Designing a sustainable rangeland information system for Turkey

    Akar, A.; Gokalp, E.

The purpose of this study is to identify the deficiencies of the rangeland information system currently used in Turkey and, as an alternative, design a sustainable rangeland information system. In the study, both the extent of changes that occurred over time in the rangelands and the factors that caused such changes were identified, and solutions were suggested to eliminate those factors. The rangelands located in the Akcaabat district of Trabzon province were selected as the study area. Land use maps were produced by using the object-based classification method. According to the results of change analyses made with this information system, it was found out that, from 1973 to 2012, a surface area of 159.8 hectares had been degraded, demonstrating that the current information system had not been successful enough in the management of rangelands. For that reason, a sustainable rangeland information system free from all deficiencies was designed.

Investigation of pear drying performance by different methods and regression of convective heat transfer coefficient with support vector machine

    Das, Mehmet; Akpinar, Ebru Kavak

In this study, an air heated solar collector (AHSC) dryer was designed to determine the drying characteristics of the pear. Flat pear slices of 10 mm thickness were used in the experiments. The pears were dried both in the AHSC dryer and under the sun. Panel glass temperature, panel floor temperature, panel inlet temperature, panel outlet temperature, drying cabinet inlet temperature, drying cabinet outlet temperature, drying cabinet temperature, drying cabinet moisture, solar radiation, pear internal temperature, air velocity and mass loss of pear were measured at 30 min intervals. Experiments were carried out during the periods of June 2017 in Elazig, Turkey. The experiments started at 8:00 a.m. and continued till 18:00. The experiments were continued until the weight changes in the pear slices stopped. Wet basis moisture content (MCw), dry basis moisture content (MCd), adjustable moisture ratio (MR), drying rate (DR), and convective heat transfer coefficient (hc) were calculated with both in the AHSC dryer and the open sun drying experiment data. It was found that the values of hc in both drying systems with a range 12.4 and 20.8 W/m2 °C. Three different kernel models were used in the support vector machine (SVM) regression to construct the predictive model of the calculated hc values for both systems. The mean absolute error (MAE), root mean squared error (RMSE), relative absolute error (RAE) and root relative absolute error (RRAE) analysis were performed to indicate the predictive model's accuracy. As a result, the rate of drying of the pear was examined for both systems and it was observed that the pear had dried earlier in the AHSC drying system. A predictive model was obtained using the SVM regression for the calculated hc values for the pear in the AHSC drying system. The normalized polynomial kernel was determined as the best kernel model in SVM for estimating the hc values.

Chemical applications, scarification and stratification effects on seed germination of rare endemic Verbascum calycosum hausskn. ex Murb. (Scrophulariaceae)

Hilooğlu, Muhip; Sözen, Emel; Yücel, Ersin; Kandemir, Ali

Verbascum calycosum is an endemic plant species having considerable narrow distribution in Erzincan (Turkey) region. This species is known from only a single population and its habitats are highly threatened due to intensive human activities and soil erosion. In this study, the germination behavior of V. calycosum under different concentrations of NaCl, HCl, KNO3, GA3 (100 and 200 μM), hot-cold stratification and mechanical scarification were investigated. Seeds were exposed to a photoperiod of 8 h light/16 h dark with a 23/18 °C thermoperiod. Germination rates increased with GA3-100 μM (39%), GA3-200 μM (54.5%), mechanical scarification (34.5%) and cold stratification treatments (+4 °C, 23.25%; -20 °C, 18.25%) on the other hand, KNO3, NaCl, HCl and stratification with hot water treatments have decreased germination rates significantly when compared to the control (12.25%). Increased germination after GA3 application and mechanical scarification indicated that seeds of V. calycosum exhibited both non-deep and intermediate physiological dormancy as well as physical dormancy due to its hard seed coat. The highest speed of germination index was obtained at cold stratification of +4 °C and -20 °C (10.3). This study represents first report about seed dormancy and germination characteristics of V. calycosum. Overall, these results will provide valuable data for ex situ conservation of this rare endemic plant.

Cress (Lepidium sativum) is an annual herb from Brassicaceae family, and in some regions. It is known as peppergrass, garden cress, garden pepper-cress, pepperwort, or poor man’s pepper. Cress is an important medicinal plant in some countries including Turkey. This plant has major scientific and therapeutic significance. Peroxidase (POD, E.C.1.11.1.7) is an oxidor-eductase enzyme produced by a number of organisms. In this study, POD enzyme was purified from cress by ammonium sulphate precipitation, gel filtration, and CM-Sephadex ion-exchange chromatographies. K m and V max values were calculated from the Lineweaver-Burk graph for H 2 O 2 and guaiacol substrates and the substrate specificity of POD was investigated. The results showed that substrate specificity of H 2 O 2 is better than substrate specificity of guaiacol for this enzyme. The inhibition effects of three cations (Al 3+ , Cu 2+ , and Cr 3+ ) and one organic compound of cetyl trimethylammonium bromide were performed and their inhibition types were determined. Finally, optimum pH, optimum temperature, optimum ionic strength, and stable pH were determined.

Kurt, Aynur; Colak, Nesrin; Bengu, Aydin Sükrü; Gundoğdu, Ali; Akpinar, Erdal; Hayirlioglu-Ayaz, Sema; Ayaz, Faik Ahmet

Grape berries are a good source of nutrients and nutraceuticals and have many benefits for human health. Growing interest in the export potential and consumption of a new grape (cv. Karaerik), cultivated as a table grape in Turkey, encouraged us to profile its major nutrient contents from six different locations. Due to its popularity, the nutritional value of this grape berry needs to be investigated to ascertain its potential economic and health benefits. The most abundant sugars in the grape berry were fructose and glucose (peel/whole fruit; averages 236.57 and 127.87, and 183.36 and 108.60 g kg -1 fresh weight, respectively), while the major organic acids were tartaric and malic acids (7.17 and 2.81, and 2.61 and 1.76 g kg -1 fresh weight, respectively). Linoleic acid (peel/whole fruit/seed; 37.14, 33.12 and 57.83%, respectively) was the predominant fatty acid, while potassium (peel/whole fruit/seed; 9331.5, 10226.33 and 5354 μg/g dry weight, respectively) was the predominant mineral, followed by phosphorus (1592.8, 2672 and 3072.67) in the berry. Our results demonstrate that the nutrient components and physicochemical parameters varied significantly among the sampling locations. The grape berry contains considerable quantities of potentially beneficial healthy nutrients worthy of further evaluation.

    Yangilar, Filiz; Çakmakçi, Songul

It was aimed in the present study to determine the mineral content, probiotic, sensory, physical and chemical characteristics of probiotic yogurt-PC (only probiotic Lactobacillus acidophilus DSMZ 20079 strains with direct vat set starter culture) obtained with the addition of corn flour (CF) at different concentrations during a 21-day storage period. It was found in the study that all probiotic yogurt formulations with CF gave high L. acidophilus viabilities between 6.89 and 7.87 log cfu g(-1) during the storage and CF addition did not affect the viability of the probiotic bacteria. The viscosity and dry matter content increased in CF added samples with increasing CF concentrations. Syneresis in PC yogurt sample was found to be significantly higher than that in other samples. The greater the CF supplementation (5%) has the lower the syneresis on the 21st day of storage. The CF+PC ( 5%) sample was determined to be a better source of Ca, Mg, P and Fe than the Control and PC yogurts. The general acceptability of yogurt involving 5% PC decreased significantly than the others. Considering all the properties in general, 2.5% CF + PC sample it is recommended to be found as probiotic yohurt.

    Yangilar, Filiz

Effects of the edible films coated with natamycin (N) solutions and enriched with essential oils (EOs) were investigated on the chemical (e.g., the rates of pH, dry matter, fat, acidity, salt, protein, water soluble nitrogen, ripening index (RI), 12% trichloroacetic acid-soluble nitrogen, and pH 4.6 soluble-nitrogen), microbial (e.g., total number of aerobic mesophilic bacteria, lactic acid bacteria in MRS, lactic acid bacteria in M17, coliform and mould) and sensory properties of Kashar cheese during ripening time (3rd, 30th, 60th and 90th days) in the present study. Antimicrobial effectiveness of the films against moulds was measured on 30th, 60th, and 90th days of storage. The highest and lowest mould counts were determined for the C (2.87 log cfu/g) and N5 (1 log cfu/g) samples, respectively, at 90 days of ripening. Compared to other cheese samples, C and N3 coated cheese samples represented higher levels of water-soluble nitrogen and RI at the end of storage. N3 coated cheese samples were preferred more by the panellists while N5 coated cheese samples received the lowest scores. Practical applications: Antibacterial and antioxidant activities of essential oils have received considerable attention in food preservation. During Kashar cheese storage, the edible film incorporating oregano and rosemary essential oils coating process showed reducing effect on the microbial growth and extended the shelf-life of Kashar cheese.

Evaluation of Waste Loquat Kernels as Substrate for Lipid Production by Rhodotorula glutinis SO28

Ortucu, Serkan; Yazici, Aysenur; Taskin, Mesut; Cebi, Kadir

This study was performed to produce lipids from the isolated oleaginous yeast Rhodotorula glutinis SO28 using loquat kernel extract (LKE) as substrate. LKE was prepared using acid hydrolysis and alkaline neutralization steps. Lipid production was performed in shaking flaks culture. Even if LKE was used as a sole source of nutritional substances, it could support cell growth and lipid synthesis in the yeast. Additional carbon, nitrogen and phosphorus sources were found to significantly alter the lipid accumulation potential of the yeast. Optimal concentrations of additional carbon (glucose) and nitrogen (ammonium sulphate) sources for lipid accumulation were determined as 15 and 0.5 g/L, respectively. On the other hand, all the concentrations of additional phosphorus source were found to significantly reduce the lipid accumulation. Optimal incubation time was determined as 132 h. Under the optimized culture conditions, the lipid concentration and lipid content of the yeast were determined as 7.82 g/L and 62 %, respectively. Fatty acid methyl ester analysis exhibited that this yeast strain could produce high proportions of C16:0 and C18 fatty acids, which are ideal for biodiesel production. This is the first report on the use of waste loquat kernels as substrate for microbial lipid production.

The healing effect of cimin grape against toxic effects of ZnOTiO2 nanocomposite

    Atici, T.; Altun Çolak, D.

Today, occurrence of nanotechnology is making life easier, increasing the individual happiness and attracts attention to the benefits to be provided by it. As with any technology, nanotechnology also has risks, which are discussed here. In this study, toxic effects of ZnOTiO2 nanocomposite on various growth parameters of Drosophila melanogaster and removal of these effects by Cimin grape (Vitis vinifera L. ssp., Cimin) were investigated. For this purpose, assessment of the number of offspring and the survival rate were performed by feeding flies with ZnOTiO2 (1 mg/ml) and Cimin grape seed and fruit extracts (5 and 10 mg/ml). In the first stage, larvae of the the same age were put into the medium of each application group and the adult individuals were recorded. In the second stage, in order to determine the number of offspring in all treatment groups, in a set of experiments the grape extracts and ZnOTiO2 were only applied to the female members, while in another set of experiments, the grape extracts and ZnOTiO2 were applied only to male members, belonging to the same application group. The total number of flies developed from the eggs laid during ten days of pair mating was counted. As a result, survival rate and the offspring number in the application groups, that were only exposed to ZnOTiO2 were found to show a decline, compared with the other groups. At the same time, the values obtained from the groups, exposed to the ZnOTiO2 and grape extracts together, were close to control group values. This result supports the healing effects of Cimin grape, which is abundant in Erzincan.

Protective role of humic acids against picloram-induced genomic instability and DNA methylation in Phaseolus vulgaris

Taspinar, Mahmut Sinan; Aydin, Murat; Sigmaz, Burcu; Yildirim, Nalan; Agar, Guleray

Picloram (4-amino-3,5,6-trichloropicolinic acid) is a liquid auxinic herbicide used to control broad-leaved weeds. Picloram is representing a possible hazard to ecosystems and human health. Therefore, in this study, DNA methylation changes and DNA damage levels in Phaseolus vulgaris exposed to picloram, as well as whether humic acid (HA) has preventive effects on these changes were investigated. Random amplified polymorphic DNA (RAPD) techniques were used for identification of DNA damage and coupled restriction enzyme digestion-random amplification (CRED-RA) techniques were used to detect the changed pattern of DNA methylation. According to the obtained results, picloram (5, 10, 20, and 40 mg/l) caused DNA damage profile changes (RAPDs) increasing, DNA hypomethylation and genomic template stability (GTS) decreasing. On the other hand, different concentrations of applied HA (2, 4, 6, 8, and 10%) reduced hazardous effects of picloram. The results of the experiment have explicitly indicated that HAs could be an alternative for reducing genetic damage in plants. In addition to the alleviate effects of humic acid on genetic damage, its epigenetic effect is hypomethylation.

The effect of ultrasonic treatments on turbidity, microbial load, and polyphenol oxidase (PPO) activity of plum nectar

Irkilmez, Mustafa Umit; Başlar, Mehmet; Sağdiç, Osman; Arici, Muhammet; Ertugay, Mustafa Fatih

This study examined the effects of ultrasound on several quality parameters of plum nectars, such as turbidity, microbial load, PPO activity, phenolic content, and physicochemical quality. The ultrasonic treatment was applied to the plum nectars at different treatment temperatures (40, 50, 60, and 70 °C) and durations (2.5, 5, 7.5, and 10 min). Ultrasound processing caused a significant increase in the turbidity level and stability of the plum nectars, which were correlated with increases in treatment temperature and duration. Similar effects were also observed in reduction of microbial load. Ultrasonic treatments at 7.5 min at 60 °C and 5 min at 70 °C reduced the total yeast and mold counts (< 1 log cfu/mL). The total mesophilic-aerobic microorganism (TMAB) count was also reduced by up to 2 log. The maximum inactivation of polyphenol oxidase (PPO) was ~46 % at 70 °C. No dramatic trends were observed for the pH, brix, and the total phenolic compound content of the plum nectar, while the flavonoid content dramatically increased. The ultrasonic treatment caused a slight positive color change by increasing the L* value of the plum nectars. In conclusion, the ultrasonic treatment was important process for increasing the turbidity quality and for inactivation of yeasts and molds, but it was not effective at inactivating the PPO enzyme.

Effects of whey protein isolate based coating enriched with Zingiber officinale and Matricaria recutita essential oils on the quality of refrigerated rainbow trout

    Oğuzhan Yıldız, Pınar; Yangılar, Filiz

The present study aimed to investigate the effect of whey protein isolate (WPI) coating enriched with essential oils (ginger and chamomile) on rainbow trout fillets during storage at 4°C. Samples were analyzed at 0, 3, 6, 9, 12, and 15 days for microbiological (total aerobic mesophilic bacteria, psychrotrophic bacteria, lactic acid bacteria, Pseudomonas), chemical analyses (pH, thiobarbituric acid reactive substances-TBARS, total volatile base nitrogen-TVB-N, peroxide value [PV]) and sensory quality (odor, taste, tissue, appearance). Bacterial growth was inhibited in samples with high concentrations of essential oils due to its antimicrobial activity. During the storage period, TVB-N, TBARS, and PVs content gradually increased in all groups, and significant differences were found between the groups (p <.05). In conclusion, the addition of of ginger and chamomile essential oils to WPI coatings showed a positive effect on the rainbow trout shelf-life. Practical applications: Whey protein is a by-product of the cheese-making industry and is already known as an excellent barrier to oxygen, aroma, and oil and can be used as a coating material for improving the oxygen barrier property of food packaging. The use of an essential oil in fresh fish preservation may be considered an alternative “natural” additive, extending the shelf life of the product. Our study has clearly shown that addition of ginger and camolime essential oil and whey protein isolate coating in fish resulted in longer shelf life, and this method could be commercially used.

Exogenous salicylic acid alleviates cold damage by regulating antioxidative system in two barley (Hordeum vulgare L.) cultivars

Mutlu, Salih; Atıcı, Ökkeş; Nalbantoğlu, Barbaros; Mete, Ebru

In this work, the time-dependent effect of exogenous salicylic acid (SA) applied before cold stress was investigated on the regulation of antioxidative respond mechanisms in two barley (Hordeum vulgare L.) cultivars (Akhisar and Tokak) differed in cold tolerance. SA (0.1 mM) was applied to 7-days old barley seedlings growing under control conditions (20/18°C). After this application, the seedlings were transferred to cold conditions (7/5°C) at different times (7, 14, 21 and 28 days) for 3 days. Then, the contents of malondialdehyde (MDA), hydrogen peroxide (H2O2) and the activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT) and peroxidase (POX) were determined in the leaves (control, cold and SA + cold) from the seedlings harvested at 10, 17, 24 and 31 days. Moreover, the effect of cold stress was evaluated on the endogenous SA level at the both cultivars by comparing to control plants. The MDA content increased in the cold treatment while it decreased in the SA treatment at all the days studied at the both cultivars. The SA + cold treatment could have a variable effect on H2O2 content at the tolerant barley (Tokak) while decreased its content at the sensitive cultivar (Akhisar). The SA + cold treatment could increase the activities of POX and SOD at both cultivars, but it increased the CAT activity at the tolerant cultivar while decreased at the sensitive cultivar. In addition, the content of endogenous SA was decreased by cold stress at all the days studied at the barley cultivars as compared to control plants. The results show that the SA treatment could be effective on the regulation of the parameters studied at cold conditions until 31 days after SA application, and the importance of exogenous SA treatment before cold stress. It was concluded that exogenous and endogenous SA could play an ameliorating role on cold tolerance by regulating reactive oxygen species and the activities of antioxidative enzymes in both cold-sensitive and cold-tolerant cultivars of barley.
