- ArticleAçık Erişim2022Is Gastric Residual Volume Measurement Really Necessary to Achieve Targeted Calories?
Koç, Alparslan
Objective: Malnutrition, energy protein, and inadequate or excessive intake of other nutrients have measurable effects on tissues, body function, and clinical outcomes of patients. We aimed to determine the time to reach target calories, nutritional failures, and complications during feeding in measured and unmeasured gastric residual volume patients under ventilation in the intensive care unit. Methods: The study was completed with 62 patients under mechanical ventilatory support in the intensive care unit. According to the consultation order, patients were divided into 2 groups. Gastric residual volume was measured in the control group (gastric residual volume, n = 31) and not in the other (non-gastric residual volume, n = 31). Nutrition nurses continuously monitored all enteral-fed patients, and the results were recorded. Results: The feeding pause of the gastric residual volume group was longer than that of the non-gastric residual volume group (P < .001). The time to reach target calories was higher in the gastric residual volume group than in the non-gastric residual volume group (P = .010). The rate of vomiting as a complication was 9.7% (3 patients) in the gastric residual volume group and 6.5% in the non-gastric residual volume group, although the difference was not significant (P = .641). The observation rate of abdominal distension was 6.5% (2 patients) in the gastric residual volume group and the non-gastric residual volume group (P = .999). The positive end-expiratory pressure (PEEP) values were higher in patients who vomited, but the difference was not significant (P = .203). In patients with abdominal distension, PEEP values were higher than in patients without distension, but the difference was not significant (P = .282). Conclusion: In conclusion, gastric residual volume measurement in patients with mechanical ventilatory support prolonged nutritional breaks and extended the time required to reach target calories compared with patients without gastric residual volume measurement.

In this study, changes in malondialdehyde level (MDA), superoxide dismutase (SOD) and catalase (CAT) activity in fish liver tissue were investigated by applying two doses of 2,4-dichlorophenoxyacetic acid (2,4-D) herbicide to Capoeta umbla fish for 72 hours. Changes in MDA level, CAT and SOD activity were determined by spectrophotometric methods. As a result of the study, it was determined that there was a statistically significant increase in MDA level, CAT and SOD activity in C. umbla liver tissue exposed to different doses of herbicide (p < 0.05). It was observed that 2,4-D herbicide disrupted the oxidant/antioxidant balance in fish liver tissue. However, these two antioxidant enzymes, which are the primary defense mechanism, appear to be resistant to the toxic effect of 2,4-D.

This study aimed to determine the antioxidant activities, phenolic compounds, and hormone contents of 12 species of edible wild plants belonging to 9 families, which are consumed as vegetables by the local people at Ergan Mountain in Erzincan in Turkey. Polygonum cognatum and Malva neglecta were determined to have more antioxidant enzyme activity, more phenolic compounds, and higher hormone content than the other species. The highest catalase (CAT), peroxidase (POD), glutathione reductase (GR), glutathione-s-transferase (GST) values for P. cognatum were determined as 45.12, 94.83, 36.76, and 1218.35 EU g−1, respectively. The highest superoxide dismutase (SOD) and ascorbate peroxidase (AxPOD) content for M. neglecta were determined as 97.53 EU g−1 and 81.93 EU g−1, respectively. P. cognatum is the species in which the highest levels of the hormones indolacetic acid (IAA), gibberellic acid (GA), salicylic acid (SA), cytokinin, zeatin and jasmonic acid were detected. The highest levels of caftaric acid (CA), catechin (CAE), ferulic acid (FA), malvidin-3-o-glucoside (MG), myricetin (MYR), rutin (RT), trans-coumaric acid (TPCA), tyrosol (TY), and vanilic acid (VA) compounds were found in M. neglecta. It was determined that Falcaria vulgaris species had the highest levels of ferulic acid (FA) and quercetin (QUE) phenolics. The results show that edible wild vegetables consumed and studied by the people of the region are an important source of natural antioxidants. The possibilities of using these wild plants as functional foods should be investigated.
- ArticleAçık Erişim2022Macromolecules Influence Cellular Competence and Expression Level of IGFs Genes in Bovine Oocytes In Vitro
Şen, Uğur; Şirin, Emre; Önder, Hasan; Özyürek, Selçuk; Kolenda, Magdalena; Sitkowska, Beata
In vitro maturation (IVM) of mammalian oocytes, which influences subsequent in vitro development of embryos, is affected by the macromolecule content in culture media for the success of oocyte maturation competence, in which the cytoplasmic and nuclear reprogramming events occur. The insulin-like growth factor family (IGFs) promotes the maturation of bovine oocytes and the expansion of cumulus cells and also inhibits apoptosis. This study was, therefore, designed to examine the effects of macromolecules (bovine serum albumin, BSA; fetal calf serum, FCS; and polyvinyl alcohol, PVA) on in vitro nuclear maturation, total cellular protein, glutathione peroxidase (GPx) enzyme activity, and the gene expression level of IGF1, IGF2, and their receptor in bovine oocytes. Oocytes obtained from bovine ovaries were cultured in bicarbonate-buffered medium 199 supplemented with 4 mg/mL BSA, 10% FCS, 1 mg/mL PVA, and without macromolecule supplement (control) during 22 h in the air with a humidified atmosphere and 5% CO2 at 38.5 °C temperature. Supplementation of BSA and FCS increased (χ2 = 9.84; p < 0.05) the percentages of oocytes that reached metaphase II compared to the control and PVA. The amount of protein per ml of cell extracts of oocytes matured in FCS supplemented culture media was higher (p < 0.05) than the oocytes in the PVA and control. The levels of GPx enzyme activity in cell extracts isolated from oocytes in each experimental group did not change over time, but the GPx enzyme activity in oocytes matured in PVA-supplemented culture media was lower (p < 0.05) than in oocytes in the other experimental groups. Transcript for the IGF1 gene was not detected in all experimental groups, but the supplementation of BSA and FCS significantly elevated the transcript level of the IGF2 gene. In addition, the maturation of oocytes with BSA-supplemented media increased the transcript level of the IGF1R gene, whereas the transcript level of the IGF2R gene was similar among macromolecule supplementation groups. The current study concluded that BSA and FCS could improve in vitro bovine oocyte development due to supporting nuclear maturation and increasing the total cellular protein content, GPx enzyme, and transcript activity.

Beans are legumes that play extremely important roles in human nutrition, serving as good sources of protein, vitamins, minerals, and antioxidants. In this study, we tried to elucidate the genetic diversity and population structure of 40 Turkish bean (Phaseolus vulgaris L.) local varieties and 5 commercial cultivars collected from 8 different locations in Erzurum-Ispir by using inter-primary binding site (iPBS) retrotransposon markers. For molecular characterization, the 26 most polymorphic iPBS primers were used; 52 bands per primer and 1350 bands in total were recorded. The mean polymorphism information content was 0.331. Various diversity indices, such as the mean effective allele number (0.706), mean Shannon’s information index (0.546), and gene diversity (0.361) revealed the presence of sufficient genetic diversity in the germplasm examined. Molecular analysis of variance (AMOVA) revealed that 67% of variation in bean germplasm was due to differ-ences within populations. In addition, population structure analysis exposed all local and commercial bean varieties from five sub-populations. Expected heterozygosity values ranged between 0.1567 (the fourth sub-population) and 0.3210 (first sub-population), with an average value of 0.2103. In contrary, population differentiation measurement (Fst) was identified as 0.0062 for the first sub-population, 0.6372 for the fourth subpopulations. This is the first study to investigate the genetic diversity and population structure of bean germplasm in Erzurum-Ispir region using the iPBS-retrotransposon marker system. Overall, the current results showed that iPBS markers could be used consistently to elucidate the genetic diversity of local and commercial bean varieties and potentially be included in future studies examining diversity in a larger collection of local and commercial bean varieties from different regions.
- ArticleAçık Erişim2022The Effect of Mammalian Sex Hormones on Polymorphism and Genomic Instability in the Common Bean (Phaseolus vulgaris L.)
Türkoğlu, Aras; Haliloğlu, Kamil; Balpinar, Özge; Öztürk, Halil Ibrahim; Özkan, Güller; Poczai, Peter
Mammalian sex hormones are steroid-structured compounds that support the growth and development of plants at low concentrations. Since they affect the physiological processes in plants, it has been thought that mammalian sex hormones may cause modifications to plant genomes and epigenetics. This study aims to determine whether different mammalian sex hormones (17 β-estradiol, estrogen, progesterone, and testosterone) in several concentrations (0, 10−4, 10−6, and 10−8 mM) affect genetic or epigenetic levels in bean plants, using in vitro tissue cultures from plumule explants. We investigated levels of DNA damage, changes in DNA methylation and DNA stability in common bean exposed to mammalian sex hormones (MSH) using inter-primer binding site (iPBS) and Coupled Restriction Enzyme Digestion-iPBS (CRED-iPBS) assays, respectively. The highest rate of polymorphism in iPBS profiles was observed when 10−4 mM of estrogen (52.2%) hormone was administered. This finding indicates that genetic stability is reduced. In the CRED-iPBS profile, which reveals the methylation level associated with the DNA cytosine nucleotide, 10−4 mM of estrogen hormone exhibited the highest hypermethylation value. Polymorphism was observed in all hormone administrations compared to the control (without hormone), and it was determined that genomic stability was decreased at high concentrations. Taken together, the results indicate that 17 β-estradiol, estrogen, progesterone, and testosterone in bean plants affect genomic instability and cause epigenetic modifications, which is an important control mechanism in gene expression.

- ArticleAçık Erişim2022Determining Genetic Diversity and Population Structure of Common Bean (Phaseolus vulgaris L.) Landraces from Türkiye Using SSR Markers
Özkan, Güller; Haliloğlu, Kamil; Türkoğlu, Aras; Özturk, Halil Ibrahim; Elkoca, Erdal; Poczai, Peter
Assessment of genetic diversity among different varieties helps to improve desired characteristics of crops, including disease resistance, early maturity, high yield, and resistance to drought. Molecular markers are one of the most effective tools for discovering genetic diversity that can increase reproductive efficiency. Simple sequence repeats (SSRs), which are codominant markers, are preferred for the determination of genetic diversity because they are highly polymorphic, multi-allelic, highly reproducible, and have good genome coverage. This study aimed to determine the genetic diversity of 40 common bean (Phaseolus vulgaris L.) landraces collected from the Ispir district located in the Northeast Anatolia region of Türkiye and five commercial varieties using SSR markers. The Twenty-seven SSR markers produced a total of 142 polymorphic bands, ranging from 2 (GATS91 and PVTT001) to 12 (BM153) alleles per marker, with an average number of 5.26 alleles. The gene diversity per marker varied between 0.37 and 0.87 for BM053 and BM153 markers, respectively. When heterozygous individuals are calculated proportional to the population, the heterozygosity ranged from 0.00 to 1.00, with an average of 0.30. The expected heterozygosity of the SSR locus ranged from 0.37 (BM053) to 0.88 (BM153), with an average of 0.69. Nei’s gene diversity scored an average of 0.69. The polymorphic information content (PIC) values of SSR markers varied from 0.33 (BM053) to 0.86 (BM153), with an average of 0.63 per locus. The greatest genetic distance (0.83) was between lines 49, 50, 53, and cultivar Karacaşehir-90, while the shortest (0.08) was between lines 6 and 26. In cluster analysis using Nei’s genetic distance, 45 common bean genotypes were divided into three groups and very little relationship was found between the genotypes and the geographical distances. In genetic structure analysis, three subgroups were formed, including local landraces and commercial varieties. The result confirmed that the rich diversity existing in Ispir bean landraces could be used as a genetic resource in designing breeding programs and may also contribute to Türkiye bean breeding programs.

Methods: One hundred twenty-two intellectually disabled children aged 4–18 years from five different special education and rehabilitation centers were included in the study. Their socio-demographic characteristics, body weight, height, body mass index (BMI), mid-upper arm circumference (MUAC), and skinfold thickness were examined, and their three-day food consumption record was taken. Results: Anthropometric scores did not differ significantly according to the severity of intellectual disability. According to evaluations made based on the World Health Organization’s 2007 percentile curves, 61.4% of the subjects with mild intellectual disability (MID), 57.1% of those with moderate intellectual disability (MOID), and 53.3% of those with severe intellectual disability (SID) were within the normal BMI range for their age. Conclusion: Rates of malnutrition and shortness in children and adolescents with SID were found to be higher than among children with MID and MOID, though the differences were not statistically significant.

Bronx Seedless and Italia (Vitis vinifera L.) are a variety preferred by consumers owing to their exciting flavour and widely cultivated in Aegean Region in Turkey. The aim was to identify the biogenic amines of these table grapes during berry ripeness. The biogenic amines were analyzed by HPLC in six different berry phenological stages. Italia grapes presented lower biogenic amine content than Bronx Seedless table grapes. The concentration of most of the biogenic amines analyzed linearly raised from the beginning of berry touch to when berries ripen for harvest stages. The most common biogenic amines in grape varieties were putrescine, followed by histamine, agmatine, and tyramine. There was also a positive correlation between all biogenic amines of the two grape varieties. The weakest correlation was found between spermine and cadaverine, whereas the strongest correlation was found among dopamine, trimethylamine, norepinephrine, tyramine, and histamine amines. The present study is the first report of a synthesis study regarding the effect of B.A.s on quality characteristics throughout berry ripeness in grape varieties containing foxy and muscat tastes. The concentration and composition of biogenic amines identified for both varieties might provide helpful information regarding human health and the vintage.

BACKGROUND: ‘Karaerik’ is a novel table grape (Vitis vinifera L.) native to Turkey and widely cultivated in areas bordering the city of Erzincan. Because of the demonstrated beneficial effects on human health of the grape phenolic composition, the aim of this work was to conduct a detailed profiling of non-anthocyanin phenolic fractions from different grape tissues of the ‘Karaerik’ table grape. Both qualitative and quantitative characterization of phenolic compounds were achieved using high-performance liquid chromatography–diode array detection–electrospray ionization–tandem mass spectrometry. Total phenolic content and oxygen radical absorbance capacity were also determined to evaluate the antioxidant properties of this table grape. RESULTS: A high number of non-anthocyanin phenolic compounds was identified in ‘Karaerik’ table grape skins and seeds, including 11 flavonols, six hydroxycinnamic acid derivatives, two stilbenes, several monomeric and dimeric flavan-3-ols and proanthocyanidins. Quercetin-type derivatives dominated the flavonol profile of grape skins, followed by myricetin type. Tartaric acid esters of three acids (caffeic, coumaric and ferulic acids) were the main hydroxycinnamic acid derivatives in this cultivar. Qualitative and quantitative differences were observed in flavan-3-ol composition among the grape tissues. Proanthocyanidins were the most abundant class of phenolic compounds in ‘Karaerik’ grapes, being mainly located in seeds. Higher antioxidant capacity values were determined in grape seeds, in correlation with the total phenolic content. CONCLUSION: These results provide useful information for a better understanding of phenolic antioxidants from the ‘Karaerik’ table grape and will contribute to promoting the varietal identity and health-related properties of this fruit.
