved TAS, indicating reduced oxidative stress. Additionally, LANO appeared to stabilize NO levels. These biochemical findings were corroborated by histopathological examination. Conclusions: The study's results indicate that LANO may be beneficial in protecting against NSAID-induced gastric damage. LANO's observed modulation of oxidative stress markers and NO levels suggests its potential role in managing gastric ulcers.

The effects of cetuximab with agomelatine on gene expression in colon cancer cells

Erbaş, Elif; Kara, Adem; Ustundag, Hilal; Albayrak, Kevser; Köse, Rukiye

This study investigated the combined effects of agomelatine, a melatonergic antidepressant, and cetuximab, an EGFR inhibitor, on the colorectal cancer cell line (Caco-2). Caco-2 cells were treated with agomelatine (0.3 μg/ml and 3 μg/ml) and cetuximab (50 μg/ml), individually and in combination, for 24 and 48 hours. Cell viability was assessed using the MTT assay. Gene expression analysis of EGFR, BCL2, PIK3CA, BAX, mTOR, and AKT3 was performed using real-time PCR. All treatment groups showed significant decreases in cell viability compared to the control (p<0.05), with enhanced effects in combined treatments. EGFR expression was significantly reduced in drug-treated groups, particularly with cetuximab (p<0.05). While changes were observed in BCL2, PIK3CA, BAX, mTOR, and AKT3 expression, these were not statistically significant (p>0.05). This study demonstrates the potential synergistic cytotoxic effects of agomelatine and cetuximab on Caco-2 colorectal cancer cells. The significant reduction in EGFR expression suggests a potential mechanism of action. These findings provide insights into combining chemotherapeutic agents with drugs addressing circadian rhythm disorders in CRC treatment strategies. Further research is warranted to elucidate the clinical implications of these observations.

Neuroprotective effect of chlorzoxazone against glutamate toxicity in rat primary cortex neuron culture

Hacimuftuoglu, Ahmet; Çıçek, Betül; Yeni, Yeşim

Glutamate (Glut) toxicity is one of the main causes of neurological diseases. Chlorzoxazone (CZ) is a muscle relaxant used to decrease pain and inflammation associated with acute and chronic twists and bruises. Here, we objected to research the neuroprotective effect of CZ applied to reverse Glut-induced neurodegeneration in the neonatal cerebral cortex through anti-inflammatory and antioxidant mechanisms. Neonatal cortical neurons were exposed to Glut and different doses of CZ (10, 20, and 40 µM) were applied to assess the effect of CZ on Glut toxicity. We then examined changes in cell viability, inflammation, and oxidative stress. Our cell viability analysis showed that CZ protected cells from Glut-induced neuronal damage. In addition, the neuroprotective properties of CZ were evaluated by examining oxidative and antioxidant parameters such as MDA, MPO, CAT, GSH, GPx, and SOD. In line with the data obtained, it was observed that the cell viability rate decreased to 60% in the Glut group. However, with CZ application, the most significant increase in cell viability was seen at the 40 μM dose (86%), while the least increase was seen at 10 μM CZ (77%). It also proved that CZ increased the activity of antioxidant parameters while reducing oxidative parameters and inflammation. Therefore, the present findings collectively demonstrated that CZ potently inhibits Glut-induced injury in neonatal cortical neurons. The present work is the initial to show the protective effect of CZ in neonatal cortical neurons exposed to Glut excitotoxicity and suggesting that CZ may be used as a therapeutic agent.

Mild TBI was induced by dropping a 450-g iron weight from a height of 1 m onto the skull of the rats. The tDCS + TBI + tDCS group was prophylactically administered 1 mA stimulation for 30 min for 7 days starting 5 days before inducing TBI. In the TBI + tDCS group, tDCS (1 mA for 30 min) was administered 2 h after TBI, on days 1 and 2. Cognitive and locomotor functions were assessed using the novel object recognition and open field tests. The calcium, glutamate, and N-methyl-D-aspartate receptor 1 (NMDAR1) levels in the hippocampus were measured using enzyme -linked immunosorbent assay. RESULTS: Although the motor and cognitive functions were substantially reduced in the TBI group when compared with the sham, they improved in the treatment groups (p<0.05). The calcium, glutamate, and NMDAR1 levels were considerably higher in the TBI group than in the sham (p < 0.001). However, they were considerably lower in the tDCS + TBI + tDCS and TBI + tDCS groups than in the TBI groups (p < 0.05). In particular, the change in the tDCS + TBI + tDCS group was higher than that in the TBI + tDCS group. CONCLUSION: Application of tDCS before the development of TBI improved motor and cognitive dysfunction. It demonstrated a neuroprotective and therapeutic effect by reducing the excitotoxicity via the regulation of calcium and glutamate levels.

Gulcin, Ilhami; Ozden, Eda Mehtap; Mutlu, Muzaffer; Mirzaee, Ziba; Bingol, Zeynebe; Köksal, Ekrem; Alwasel, Saleh; Goren, Ahmet C.

Background This study investigated the antidiabetic and antioxidant properties of hemp seed oil using various bioanalytical methods. Furthermore, this study determined the suppressive properties of hemp seed oil on alpha-amylase, acetylcholinesterase and carbonic anhydrase II that purified by the sepharose-4B-L-Tyrosine-sulfanilamide affinity chromatoghraphy, all of which are related to different metabolic diseases. Moreover, the phenolic concentration in the essential oil was quantified through LC-HRMS chromatography. Thirteen distinct phenolic compounds were detected in hemp seed oil. Additionally, both the chemical components and quantity of essential oils within hemp seed oil were assessed through GC-FID and GC/MS analyses. Results The predominant essential oils in hemp seed oil included linoleoyl chloride (34.62%), linoleic acid (33.21%), and 2-4-di-tert-butylphenol (5.79%). Hemp seed oil's ability to scavenge radicals was studied through the use of 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) and 1,1-diphenyl-2-picrylhydrazil bioanalytical radical scavenging methods. The results unveiled its potent radical-scavenging properties, with an 46.20 mu g/mL for 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radicals and IC50 of 9.76 mu g/mL for 1,1-diphenyl-2-picrylhydrazil radicals. The investigation also extended to explore the reducing capabilities of Fe3+-2,4,6-tri(2-pyridyl)-S-triazine, copper (Cu2+), and iron (Fe3+). Hemp seed oil demonstrated notable inhibitory effect against alpha-amylase (IC50: 545.66 mu g/mL), achethylcholinesterase (IC50: 28.00 mu g/mL), and carbonic anhydrase II (IC50: 322.62 mu g/mL). Conclusions This interdisciplinary research will prove valuable and set the stage for future investigations into the antioxidant characteristics and enzyme inhibition patterns of plants and plants oils that hold medical and industrial significance.

BACKGROUND In cases of coronavirus disease 2019 ( COVID- 19), favipiravir is commonly included to the therapy regimen. Drug interactions between favipiravir and other COVID-19 therapy drugs are frequently researched. However, no research on possible drug interactions between Favipiravir and radiocontrast agents, which have become almost crucial in diagnostic processes while not being part of the treatment, has been found. AIM To determine potential medication interactions between Favipiravir and radiocontrast agents. METHODS The study comprised patients who were taking Favipiravir for COVID-19 therapy and underwent a contrast-enhanced computed tomography (CT) or magnetic resonance imaging (MRI) test while taking the medicine. The computerized patient files of the cases included in the study, as well as the pharmacovigilance forms in the designated hospital, were evaluated for this purpose. RESULTS The study included the evaluation of data from 1046 patients. The study sample's mean age was 47.23 +/- 9.48 years. The mean age of cases with drug interactions was statistically significant greater than that of cases with no drug interactions (P = 0.003). When evaluated with logistic regression analysis, a 1-year raises in age increases the risk of developing drug interactions by 1.63 times (P = 0.023). There was no statistically significant difference in the occurrence of medication interactions between the sexes (P = 0.090). Possible medication interactions were discovered in 42 cases (4%). CONCLUSION The findings of this study revealed that the most notable findings as a result of the combined use of contrast agents and favipiravir were increased creatinine and transaminase values, as well as an increase in the frequency of nausea and vomiting. The majority of drug interactions discovered were modest enough that they were not reflected in the clinic. Drug interactions become more common as people get older.
